2.Immunoaffinity Chromatography Method for Purification of Human Protein C

A special monoclonal antibody(McAb)to protein C(PC)was prepared,itrevealed the different conformation in the absence and presence of Ca~(++).This McAbwas coupled to Sepharose-4B and the gel was loaded into the column for thepurification of PC.The immobilized antibody was capable of binding PC fromthe plasma in the absence of Ca~(++).This immunopurification resulted in a 12 000-fold after one step proceduce.The specific activity of the PC product ranged be-tween 180-230 U/mg and behaved as two bands(?-PC and?-PC)on nonreducedSDS-PAGE and five bands(?-S-PC,?-S-PC,?-heavy chain,?-heavy chainand light chain)on reduced SDS-PAGE.
3.Preliminary Report on Preparation of Monoclonal Antibodies to Human Plasma Protein C

Eight cell strains of mouse hybridoma named PC 01 to PC 08 have been produced by immu-'nizing the mice with human plasma protein C (HPC) which is purified using physico-chemical method. Each of them has long-term stable ability to secret the monoclonal antibodies. Among them,PC 03 is a conformation-specific monoclonal antibody,and its binding to HPC can be pro-motad distinctly by divalent metal ion. However,the monoclonal antibodies of PC02 and PC03 can noticeably advance the enzymatic activity of HPC,whereas the monoclonal antibody of PC04 has capacity to inhibit the enzymatic activity of HPC.
4.Expression of hepatocyte growth factor and vascular endothelial growth factor in acute myeloid leukemia and their relationship with angiogenesis

Objective To investigate the expression of hepatocyte growth factor (HGF)and vascular endothelial growth factor (VEGF)in acute myeloid leukemia(AML) and its significance in the progress of AML.Methods Serum levels of HGF and VEGF in AML patients,including 25 de novo cases,17 cases with complete remission( CR),16 cases without remission and 12 cases with relapse,were measured by enzyme-linked immunosorbent assay( ELISA ).The data were compared with those of normal controls.Results The serum levels of HGF were( 1357.29 ± 358.64)ng/L in de novo patients,(1175.93 ± 306.71 )ng/L in patients without remission and( 1261.21 ± 340.83 )ng/L in patients with relapse,which were significantly higher than those in normal control(232.62 ± 99.13 ) ng/L and CR patients ( 256.65 ± 94.32 ) ng/L( F =78.35,P < 0.01 ).The serum levels of VEGF were ( 253.84 ± 49.14 ) ng/L in de novo patients,( 245.87 ± 54.68 ) ng/L in patients without remission and(264.75 ± 62.52) ng/L in patients with relapse,which were significantly higher than those in normal control ( 97.61 ± 16.19 ) ng/L and CR patients ( 99.76 ± 15.93 ) ng/L ( F =68.65,P < 0.01 ).Compared with de novo group,the serum levels of HGF and VEGF in CR patients decreased significantly but showed no significant difference from those in normal controls(P >0.05).The expression of HGF and VEGF in acute myeloid leukemia was positively correlated with each other ( r =0.49,P < 0.05 ).Conclusion The expression of HGF and VEGF was closely related with the incidence and development of acute myeloid leukemia.Preventing HGF expression may be a possible strategy for leukemia therapy in the future.
5.Practicable value of DAP (dose-area product) database in occupational radiation protection in cardiovascular interventional procedures

Objective To evaluate the efficacy of the DAP database in reducing radiation hazards during cardiovascular intervention.Methods The study conducted a retrospective analysis of all consecutive cardiovascular cases in the database from April 1st,2016 to June 30th,2016.Based on the imaging systems,an inage intensifier (Ⅱ) and a flat panel detector (FPD),the cases of patients undergoing coronary angiography (CAG) and percutaneous coronary interventions (PCI) were collected to observe Body Mass Index (BMI),fluoroscopy time (FT) and dose area product (DAP).Results The FT and DAP values in CAG were (3.26-±2.59) min and (1 938±1 296) μGy·m2 for the Ⅱ system and (3.00 ± 2.89) min and (3 718 ± 2 859) μGy· m2 for the FPD system,respectively,whereas,the FT and DAP values in PCI were (17.81 ± 12.71) min and (8 899 ±7 032) μGy·m2 for the Ⅱ system and (21.99 ± 15.91) min and (19 526 ± 14 134) μGy · m2 for the FPD system,respectively.The differences in DAP values (x ± s) for CAG and PCI between the angiography systems were significant(t =-10.664,11.239,P < 0.05).The DAP and total DAP values during PCI for fluoroscopy from the FPD systen were 3 596 755.60 and 4 881 484.50 μGy · m2,respectively.Conclusions The DAP values in CAG and PCI using FPD system are much higher than the relevant data.The use of an FPD system resulted in higher DAP values compared to the Ⅱ system in cardiovascular interventional procedures.Reduction in fluoroscopy time may decrease radiation hazards during PCI.The DAP database allows for a fast,convenient and enough objective data to discovery timely difference and variation in dose in cardiovascular interventional procedures.
6.Study on Preparation Process of Syrigopicroside Dropping Pills

