1.Changes in the myocardial and liver tissues of exercise-induced fatigue rats after acupuncturing different acupoints

BACKGROUND:According to the theory of Traditional Chinese Medicine, stimulating certain acupoints of the body after exercise may improve its antioxidant capacity, but different acupoints have different effects. OBJECTIVE:To compare the effect of different acupuncture combinations on the antioxidant capacity of myocardial and liver tissues in rats after exhausted swimming exercise, and screen out the optimal acupoint combination for exercise-induced fatigue. METHODS:Seventy Sprague-Dawley male rats were divided into five groups: control group (n=10), model group (n=15),Sanyinjiao +Zusanli group (n=15),Shenshu +Zusanli group (n=15),Dazhui+Zusanli group (n=15). Except the control group, rats in the other four groups were subject to exhausted swimming. In the three acupuncture groups, rats were given acupuncture at corresponding acupint combinations at 15 minutes after exercise. RESULTS AND CONCLUSION:Compared with the control group, the activities of superoxide dismutase and glutathione peroxidase in the myocardial and liver tissues were significantly reduced in the model group, while the malonaldehyde content in cardiac muscle was significantly increased. Compared with the model group, the activities of superoxide dismutase and glutathione peroxidase in the cardiac muscle were significantly increased inSanyinjiao +Zusanli group, the superoxide dismutase in liver tissue was significantly increased inShenshu+Zusanli group, the malonaldehyde content in the myocardial tissue was significantly decreased in Sanyinjiao +Zusanli group and Dazhui +Zusanligroup, the malonaldehyde content in the liver tissue was significantly decreased in Sanyinjiao +Zusanligroup. Different acupuncture combinations have different effects on the activities of superoxide dismutase and glutathione peroxidase, as wel as the malonaldehyde content in the myocardial and liver tissues of fatigued rats. Acupunture could increase the antioxidant ability in cardiac muscle of rats and the best combination might beDazhui+Zusanli; acupoint combinations ofShenshu +Zusanli andSanyinjiao +Zusanli are specific for liver tissue.
2.Astragalus mongholicus-induced differentiation of rat bone marrow mesenchymal stem cells

BACKGROUND: Extract of astragalus mongholicus has good effects on antioxidization and oxygen free radical scavenge. Bone marrow mesenchymal stem cells (BMSCs) have multiple directional differentiation potential and superiority of autologous transplantation. This can provide a new way for treating neurodegenerative diseases.OBJECTIVE: To explore the differentiation of BMSCs of rats following astragalus mongholicus induction.DESIGN: A cytological in vitro study.MATERIALS: One clean male rats aged 6 weeks were purchased from Experimental Animal Center of Chinese Academy of Medical Sciences. Astragalus mongholicus injection (lot number 060105) were obtained from Dali Pharmaceutical Co., Ltd.METHODS: Rat BMSCs were in vitro isolated, cultured and purified by the whole bone marrow method. At the fourth passage, BMSCs at a density of 4×10<'8>/L were incubated in 12-well plate with a coverslip. After 80% 90% cells were confluent, the medium was changed. BMSCs were then incubated in DMEM, supplemented with 200 g/L astragalus mongholicus injection and 15% fetal bovine serum, for 6 days. BMSCs in the control group were not treated with astragalus mongholicus injection.MAIN OUTCOME MEASURES: Morphological changes in BMSCs were observed under the inverted microscope before and after induct.ion. Expression of specific markers was determined by immunocytochemical staining following induction.RESULTS: Primary cultured BMSCs were round. At the fourth passage, BMSCs were spindle. BMSCs after induction showed processes, which became more with prolonged time. Some processes were branch-shaped, forming a network structure. Results of immunocytochemical staining demonstrated that many cells were positive for nestin, neuron specific enolase and glial fibrillary acidic protein on day 3, and abundant cells were positive for microtubule-associated protein-2 on day 6.CONCLUSION: Astragalus mongholicus induces differentiation of BMSCs into neural stem cells, and then promotes the differentiation into neurons or glial cells, and makes differentiated cells mature and aging.
3.Advances in Application of Blue Laser Imaging in Diagnosis of Early Upper Gastrointestinal Cancer

