1.Health-Related Quality of Life of Children with Subclinical Epileptiform Electro encephalographic Discharges

Objectives To study health-related quality of life(HRQOL) of children with subclinical epileptiform electroencephalographic discharges.Methods Subjects were 30 children with subclinical epileptiform electroencephalographic discharges,50 children with clinical seizures,28 children without clinical seizures and 30 controls.Guardians completed a valid epilepsy-specific HRQOL questionnaire for children,the quality of life in children epilepsy questionnaire(QOLCE).Results Children with subclinical epileptiform electroencephalographic discharges have 8 subscales scores lower than control group,2 subscales scores higher than group with clinical seizures.Conclusions Quality of life of children with subclinical epileptiform electroencephalographic discharges is better than that of children with clinical seizures.However cognitive,behavior,daily life and social activity are impaired by subclinical epileptiform electroencephalographic discharges and lowered the HRQOL of children.
2.The effect of exposure to 50 Hz magnetic fields on the proliferation of different cell densities of human MG-63 osteosarcoma cell line in vitro

Objective To study the effects of 50 Hz magnetic fields on the in vitro proliferation of human osteosareoma cell line MG-63 with different cell densities.Methods Four different magnetic intensities(1 mT, 2 mT,3 roT,4 mT)were used to stimulate the cells,and the experiment was repeated with different cell densities. The method of MTT was employed to evaluate the level of proliferation.Results Fifty Hz magnetic fields signifi- cantly affected the level of proliferation of human osteosareoma cell line MG-63,and the 2 mT intensity exerted the greatest influence on it.The effects of the magnetic field differed with different cell densities.Conclusion The effect of 50 Hz magnetic fields on the in vitro proliferation of human osteosarcoma cell line MG-63 was not only relat- ed to the magnetic intensity,but also the cell density,
4.The effect of electromagnetic fields on gene expression profiles in bone marrow mesenchymal stem cells

Objective To screen for differential expression of genes in bone marrow mesenchymal stem ceils (MSCs) stimulated by electromagnetic fields (EMFs) and to study the mechanism of differentiation of the bone mar- row MSCs induced by EMFs.Methods Mouse bone marrow MSCs were isolated and cultured in vitro.The third- passage cells were stimulated by EMFs,then the total RNA was extracted,purified to cDNA and reversely transcribed to yield a cRNA probe.The cRNA probes from stimulated and control cells were labelled with Cy5 and Cy3,respec- tively.The two samples were hybridized with a mouse oligo microarray,and the hybridization signals were scanned. The reverse transcriptase polymerase chain reaction (RT-PCR) was used to identify the interesting genes:Duspl and Rabl.Results A total of 153 differentially expressed genes were found comparing the stimulated group and the control group.There were 74 up-regulated genes and 79 down-regulated genes in the stimulated group.Semi-quantita- tive RT-PCR confirmed that the Duspl and Rabl mRNA expression levels of the stimulated group were up-regulated. Conclusion The gene expression profiles of the bone marrow MSCs were altered after EMF exposure.The genes are involved in cell proliferation and differentiation,transcription control,metabolism and response to stress.These re- sults provide deeper insight into the mechanism of differentiation of the bone marrow MSCs.
5.Detection the serum IgM and IgG specific for TORCH in 1307 women in the period of pre-pregnancy and pregnancy.

