1.In-Vitro Inhibitory Effect of Oleum Curcumae Aromaticae on Leukemia Cell Strain

[Objective] To evaluate the therapeutic effect of Oleum Curcumae Aromaticae (OCA) on leukemia cell strain HL-60 and K562 by observing its sensitivity in inhibiting HL-60 and K562. [Methods] Subculture of HL-60 and K562 cells was performed on the 96-hole culture plate and OCA 100?L at the final concentration of 400, 200, 100 and 50mg/L respectively was added. The blank control was added with RPMI-1640 culture fluid. After 24, 48 and 72 hours of treatment, MTF 10?L (5mg/mL) was added and the inhibitory rate on the cells was assayed by detecting the absorption value at 570nm wavelength. 2.Protective effect and mechanism of pinacidil on mitochondrial respiratory function in intestinal mucosa of scalded rats

Objective To investigate the protective effect of pinacidil on the intestinal mucosa of scalded rats and the mechanism. Methods A total of 24 healthy SD rats were randomly divided into normal control group, burn group, and pinacidil treated group. Rats in burn group and pinacidil treated group were inflicted with 30% TBSA Ⅲ degree burn and resuscitated intraperitoneally with Ringer's solution immediately after burn. Pinacidil was injected intraperitoneally into rats in pinacidil treated group at the dose of 2 mg/kg. Mitochondrial respiratory function intestinal mucosa and levels of reactive oxygen species (ROS), maleic dialdehyde (MDA), and superoxide dismutase (SOD) in plasma were determined at 6 h after burn. Results Mitochondrial respiratory function control rate (RCR), ST3, and SOD levels in pinacidil treated group were evidently higher than those in the burn group. However, ST4, MDA, and ROS levels in pinacidil treated group were significantly lower than those in the burn group, but ST4 was not significantly different from that in the normal control. Conclusion Pinacidil can attenuate the damage of intestinal mucosa in scalded rats.

To further uncover the scientific significance and molecular mechanism of the Chinese herbs with pungent hot or warm natures, endogenous and exogenous expression systems were established by isolation of dorsal root ganglion (DRG) neurons and transfection of HEK293 cells with TRPV1 channel gene separately. On this basis, the regulation action of capsaicin, one main ingredient from chili pepper, on TRPV1 channel was further explored by using confocal microscope. Besides, the three-sites one-unit technique and method were constructed based on the brown adipose tissue (BAT), anal and tail skin temperatures. Then the effect of capsaicin on mouse energy metabolism was evaluated. Both endogenous and exogenous TRPV1 channel could be activated and this action could be specifically blocked by the TRPV1 channel inhibitor capsazepine. Simultaneously, the mice's core body temperature and BAT temperature fall down and then go up, accompanied by the increase of temperature of the mice's tail skin. Promotion of the energy metabolism by activation of TRPV1 channel might be the common way for the pungent-hot (warm) herbs to demonstrate their natures.

Many Chinese drugs (CHD) have showed their significant effects of integral immune-regulation, and lots of researches have conducted in recent years for exploring their mechanism from different levels, like cytological, molecular and genetic levels. In this paper, the relation between immune-regulation of CHD and Toll-like receptors/nuclear factor-kappaB (TLRs/NF-kappaB) signaling pathway was introduced in brief based upon the achievements of previous researches. It was pointed out that the two are closely related, to explore mechanism of CHD in this way is meaningful not only for further deepening the theoretical understanding of CHD's pharmacological immunoregulation, but also be practically facilitate for enhancing therapeutic efficacy of CHD and developing new CHD.

9.Analysis of magnetic susceptibility of normal brain of young adults by quantitative susceptibility mapping

Objective To explore the changes of susceptibility of different sides and gender in healthy young adults with quantitative susceptibility mapping (QSM).Methods Totally 41 healthy young right-handed adults underwent conventional brain MRI and QSM scan,and the susceptibility maps were obtained by the image post-processing software.Then the ROI of the bilateral frontal grey matter (FGM),frontal white matter (FWM),caudate (CA),globus pallidus (GP),putamen (PU),thalamus (TH),substantia nigra (SN),red nucleus (RN),dentate nucleus (DN),pons (PO),corpus callosum (CC) were manually drawn to obtain magnetic susceptibility on the susceptibility map.The magnetic susceptibility of each ROI was compare between both sides,as well as gender by Mann-Whitney test.Results The magnetic susceptibility of the bilateral ROI of GP was the highest,and SN was followed,FWM was minimum.The susceptibility of bilateral FGM,FWM,CA,GP,PU,TH,SN,RN,DN,PO,CC had no statistically significant differences (all P>0.05).The magnetic susceptibility in CA of different gender had statistically significant difference (P<0.05).Conclusion The brain magnetic susceptibility.can be measured by QSM,and it can assess brain iron content quantitatively.
10.The expression of the early growth response gene-1 after focal cerebral ischemia/reperfusion in rats

Background To investigate the expression of the early growth response gene-1 ( Egr-1 ) mRNA after focal cerebral ischemia / reperfusion in rats.Methods Ten healthy male SD rats weighing 200 ~ 250 g were used to create model of focal cerebral ischemia. The expression of Egr-1 after focal cerebral ischemia/reperfusion in rats was determined using in situ hybridization and RT-PCR.Results (1) The result of the in situ hybridization: A trace amount of Egr-1 mRNA expressed in the neurons and the glial cells in the sham operated group. The expression of Egr-1 mRNA at the ischemic side increased dramatically following ischemia and reached peaks after 4 hours' reperfusion. Egr-1 expression started to subside following 22 hours' reperfusion and further decreased following 166 hours' reperfusion, which was still significantly higher than that in the sham operated group. (2) The result of RT-PCR: The expression of Egr-1 mRNA at the ischemic side was significantly higher than that in the sham operated group at all time points after ischemia/reperfusion in the rats(P <0. 01). Expression of Egr-1 increased 2 h after ischemia and reached the peak 4 h following reperfusion, and then decreased dramatically at 46 h after reperfusion which was still higher than that in the sham operated group (P < 0. 01). As the ischemia/reperfusion period prolonged, the expression of Egr-1 mRNA increased gradually, but still detectable even 166 h following reperfusion. The expression of Egr-1 was significantly higher than that in the sham operated group at all time points (P <0. 01).Conclusions The expression of Egr-1 mRNA increase in the neurons and the glial cells after ischemia/reperfusion, which may have protective effects on ischemic brain tissues.
