1. Survey of tumor marker prescription behavior in lung diseases based on IMB model

Objective: To explore the influencing factors and pathways of clinicians' tumor marker prescription behavior in lung diseases, so as to find out the methods and measures to promote clinicians to optimize the tumor marker prescription. Methods: From July 24, 2017 to August 10, 2017, clinicians with willingness and the right of prescription mainly from hospitals in Shanghai and Beijing were invited to participate in survey on influencing factors of tumor marker prescription behavior in lung diseases. Based on the classical theoretical framework of IMB model, an IMB theoretical model of clinicians' tumor marker detection behavior was proposed. Statistical description and correlation analysis were performed by SPSS 24.0, and the IMB theoretical model was validated and corrected by AMOS 24.0. Results: The personal intention (r=0.251, P=0.000) and personal interest protection (r=0.178, P=0.007) had a significant positive correlation with information. Meanwhile, the personal intention (P=0.000) and personal interest protection (P=0.013) had a significant positive effect on behavioral skills, and the behavioral skills had a significant positive effect on tumor marker prescription behavior (P=0.000). There was no direct effect of information on behavioral skills, and motivation had no direct effect on tumor marker prescription behavior. Behavioral skills played a mediating role in the process of motivation influencing the tumor marker prescription behavior. Conclusion: The large percentage of irrational tumor marker prescription in lung diseases suggests the universality of inappropriate testing, which needs the joint efforts of governments, hospitals, clinicians and patients on improvement.
2.Ultrasound and contrast agents enhance VEGF siRNA-mediated anti-cancer effect on human nasopharyngeal carcinoma

Objective To study the enhancing effect of ultrasound plus microbubble on small interference RNA(siRNA)transfection in vitro and in vivo.Methods Human vascular epithelial growth factor(VEGF)siRNA with 2'deoxy modification(VEGF 2'-eoxy siRNA,VdsR)was used.The human CNE cells(fromnasopharyngeal carcinoma)line was used for in vitro cell-based experiments and in vivo mouse xenograft model.Two different microbubble agents.BR14 and Levovist.were used together with the RNA transfection reagent RNA-mate.ELISA and RT-PCR assays were used to assess VEGF gene expression.Immunohistochemical staining (IHC)was performed to assess CD31 expression in xenograft tumors.Results VdsR transfection in CNE cells abolished VEGF expression as determined by ELISA experiments.In the first mouse xenograft experiment,ultrasound exposure dramatically enhanced VdsR-mediated tumor inhibition.In the second mouse xenograft experiment,when VdsR was mixed with the microbubble reagents and then injected into xenografts,ultrasoundexposure significantly reduced tumor growth in BR14-mixed VdsR group but not in the Levovist-mixed VdsR group compared to the control.RT-PCR experiments demonstrated that VEGF expression in ultrasound-exposed tumors was significantly lower than that in the control.Meanwhile,VEGF expression in the tumor tissue treated by BRl4-mixed VdsR declined as compared with the controls.Tumor vascular density as measured by CD31 immunostaining was significantly decreased in ultrasound-exposed tumors compared to the control.Conclusions Ultrasound exposure and/or microbubble can significantly enhance delivery and the efficiency of VdsR-mediated anti-tumor effects,and should be a location-specific enhancement approach for siRNA-based anti-cancer therapy.
3.Eudesmane sesquiterpenes from twigs of Manglietia hookeri.

Chemical constituents from the acetone extract of twigs of Manglietia hookeri were isolated and purified by various column chromatographic methods over silica gel and sephadex LH-20, and preparative TLC. The structures of these compounds were identified on the basis of physicochemical properties and spectral analysis, including NMR and MS spectra. Six eudesmane sesquiterpenes were obtained and their structures were identified as trans-eudesmane-4, 11-diol(1), β-eudesmol(2), (-) -10-epi-5β-hydroxy-β-eudesmol (3), epi-carrisone (4), 6-hydroxy-eudesm-4(14) -ene(5) and gynurenol(6). All the compounds were isolated from this plant for the first time. Furthermore, the 13C-NMR data of compound 3 were reported for the first time.

