1.Effects of iptaklim hydrochloride on glutamate induced cytotoxicity in cultured pheochromocytoma (PC12) cells

AIM To investigate the protective effects of iptakalim hydrochloride (Ipt) against glutamate induced neurotoxity. METHODS Neurotoxity injury was induced by with glutamic acid in PC12 cells. The cell viability was measured by MTT assay. RESULTS Ipt at the concentrations of 1 ? 1(T-10 - 1 ? 1(T4 mol ? L-1 markedly mitigated glutamate-induced neurotoxicity. The protec- live effect was nullified by glibenclamide, an ATP-sensitive potassium channel antagnist. CONCLUSION Ipt provides neuroprotection on glutamate-induced cytotoxixity in cultured PC12 cells and this effect may be related with ATP-sensitive potassium channels.
2.Sequence analysis of muscarinic receptors M1, M3 and M5 expression by cultured rat astrocyte cells

Aim To clone the muscarinic receptors M1, M3 and M5 sequences of astrocyte cells,and compare the gene and protein sequences with those of neurons. Methods Specific primers were designed to clone the M1, M3 and M5 sequences of astrocyte cells by RT-PCR according to those of neurons,then sequenced the sequences. Result By comparing the M1, M3 and M5 sequences expression by astrocyte cells with those by neurons and we found four, eight,and one different bases and one, four and one different amino acids in M1, M3 and M5 between astrocyte cells and neurons respectively. Conclusions The gene and protein sequence differences are evident in M1, M3 and M5 between astrocyte cells and neurons.
3.New light on neuronal ATP-sensitive potassium channels

ATP sensitive potassium (K ATP ) channels widely expressed in many tissue and cell types including neurons are heteromultimers of sulfonylurea receptor (SUR) and inward rectifier K + channel (Kir6) subunits associated with 1∶1 stoichiometry as a tetramer (SUR/Kir6) 4. The activity of K ATP channels is modulated by intracellular ATP concentrations. Neuronal K ATP channels are involved in the mediation of glucose sensing and metabolic stress. Under physiological condition, they also possess important pathophysiological functions in acute brain ischemic and chronic neurodegenerative diseases.
5.Laparoscopic adrenalectomy for adrenal tumors by transperltoneal or retroperltoneal approaches:a report of 41 cases

Objective To evaluate the effect of laparoscopic adrenalectomy for adrenal tumors by transperito-neal or retroperitoneal approaches. Methods Atotal of 41 laparoscopic adrenalectomy for adrenal tumor patients were pertormed,inchiding 16 patients by transperitoneal approach and 25 patients by retroperitoneal approach. The tumor size,operative time,intraoperative blood loss, postoperative hospital stay, postoperative ambulatory day and postopera-tive complications were observed. Results Of the 41 cases ,3 cases were transferred to open surgery and the other 38 cases were achieved successful operations. No significant differences( all P>0.05) were found in tumor size,intraop-erative blood loss, postoperative hospital stay, postoperative ambulatory day between the transperitoneal group and ret-roperitoneal group ,but there were significant differences( P<0.05) in operative time and intraoperative blood loss be-tween the two group. Conclusion Both the transperitoneal and retroperitoneal laparoscopic approaches are feasible and safe for adrenal tumors, and the latter has more advantages in operative time, but the operative approach still should be chosen based on the pathological changes,tumor size and position as well as the individual conditions of the patients.

7.Research progress on the anticancer mechanism of thalidomide and the function of its target protein CRBN

Thalidomide and its analogues ( lenalidomide and po-malidomide) are small glutamic acid derivatives with strong im- <br> munomodulatory effects, belonging to immunomodulatory drug ( IMiD ) class . In addition , these thalidomide analogues demon- <br> strate an overlapping and diverse range of biological activities, including anti-angiogenesis and anti-teratogenicity. Importantly, the IMiDs exert anticancer effects such as inhibiting tumor cell proliferation and promoting tumor cell apoptosis and play impor-tant roles especially in clinical treatment of multiple myeloma. Recently, the screening and discovery of thalidomide binding protein, CRBN provides new clues to the research of its pharma-cological mechanisms and the develop ment of new generation of <br> thalidomide derivatives with less toxic side effects and more anti-tumor potency. This review briefly discusses the underlying mechanism of thalidomide and their derivatives’ action and their new identified target, as well as their contribution to the treat-ment of multiple myeloma.
8.Application of bone transmission and fascia skin flap recover large defeciency of bone and soft tissue of lower limb

