1.Antidepressant effect of Epimedium brevicornum extracts

Objective To study antidepressant effects of Epimedium brevicornum extracts.Methods(Behavioral) despair models of male mice,tails suspension test(TST),and forced swimming test(FST) were used to evaluate the effects of E.brevicornum extracts on behavioral,monoamine oxidas(MAOA) and monoamine oxidase B(MAO-B) activities in brain and liver tissue,and MDA level in liver tissue of mouse.Reserpine antagonistic model was also used to investigate possible antidepressant mechanisms of E.brevicornum extracts.Results The extracts of E.brevicornum(25,50, 100,and 200 mg/kg) significantly reduced the duration of murine immobility in TST and FST,inhibited MAO-A and MAO-B activities in brain and liver of mice,and reversed the elevated liver MDA level in mice in TST.There was no significant amelioration in the decreases of body temperature in mice of Reserpine antagonistic model.Conclusion(E.brevicornum) extracts possesses the definite antidepressant properties.
2.The lipid-lowering effect and possible mechanism of apobec-1 in nephrotic syndrome rabbits

Objective To observe the effect of liver acquired expression of apobec-1 on blood lipid metabolism, hepatic low density lipoprotein receptor (LDLR), and hepatic low density lipoprotein receptor related protein (LRP) in nephrotic syndrome(NS) rabbits and to explore the lipid-lowering effect and possible mechanism.Methods Thirty healthy ordinary level male new Zealand rabbits were randomly divided into 3 groups:sham operation group (n =10), N S group (n =10), and apobec-1 treatment group (n =10).Adaptive feeding was given for 1 week, then NS group and apobec-1 treatment group underwent left nephrectomy,and 1 week later,adriamycin (4 mg/kg) was used to construct the NS model by way of ear vein injection.In the 11th week after operation, apobec-1 recombinant adenovirus (1 × 1013 virus/kg) was injected through ear vein in apobec-1 treatment group.The 12th week after operation, all rabbits were sacrificed.Right kidney, liver, blood and 24 hour urine were collected.In 3 groups, 24 hour urinary protein (24UPr), albumin (Alb), blood urea nitrogen (BUN), creatinine (Cr), blood lipid were detected.Renal pathology was observed by means of HE staining.Expressions of liver LDLR, LRP were observed by using Western blot.Results (1) There were significant differences among the 3 groups in 24UPr (F =42.778, P =0.000), Alb (F =3.819, P =0.034), BUN (F =6.562, P =0.005), Cr (F =16.076, P =0.000), total cholesterol (TC) (F =17.531, P =0.000), total triglyceride (TG) (F =6.192, P =0.006), very low density lipoprotein (VLDL-C) (F =6.192, P =0.006), low density lipoprotein (LDL-C) (F =34.924, P =0.000) and apolipoprotein B100 (ApoB100) (F =5.180, P =0.012) and apolipoprotein B48 (ApoB48) (F =6.161, P =0.006).(2) Compared with the sham operation group, NS group showed that 24UPr, BUN, Cr, TC, TG, VLDL-C, LDL-C, ApoB100 increased, but Alb decreased, and there was statistical significance (all P < 0.05).(3) Compared with NS group, apobec-1 treatment group showed that TC, TG, VLDL-C, LDL-C, ApoB 100, ApoB48 decreased, and there were statistical significances (all P < 0.05).(4) Compared with the sham operation group, apobec-1 treatment group showed that 24UPr, BUN, Cr increased, but Alb, apolipoprotein B48 (ApoB48) decreased, there were statistical significances (all P < 0.05).(5) There were significant differences of hepatic protein expression among the 3 groups in LRP (F =44.180, P =0.000), LDLR (F =63.141 ,P =0.000).Compared with the sham operation group and NS group, apobec-1 treatment group showed that the expression of LRP was up-regulated (P < 0.01,0.05), while the expression of LDLR was down-regulated (all P < 0.05).Conclusions When liver acquired expression of apobec-1 in NS, it could up-regulate LRP,accelerate the elimination of ApoB48-lipoproteins, and produce a certain lipid-lowering effect.
3.Experimental study of Apobec-1 recombinant adenovirus in treatment of renal hyperlipemia

