1.In vitro antimicrobial effect of polymethylmethacrylate denture base with silver-containing antimicrobial agents of nanometer level

Objective:To study the antimicrobial effect of polymethylmethacrylate (PMMA) denture base with silver-containing antimicrobial agents of nanometer level against Streptococcus mutans and Candida albicans in vitro. Methods:Minimum inhibitory concentration (MIC) of the silver-containing antimicrobial agent against S. mutans and C. albicans was examined. According to the MIC, 4 concentrations(1,2,5 and 10 mg/ml) of the agent were selected for the preparation of antimicrobial PMMA resin base. Then, the antimicrobial effect of the resin base was examined by in vitro bacteria culture and the most probable number(MPN) counting. Results:The MICs of the agent against S. mutans and C. albicans were 0.1 mg/ml and 1 mg/ml respectively. With the agent at 1, 2, 5 and 10 mg/ml, the inhibition ratios of the base against S.mutans were 67.4%,71.3%,99.0 and 99.5% respectively, that against C. albicans were 25.8%,54.8%,90.3% and 93.0%, respectively. Conclusion:The polymethylmethacrylate resin base with silver-containing antimicrobial agent of nanometer level at 5 mg/ml has ideal antimicrobial activity against S. mutans and C. albicans.
2.Durability of in vitro antimicrobial effect of polymethylmethacrylate denture base containing silver-supported antimicrobial agent of nanometer level

Objective: To study the durability of the antimicrobial effect of polymethylmethacrylate (PMMA) denture base containing silver-supported antimicrobial agents of nanometer level against Streptococcus mutans and Candida albicans in vitro. Methods:The antimicrobial PMMA resin bases with the antimicrobial agent STR-1 at concentration of 5 mg/ml were prepared. Then the samples were divided into 4 groups: positive control group, the group immersed in distilled water at 57 ℃ for 14 days, the group irradiated by ultraviolet for 8 hours, and the group irradiated by ultraviolet for 8 hours after immersed in distilled water at 57 ℃ for 14 days. Then the inhibition ratios of the 4 groups against Streptococcus mutans and Candida albicans in vitro were tested. Results:The inhibition ratios of the 4 groups against Streptococcus mutans were 99%, 96%, 98%, and 90% in turn. The inhibition ratios of the 4 groups against Candida albicans were 91%, 82%, 90%, and 80% respectively. Conclusion: The antimicrobial effect of PMMA denture base containing silver-supported antimicrobial agents STR-1 of nanometer level at concentration of 5 mg/ml against Streptococcus mutans and Candida albicans in vitro is durable.
3.Effects of ketamine on preventing the increase of postoperative morphine requirement induced by fentanyl

AIM: To observe the effects of ketamine on preventing the increase of postoperative morphine requirement induced by fentanyl.METHODS: Sixty women undergoing total abdominal hysterectomy by spinal anaesthesia were assigned to 4 groups consisting of placebo(normal saline,C),fentanyl(3 bolus of 1 ?g/kg,at 15 min intervals,F),ketamine(infusion of 15 ?g?kg-1?min-1 ketamine from the skin incision until 20 min before the end of the surgery,K),ketamine and fentanyl(infusion of 15 ?g?kg-1?min-1 ketamine from the skin incision until 20 min before the end of the surgery plus 3 bolus of 1 ?g/kg fentanyl,at 15-min intervals,FK).The cumulative morphine consumption,pain score,and adverse effects(nausea, vomiting,hallucination,dizziness and itching) were recorded at 1,3,6,12,24,48 h postoperatively.RESULTS: There were no significant differences in age,weight,duration of surgery and the post-operative sensory block time.The cumulative morphine consumption in group F was significantly higher than those in group C at 3,6,12 h postoperatively(P
4.Comparison of response evaluation criteria in solid tumors (RECIST), European Association for the study of the liver (EASL) and modified RECIST criteria in evaluation of tumor response after transarteriai chemoembolization of primary liver cancer