4.Effect of electroacupuncture on calcium-activated chloride channel currents in interstitial cells of Cajal in rats with diabetic gastroparesis

Objective: To investigate the mechanisms of electroacupuncture (EA) at Zusanli (ST 36), Liangmen (ST 21) and Sanyinjiao (SP 6) in intervening diabetic gastroparesis (DGP) based on calcium-activated chloride channel. Methods: Forty Sprague-Dawley rats were randomly divided into four groups, including a normal control group (group A), a model group (group B), an EA group (group C) and a metoclopramide group (group D), with 10 rats in each group. A single intraperitoneal injection of 2% streptozotocin (STZ) combined with 8-week high-glucose high-fat diet was used to establish a DGP rat model. After intervention, gastrointestinal propulsive rate was observed; the expression level of transmembrane protein 16A (TMEM16A) was examined by immunohistochemistry; the Ca2+ concentration in interstitial cells of Cajal (ICCs) was detected by immunofluorescence; and whole-cell patch-clamp technique was applied to detect the current intensity of calcium-activated chloride channel (ICaCC) in ICCs in gastric antrum. Results: After modeling, the blood glucose levels in group B, group C and group D were significantly increased compared with group A (all P<0.01); after intervention, compared with group B, the blood glucose levels in group C and group D were significantly decreased (P<0.05, P<0.01); the intra-group comparison of blood glucose level between after modeling and after intervention found significant difference only in group C (P<0.01). The gastrointestinal propulsive rates in group B, group C and group D were significantly different from that in group A (P<0.01 or P<0.05); the gastrointestinal propulsive rates were markedly higher in group C and group D than in group B (P<0.01, P<0.01). The expressions of TMEM16A in group B and group C were decreased compared with group A (P<0.01, P<0.05); the expressions of TMEM16A in group C and group D were increased compared with group B (P<0.01, P<0.05). The fluorescence intensity of Ca2+ was significantly lower in group B than in group A (P<0.01); the fluorescence intensity of Ca2+ was significantly higher in group C and group D than in group B (P<0.01, P<0.05). ICaCC in ICCs in group B was significantly decreased compared with group A; ICaCC in group C and group D were increased compared with group B. Conclusion: EA at Zusanli (ST 36), Liangmen (ST 21) and Sanyinjiao (SP 6) can significantly improve gastrointestinal motility in DGP rats by up-regulating the ICaCC in ICCs.
5.Correlation between Level of Nitric Oxide/Nitricoxide Synthase and Ultra-Structure Changes of Placenta Exposured in Lead Gestation Period in Rats

Objective To study the correlation between level of nitric oxide/ nitricoxide synthase(NO/NOS) on placenta homogenate and ultra-structure changes of placenta in pregnancy lead exposure in rats.Methods Seventeen normal pregnant rats and 46 rats of exposured in lead which were divided into A,B,C groups were studied.The level of NO/NOS of placenta were measured by nitrate reductase and NOS kit.Placentas were randomly selected from each group to detect ultra-structure by electron-microscope.Results There were significant difference among A,B and control groups on level of NO/NOS(all P0.05).Compensation hyperplasy or minor injury were observed in lead exposure of stage groups.Lead exposure during whole gestation period,the lead level was maxmum,and decompensation were observed on placental construction.Conclusions There is a close correlations between level of lead,NO/NOS and pathological change of placental tissue,and both of them may play an important role in the pathogenesis of peripartum lead exposure.
6.AS-PCR assay for 20 mtDNA SNP typing and haplotype frequency.

OBJECTIVE:
To develop a multiplex allele-specific PCR (AS-PCR) assay with three-color fluorescence labeling for mitochondrial DNA (mtDNA) SNP typing.
METHODS:
Based on the principle of AS-PCR, the primer sets were designed for 20 SNP located on the coding region of mtDNA and divided into 2 groups labeled with FAM and HEX fluorescence, respectively. A primer set included two forward (reverse) allelic specific primers with different sizes and a generic reverse (forward) primer. Blood samples from 200 unrelated individuals were analyzed by AS-PCR and capillary electrophoresis. Three random samples at least for each SNP site were examined and verified by direct sequencing. The haplotype frequency was investigated.
RESULTS:
Distinct electropherograms of 200 blood samples were obtained successfully. The typing results of direct sequencing were identical to those obtained from AS-PCR. The minimum detectable DNA concentration was 0.2 pg under the system of 10 microL. The sensitivity of the DNA concentrations ranged from 0.5 to 5 pg. The 200 individuals were assigned into 15 haplotype, and the haplotype diversity was 0.906 0.
CONCLUSION
AS-PCR is a simple, rapid and efficient method for mtDNA SNP typing, and can be applied to forensic practice.

