1.Detection of truncating mutations of APC gene by digital protein truncation test labeled with fluorescent Lys-t-RNA

Objective To investigate the relationship between truncating mutations of APC gene and sporadic colorectal cancer,and analyze the feasibility of non-radioactive protein truncation test (PTT) in the detection of mutations of APC gene.Methods Ninety-six samples of sporadic colorectal cancer tissues ( including 44 patients with colonic cancer and 52 patients with rectal cancer) were obtained from the General Hospital of Ningxia Medical University from September 2008 to September 2010.The mutation cluster region of the APC gene was screened using digital PTT labeled with fluorescent Lys-t-RNA,with a polymerase chain reaction fragment amplified from genomic DNA serving as a tenplate for in vitro translation.The occurrence of gene mutation was determined according to the emergence of truncated peptides.The mutation cluster region of the APC gene in 46 samples of colorectal cancer tissues was analyzed by direct sequencing.The detection rates of the 2 methods were compared by chi-square test.Results Thirteen (26%) truncated peptides were detected in the 50 samples of colorectal cancer tissues.The mutation type of 4 samples is nonsense mutation,which resulted in emergence of truncated gene products.Eleven (24%) truncated peptides were detected in the 46 samples of colorectal cancer tissues.There was no significant difference in the detection rates between PTT and direct sequencing ( x2 =0.033,P > 0.05 ).Conclusions Truncating mutations of APC gene are common alterations in sporadic colorectal cancer in the Chinese.Digital PTT labeled with fluorescent Lys-t-RNA is rapid and high-sensitive in screening gene mutations.
2.Prevention of aseptic loosening prothesis of alendronate in beagle:abiomechanical study

Objective To investigate effect of alendronate on prevention of aseptic loosening prothesis.Method Twelve beagles which had been implanted a titanium plug in femur were divided into two groups randomly,one received alendronate,and the other placebo as control.After 12 weeks,all beagles were sacrificed.specimens of femur were studied by biomechanical test.Independent sample t test was used for statistical analyses.Results The pull-out strength of the experimental group was higher than that of the control (P < 0.05),and the torsion strength was also higher in the experimental group(P < 0.05).Conclusion The therapy of alendronate can change the bone strength,inhibit aseptic loosening of prosthesis in beagle and could be a good drug for the prevention and treatment of aseptic loosening prothesis.
3.Application of Percolse, Angio-Seal Vascular Closure Devices or Manual Compression in Emergency Percutaneous Coronary Intervention

ObjectiveTo assess the value of Percolse, Angio-Seal vascular closure devices in emergency percutaneous coronary intervention. Methods603 patients undergoing emergency percutaneous coronary intervention were divided into manual compression group (n=160), Perclose group (n=237) and Angio-Seal group (n=206). The time of hemostasis, time of immobilization and the incidence of vascular complications were observed in each group. ResultsThe time of hemostasis, time of immobilization in vascular closure devices groups were significantly shorter than those in manual compression group (P<0.01). The incidence of vascular complications were significantly lower than those in manual compression group (P<0.01). There was no difference between Perclose group and Angio-Seal group (P>0.05). The rate of successful hemostasis was not significantly different among each group (P>0.05). ConclusionThe Perclose and Angio-Seal vascular closure device is safe and reliable hemostasis in emergency percutaneous coronary intervention.
4.Advances and orientation of dermatological research in PLA

Objective To summarize the progress in dermatological researches in the past 5 years achieved domestically and abroad,and venture to propose the developmental orientation and problems to be emphasized in dermatology in the next 5 years. Methods The achievements and progresses gained in dermatology in the recent 5 years were retrieved by employing the well-know informatics technology. Results Remarkable achievements have been gained in dermatological researches during recent 5 years,especially those concerning psoriasis,connective tissue diseases,vitiligo and dermatomycosis. Two professional journals in dermatology were founded by military medical service in recent 5 years. Several awards for the achievements in dermatology were rewarded,such as,one each,the first-and second-class Army Science and Technology Progress Award,second-class Army Medial Achievements Award,second-class Chinese Science and Technology Award in medicine,provincial level second-class Science and Technology Progress Award,etc. Apart from those awards,researches on investigation and prevention of skin diseases during military training had made important breakthrough. Conclusions In the next 5 years,the points of focus in dermatological research should still be put on serving the basic army units,raising the level of prevention and treatment of skin diseases in the course of training,ensuring the psychological health of soldiers,supporting the important research items and promoting further the development of dermatology.
5.Clinical analysis of 166 aged women with breast cancer

