1.Quality study on differently-processed products of Leonurus heterophyllus sweet

Objective: To investigate the effects of processing on contens of saponin of Leonurus heterophyllus sweet. Methods: The determinations of saponin were carried out by TLC. Results: There were obvious differences between the crude and the different processed products of Leonurus heterophyllus sweet in contents of saponin, and there were no obvious changes in constituents of saponin.Conclusion: The different processing methods and temperature had great effects on the contents of saponin of Leonurus heterophyllus sweet.
2.Involvement and modulation effects of oxytocin and opioid receptor on evoked discharge of hippocampal CA1 neurons in rats

BACKGROUND: Central oxytocin (OT) may be a neurotransmitter or neuromodulator and play an important role in learning and memory, sexual behaviour, pain modulation and opiate tolerance and dependence. To research the interactions between oxytocinergic and opioidergic system in hippocampus has some significance.OBJECTIVE: To investigate the effects of OT administered intracerebroventricularly on evoked discharge of left dorsal hippocampal CA1 neurons in rats and the possible interactions between oxytocinergic and opioidergic system.DESIGN: A randomised controlled study.SETTING: Department of Physiology of Guangdong Medical College; Department of Physiology and Pathology of Medical College of Wuhan University.MATERIALS: The study was conducted in the Physiology Department of Medical College of Wuhan University from September 2002 to September 2003. A total of 36 male Sprague-Dawley rats were selected and randomly divided into six groups: control (NS), OT groups (0.2 mg/L, 2 mg/L and 20 mg/L), [d (CH2)5-OVT]+OT (2 mg/L), naloxone+OT (2 mg/L), with 6 rats in each group.METHODS: Single-unit recording was performed with glass microelectrode. The glass microelectrode was inserted by a micromanipulator into hippocampal CA1. The electrical activity was amplified by a microelectrode amplifier and then recorded by the biological experimental system,monitored at the same time with oscilloscope. When recording the neural discharge, electrical stimulation of the sciatic nerves was performed once 5minutes through a double stainless electrode. 5 μL oxytocin in dosage of 0.2, 2 and 20 mg/L were injected slowly into lateral ventricle via microlitre syringe. [d(CH2)5-OVT]+OT (2 mg/L) group: 2.5 μL [d(CH2)5-OVT](80 mg/L) was injected into lateral ventricle and then 2.5 μL oxytocin (2 mg/L). Naloxone+OT (2 mg/L) group: 2.5 μL naloxone (400 mg/L) was injected into lateral ventricle and then 2.5 μL oxytocin (2 mg/L). According to frequency of discharge, effect of oxytocin at various dosages on discharge induced by neurons in hippocampal CA1 area and [d (CH2)5-OVT]and naloxone on oxytocin was assayed. MAIN OUTCOME MEASURE: Changes of discharge frequency after stimulation.RESULTS: Data of totally 36 rats were entered the final analysis. ① OT (0.2 mg/L, 2 mg/L and 20 mg/L) administered by intracerebroventricularly could decrease the evoked discharge of hippocampal CA1 neurons in a dose-dependent manner. ② The inhibitory effects of OT (2 mg/L) could be blocked by pretreated intracerebroventricularly injection of [d (CH2)5-OVT](80 mg/L, 2.5 μL). ③ Intracerebroventricular injection of naloxone (400 mg/L, 2.5 μL) could attenuate the effects of OT (2 mg/L) significantly.CONCLUSION: OT can inhibit the electrical activities of hippocampal CA1 neurons to external electrical signal through activating the oxytocin receptor. Moreover, central opioid receptor is involving in the inhibitory effects of OT.
3.The study of the association between COMT polymorphisms and methylphenidate response in the treat-ment of attention deficit hyperactivity disorder