Objective To explore the effect of the liver acquired expression of apolipoprotein B editing catalytic polypeptide-1 (Apobec-1) on hyperlipidemia of renal disease. Methods Thirty healthy ordinary level male New Zealand rabbits were randomly divided into three groups: sham operation group, nephropathy group, and Apobec-1 treatment group (Each group has 10 rabbits). Adapt feeding for one week, nephropathy group and Apobec-1 treat-ment group underwent left nephrectomy, and one week later, adriamycin (4 mg / kg) was used to construct the ne-phropathy model by ear vein injection. The eleventh week after operation, apobec-1 recombinant adenovirus (1 × 1013 Virus/ kg) was injected by ear vein in apobec-1 treatment group. Results ① Compared with the sham operation group, nephropathy group showed that 24UPr, BUN, Cr, total cholesterol (TC), total triglyceride (TG), very low density lipoprotein (VLDL-C), low density lipoprotein (LDL-C), apolipoprotein B100 (ApoB100) were increased(P < 0. 05), but Alb was decreased(P <0. 05). ② Compared with the nephropathy group, Apobec-1 treatment group showed that TC, TG, VLDL-C, LDL-C, ApoB100, ApoB48 were decreased(P < 0. 05). ③ Compared with the sham operation group, Apobec-1 treat-ment group showed that 24UPr, BUN, Cr were increased( P < 0. 05), but Alb, ApoB48 were decreased( P <0. 05). ④ Compared with the sham operation group and nephropathy group, Apobec-1 treatment group showed that the expression of Apobec-1 and ApoB48 were up-regulated (P < 0. 01). Conclusion When liver aquires expres-sion of Apobec-1 in hyperlipidemia of renal disease, it can reconstruct ApoB mRNA, increase the synthesis of ApoB48-lipoprotein, and play a certain lipid-lowering effect.
4.The Evaluation of Therapy Efficacy of Gamma Knife on Pituitary Adenoma by Using MR

Objective To explore the therapy efficacy of gamma knife on pituitary adenoma based on the changes of MR imagings and hormone levels before and after gamma knife treatment .Methods MR imagings and hormone levels, clinical material were retrospectively analysed on 45 patients(altogether 59 times MR examinations)followed up from 1~28 months. Both plain and enhancement MR scans were performed on each MR examination. Results 59 cases were followed up for 1~28 months . The tumor size shranked in 31 cases (52.5%), no changs in 25 cases (42.4%), increased in 3 cases(5%). Of the 59 cases , intratumoral cyts were observed in 15 cases (25%). Functional pituitary adenomas were followed up in 23 cases. The hormone levels droped in 13 cases (56.5%), the number of hormone levels droped in about the sixth month after gamma knife treatment was 8.6% , in 1 year was 13% and in 2 years was 34.8%. Conclusion Gamma knife can effectively keep the pituitary adenomas from their growing up; avoid the overproduce of functional pituitary adenomas . The treatment effects of gamma knife can be enlarged with the time gone.
5.Expressions of Uroplakin gene family in interstitial cystitis

Objective To detect gene expressions of Uroplakin (UP) family in interstitial cystitis (IC). Methods Gene expression of UP Ia, Ib, II, III, and III-δ4 was quantitatively measured in bladder biopsy samples from patients with IC (n=29) and control subjects (n=16) using real-time RT-PCR. Results UP III-delat4, a splicing variant of UP III, was significantly up-regulated to 4.80 in IC samples (P<0.001). When IC patients were divided into two groups, one with and one without ulcerative changes, the UP Ia, UP Ib, UP II, UP III and III-δ4 genes were significantly up-regulated to 4.99, 3.31, 2.75, 15.38, 11.48 in nonulcerative IC patients, but decreased to 0.04, 0.29, 0.09, 0.11, 0.34 in ulcerative IC patients (P<0.01). The up-regulation of UP III-δ4 was more prominent than that of UP III: 26.5-fold versus 5.6-fold compared to the median values of normal subjects. Conclusion Although the clinical implications of the over-expression of UP III and III-δ4 in non-ulcerative IC bladders remain to be clarified, from the diagnostic viewpoint, UP III-δ4 is a potential marker for identifying nonulcerative IC.
6.Expression of CNP/NPR-B axis and NPR-C/NEP elimination pathways in rats with unilateral ureteral obstruction

Objectives To study the expression features of C-type natriuretic peptide (CNP)/natriuretic peptide receptor-B (NPR-B) axis and two parallel elimination pathways, natriuretic peptide receptor-C (NPR-C) and neutral endopeptidase (NEP) in unilateral ureteral obstruction (UUO) rats. Methods CNP, NPR-B, NPR-C, NEP, Col-IV and type IV collagen (Col-IV) mRNA and proteins were determined by in situ hybridization, real-time PCR, immunohistochemistry and western blot in UUO rats at 24h, 72h, 1w, 2w, 3w, 1m, 2m and 3m. Results CNP expression tended to be higher immediately after ligation and de-clined along with the progression of disease, occurring predominantly in tubular epithelial cells. A high-level CNP may attribute to the elevated expression of NPR-B in the early phase of UUO. Conclusions NEP and NPR participate in the regulation of CNP expression in tubulointerstitial fibrosis. The gradual increased expression of NPR-C and NEP may cause the subsequent de-cline of CNP.
7.MRI features of intracranial primitive neuroectodermal tumors in adults: comparing with histopathological findings.

