BACKGROUND: Electrostatic layer-by-layer (LbL) self-assembly is a simple, effective and novel approach to carrying macromolecules into microcapsule carriers.OBJECTIVE: To prepare chitosan (CS) microcapsules to control the releasing of heparin (Hep) by LbL.METHODS: CS microspheres were fabricated by a sodium sulfate-based precipitation process and then used as positively charged templates for polyelectrolyte multilayer coatings to carry heparin by the LbL self-assembly technique. The polycation was CS and the polyanion was Hep. The microcapsule was formed via coating {CS/Hep}3 on CS hydrogel template. The microcapsules composed of {CS/ Hep}3 on CS hydrogel templates were confirmed by fluorescence inverted microscopy, confocal laser scanning microscopy and laser particle size analysis. The assembly of polyelectrolyte layers on CS hydrogel microspheres was monitored by Zeta Potential Analyzer.RESULTS AND CONCLUSION: The resulting microcapsules were about 1 μm in average diameter, and allowed spontaneous loading of heparin through electrostatic interaction, with the encapsulation efficiency and carrying capacity of 83.8% and 3.05%, respectively.

Objective Current research proposed that lid-wiper epitheliopathy may be a early symptom of dry eye.At present,there are seldom studies on lid-wiper epitheliopathy in both home and abroad.This study was to investigate the effect of artificial tears without antiseptic in the treatment of lid-wiper epitheliopathy and discuss the method of treatment of lid-wiper epitheliopathy. MethodsThis is a case-observative study.Seventy patients with lid-wiper epitheliopathy and with negative results in tear film break-up test,SchirmerⅠtest and corneal fluorescine staining were enrolled in the Peking University First Hospital during September 2006 and June 2008.The treatment group included 54 eyes of 54 patients who received topically free-antiseptic artificial tears (dextran 70 eye drops) four times per day for two weeks,and the control group included 16 eyes from 16 patients who were not treated.Dry Eye Questionnaire were taken from the patients before the administration of drug and 2 weeks after treatment.Fluorescein and Lissamine green double-staining was used before and after treatment for the evaluation of lid-wiper epitheliopathy.The disease was graded into 0 (absence) to 3(severe),and the symptoms and dye were scored following the Standard Patient Evaluation of Eye Dryness (Korb criteria).Informed consent and good compliance were obtained before initial and during the therapy duration. ResultsIn the treatment group,the scores of symproms and staining were significantly different between before and after administration of drug (12.91±2.55 vs 2.78±2.27 in symptom,t=22.70,P＜0.05;1.79±0.78 vs 0.19±0.31 in dye,Z=-6.40,P＜0.05),indicating the symptoms and dye results were obviously improved after therapy.In the control group,no significant differences were found in the symptom score and dye score in the corresponding time period (12.63±2.60 vs 12.94±2.11,t=-1.10,P＞0.05 in symptom;1.97±0.74 vs 1.98±0.72,Z=-0.58,P＞0.05 in dye).According to the grade of disease,the magnitude of the staining after treatment was considerably improved in comparison with before treatment (Z_1=-3.47 for mild,Z_2=-4.04 for moderate,Z_3=-3.74 for severe),showing statistically significant differences between them (P＜0.05).ConclusionThe use of artificial tears is an efficacious treatment for treating lid wiper epitheliopathy.The topicaly utilization of artificial tears can alleviate the symptoms and pathological changes of lid wiper epitheliopathy.

Taking Naegleria australiensis, a species of pathogenic free-living amoeba (isolated in Shanghai in 1986), as antigen, two cell lines which provided potentially permanent source of monoclonal antibodies were established by lymphocytic hybridoma technique. The results of identification showed that: (1) the two cell lines could secret two different kinds of McAbs; (2) both of the McAbs were IgG (by gel diffusion); (3) McAbs produced in BALB/c mice were at high concentrations. One of them had a titer of ≥1 : 8 192 and the other≥l : 1 024 (by enzyme linked immunosorbent assay). A decline of the titers after purification by salting-out method was shown. One of the purified McAbs had a titer of≥1: 2 024 and the other≥l : 128.We have adopted two ways of recovering cryopreserved cells : ordinary recovering and "direct" recovering. The latter way was more practical because it could reduce the cycle of antibody production, and avoid contamination and chromosome variation. Experiments with different doses of cells revealed that, if the latter way was used, the optimal dose was 3 to 5 ? 106 cells per mouse.

