Chinese Traditional and Herbal Drugs 2019;50(15):3541-3546
Objective: In order to investigate the effect of Shufeng Jiedu Capsule (SJC) compatibility on anti-inflammation of pneumonia model rats. SJC whole prescription and its components with effect of heat-clearing and toxin-removing and exterior-releasing were studied. Methods: The pneumonia model of rats induced by Streptococcus pneumoniae was used, divided into control group, model group, cephalexin group, SJC whole prescription group (whole prescription group), SJC exterior-releasing group (exterior-releasing group), and SJC heat-clearing and toxin-removing group (heat clearing and detoxifying group). After administration for 6 d, the colony count of peripheral blood and alveolar lavage fluid (BALF), the number of white blood cell (WBC) and its classification in peripheral blood and alveolar lavage fluid, the contents of nuclear transcription factor-kappa B (NF-κB) and cyclooxygenase (COX-1 and COX-2) were tested to evaluate the compatibility rationality. Results: Compared with model group, the whole prescription group, exterior-releasing group, and heat-clearing and toxin-removing group significantly reduced the colony count of peripheral blood and alveolar lavage fluid, significantly reduced the number of WBC in peripheral blood, significantly increased the proportion of eosinophils and reduced the proportion of basophils. The Q values of rats in the whole prescription group were beyond 1 on two indicators, which were the number of WBC and the proportion of eosinophils. The number of WBC in alveolar lavage fluid and the contents of NF-κB and COX-2 decreased significantly in the whole prescription group, exterior-releasing group and heat-clearing and toxin-removing group. Conclusion: SJC has significant anti-inflammatory effect and therapeutic effect on Streptococcus pneumoniae infection in rats. The exterior-relieving component and the heat-clearing and toxin-removing component have significant synergistic effects.

OBJECTIVETo evaluate the early clinical and radiographic outcome of scaphoid non-unions treated with Acutrak headless compression screw.
METHODSFrom January 2008 to July 2011,21 patients with scaphoid non-union were treated in our department. There were 18 males and 3 females with a mean age of (23.6 +/- 4.6) years; 12 cases were on right hand and 9 were on left. According to Herbert-Fisher classification, there were 10 cases with type D1, 7 cases with type D2, 3 cases with type D3, and 1 case with type D4. The mean time from injury to operation was (12.4 +/- 2.7) months. All patients were treated with Acutrak headless compression screw fixation (6 cases received 2 screws fixation, 15 cases received 1 screw fixation, and Matti-Russe bone grafting was applied in 7 cases). The carpal height, the scaphoid index and changes of the scapholunate angle were assessed before and after the operation. Range of motion and grip strength were recorded and the wrist function was assessed according to the Patient-Rated Wrist Evaluation (PRWE).
RESULTSAverage duration of follow-up was (21.3 +/- 3.6) months. All the patients attained radiological union in a mean time of (13.3 +/- 2.4) weeks following the operation. No obvious complications were recorded. The surgical treatment allowed the preoperative mean scaphoid index of 0.61 +/- 0.13 and the preoperative mean scapholunate angle of (59.4 +/- 6.8) degree to be improved to 0.69 +/- 0.10 and (44.3 +/- 8.2)degree postoperatively, respectively. There was a substantial improvement in grip strength and pain amelioration after surgery. The preoperative mean PRWE score of 45.2 +/- 4.7 was improved to 76.1 +/- 5.2 postoperatively. All patients returned back to the original work, the average time from surgery to work was (6.0 +/- 1.1) months.
CONCLUSIONFor scaphoid non-unions, Acutrak headless compression screw fixation can provide anatomical reduction, provide satisfactory results with a high union rate, well return of function and minimal complications in the early stage.

The influenza A virus matrix protein2 gene(M2)which deleted transmembrane region was amplified by overlap extending PCR,and the multiepitope gene of hemagglutinin(HA)was PCR amplified with seven continuous synthesized segments by designing primer. The two gene segments were separately cloned into pMD18T vector to sequence analysis and prokarytic expression vector pET28a+ to construct the recombinant plasmid pET28a+M2dHA.The recombinant plasmid was transformed into E.coli BL21(DE3),and the high expression strain was obtained by screening monoclones. The recombinant protein existed as inclusion bodies, which accounted about 45% of the total cellular protein. The inclusion bodies were washed with 1% Triton X100 solution twice, and dissolved in 8 mol/L urea solution. The solution protein was purified by Ni+2 affinity chromatography, and refolded by dilution renaturation, then purified by Q Sepharose FF cation exchange column. The purity of the protein was over 90% by HPLC analysis. The result of Western blot showed it has good antigenicity and specificity. These results strongly supported for the further study of the broadspectrum influenza virus subunit vaccine.

Objective To understand the genetic polymorphism of Salmonella and Staphylococcus aureus in Guangdong province, as well as to explore methods for identifying and tracing the source of these two foodbome pathogens. Methods Using the automated ribotyping system, two foodbome pathogens were tested with either EcoR Ⅰ or Pvu Ⅱ restriction enzymes. BioNumerics software was then applied for image analysis, database establishment and other corresponding analysis. Results Digestion of 32 Salmonella isolates with Pvu Ⅱ yielded 19 different ribotypes, and digestion of 14 Salmonella isolates with EcoR Ⅰ yielded 2 different ribotypes. Staphyloccus aureus isolates showed greater genetic diversity, whereas EcoR Ⅰ digestion of 49 different isolates yielded 31 different ribotypes. Conclusion Unique Salmonella and Staphylococcus aureus isolates could be identified through ribotyping. Although Salmonella serotyping and ribotyping were not strongly correlated, the combination of both restriction enzymes could be used to more effectively identify the genetic relationship among different strains as well as the source of food poisoning. Thus, not only could the genetic relationships amongst the different strains be inferred through ribotyping skills, the source of food poisoning and mode of transmission could also be determined under the use of this method.