Objective To explore the impacts of catechol-O-Methyltransferase (COMT) gene polymorphisms on the outcomes of methylphenidate treatment for attention deficit hyperactivity disorder (ADHD) in children. Methods One hundred seventy-seven ADHD children of Chinese Han descent received open-labelled dose titration with methylpheni?date to achieve optimal response in 2~4 weeks. The behavior changes were evaluated by using ADHD diagnostic scale (parent version) before and after treatment. COMT gene rs4680 (Val158Met) and rs165599 were genotyped using fluores?cent real-time PCR. The genotype distribution and treatment outcomes including remission, response and non-response were analyzed. Results The treatment response differed significantly among patients with different genotypes of rs4680 (P<0.01). Thirty-eight (40.9%) patients with G/G genotype had remission and 19 (20.4%) had good response;Thirty-five (48.6%) patients with G/A genotype had remission and 16 (22.2%) had good response. In contrast, only one patients with A/A genotype had remission and none of them had good response. The changes of the ADHD diagnostic scale total scores, inattention scores and hyperactivity impulsive scores were statistically different among different genotypes of rs4680 (P<0.05). The hyperactivity impulsive scores were significantly higher in the G allele carrier than A allele carrier (P=0.04). The treatment response was not significantly different among different rs165599 genotypes (P>0.05). Conclu?sion COMT gene rs4680 (Val158Met) polymorphism is associated with methylphenidate response in a Han Chinese popu?lation. Patients with G allele is more likely to benefit from methylphenidate treatment in comparison with A/A genotype.
5.Factor analysis of hospitalization expenses for lens surgery

Objective To identify the common factor affecting hospitalization expense of lens surgery operation, and propose effective measures to curb unreasonable increase of medical costs.Methods Collection of inpatients information of the year 2014 from a tertiary hospital in Beijing, for factor analysis of the hospitalization expenses of 555 inpatients categorized as DRGs of CB39 (lens surgery), using the BJ-DRGs platform.Results The highest factor of lens surgery expenses is disposable medical surgery materials, accounting for 42.77%;followed by surgery fee and medication fee, accounting for 23.19% and 17.15% respectively.Conclusion For the hospitals, medicine and medical material rank the highest in cost control, and they are recommended to work on them by means of clinical pathways, medical norms and purchasing policy, and to explore new means of payment in the meantime.

The first genetic linkage map of Salvia miltiorrhiza was constructed in 94 F1 individuals from an intraspecific cross by using simple sequence repeat (SSR), sequence-related amplified polymorphism (SRAP) and inter-simple sequence repeat (ISSR) markers. A total of 93 marker loci in the linkage map, consisting of 53 SSR, 38 SRAP and 2 ISSR locus were made up of eight linkage groups, covered a total length of 400.1 cm with an average distance of 4.3 cm per marker. The length of linkage groups varied from 3.3 -132 cm and each of them included 2-23 markers, separately. The result will provide important basis for QTL mapping, map-based cloning and association studies for commercially important traits in S. miltiorrhiza.
7.Relationship of expression of osteopontin and CD44 variant isoforms in multiple myeloma patients with progress of multiple myeloma

Background and purpose: Osteopontin (OPN) is a glycophosphoprotein that is expressed by numerous human cancer cells. The function of OPN in skeletal modeling and remodeling, bone resorption, angiogenesis and tumor cell metastasis and progression through binding with integrin and CD44 receptors were studied. Our purpose of the study was to detect the level of osteopontin(OPN) and CD44 variant isoforms(CD44v6) in multiple myeloma (MM) patients, and to explore the relationship between OPN and CD44v6 with the progress of MM. Methods: 32 MM patients were admitted to our hospital from Sep. 2007 to Dec. 2008. The patients were divided into two groups, group A (untreated and relapsed MM patients) and B (stable MM patients), and the control group including 15 subjects were the benign anemia patients or healthy people who suffered bone fracture. Bone marrow mononuclear cells (MNCs) and bone marrow stromal cells (BMSCs) from MM patients and subjects were investigated as potential OPN and CD44v6 producers. The level of OPN and CD44v6 of the conditioned media from MM patients and subjects were analyzed by ELISA. Results: The OPN level in group A (19 cases) was significantly higher than group B (13 cases) and control group (P<0.05). The CD44v6 level of 14 patients in group A was significantly higher than that of 10 cases in group B and control group (P<0.05); The OPN level of MM patients was correlated with the level of CD44v6 (r=0.52, P=0.000), the percentage of plasma cells in the bone marrow (r=0.74, P=0.000), M protein (r=0.53, P=0.014), and β2-microglubin (r=0.62, P=0.002). Conclusion: The increase of OPN and CD44v6 is associated with progress and pathogenesis of MM,and may be involved with tumor burden, stage and tumor invasion.
8.Factors affecting postmenopausal osteoporosis in Dali Bai Autonomous Prefecture

