2.An experimental study on distribution of musk into the brain through blood brain barrier

OBJECTIVE: To study the possible pathway of the effect of musk on brain disorder, distributing into the brain through blood brain barrier. METHODS: We used the musk ketone (muscone), a main composition of musk, to inject through the tail vein of the rats into the blood and took the brain and other organs at different times to make samples. Then gas chromatography was used to measure the distribution of muscone in the brain and other organs. RESULTS: Muscone could pass through the normal rat's blood brain barrier into the brain and soon reached the highest peak and remained in higher concentration, and more slowly metabolized as compared with other organs. CONCLUSION: Musk distributing into the brain through blood brain barrier provides the basis for its effect in treating brain disorders. Chromatography is an effective method to study the active composition of Chinese herbal medicine distributing through the blood brain barrier into the brain.
3.The diagnosis and treatment for major arterial injuries of the extremities

Objective To investigate the diagnosis and treatment of major arterial injuries of the extremities. Methods In the period between September 1999 and June 2002, 129 patients with 176 arterial injuries were admitted to our hospital. 122 patients of them with 162 arterial injuries underwent microsurgical repair. Results The limbs of 116 patients with 154 arterial injuries survived. The total survival rate of affected extremities was 89.9% ; the blood vessel recanalization rate was 95.06% . Conclusion Careful physical examination and advanced vascular Doppler equipment can avoid missed diagnosis, and microsurgical technique in vascular repair can successfully save limbs.

Objective To evaluate the value of measurement of the width of pars compacta of substantia nigra(SNc) and SNc's ratio to midbrain diameter in diagnosing Parkinson's disease(PD) and differentiating PD from vascular parkinsonism(VP) on routine MRI.Methods The width of SNc, midbrain diameter, and SNc's ratio to midbrain diameter were measured in 60 patients with PD (38 patients with Hoehn-Yahr stage Ⅰ~Ⅱ, 14 patients with Hoehn-Yahr stage Ⅲ, and 8 patients with Hoehn-Yahr stage Ⅳ), 60 patients with VP (26 patients with Hoehn-Yahr stage Ⅰ~Ⅱ, 20 patients with Hoehn-Yahr stage Ⅲ, and 14 patients with Hoehn-Yahr stage Ⅳ), and 60 age-matched heathly control subjects by means of routine axial T 2-weighted MRI. Objective To investigate the effect of rhein on endothelial plasminogen activator inhibitor-1 (PAI-1) mRNA expression and protein synthesis induced by transforming growth factor ?1 (TGF-?1), and explore the protective mechanism of rhein on human endothelial cells. Methods A human umbilical endothelium derived cell line (ECV-304) from ATCC was used in this study. The endothelial PAI-1 mRNA expression and protein synthesis were detected by Northern blot and flowcytometry analysis respectively. The activity of phospho-p44/p42 MAPK induced by TGF-?1 was determined by immunoprecipitation analysis and Western blot. Results TGF-?1 (2?g/L) could increase rapidly endothelial PAI-1 mRNA expression and persist for 24 hours. The PAI-1 mRNA expression induced by TGF-?1 in endothelial cells was inhibited by incubating with rhein. Rhein could also inhibit the endothelial PAI-1 protein synthesis. Further study revealed rhein showed a significantly inhibitory effect on the activity of phospho-p44/p42 MAPK induced by TGF-?1 in human endothelial cells. Conclusions Rhein shows a protective action on endothelial cells by inhibiting PAI-1 overexpression, and this may provide beneficial effects on the treatment of diabetes mellitus and its complications.
8.Nursing care to hemodialysis patients with hypocalcemia after parathyroiddectomy

Objective To explore the postoperative care to hemodialysis patients with hypocalcemia after parathyroidectomy with refractory secondary hyperparathyroidism. Methods The nursing measures included timely correction of hypocalcemia, nursing of muscular spasm, dietary nursing and rational medication. Five of them developed numbness in the limbs, which was alleviated by administration of venous transfusion of calcium gluconate. After six months, the blood calcium in 6 patients resumed to normal level. For 1 case with lower blood calcium, the level of blood calcium maintained at 1.8~2.0mmol/L after supplement with high dose of calcium and high -calcium hemodialysis. Conclusions The hemodialysis patients with refractory secondary hyperparathyroidism after parathyroiddectomy are susceptible to hypocalcemia. The timely correction of hypocalcemia and related nursing measures are of significance for their recovery.
9.Screening of invasion and metastasis related genes regulated by Stat3 in pancreatic cancer SW1990 cell