7.Determination and analysis of silica particles surface alumino-silicate occlusion.

OBJECTIVETo investigate the methods of determining aluminum silicate coated on the surface of silica particles and analyze the role of surface occlusion on development of silicosis.
METHODSRespirable dust samples were collected on filters using 2 L/min flow in tungsten mines and pottery factories of Jiangxi province, and tin mines of Guanxi province. Dust particles were analyzed by a multiple-voltage scanning electron microscopy-energy dispersive X-ray spectroscopy (MVSEM-EDS) using 20 KeV and 5 KeV electron beam accelerating voltages. Changes in the silicon to aluminum X-ray line intensity ratio between the two voltages are compared particle by particle. This provided an index that distinguishes a silica particle that was homogeneously aluminum-contaminated from a clay coated silica particle.
RESULTSThe total of 3,982 dust particles from 47 dust samples of seven pottery factories, three tin mines and three tungsten mines were analyzed in this study. Significant difference of aluminum silicate coated on the surface of silica particle was shown between particles from pottery factories and tin mines. The average sample percentage of respirbale-sized silica particles alumino-silicate occlusion in the pottery factories (45%) was higher than that in the tin mines (18%) and tungsten mines (13%).
CONCLUSIONHigher percentages silica particles alumino-silicate occlusion is observed in the pottery factories than that in metal mines. These surface analysis results help to understand differences in risk of silicosis when exposure is normalized to cumulative respirable surface silica dust.

9.Prevalence and risk factors of bronchial asthma among Li nationality in Hainan province.

OBJECTIVETo study the risk factors of bronchial asthma of Li nationality in Hainan.
METHODSA total of 13 050 subjects of Li nationality were selected by random unequal ratio stratified cluster sampling method from southern, central and western part of Hainan and investigated with Hainan Epidemiological Asthma Survey Questionnaire of Li Nationality. There were 441 cases of bronchial asthma, and 1296 cases of control that were sampled by random number table method. The logistic regression method was used to analyze risk factors.
RESULTSThe asthma prevalence of Li nationality in Hainan was 3.38%(441/13 050). The main risk factors of asthma were family asthma (OR = 4.323, 95%CI = 3.259 - 5.735), hypersensitiveness (OR = 7.775, 95%CI = 5.686 - 10.632), smoking (OR = 1.494, 95%CI = 1.174 - 1.902), cooking fuels and living environment. Cold air change (OR = 1.604, 95%CI = 1.286 - 2.001) and respirable dust or irritant gas (OR = 2.123, 95%CI = 1.702 - 2.648) were the important incentives.
CONCLUSIONThe main risk factors of asthma among Li nationality were family asthma, hypersensitiveness, smoking, cooking fuels by means of fuel oil, hay or wood, living environment by means of couch grass room and human-livestock mix live, cold air change, respirable dust or irritant gas.

10.Effect of epidermal growth factor signal pathway on integrin alpha5 beta1 in prostate cancer cell line DU145.

OBJECTIVETo investigate the molecular mechanism of epidermal growth factor (EGF) signal pathway on the expression of integrin alpha5 beta1 in prostate cancer cell line DU145.
METHODSUsing flow-cytometry, the effects of EGF and the mitogen-activated protein kinase (MAPK) signal pathway inhibitor PD98059 on the expression of integrin alpha5 and beta1 subunits on DU145 cell surface were analyzed. RT-PCR and Western blot methods were used to examined the expression of mRNA and cell total protein of integrin alpha5 and beta1 subunits. And the metastatic phenotypes in DU145 cell were investigated.
RESULTSThe expression levels of integrin alpha5 beta1, which was the receptor for fibronectin, were changed. EGF up-regulated the protein and mRNA expression of beta1 subunit on DU145 cell surface, 231% and 248% (P < 0.01) compared to the control respectively, and it could significantly promote the ability of DU145 cell adhesion to fibronectin and migration. However PD98058, which was the inhibitor of MAPK signal pathway, down-regulated the protein and mRNA expression of beta1 subunit, 60% and 63% (P < 0.01) compared to the control respectively, and it had the contrary function on the adhesion and migration ability of DU145 cell. But both had no effect the expression of alpha5 subunit.
CONCLUSIONSEGF might promote the metastatic ability mainly by up-regulating the expression of beta1 subunit by activating MAPK signal pathway in DU145 cells. Their regulation effects are on the mRNA transcriptional level.