Objective To compare the concordance among RECIST, EASL and modified RECIST criteria for the evaluation of tumor response after transarterial chemoembolization of primary liver cancer.Methods Fifty patients with primary liver cancer underwent 2 TACE cycles separated by 30-40 days.Triphasic helical CT or MRI scans were performed at baseline, at 4 weeks after TACE procedure, and 2 independent radiologists evaluated tumor response according to above-mentioned three different criteria. Chisquare test was used to compare the response rate, and kappa coefficients were used to evaluate the coherence. Results When tumor responses were evaluated using the RECIST-EASL and modified RECIST criteria, the numbers of the patients achieved complete response, partial response, stable disease,progressive disease were 0, 10, 30, 10; 6,21,14,9; 6,21,13,10 respectively. The objective response rates for three different criteria were 20%, 54%, 54% respectively ( P < 0. 01 ). Kappa coefficients between RECIST and EASL, between RECIST and modified RECIST, between EASL and modified RECIST were 0. 382, 0. 170, and 0. 857 (P = 0. 000). Conclusions RECIST criteria underestimates the extent of tumor response after TACE in primary liver cancer. Both EASL and modified RECIST criteria appear to agree with each other in determining treatment response. Furthermore, the modified RECIST is more convenient in clinical practice compared with EASL criteria.
5.Biocompatibility test of polymethylmethacrylate denture base resin containing silver-supported antimicrobial agent STR-1 at nanometer level

Objective: To evaluate the biocompatibility of polymethylmethacrylate(PMMA) denture base resin containing silver-supported antimicrobial agent STR-1 of nanometer level in vitro. Methods: According to the national standards for biological evaluation of dental materials, the cytotoxicity of denture base resin containing STR-1 at concentrations of 5 g/L and 10 g/L was examined by molecular filtrating method, and the hemolysis of STR-1, denture base resin containing STR-1 at concentrations of 5 g/L and 10 g/L was also surveyed. Results: The control denture base resin without containing STR-1 and the denture base resins containing STR-1 at concentrations of 5 g/L and 10 g/L were not cytotoxic to L929 cells. Two hours and 24 hours after cell culturing, the filter membranes of the control and experimental groups were stained evenly with blue color. The staining intensity was not decreased and the fading areas were 0 mm~2 during the culturing. The cytotoxicity grades were 0. The hemolysis rates of the antimicrobial agent STR-1 and the denture base resins containing STR-1 at concentrations of 5 g/L and 10 g/L were 1.7%, 3.5% and 3.7% respectively. They were less than the national guild standard 5% which represent no hemolysis. Conclusion: The PMMA denture base resins containing silver-supported antimicrobial agents STR-1 of nanometer level at concentrations of 5 g/L and 10 g/L exhibit good biocompatibility.
6.Research on algorithm of independent component analysis with two channel electrocardiograms.

According to the independent component analysis (ICA) theory, in order to seperate the fetal electrocardiograms (FECG) from the observed data, we must use the larger number of observed signals than that of independent components. This requires that the number of the sentors on the electrocardiograph must be larger than a particular value, which is in practice hard to be satisfied. So we proposed another algorithm using fewer channels of abdominal electrocardiograms, which combines ICA and adaptive noise cancellation to extract the FECG from two leads of abdominal electrocardiograms. The experiment results showed that we could obtain clear FECG by the method we proposed.

7.Study on the intervention effect of Qileng decoction (芪棱汤) on mitochondria mediated and caspase-9 dependent apoptopic pathway

Objective: To evaluate the effects and partly mechanism of Qileng decoction(QLD,芪棱汤) resisting focal cerebral ischemia/reperfusion(I/R) injury in rats by apoptosis and signal transduction pathway.Methods: The rats were randomly divided into three groups including sham operation group,normal saline(NS) control group and QLD group.The model of focal cerebral I/R injury was induced by using modified thread embolizing in rats.Rats were evaluated by neurologic function score at 2 hours after ischemia and 2,4,6,12,24 and 48 hours after cerebral reperfusion,and the pathological changes of nerve cells and mitochondria ultrastructure at pallium and hippocampus CA1 region were observed at 24 hours after reperfusion.Immunohistochemical method was performed to examine the expression of cytochrome C(cyt C) and caspase-9 at different time points after reperfusion.Apoptosis of nerve cells in ischemic penumbra(IP) was also characterized by terminal deoxynucleotidyl-transferase mediated dUTP-biotin nick end labeling(TUNEL) method.Results: Compared with NS control group,neurologic function scores at different reperfusion time points were improved and the pathological changes were ameliorated at 24 hours after cerebral I/R in QLD group.Mitochondria hydropsia was alleviated,mitochondrial cristae fragmentation and granulum basale shedding were diminished,and mitochondrial basical morphology was retained.Meanwhile,apoptosis index(AI) was decreased and the expressions of cyt C and caspase-9 were reduced in IP in QLD group.Conclusion: QLD intervenes in mitochondria mediated and caspase-9 dependent apoptopic pathway.QLD lowers AI and plays a role of protecting nerve by maintaining mitochondrial basical form,stabilizing mitochondrial membrane and inhibiting the release of cyt C and activation of caspase-9.The above actions are possibly some parts of mechanisms of QLD resisting focal cerebral I/R injury.
8.Discovery of biomarkers from serum in hepatocellular carcinoma using protein chip technique

