2.Paracrine effects of bone marrow mesenchymal stem cells on biological function of osteoblasts
Chinese Journal of Tissue Engineering Research 2014;(10):1477-1483
OBJECTIVE:To investigate the paracrine effects of bone marrow mesenchymal stem cells on osteoblast biological function. METHODS:Bone marrow mesenchymal stem cells were isolated using standard Ficol-Paquedensity gradient centrifugation. Mesenchymal stem cellconditioned medium was prepared to cultivate osteoblasts, MG63. Proliferation of MG63 cells was analyzed by cellcounting kit-8. Migration of MG63 cells was analyzed by cellscratch method. Alkaline phosphatase activity of MG63 cells was analyzed by microplate test kit. Real-time PCR was performed to evaluate osteoblast differentiation markers, alkaline phosphatase, col agen type I and osteocalcin. Alizarin red staining was performed to evaluate osteoblast mineralization. RESULTS AND CONCLUSION:The cells were strongly positive for CD44, CD73 and CD90, but negative for CD34. MG63 cells cultured in the conditioned medium showed better proliferation and migration than those cultured in the Dulbecco’s modified Eagle’s medium. The activity and mRNA expression of alkaline phosphatase were much higher after induction of 4, 7 days (P<0.01). There was no significant difference in expression of col agen type I and osteocalcin after induction of 4 days, but they were significantly higher than those in the control group after induction of 7 days (P<0.05). Alizarin red staining showed that the number of calcium nodules was increased and the mineral apposition was enhanced after induction of 21 days with the conditioned medium. These findings suggest that the paracrine substance of bone marrow mesenchymal stem cells can significantly promote osteoblast proliferation, migration, differentiation and mineralization.

Objective To study the ultrasound-guided treatment by real-time biplane imaging in animals and patients. Methods In 6 dogs gallbladders were substituted as hepatic cysts. The shapes of gallbladders and spatial positions of needle tips in gallbladders were studied by real-time biplane imaging before and during treatments. During treatments of 3 renal cysts the changes of cysts and the spatial relations between needle tips and renal cysts were observed to confirm that the needle tips were always in the cysts. In 2 cases of hepatocellular carcinoma boiloff of the tissue around the antenna was observed by real-time biplane imaging during microwave coagulation. Results It was rapid and convenient to acquire the shape information of objects and locate the position of puncture by real-time biplane imaging. And it was more convenient to observe the spatial relation between needle tips and objects of treatment than by conventional single-plane imaging. Conclusions The method of real-time biplane imaging has great advantages in ultrasound-guided treatment.

International Journal of Cerebrovascular Diseases 2009;17(4):284-287
Objective To investigate the prognostic value of plasma brain natriuretic peptide (BNP) in patients with acute cerebral infarction. Methods Sixty-five patients with first-ever acute cerebral infarction and 32 healthy controls were recruited. The neurological deficits in patients with cerebral infarction were evaluated by using Chinese Stroke Scale (CSS).Plasma BNP levels were detected by immunofluorescence technique. Death and recurrent stroke events were followed up. Plasma BNP levels were compared between an event group and a non-event group, and the relationship between plasma BNP levels and poor prognosis was analyzed. Results Plasma BNP levels in patients with acute cerebral infarction was significantly higher than those in healthy controls (238.7 ± 131.6 pg/ml vs 38.7±23.8 pg/ml, P <0.01). Nine patients (13.8%) died, and 8 (12.3%) had nonfatal recurrent stroke during the follow-up period. Compared to the non-event group, the baseline plasma BNP levels in the death/recurrent stroke event group were significantly higher (304.0 ± 134.9 pg/ml vs 214.4 ± 120.9 pg/ml,P < 0.01). There was significant correlation between plasma BNP levels and CSS scores (r = -0.359, P <0.05). After performing multivariate analysis of various risk factors, it found that BNP levels (OR = 3.5, 95 % CI 2.1 to 5.8, P < 0.01), advanced age (OR = 4.1,95% CI 1.7 to 9.2, P <0.01) and CSS scores (OR =2.6, 95% CI 1.6 to 4.3, P <0.01)were the independent predictors of poor outcome. Conclusions The increased BNP levels are the recent death and recurrent independent predictors in patients with acute cerebral infarction.

Journal of Interventional Radiology 2017;26(2):157-160
Objective To evaluate the feasibility and safety of establishing benign proliferative esophageal stenosis model by using stent implantation in experimental rats. Methods A customized self-expanding,metallic and straight tubular stent was used in this experiment (5 mm in diameter and 15 mm in length),on both sides at the stent's middle part there was a protruding barb that was used as a fixation device.Twelve healthy Sprague Dawley (SD) rats were randomized divided into group A (blank control group) and group B (stent implantation group),with 6 rats in each group.Esophageal stent implantation was employed in the rats of group B,and esophageal radiography was separately performed immediately, one and 4 weeks after stent implantation. All the experimental rats were sacrificed 4 weeks after stent implantation. The normal esophageal tissue of the rats in group A and the esophageal tissue at stent site of the rats in group B were collected and sent for pathological examinations, including gross morphology, light microscopy, etc. Results Successful stent implantation was achieved in all rats of group B,and the esophageal radiography performed immediately, one and 4 weeks after stent implantation showed no esophageal stent displacement;no severe complications occurred during the operation or follow-up period.Compared with group A,esophageal radiography reexamination performed 4 weeks after stent implantation in group B revealed that esophageal stricture at stent segment, caused by benign tissue hyperplasia, could be observed. The esophageal stent segment was taken out,its lumen was obviously narrowed under gross observation, and typical benign hyperplasia could be seen under optical microscope examination. Conclusion Using esophageal stent implantation to establish esophageal stenosis model is safe and feasible in experimental rats. The use of esophageal stent with barbs can significantly reduce the incidence of stent displacement.