[Objective]To research the effect of using Ilizarov technique in bone transmission and fascia skin flap for large defeciency of bone and soft tissues of lower limb.[Method]There were 12 patients in our group,11 patients had severe compound trauma of the leg,one in arm.Bone defeciency range was 4~12 cm,average 8 cm,cotaneous defeciency was 4?5 cm2~14?5 cm2,and all of them had acute infection.Nine had been treated with leg lateral fascia skin flap,1 had been treated with leg fascia skin flap,1 had been treated with the opposide leg skin flap,1 had been treated with the arm fascia skin flap,in 12 bone transport,elongation range was 4~12 cm,average 8 cm.Eight had been used double arms external fixation device,4 cases had been used circle shape external fixation device.[Result]Totally 12 patients were followed up for 2~10.8 years(average,4.2 years) elongation part and compressed part healed naturally,all of the fascia shin flap were alive.No bone ununion and chronic osteomyelitis appeared.[Conclusion]Using Ilizarov bone transport technique and fascia skin flap is a good way for treating large defeciency of bone and soft tissue.Its advantages:(1)The wound limb can maintain stability by external fixatian device.The methed of the bone transportation can fill big section bone deficiency;(2) In the same time the fascia shin flap may be transported,and repair soft tissue deficiency,provide good condition to bone's healing and control infection;(3)The external fixation device can provide distraction and compression functions,this function makes elongation part and compressed part obtain the reasonable biomechanics condition which would be good to bone healing;(4)The course of bone transmission can stimulate bone repair mechanisms,to enhance the wound limb metabolism,increase blood supply circularation,the grown bone factor aggregation,enhance the body's initiative repair mechanisms and infection controlling mechanism;(5)In the protection of the external fixation device,the patient's wound limb can do early rehabilitation,thus can avoid osteoporosis and joint dyfunction because of the lone-term immobility;(6)To use these methods,the treatment period will be shortened,antibiotic application will be decreased,the treatment cost will be lower than other methods and complications will be decreased as well.
9.Effect on antagonism of muscarinic acetylcholine receptors antagonists by desensitization of nicotinic acetylcholine receptors

Aim To investigate the effect on antagonism of benthiactzine and atropine against the function of muscarinic acetylcholine receptors by desensitization of nicotinic acetylcholine receptors. Methods The whole cell recording configuration of patch-clamp technique was used and the cell model was rat SCG neurons. To identify antagonists' antimuscarinic effect, mAchRs mediated IM-inhibition was measured and nicotine and oxotremorine were used. Results After de sensitization of nAChRs,the antimuscarinic effect ofboth benthiactzine and atropine decreased compared to the normal condition. The decreased antimuscarinic effect of benthiactzine was weaker than that of atro-pine. The antimuscarinic effect of both benthiactzine and atropine recovered gradually with the recovery of nAChRs from desensitization. Conclusion Desensiti-zation of nAChRs weakens the antagonists' antimuscarinic effect.
10.Effects of iptakalim hydrochloride on cerebral neuronal sodium, calcium and potassium channels

AIM To investigate the effects of iptakalim hydrochloride (Ipt) on cerebral neuronal sodium, calcium and potassium channels. METHODS The effects of Ipt on sodium, calcium and potassium channels in cultured hippocampal neurons was observed using whole-cell recording technique. RESULTS Ipt had no significant effect on Na +, Ca 2+ currents, but significantly increased the outward K + currents. This enhancement of Ipt was inhibited by synchronously application of glibenclamide. Upon the administration of Ipt 1, 10, 100 ?mol?L -1, the outward K + current was significantly increased to (121.9?8.1)%, (114.7?7.1)%, (109.1?10.0)% respectively (n=6~9, P