Objective To evaluate the effect of operation on aged women with breast cancer. Methods The clinical data of the 166 patients recruited from 1989 to 2004 were retrospectively analyzed. Results One hundred and sixty six patients were all treated by operation. The modified radical mastectomy was performed in 98 cases(59.0%), radical masteetomy in 21 cases(12.7%), mastectomy in 29 cases (17.5%), palliative operation in 18 cases (10.8%). After operation, one hundred and twenty six patients received chemotherapy, twelve radiotherapy and 73 endocrine therapy. The rates of postoperative complications were 10.8%, no patients died during operation. The overall postoperative 3-and 5-year survival rates were 82.2% and 65.2% respectively. Conclusions The effect of surgical treatment and its combination with other adjunct therapies is satisfactory for aged women with breast cancer. Different types of operations should be performed for different patients. Ageing is not a contraindication of the operation. Surgery should be combined with medical therapy to reduce the concomitant diseases and postoperative complications.
6.Effect of iptkalim on myocardial enzymes and free radicals metabolism with hypoxic pulmonary hypertension.

OBJECTIVETo explore the effects of iptkalim on myocardial enzymes and free radicals metabolism with hypoxic pulmonary hypertension (HPH), in order to provide evidence for the mechanism of iptkalim on clinical treat.
METHODS110 young men stayed at high altitude above 5 000 m were divided into iptkalim group (n = 74) and placebo group (n = 36), aminotransferase (ALT), aspartate aminotransferase (AST), gamma-glutamyltransferase (gamma-GT), creatine kinase (CK), lactate dehydrogenase (LDH), superoxide dismutase (SOD), malonaldehyde (MDA), nitric oxide(NO) and nitric oxide synthase(NOS) were detected before and after took medicines for 6 mouths.
RESULTSAfter took medication for 6 mouths, ALT, AST, gamma-GT, CK and LDH were reduced, SOD, NO, and NOS were increased, MDA were reduced, there were very significant difference (P < 0.05).
CONCLUSIONOxygen free radicals have taken part in the process of HPH, iptkalim have the effect of anti-peroxidation of lipid and protect myocardial cells stress injured by hypoxia which related with mitochondrial membrane and cell membrane's K(ATP) channel activation.

7.Cervical radiculopathy treatment experts' consensus to establish radiculopathy type by using the modified Delphi meth-od

Objective Cervical radiculopathy treatment experts' consensus to establish radiculopathy type by using the modified Delphi method. Methods Use document retrieval method to review information and articles about the treatment guidelines and articles of cervical radiculopathy including domestic and international areas, established a protocol about clini?cal consensus of the treatments for cervical radiculopathy. This protocol included 23 questions (the effective proportion of non?operating therapy, neck immobilization, physiotherapy, pharmacologic treatment, surgical indications, contraindications, anteri?or surgical decompression, anterior surgical implants). We performed a modified Delphi survey in which current professional opinions from experienced experts, representing from almost all of the Chinese provinces, were gathered. And then we modi?fied the protocol according to those professional opinions. Three rounds were performed and finally we established consensus. Consensus was achieved with ≥70% agreement. Results The panel included 30 experienced experts. The recycling question?naire's quantity of three rounds were 30(100%), 24(80%) and 16(53.3%) respectively. After three expert assessments, there were 18 questions which achieved with≥70%agreement and these questions accounted for 64.3%(18/28) of all the questions. Consen?sus of the treatments for cervical radiculopathy was reached on 7 aspects, including:the effective proportion of non?operating thera?py (1 question), neck immobilization (1 question), physiotherapy (1 question), pharmacologic treatment (5 questions), surgical indi?cations (3 questions), contraindications (4 questions), surgery (3 questions). Conclusion This modified Delphi study had reached a consensus concerning several treatment issues on cervical radiculopathy which had strong representativeness of experts and good convergence of opinions. In the absence of high?level evidence, at present, these experts' opinion findings will guide health care providers to define appropriate treatment in their regions. Areas with no consensus provide excellent insight for future research.
