BACKGROUND:How to obtain a sufficient number of cells is one of the key issues in the celltransplantation therapy, and studies have shown that stem cellproliferation can be promoted by reasonable stimulus.
OBJECTIVE:To investigate reduced glutathione effects on biological characteristics of human umbilical cord blood mesenchymal stem cells.
METHODS:The cells were divided into two groups:the control group consisted of the normal human umbilical cord blood mesenchymal stem cells, and in the experimental group, human umbilical cord blood mesenchymal stem cells were treated with 0.15 g/L reduced glutathione.
RESULTS AND CONCLUSION:At days 5, 7, 9, cells treated with 0.15 g/L reduced glutathione showed higher absorbance values than those in the control group (P<0.05). Flow cytometry showed reduced glutathione had no effects on CD29, CD44, CD45, CD105 expression. Real-time PCR results showed reduced glutathione was capable of promoting extracellular signal-regulated kinase mRNA expression (P<0.05). Findings from this study showed that 0.15 g/L reduced glutathione can promote the proliferation of human umbilical cord blood mesenchymal stem cells.

BACKGROUND:In recent years, studies have shown that mesenchymal stem cells can secrete various growth factors, and has certain application prospect in the treatment of liver fibrosis.
OBJECTIVE:To explore the different sources of mesenchymal stem cells in the treatment of liver fibrosis in rats. METHODS:Fifty Sprague-Dawley rats were given intraperitoneal injection of carbon tetrachloride to construct the model of liver fibrosis. Another 10 normal Sprague-Dawley rats were used as normal controls. Model rats were randomly divided into five groups, 10 rats in each group, including model control group, normal saline group, bone marrow mesenchymal stem cellgroup, umbilical cord mesenchymal stem cellgroup, and umbilical cord blood mesenchymal stem cellgroup. After 8 weeks of modeling, different sources of mesenchymal stem cells at a density of 2×106 were injected via tail vein into model rats. After 12 weeks, the rats were kil ed, and serum alanine aminotransferase, aspartate aminotransferase and albumin levels were detected as wel as the expression of type I col agen and glial fibril ary acidic protein in liver tissue was detected by fluorescence quantitative PCR method.
RESULTS AND CONCLUSION:We have successful y established the SD rat model of hepatic fibrosis. Bone marrow mesenchymal stem cells aggravated hepatic fibrosis, umbilical cord blood and umbilical cord mesenchymal stem cells could reduce the level of hepatic fibrosis in rats. Umbilical cord mesenchymal stem cells had the most obvious effect that significantly reduced expressions of alanine aminotransferase, aspartate aminotransferase and type I col agen and glial fibril ary acidic protein. The results showed different sources of mesenchymal stem cells have different effects on rat fibrosis. Bone marrow mesenchymal stem cells aggravate hepatic fibrosis, and umbilical cord and umbilical cord blood stem cells can al eviate hepatic fibrosis in rats.

This study was aimed to establish a rabbit model of bridging artery defect with autogenous vein under equal arterial and venous compliances by selecting an appropriate anastomosing tension based on assessing the influence of vessel's longitudinal strain on compliance. The exponential form P = M1 x [eM2(v-v0)-1] was employed to fit the blood pressure-unit volume curves under different stretch ratios(lambda) obtained from real curves on 13 femoral arteries and 12 veins whose uniaxial loading tests at longitudinal directions had been performed, then the quantic M = a1 lambda 5 + a2 lambda 4 + a3 lambda 3 + a lambda 2 + a5 lambda + a6 was done to fit M-lambda data. Results showed the differential form dv/dp = 1/(M1 x M2 + 11.78 M2) derived from P = M1 x [eM2(v-v0)-1], and lambda reflected the relationship between compliance and stretch ratio (or longitudinal tension) under average artery pressure 11.78 KPa well. It was found that under longitudinal tension 1.19 g, the arterial and venous compliances were equal(0.031 microliter/KPa) while the respective stretch ratios were 1.32 and 1.67.

Objeetive To investigate the clinical efficacy of treating stomach cance belonging to spleen and stomach deficient cold type with self-designed formula and chemotherapy. Methods 100 patients of stomach cancer belonging to stomach and spleen deficient cold were randomly grouped into a control group and a treatment group, with 50 cases in each group. The control group was only treated with chemotherapy, and the treatment group was additionally treated with self-designed Chinese formula on the basis of the control group. After 3 months' treatment, the curative effect, the improvement of life quality, the changes of body weight, the improvement of peripheral blood and immune function, and the occurrence of adverse reaction were observed. The survival rate of patients after 1,3 and 5 years' treatment was investigated. Results ① Short term curative effect:the total effectiveness of the control group and the treatment group was 60.0% (30/50) and 30.0% (15/50) respectively, with the treatment group being better than the control group(Z=-2.100,P<0.05);② life quality:theimprovement rate of life quality in the treatment group and the control group was 64.0% (32/50) and 20.0% (10/50) respectively, with the treatment group being better than the control group (Z=-3.259,P<0.05); ③ body weight:the improvement rate of body weight in the treatment group and the control group was 24.0% and 16.0% respectively, and there was no significant difference (Z=-1.815,P>0.05) ; ④ peripheral blood:the peripheral blood abnormality rates of treatment group was obviously lower than the control group (Z=-4.286,P<0.05) ;⑤ immune function changes:CD4+、CD8+、and the activity of NK cell of two groups after the treatment [the treatment group was (40.8 ± 4.1)%,(26.1-3.2)%,(83.57-2.52)% respectively; the control group was (31.7±2.9)%、(33.8±3.0)%、(92.88±3.83)% respectively] wasobviously improved compared with the same group before the treatment [the treatment group was (34.2±3.5)%,(31.0±2.0)%,(85.01±2.92)% respectively; the control group was (34.0±3.2)%,(30.9±2.2)%,(85.02± 3.48) % respectively].The immune indexes (CD4+、CD8++、CD4+/CDg+and the activity of NK cell)in the treatment group presented significant difference (P<0.01) than the control group after the treatment.⑥adverse reaction:compared with the control group, the occurrence rate of adverse reaction of the treatment group was obviously lower than the control group, and there was significant difference (Z=-5.297,P<0.05).⑦survival rate:the survival rate of patients after 1,3 and 5 years' treatment in the treatment group was better than the control group, and there was significant difference (x2=5.263,12.96,7.895,P<0.05).Conclusion The combination of self-designed Chinese formula and chemotherapy could effectively relieve the clinical symptoms, improve the immunity, reduce the occurrence of adverse reaction, and improve the life quality of patients with stomach cancer belonging to stomach and spleen deficient cold type.

Human dental pulp stem cells (DPSCs) have emerged as an important source of stem cells in the tissue engineering, and hypoxia will change various innate characteristics of DPSCs and then affect dental tissue regeneration. Nevertheless, little is known about the complicated molecular mechanisms. In this study, we aimed to investigate the influence and mechanism of miR-140-3p on DPSCs under hypoxia condition. Hypoxia was induced in DPSCs by Cobalt chloride (CoCl