OBJECTIVE To investigate the expression and significance of Fas, FasL and FADD in laryngeal carcinoma. METHODS Immunohistochemical method was used to examine the expression of Fas, FasL and FADD in laryngeal carcinoma specimen and adjacent normal tissues. Objective To study the correlation between organizational commitment of nurses and their intention to drop out. Methods Toally 304 contract nurses were recruited in this investigation. The questionnaires including demographic data, scale of organizational commitment and intention to drop out scale were used. Results The score of intention to drop out was 16.94 ± 2.16. The score on organizational commitment was 71.19 ± 6.65. There was a positive correlation between intention to drop out and organizational commitment among the contract nurses. Conclusions Improving contract nurses′organizational commitment can be an effective mean to reduce contract nurses′intention to drop out. It is an important means to promote the development of the nurse team and provide a reference for the scientific development of nursing.

Objective To investigate the expressions of apoptosis stimulating protein of P53 (ASPP)family in colorectal carcinoma tissue and to explore their relationship with the clinicopathological characteristics of colorectal carcinoma,and to clarify the effect of ASPP family in the development of colorectal carcinoma.Methods 45 cases of colorectal carcinoma tissue and 20 cases of healthy controls were selected. Among 45 cases of colorectal carcinoma tissue, there were 1 1 cases of well differentiated colorectal carcinoma, 2 1 cases of moderately differentiated colorectal carcinoma,and 13 cases of poorly differentiated colorectal carcinoma;7 cases of T1 stage colorectal carcinoma,8 cases of T2 stage colorectal carcinoma,25 cases of T3 stage colorectal carcinoma,and 5 cases of T4 stage colorectal carcinoma;19 cases of N1 stage with lymph node metastasis,26 cases of N0 stage without lymph node metatasis.The expressions of ASPP1,ASPP2,and iASPP in 45 cases of colorectal carcinoma tissue and 20 cases of normal colon tissue were detected by immunohitochemistry SP method,and the correlations between the expressions of ASPP family and the pathologic typing,infiltrative depth,and lymph node metastasis of colorectal carcinoma were analyzed. Results ①The immunohitochemical staining results showed that the ASPP family members expressed in colorectal carcinoma tissue and normal colon tissue, and there were no significant differences in ASPP1 and ASPP2 positive rates between colorectal carcinoma tissue and normal colon tissue (P>0.05);the positive expression rate of iASPP in colon cancer tissue was higher than that in normal colon tissue (P<0.01).② The expression of ASPP1 in colon caner tissue had no correlation with the differentiation degree of tumor cells (rs=0.163,P>0.05);the expression of ASPP2 positive rate was decreased when the differentiation degree of tumor cells reduced,they had positive correlation (rs=0.454,P=0.002);the expression of iASPP in colon cancer tissue had no correlation with the differentiation degree of tumor cells (rs=-0.171,P>0.05).③ The expression of ASPP1 in colon caner tissue had no correlation with the infiltrative depth of tumor (rs=-0.268,P>0.05);the expression of ASPP2 positive rate was decreased when the tumor infiltrative depth increased,they had negative correlation (rs=-0.348,P<0.05);the expression of iASPP in colonic carcinoma caner tissue had no correlation with the infiltrative depth of tumor (rs=0.231,P>0.05).④The expressions of ASPP1,ASPP2, and iASPP in colon caner tissue had no correlation with lymph node metastasis (rs=0.089,rs=0.044,rs=0.210, P>0.05).Conclusion The expression levels of iASPP in colon cancer and normal colon tissues are different,it may be useful for the diagnosis, differential diagnosis and evaluation in benign and malignant colorectal diseases. The expression of iASPP is negatively correlated with the pathologic typing and neoplasm staging of colorectal carcinoma,it indicates that iASPP can be used as a indicator in judging the prognosis of colorectal carcinoma.

