1.Normal development of refractive state and ocular dimensional components in guinea pigs

Objective To study the emmetropization in guinea pig eyes during the normal development from birth in order to provide theoretical basis to use guinea pigs as model for research on near-sightedness.Methods Sixty-four guinea pigs were assigned to 8 groups (n=8,4 male/4 female).Each group underwent a series of ocular examinations at one of the 8 time-points (0,1,2,3,5,7,9 and 11 weeks),including refraction(R),radius of corneal curvature (RCC),depth of anterior segment(AS),thickness of crystalline lens (CL),length of vitreous chamber (VC)and axial length (AL).Pooled results from both eyes of the same animals with mixed sexes can be used for further assessment of the emmetropization with the associated biometrical changes of the eye.Correlations were made between RCC,AS,CL,VC,AL and R. Results There were no significant differences between the right eye and left eye or between male and female in results of all the examinations. The refraction at birth was (+5.25?0.22 )D in guinea pigs.This value rapidly decreased during the first 3 weeks,approaching(+1.34?0.61) D by 11 weeks.There was no significant difference in refraction between 9 and 11 weeks (P= 0.215). The RCC was (3.23?0.01) mm at birth,AS was (1.20?0.00) mm at birth,CL was (2.72?0.02) mm at birth and VC was (3.27?0.01) mm at birth,they increased within the first 3 weeks despite a transient decrease in the RCC within the first week.Such an increase continued except the AS which became constant after 3 weeks.The VC was more correlated to the emmetropization (r=-0.818,P
2.Comparative studies on genetic resource of characteristics of Alisma.

Based on the introduction and cultivation of Alisma germplasm which were from Fujian, Jiangxi and Sichuan provinces, the biological characteristics, morphological characteristics and quality were observed and studied. After three-year continuous experiment and monographic study, there were remarkable difference in the biological characteristics, morphological characteristics and product quality of Fujian Alisma, Sichuan Alisma and Jiangxi Alisma. Fujian Alisma and Jiangxi Alisma were the same plant species of A. orientalis, whereas Sichuan Alisma and Fujian Alisma were the different plant species of A. plantago-aquatica. The study results will provide the theoretical and practical basis for the genuine medicinal materials research and good agricultural practice (GAP) of Alisma.

3.Retrospective analysis of the effect of oral nutritional supplements during labor on delivery outcomes in low-risk pregnant women

Objective:To explore the effect of oral nutritional supplements (ONS) during labor on delivery outcomes in low-risk pregnant women and the risk factors of cesarean section.Methods:A retrospective analysis was conducted in a total of 206 full-term pregnant women with singleton and cephalic presentation at the delivery center in our hospital from March 15th to May 15th, 2022. Standard diet education was given to all those women by midwives. Pregnant women who received the enteral nutrient solution prepared by the Department of Clinical Nutrition during labor were in the ONS group ( n = 110), while those who did not were in the control group ( n = 96). The baseline characteristics and delivery outcomes were compared between the two groups, and the risk factors of cesarean section were also analyzed. Results:There were no significant differences in terms of maternal age, height, baseline weight, baseline body mass index (BMI), weight gain during pregnancy, prenatal BMI, gestational week, intraspinal labor analgesia, oxytocin, gastrointestinal intolerance, neonatal length, and weight between the two groups ( P > 0.05). However, the total oral energy intake during labor in the ONS group was significantly higher than that in the control group ([1 349.99± 569.51] kJ vs [249.59 ± 455.19] kJ, P < 0.01). The rate of vaginal delivery in the ONS group was significantly higher than that in the control group (93.6% vs 81.3%, P = 0.01), and the duration of the first stage of labor ([487.06 ± 232.94] min vs [416.17 ± 191.13] min, P = 0.03) was also significantly longer in the ONS group. There were no significant differences between the two groups in terms of the duration of the second and third stages of labor, perineal laceration, cervical laceration, vaginal laceration, amount of bleeding, hospital stay, and Apgar score after birth ( P > 0.05). Multivariate logistic regression analysis showed that maternal age was a risk factor for changing to cesarean section in women with low-risk pregnancies ( OR 1.20, 95% CI 1.03 to 1.40, P = 0.02), while ONS during labor ( OR 0.31, 95% CI 0.11 to 0.85, P = 0.02) and intraspinal labor analgesia ( OR 0.10, 95% CI 0.04 to 0.26, P < 0.01) were protective factors. Conclusions:ONS during labor can significantly increase the vaginal delivery rate, without increasing the incidence of gastrointestinal intolerance in low-risk pregnant women. Maternal age is a risk factor for cesarean section while ONS during labor and intraspinal labor analgesia are protective factors in women with low-risk pregnancies.
4.Analysis of diagnosis and treatment of Lesch-Nyhan syndrome

Objective:To explore the etiology, clinical diagnosis and treatment strategy of Lesch Nyhan syndrome.Methods:We retrospectively analyzed 2 patients with severe dyskinesia, mental retardation and complicated renal calculi who were admitted to the first people's Hospital of Zhengzhou in August 2019. Case 1, male, 9 years old, had multiple urinary calculi for 1 year. The patient came to the local hospital because double multiple kidney stones and bladder stonesa year ago. The patient had been treated with transurethral holmium laser lithotripsy for bladder stones. The results of infrared spectrum showed that the bladder stone was anhydrous uric acid stone. A week ago, color Doppler ultrasound showed multiple kidney stones and bladder stones. The patient was underdeveloped, mentally retarded and had a full-term cesarean section. There was no history of hypoxia, asphyxia and rescue of the patient. He had the following clinical manifestations: In the waking state, he was no language response to any stimulation. The nasolabial fold on the right was shallow and the corner of the mouth was oblique to the left. He lost the large movements such as lifting head, sitting alone, standing. The trunk showed torsion spasticity, limb muscle strength 2-3, limbs showing spastic hypertonia, limb joints stiff, hands showing fist-like, no involuntary movement and muscle fasciculation. The biceps reflex and knee tendon reflex were not elicited, and the pathological reflex was positive. Serum uric acid was 517 μmol/L. The Case 2 came from the same family, male, 6 years old, had the similar symptoms to his elder brother case 1. The family members complained on behalf of the child about intermittent fever for more than 2 years. The imaging examination of case 2 revealed kidney stones. Serum uric acid was 373 μmol/L. Whole Exome Sequencing and Sanger Sequencing were used to find the genetic causes of the two siblings. The NCBI-Homologene database was used to find the homologous sequence of the human HPRT1 gene, and the human HPRT1 gene sequence was compared with other species to analyze the protein conservation. The online website PredictProtein (http: //www.predactprotein) was used to predict the two-dimensional structure of the HPRT1 gene. The reported cases were summarized and same with the treatment plan.Results:A De novo mutation [c.571T>G(p.Tyr191Asp)] was found in the HPRT1 gene of the child, which was inherited from the mother. Lesch Nyhan syndrome can be diagnosed by the results of gene examination combined with clinical manifestations. The amino acid Tyr at the 191 position and the amino acids before and after it were highly conserved. Amino acid 191 was involved in the β-strand of the protein. We treated the patients with the lowest dose of allopurinol and children's conventional dose of potassium sodium bicitrate granules, and low purine diet. After 3 months of treatment, the serum uric acid was decreased, and the urinary calculi did not increase significantly.Conclusions:Combining with the clinical manifestations of children, HPRT1 gene might be the cause of pediatric disease and the two siblings could be diagnosed as Lesch-Nyhan syndrome. For such patients, the lowest dose of allopurinol and children's conventional dose of potassium sodium hydrogen citrate granule combined with diet could be more effective.