Objective:
To investigate the prevalence of postmenopausal osteoporosis (PMOP) and analyze its influencing factors among women at ages of 50 to 59 years in Dali Bai Autonomous Prefecture, Yunnan Province, so as to provide insights into the prevention of PMOP among menopausal women.
Methods:
Bai Ethnic menopausal women at ages of 50 to 59 years who received healthy examination at the Center of Healthy Examination, Dali Prefecture People's Hospital from June 2017 to May 2021 were selected as the study subjects, and subjects' demographic characteristics, living habits, history of diseases, family history of osteoporosis and history of parturition were collected using self-designed questionnaires. The height, body weight and bone density were measured, and fasting blood glucose, vitamin D3, blood lipids and liver functions were detected. The factors affecting the development of PMOP were identified using a multivariable logistic regression model.
Results:
Totally 2 000 questionnaires were allocated, and 1 584 valid questionnaires were recovered, with an effective recovery rate of 79.20%. The respondents had a mean age of ( 56.22±2.61 ) years, and mean body mass index ( BMI ) of ( 24.62±2.35 ) kg/m2. There were 497 respondents ( 31.38% ) with a family history of osteoporosis, and the prevalence of PMOP was 20.64%. Multivariable logistic regression analysis identified age ( OR=1.135, 95%CI: 1.074-1.196 ), age of menarche ( OR=1.138, 95%CI: 1.059-1.217 ), duration of menopause (OR=1.425, 95%CI: 1.228-1.622), number of parturition ( >2, OR=5.036, 95%CI: 2.972-7.101 ), smoking ( OR=2.594, 95%CI: 1.767- 3.421 ), alcohol consumption ( OR=2.051, 95%CI: 1.503-2.598 ), family history of osteoporosis ( OR=2.540, 95%CI: 1.769-3.311 ), hypertension ( OR=1.492, 95%CI: 1.406-1.578 ), diabetes ( OR=1.774, 95%CI: 1.581-1.967 ), total cholesterol ( OR=1.483, 95%CI: 1.251-1.716 ), triacylglycerol ( OR=1.801, 95%CI: 1.576-2.026 ), low-density lipoprotein cholesterol ( OR=1.614, 95%CI: 1.498-1.731 ), fasting blood glucose ( OR=1.192, 95%CI: 1.077-1.307 ), BMI ( OR=0.934, 95%CI: 0.862-0.993 ), outdoor activity ( ≥1 time/week, OR: 0.413-0.549, 95%CI: 0.329-0.637 ), age of menopause ( OR=0.909, 95%CI: 0.841-0.977 ), daily intake of calcium ( ≥600 mg, OR: 0.493-0.644, 95%CI: 0.389-0.786 ), vitamin D3 level ( ≥20 ng/mL, OR: 0.604-0.719, 95%CI: 0.523-0.853 ) and high-density lipoprotein cholesterol ( OR=0.658, 95%CI: 0.550-0.767 ) as factors affecting the development of PMOP.
Conclusions
The prevalence of PMOP in Dali Bai Autonomous Prefecture is similar to the nationwide level in China, and old age, smoking, alcohol consumption, a family history of osteoporosis and high blood lipid levels may increase the risk of PMOP.

Allotransplantation of peripheral blood-derived hemopoietic stem cells is the best therapy for acute leukemia. With increases of only-child families, sibling donors are decreasing. Moreover, the probability is low and time consuming is long to search a matched hemopoietic stem cell donor from the Chinese Marrow Donor Program. Haploidentical stem cell transplantation would bring a hope for donor resource. However, difficult transplantation and high incidence of graft versus host disease are two risks. Based on modified regimen and cyclosporine A+short-term amethopterin, mycophenolic acid, interleukin-11, anti-lymphocyte immunoglobulin and hemopoietic stem cells mobilized by recombinant human granulocyte colony-stimulating factor can prevent graft versus host disease. This therapy succeeded in two cases with no severe graft versus host disease.
10.Mobilization and collection of peripheral blood stem cells by using rhG-CSF in healthy donors

Objective To analyse the effect of mobilization and collection's time of peripheral blood stem cells(PBSC) from 8 cases of healthy donors. Methods The 10 donors were studied by self-control design.The number of aphereses was two times every donors. Healthy donors received rhG-CSF according to two different PBSC collection starting time: group 1:PBSC collection was starts 2 hours(2 h) after the fourth day or the fifth day of rhG-CSF. group 2:PBSC collection was starts 4 hours(4 h) after the fourth day or the fifth day of rhG-CSF.(The first dose of rhG-CSF was given on day 1, considering day 0 as the day before starting mobilization). In this study we have compared with two groups of apheresis product. Results The MNC count was significantly higher for donors 4 h collection (groups 2) then 2 h. ( groups 1)(P