Objective To establish the optimum preparation process for Syrigopicroside Dropping Pills.Methods The preparation conditions,such as the proportion between the extracts and matrix,dripping temperature,the ratio of matrix,were studied with orthogonal test.The obtained results were tested by three indexes,such as the variation coefficient of weight of pill,time limit of dissolution and appearance of the dropping pills with analysis of variance.The preparation process was optimized.Results The optimal technical conditions were as follows:the ratio of PEG4000 and PEG6000 as matrix was 4∶1,the ratio of syrigopicroside to matrix was 1∶4,and the dripping temperature was 80 ℃.Conclusion The preparation technology can meet the requirement for dripping pill specified in China Pharmacopeia(2005 Edition).
7.The Value of MR Imaging in Diagnosis of the Medial Collateral Ligaments Injury of Knee Joint

Objective To assess the value of MRI imaging in diagnosis of the medial collateral ligament(MCL) injury of the knee joint.Methods MR findings of 20 cases of clinically suspected MCL injury of the knee joint were retrospectively reviewed and compared with operative findings of the knee joint.Results In 20 cases of MCL injury,there were 2 cases of gradeⅠinjury,9 gradeⅡand 9 grade Ⅲ.Taking the results of operation as standard,the accuracy of diagnosis of grade Ⅱand Ⅲ injury of the MCL was 88.9% and 100% respectively.Conclusion MRI can accurately display the MCL injury of the knee joint,it is important for the preoperative treatment planning.
8.Determination of the Related Substances in Nortriptyline Hydrochloride by HPLC

Objective: To establish a determination method for the related substances in nortriptyline hydrochloride by HPLC. Methods:An Agilent C18 (250 mm × 4. 6 mm,5 μm) column was adopted with a mobile phase consisting of acetonitrile-0. 2% trieth-ylamine(pH 3. 0)(34 ∶66). The flow rate was 1. 0 ml·min-1, the detection wavelength was set at 210 nm, the column temperature was 40℃ and the injection volume was 20 μl. Results:The resolutions of the related substances were acceptable. The detection limit and the quantitation limit of the samples was 0. 40 ng and 1. 35 ng, respectively. Conclusion: The method is sensitive, accurate and specific, and can be used to determine the related substances in nortriptyline hydrochloride.
9.The survey of gross radioactivity level in drinking water in Inner Mongolia Region

Objective To ascertain the gross α/β levels in drinking water in Inner Mongolia and to estimate the annual effective dose to the local residents from radionuclides in drinking water.Methods A total of 768 water samples were collected from 101 counties distributed over 12 cities of Inner Mongolia.Low background α/β measuring instrument was used to measure the radioactivity level;On this basis,use EPA Federal Guidance Report 11 universal method to estimate the annual effective dose to the local residents via intake of radionuclides from drinking water.Results The gross α radioactivity range was 0.016-1.003 Bq/L for tap water,O.016-0.975 Bq/L for factory water,0.017-1.544 Bq/L for river water,0.120-0.672 Bq/L for lake water,0.016-0.492 Bq/L for reservoir water,0.016-1.139 Bq/L for well water,0.032-3.156 Bq/L for spring water,respectively.The gross β radioactivity range was 0.030-0.828 Bq/L for tap water,0.031-0.571 Bq/L for factory water,0.066-0.873 Bq/L for river water,0.169-2.268 Bq/L for lake water,0.046-0.519 Bq/L for reservoir water,0.071-0.526 Bq/L for well water,0.087-1.063 Bq/L for spring water,respectively.Conclusions In Inner Mongolia,the gross α/β mean value in tap water is less than the World Health Organization-recommended value and the average annual effective dose from tap water is also less than the WHO-recommended value O.1 mSv/a.The gross α/β radioactivity from the other water samples is also within the range of the nationwide average.
10.Cleaning Efficacy of Automatic Cleaning Machines Evaluation and Impact Factors

OBJECTIVE To evaluate the cleaning efficacy of the automatic cleaning machine and the risk factors to ensure the effect of disinfection and sterilization. METHODS Cleaning efficacy of the automatic cleaning machines was detected by STF cleaning test card.It was put in the first cabin of every automatic cleaning machine.And it was also tested for these factors in determining cleaning qualities such as enzyme concentration,water temperature and pieces of instruments washed. RESULTS The cleaning efficacy of the automatic cleaning machines was 98.9% in the continuous two years.The qualified rate was 98.6% for surgical instruments,99.4% for all instrument in clinical sections. CONCLUSIONS The machine is suitable for the cleaning of surgical instruments.It also raises working efficiency,cutting down the cost of repairs and maintenance,reducing the chances of injury and infections in nurses who performed manual cleaning precedures.The procedures of the STF cleaning test card in detecting the cleaning efficacy is simpler,safer,and suitable for clinical applications.