Blue laser imaging (BLI) is a new endoscopic system equipped with the laser beam emitting two different wavelengths.It produces bright and high resolution images for observation of microvascular and microsurface patterns of esophageal and gastric mucosa, helping the diagnosis of early upper gastrointestinal cancer.Compared with the existed endoscopic techniques, BLI shows its unique advantages.In this article, advances in application of BLI in diagnosis of early upper gastrointestinal cancer were reviewed.
4.Distribution and Drug Resistance of Pathogens in a Hospital During Last Three Years

OBJECTIVE To investigate the distribution and resistance of commonly encountered pathogens,and provide reference of antimicrobial agents.METHODS For the clinical specimens during the last three years,flora identification and bacteriostatic test were operated with Microscan WalkAway-40.RESULTS Gram-negative bacteria were 252,507 and 742 strains,Gram-positive bacteria were 142,166,and 243 strains,fung were 0,26,and 229 strains from 2002 to 2004,respectively,the most commonly encountered Gram-negative bacteria were Escherichia coli,Klebsiella pneumoniae,Pseudomonas aeruginosa,Enterobacter cloacae,and Acinetobacter baumannii;the Gram positive bacteria were Staphylococcus aureus,S.epidermidis,S.haemolyticus,and Enterococcus.CONCLUSIONS It is important for reasonable drug application to know the distribution and resistance of commonly encountered bacteria.
5.Antibiotics Resistance of Staphylococcus aureus Isolated from Nosocomial Infection Patients and Detection of Resistance Genes

OBJECTIVE To investigate the antibiotics resistance and the presence of resistant gene of Staphylococcus aureus isolated from nosocomial infection patients in our hospital.METHODS Antibiotic susceptibility tests were conducted by Microscan WalkAway-40,the antibiotics resistant genes of aac(6')/aph(2″),aph(3')-Ⅲ,ant(4',4″),TEM,tetM,macA,erm and qacA/B of 16 MRSA and 24 MSSA strains were detected by PCR.RESULTS The percentage of MRSA in this investigation was 59.4%,the positive rate of ?-lactamases of 192 S.aureus strains was 98.9%,resistant rate of S.aureus to ?-lactam antibiotics was 52.1-99.0%,and to erythromycin,clindamycin,clarithromycin,ciprofloxacin,norfloxacin,gentamicin and tetracycline was 52.1%,78.1%,85.5%,56.3%,61.1%,55.2%,and 69.8%,respectively,but to nitrofurantoin and sulfamethoxazole/trimethoprim were relatively lower(1.1% and 10.5%) than the others.VISA,h-VISA and VRSA were not measured.To the most of antibiotics,the resistant rate of MRSA was higher than MSSA.The presence of resistant gene of MRSA and MSSA was different.There was more positive percentage of gene in MRSA than in MSSA.CONCLUSIONS Antibiotics resistance of S.aureus isolated from nosocomial infection patients in our hospital is severe.Compared to MSSA,MRSA has higher antibiotic resistant rate and more complicated drug-resistant mechanism.Some measures should be done to reduce the antibiotic resistance of S.aureus.
6.Protective mechanism of exogenous hydrogen sulfide on inflammatory and oxidative damage of myocardium due to stress after amputation in rats

Objective To investigate the protective mechanism of hydrogen sulfide(H2S) on inflammatory and oxidative damage of myocardium due to stress after amputation,and explore the relationship between H2S signal pathway and inflammatory and oxidative damage.Methods Twenty-eight adult male Wistar rats(weighing 260-300g) were randomly divided into 4 groups(7 each):control group,trauma group,NaHS group and PPG group.Rat model of traumatic stress was established in the rats in the 3 latter groups,and the animals were sacrificed 6 hours after modeling.The rats in control group were also sacrificed at the same time point.Rats in NaHS and PPG group respectively received intraperitoneal injection of NaHS(28?mol/kg) or PPG(50mg/kg).The level of H2S in plasma was measured by sensitive sulphur electrode,the activity of CSE in myocardium was determined by methylene blue method,myeloperoxidase(MPO) in plasma and myocardium was measured with ultraviolet spectrophotometry,and malondialdehyde(MDA) in plasma and myocardium was measured by TBA method.Results Compared with the control group,the content of H2S in plasma decreased,MPO and MDA in plasma and myocardium increased 6 hours after amputation(trauma group).Compared with the trauma group,the contents of H2S and MPO in plasma and the activity of CSE in myocardium increased,MDA in plasma and MPO and MDA in myocardium decreased in NaHS group,while the contents of H2S and MPO in plasma and the activity of CSE in myocardium decreased,MDA in plasma and MPO and MDA in myocardium increased in PPG group.Conclusion Inflammatory and oxidation injuries may occur in rat's myocardium after amputation,and H2S can offer a protective effect on myocardium by decreasing inflammatory and oxidative damage.
7.Genotyping of SCCmec and Detection of Antibiotic-resistant Genes in Meticillin-resistant Staphylococcus aureus