Objective To screen the genes related with signal transducers and activators of transcription 3 (STAT3) regulating pancreatic cancer invasion and metastasis by gene chips.Methods Human pancreatic cancer cell line SW1990 stably expressing low level of Stat3 was established by lentivirus transfection,while cells transfected with mock plasmid and cells without transfection served as control groups.The differences of invasion and metastasis related genes expression among the three groups were screened by gene chips.STAT3 mRNA and protein expression was measured by real-time PCR and Western blot.Three differentially expressed genes (MMP-7,IL-1β and IgTα7) were verified.ResultsThe expression level of STAT3 mRNA was 0.391 ± 0.037 after pancreatic cancer SW1990 cell trarsfected with STAT3 targeted lentivirus,which was significantly lower than those in mock plasmid group (1.002 ± 0.015) and nontransfected group ( 1.206 ± 0.042,P < 0.05 ) ; the expression level of STAT3 protein was 182.38 ± 65.32,which was significantly lower than those in mock plasmid group (223.40 ±58.40) and non-transfected group (212.33 ±53.69).Eight invasion and metastasis related genes of SW1990 lowly expressing Stat3 were upregulated,while 3 genes were down-regulated.By verification,the mRNA level of MMP-7 and IL-1β were lower than in control group transfected with mook plassmid(0.287 ± 0.115 vs 1.010 ± 0.124,t =19.45,P =0.000;0.490 ± 0.10 vs 1.002 ± 0.002,t =13.83,P =0.000),but the mRNA level of IgTα7 was not decreased (1.173 ±0.280 vs 0.998 ±0.003,t =4.236,P =0.094).Meanwhile,the protein level of MMP-7 was significantly down-regulated when Stat3 was knocked down.ConclusionsStat3 causes changes of expressions of many invasion and metastasis-related genes of SW1990,and MMP-7 may be the main target gene regulated by Stat3.
10.Effect of high mobilityg group box-1 protein on interleukin-2 and interleukin-2 receptor α expressions in human T lymphocytes

Objective To investigate the effect of high mobility group box-1 protein (HMGB1) on inter-leukin-2 (1L-2) and interleukin-2 receptor α (IL-2α) expressions in human T lymphocytes and its potential regulat-ing mechanism in vitro. Method Human T lymphocytes were isolated and suspended, the cells were cultured with 20 μg/mL phytohemagglutinin (PHA) in 5% CO_2 at 37 ℃, recombinant human HMGB1 (rhHMGB1, 0, 10, 100, 1000 ng/mL) was added with the PHA and cultures were centrifuged at 12 and 48 hours for cell collect-ing. Reverse transcription polymerase chain reaction (RT-PCR) amplification was perfomed to determine gene ex-pressions of IL-2, IL-2Rα. IL-2, sIL-2R protein levels in cell culture supematants were measured by ELIZA. Re-sults After coincubated with rhHMGB1 (10, 100, and 1000 ng/mL) for 12 hours, IL-2 levels in cell culture su-pernatants respectively were 0 . 064 ± 0. 017 μg/L, 0.076±0.033 μg/L, and 0.061 ±0.02 μg/L, which were significantly higher compared with the untreated cells (0.045±0.011 μg/L, P < 0.05 or P < 0.01). Mean-while, IL-2 mRNA expression was markedly up-regulated following rhHMGB1 stimulation in various doses (F = 4.6872, P < 0.01). At 48 bourn, however, both IL-2 mRNA expression and protein production tended to de-crease along with an increased dose of dd-IMGB1 stimulationn. IL-2/sIL-2R ratio in 1000 ng/mL rhHMGB1 was markedly lower than that in 10 ng/ml rhHMGB1 (0.036±0.015 vs.0.055±0.017, P <0.05), together with down-regulation of IL-2Rα mRNA expression(P <0.01). Conclusions These data indieated that HMGBI could marked influence the IL-2/IL-2R expression in human T lymphocytes. With the increase in stimulating doses and prolongation of time, HMGBI might down-regulate T cell-mediated immune response of human lymphocytes.