Objective To screen serum biomarkers in patients with hepatocelhlar carcinoma by using surface-enhanced laser desorption and ionization time-of-flight mass spectrometry(SELDI-TOF-MS)technique and to evaluate its clinical implication in the patients whose alpha-fetoprotein were negative in the sera. Methods Proteomic spectra were generated by mass spectroscopy in 112 cases, including 57 cases AFP-negative hepatocellular carcinoma,and 55 cases of healthy control. The consequence was analyzed and the characteristic preteomic peaks was selected by using Biomarker Wizard. Results Seven low expressed potential biomarker were indentified with the mass-to-charge ratio of 4.2×103, 4.1×103, 6.7×103, 5.7×103, 6.5×103, 6.9×103, 5.8×103. The sensitivity for diagnosing hepatic cancer were 88.23 % and specificity was 92.31%. These peaks were not correlated with age, sex, tumor mass size, pathology grading and cirrhosis in hepatocellular carcinoma. Conclusion SELDI-TOF-MS offers a unique platform for the proteomic detection of hepatocellular carcinoma.It also offers an auxiliary diagnosis method to the patients whose alpha-fetoprotein are negative in the serum.
9.Correction of adhesive folded upper eyelid after blepharoplasty

Objective Adhesion or too highly located folds upper eyelid and even hlepharoptosis are common complications after double eyelid operation. The method was designed to correct these de-formities. Methods The incision of upper eyelid was designed under the scar. Skin was resected between the former and the new double line, if there was excessive skin. Completely relieve the adhesion until nor-mal tissue was exposed. Expose lateral extension of retro-orbicularis oculi fat pad. An upper pedicle fat flap was formed after cutting tissues off along bilateral borders. Appropriately and transversely cutting lateral portion of the pedicle made it rotate inside downward with enough blood supply. The fat flap was fixed with released orbital fat and the lower edge of fat flat fixed with upper border of supratarsal to restorate the volume. For severe condition, fat granule injection could be used after the skin was sutured. Results 42 cases (80 eyes) were treated in the past two years. The deformities of 32 cases were corrected by the application of retro-orbicularis oculi fat pad. Fat granule injection was applied for the other 10 pa-tients meanwhile. 30 cases (60 eyes) were followed up for 6 to 24 months. The results were satisfactory. Conclusion The methods are performed easily and the results are satisfied. It is worth of recommenda-tion.
10.Fucoidan Promotes the Reconstruction of Skin Equivalents.

In this study we investigated the effects of fucoidan on the proliferation of fibroblasts and the reconstruction of a skin equivalent (SE). Fucoidan significantly stimulated the proliferation of CCD-25Sk human fibroblasts and Western blot analysis demonstrated that fucoidan markedly increased the expression of cyclin D1 and decreased the expression of p27. Fucoidan was used to reconstruct SE. Immunohistochemical staining showed that the addition of fucoidan to dermal equivalents increased expression of proliferating cell nuclear antigen (PCNA) and p63. In addition, expression of alpha6-integrin was significantly increased by fucoidan, whereas expression of beta1-integrin, type 1 collagen, elastin, fibronectin did not markedly change. These results suggest that fucoidan has positive effects on epidermal reconstruction and will therefore be beneficial in the reconstruction of SE.