OBJECTIVE To investigate the staphylococcal cassette chromosome mec(SCCmec) genotype characteristics and antibiotic-resistant genes in meticillin-resistant Staphylococcus aureus(MRSA) isolated from Lianyungang.METHODS The SCCmec of clinically isolated MRSA strains were genotyped with a novel multiplex PCR strategy reported by Zhangetal.Antibiotic-resistant genes of aac(6′)/aph(2″),aph(3′)Ⅲ,tetM,erm,TEM,and ant(4′,4″) were analyzed by traditional PCR.RESULTS The isolates were almost SCCmec Ⅲ positve,only one isolate couldn′t be typed.The positive rates of aac(6′)/aph(2″),aph(3′)Ⅲ,tetM,and erm were 98%,46%,72% and 86%,respectively.TEM and ant(4′,4″) tested were all negative.CONCLUSIONS Almost all genotypes of MRSA prevailing in Lianyungang carry the SCCmec Ⅲ gene.There are high positive percentages of antibiotic-resistant genes of aac(6′)/aph(2″),aph(3′)Ⅲ,tetM and erm in the isolates.The novel multiplex PCR strategy recommended by Zhang et al can be applied into genotyping study of MRSA SCCmec effectively.
8.Nuclear magnetic resonance-based metabonomics features before and after 800 meters race

BACKGROUND:Metabolic characteristics of the 800 meters race is not very clear, which creates a lot of obstacles for the exercise training of 800 meters race. OBJECTIVE: To investigate the characteristics of material and energy metabolism of 800 meters race, to improve the scientific training of the 800 meters race and to verify the feasibility of nuclear magnetic resonance-based urine metabonomics in sports training monitoring. METHODS: Seven middle distance athletes were subject to an 800 meters race at ultimate intensity, and their urine metabonomics analysis was conducted in combination with the Human Metabolism Database and other databases before and after exercise. RESULTS AND CONCLUSION:OPLS-DA model showed that the metabolic samples before and after movement presented the cluster-like distribution. The change was mainly caused by the folowing metabolites, including leucine, lactic acid, creatinine, succinic acid, hypoxanthine, and adenosine. Through the analysis we could infer that the 800 meters race is a sport event mainly dependent on glycolysis for energy supply, working together with raw phosphate and aerobic oxidation for energy supply. The amino acid metabolism and methane metabolism of the athletes were more vigorous after 800 meters running, and moreover, there were certain oxidative stress during the 800 meters race. These findings indicate that nuclear magnetic resonance-based urine metabonomics takes the advantages of invasive, high sensitivity, less sample requirement, simple processing.

10.18F-FDG PET/CT in Differential Diagnosis of Incidental Benign or Malignant Thyroid Focal Hypermetabolic Lesions

Purpose To investigate the significance of PET/CT in differential diagnosis of incidental thyroid benign or malignant lesions with high fluorodeoxyglucose (FDG) uptake.Materials and Methods Forty cases with focal high FDG uptake lesion in thyroid accidently detected by PET/CT were retrospectively collected. The subjects were dived into physiological uptake group (n=18), benign lesion group (n=16) and malignant lesion group (n=6) according to the pathology or follow-up examinations. The average SUVmax of different groups was analyzed, and ROC curve was used todetect benign or malignant lesions.Results The average SUVmax was 3.7±1.1 in physiological uptake group, 4.9±2.3 in benign lesion group and 9.5±4.8 in malignant lesion group. Significant difference was noticed among the three groups. The average SUVmax in malignant lesion group was higher than that of the other two groups with statistical difference (P<005). No significant difference was observed between physiological uptake group and benign lesion group (P>0.05). ROC analysis showed that the sensitivity and specificity for thyroid malignant tumor were 100.0% and 87.2% at SUVmax of 8.5.Conclusion18F-FDG PET/CT plays an important role in differentiating benign lesions of thyroid from malignance, but it is not helpful to identify benign lesions from physiologic uptake.