Objective To clarity the role of VEGF and its receptors in laryngeal squamous cell carcinoma development and metastasis. Methods The protein and mRNA expression of VEGF and its receptors (Fit-1 and KDR) were examined by Western-blotting and semiquantitative reverse transcriptase polymerase chain reaction analysis in laryngeal squamouse cell carcinoma (20 specimens) and the adjacent non-neoplastic laryngeal tissues (18 specimens). Results The protein and mRNA expression levels of VEGF and its receptors(Fit-1 and KDR) were more abundant in laryngeal squamous cell carcinoma than that in the adjacent non-neoplastic laryngeal tissues (ordedy 4.32±2.21, 2.00±0.91. 1.20±0.55, 0.29~0.31. 2.50±1.69, 0.85±0.28. P<0.01). The expression of VEGF and its receptors(Fit-1 and KDR) in laryngeal squamous cell carcinoma was significantly higher in lymph node positive group than that in lymph node negative group (P<0.01 or P<0.05). Positive correlation was observed between VEGF and KDR in the laryngeal squamouse cell carcinoma and adjacent non-neoplastic laryngeal tissues (P<0.01). There was no correlation between VEGF and Fit-1 expression(P>0.05). Conclusion The intense expression of VEGF, Fit-1 and KDR in laryngeal squamous cell carcinoma provides strong evidence linking VEGF, Fit-1 and KDR expression to the angiogenesis associated with laryngeal squamous cell carcinoma. The positive correlation between VEGF expression and KDR expression clearly demonstrates that KDR is the main signaling receptor for VEGF in laryngeal squamouse cell carcinoma. The VEGF/KDR system plays a functional role in the progression of laryngeal squamous cell carcinoma.

Objective To explore the insertion of the teaching contents of nursing in rural doctors'higher education,so as to increase their understanding of medical knowledge and improve their medical skills.Method 90 rural doctors of Grade 2014 were chosen as the research object,and according to their willingness to participate,they were divided into control group and observation group.The students in the control group were mainly taught by the relevant knowledge of clinical medicine,while nursing teaching content was inserted in the teaching of observation group.In the teaching process,firstly,the teaching contents of nursing were reorganized,and then the teaching content of clinical medicine was taken as the main line,according to the development of teaching content,flexibly inserting nursing teaching part.The results of the final examination of the internal medicine of the two groups were compared and the specific learning experience of rural doctors was investigated.SPSS 20.0 was used to analyze the data,the count data was expressed as percentage,and chi square test was performed.Results The number of students in the control group and the observation group whose subjective score was more than 40 was 17.65％ and 48.72％ respectively.The number of students scoring ＜30 points accounted for 23.53％ and 5.13％ of the total number of students,and there was a statistically significant difference between the two groups(P＜0.05).The observation group had a better learning experience,and had little influence on the learning of clinical medical knowledge.Conclusion In the stage of education for rural doctors,inserting the teaching contents of nursing can improve the teaching effect.

Background Dry eye is the most common ocular surface disease.Anterior segment pattern of optical coherent tomography (OCT) is able to be used to assess the tear meniscus directly and accurately.Objective This study was to evaluate the correlation between measuring outcomes of tear meniscus by anterior segment module of OCT with signs and symptoms of dry eye.Methods A retrospective serial cases-observational study was designed.Thirty-six patients who completed Ocular Surface Disease Index (OSDI) and got a score more than 20 were enrolled in the First Hospital of Peking University from September to October,2010 under the informed consent.The upper and lower tear meniscus height (U/LTMH,μm),depth (U/LTMD,μm) and cross sectional area (U/LMA,mm2) were measured with the anterior segment module of OCT (RTVue.pitch).A series of clinical tests were performed,including tear film interferometry,break-up time (BUT),fluorescein corneal staining,lissamine green conjunctival staining and Schirmer Ⅰ test (S Ⅰ t).The correlations of measuring outcomes of tear meniscus by anterior segment module of OCT with dry eye-related examinations were analyzed with Spearman correlation coefficients.Results The values of UTMH,UTMD,UMA were (232±56) μm,(156±53) μm,(0.021 ±0.008) mm2,respectively,and the values of LTMH,LTMD,LMA were (222 ± 72) μm,(163 ± 65) μm,(0.020 ± 0.006) mm2,respectively.No significant differences were found in the measuring values between the upper and the lower tear meniscus (all at P＞0.05).The quantitative values of tear meniscus were not significantly correlated with the OSDI scores or tear film interferometry grade.Significant correlations were seen between UTMH,UTMD,UMA and BUT (r=0.46,P=0.01 ;r=0.40,P=0.02;r=0.43,P=0.01).LTMH,LTMD,LMA were positively correlated with BUT (r=0.61,P=0.00;r=0.64,P=0.00;r=0.64,P=0.00) and S Ⅰ t (r=0.54,P=0.02 ;r=0.46,P=0.01;r=0.55,P=0.01) as well as negatively correlated with fluorescein cornealstaining (r=-0.37,P=0.04;r=-0.37,P=0.04;r=-0.41,P =0.05) and lissamine green conjunctival staining (r =-0.56,P =0.01 ; r =-0.37,P =0.04 ; r =-0.47,P =0.00).Conclusions Anterior segment module of OCT is an effective approach to the measurement of tear meniscus.The measuring outcomes of anterior segment of OCT are associated with dry eye-related signs.