The MRI appearances of 7 adult patients with pathologically proven intracranial primitive neuroectodermal tumors (PNET) were retrospectively analyzed. The MRI features were compared with findings in pathology and surgery. In this group, the tumor masses were most commonly found in the semisphere of cerebrum and in the vermis of cerebellum. They were relatively large and 4 were in lobulated shape. All of them had well-defined margins. MR images showed the tumors to be mildly or obviously hypointense on T1-weighted images and hyperintense on T2-weighted images. Most masses had heterogeneous appearances with some cystic and necrotic areas. Intratumoral haemorrhage and focal calcification were occasionally seen. Mostly, there was no or only mild surrounding edema. Marked inhomogeneous contrast enhancement on MRI was seen in 6 cases except one. Two patients with multiple intracranial metastases were revealed on MR images. In this series, there was good correlation between MRI features and findings in pathology and surgery. These results showed that certain MRI features might suggest the diagnosis of intracranial PNET in adults. MRI is an effective technique to detect these tumors and is helpful to treatment planning and follow-up.

8.MRI and pathologic correlation of cardiac myxomas

Objective To investigate the MRI features of cardiac myxoma by correlated with its pathological findings. Methods MRI features of 22 cases of pathologically confirmed cardiac myxomas were retrospectively reviewed. Results Of 22 cases, 21 are solitary, 12 located in left atrium, 6 located in right atrium, 2 located in left ventricle and 1 located in right ventricle. The other one occupied multiple chambers. MRI: 19 are heterogeneous and 3 are homogeneous. Cine-MRI: 18 attach to the endocardium with a pedunculated stalk and 4 are sessile and with a broad attachment. Thirteen cases had secondary valve insufficience or stenosis. Nine have compromised cardiac function. Nineteen cases demonstrated mild to moderate heterogeneous enhancement after Gd-DTPA administration and 3 ease showed no enhancement. Four cases had gadolinium first-pass perfusion study and showed a slow and continuous increasing time-intensity, lower than normal myocardium. The pedicles and wall showed delay enhancement. Pathologic findings: 21 are oval and lobalar configuration, 1 is grape-like. Ten cases had fresh hemorrhage and 5 had chronic hemorrhage. Fourteen had necrosis , 2 had cystic change and 4 had calcification. Blood vessels or inflanmmtory cells could be detected in 19 cases. Conclusions MRI can evaluate the size, location, morphology, especially the vascularity, histologic features and cardiac function of cardiac myxomas.
9.Clinical features of differentiated thyroid carcinoma diagnosed during pregnancy

Objective To analyze the clinical features of differentiated thyroid cancer diagnosed during pregnancy or one year after delivery.MethodsA total of 234 women with differentiated thyroid cancer were treated from January 2001 to January 2011.Cases were divided into three groups according to the time of tumor diagnosis( group 1,no pregnancy history; group 2,during pregnancy or in the first year after delivery;group 3,longer than 1 year after the delivery).ResultsThe thyroid nodule was asymptomatic and discovered on routine examination more often in group 2(78.7% ) than other two groups (P =0.02).There were no significant difference among three groups in age,hospitalization time,operation time,operation method,tumor type,primary tumor stage and mortality. But more patients in group 2 were suffering from extrathyroid invasion ( P =0.024 ),multi-focal cancer ( P =0.023 ),larger cervical lymph node ( P =0.021 ),later regional lymph node stage (P =0.020 ) and higher local recurrence (P =0.001 ).By stepwise logistic regression analysis,pregnancy is one of the factors impacting on local recurrence rate ( P =0.045 ).ConclusionsPregnancy does not affect the mortality of the differentiated thyroid cancer,but the incidence of extrathyroid invasion,multi-focal cancer,cervical lymph node metastasis,local recurrence is higher in patients with differentiated thyroid cancer diagnosed during pregnancy.
10.Percutaneous microwave coagulation for treating peripheral non-small-cell lung cancer

Objective:To evaluate the clinical value of percutaneous microwave coagulation therapy for peripheral non-small-cell lung cancer. Methods:We evaluated 35 patients with non-small-cell lung cancer who received percutaneous microwave coagulation therapy and 35 patients who received radiotherapy from March 2004 to September 2006;the patients were sex-matched, age-matched, and had the same pathology and clinical staging. Clinical effects were observed and assessed. Survival rate were calculated using the Kaplan-Meier method. The difference in survival rate between the two treatment methods was analyzed using a log-rank test. Results:The 1-year, 3-year, and 5-year survival rates for the microwave coagulation therapy group (71.4%, 40.0%, and 20.0%, respectively) were significantly higher than those for the radiation therapy (51.4%, 22.9%, and 11.4%, respectively) (P<0.05). Conclusion:Percutaneous microwave coagulation therapy is a minimally invasive, safe, and effective alternative for patients with peripheral non-small-cell lung cancer who cannot undergo routine surgery because of poor heart and lung function or fear of surgical trauma.