OBJECTIVEThe objective is to obtain the information of women in the period of prepregnancy and pregnancy infected by TORCH (Toxoplasma, Rabella Virus, Cytomegalovirus, and Herpes Simplex Virus) in Kunming for preventing fetus from infection by TORCH in uterus.
METHODSThe serum IgM and IgG specific for TORCH from 1307 women in the period of pre-pregnancy and pregnancy were measured with ELISA and The reagent is the product of virion/serion.
RESULTSThe serum IgM specific for TORCH were detected in 2.83% of 1307 pregnant women for Toxoplasma, 2.37% for Rebulla Virus, 0.46% for Cytomegalovirus, 2.45% for Herpes Simplex Virus. The total positive rate of serum IgM specific for TORCH was 1.45%. The serum IgG specific for TORCH were detected in 3.98% of 1307 pregnant women for Toxoplasma, 72.3% for Rebulla Virus, 97.78% for Cytomegalovirus, 80.34% for Herpes Simplex Virus. The total positive rate of serum IgG specific for TORCH was 63.60%. There was no significant difference among the women with different pregnant situation in terms of the serum IgM and IgG specific for TORCH.
CONCLUSIONThere are some of women in the period of pre-pregnancy and pregnancy with current infection by TORCH in Kunming. It is recommended to screen for infection by TORCH in women in the period of pre-pregnancy and pregnancy to prevent fetus from infection by TORCH in uterus.

6.Preparation and quality evaluation of quercetin self-emulsifyied drug delivery systems.

OBJECTIVETo prepare the quercetin self-emulsified formulation and evaluate its quality.
METHODThe quercetin self-emulsified formulation was optimized based on the quercetin solubility in different oils, and the self-microemulsified efficiency of various combinations of emulsifier and co-emulsifier evaluated using the pseudo-ternary phase diagram. The microemulsion of morphology, size and zeta potential were examined. The quercetin of solubility in self-emulsified system was tested and the formulation stability was investigated by accelerated experiment.
RESULTThe blank self-emulsified system was composed of ethyl oleate/Cremophor EUL/butanol with weight ratio of 10: 54: 36. After being dilutied with water, the morphology of microemulsion was homogeneous small spherical drops observed under the electro-microscopy. The particle size and the zeta potential were 16.3 +/- 4.6 nm and 2.1 +/- 0.8 mV, respectively. The solubility of quercetin in self-emulsifing system was (62.42 +/- 0.11) mg x mL(-1), increased 2 229 folds compared with that of in water. The quality of quercetin self-emulsified formulation was stable during the 3 months storage at 40 degrees C.
CONCLUSIONThe solubility of quercetin is significantly increased in self-emulsified system and the formulation is stable and easy to prepare.

7.Gene mapping in Avellino corneal dystrophy pedigree

AIM:To point the susceptible gene in Avellino corneal dystrophy family with autosomal dominant inheritance.
●METHODS: Genomic DNA was extracted from the peripheral blood samples of all individuals of the pedigree. Several microsatellite makers were selected for gene scan in the hot regions of mutation. Linkage analysis was carried out using a Linkage software package. The haplotype data were processed using Cyrillic software to define the region of the disease gene.
●RESULTS: ln our pedigree, significant evidence of linkage was obtained at marker D5S396 and D5S393 [LOD score (Z)=3. 01, recombination fraction (θ)=0. 00]. The haplotype analysis of our pedigree was located between the microsatellite markers D5S808 and D5S638.
●CONCLUSION:The pathogenic gene of the Avellino corneal dystrophy pedigree is traced to a 11. 2 cM region in the chromosome 5q.
8.Identification of novel genetic markers in Mycobacterium tuberculosis Beijing genotype strains

Objective To identify a unique protein as a novel genetic marker for rapid molecular typing of Mycobacteriutn tuberculosis Beijing genotype strains by comparing the proteome of Beijing genotype strains with non-Beijing strains.Methods Fifty-six clinical isolates of Mycobacterium tuber- culosis were analyzed by spoligotyping to determine genotypes.The two-dimensional electrophoresis (2 DE)was used to compare the global protein patterns between Beijing genotype strains and non Bei jing strains.Differential expressed proteins were measured by matrix assisted laser desorption ioniza tion lime of flight mass spectrometry(MALDI-TOF-MS).The data obtained from peptide mass fingerprinting were compared in protein database.The genes encoding differential expressed proteins and their upstream were sequenced.Results Forty nine of the 56 isolates were Beijing genotype strains and 7 isolates were non-Beijing strains.A unique protein Rv0927c was identified,which is absent in Beijing genotype strains compared with 7 non Beijing strains and H37Rv.There were two characteristic mutations in Beijing genotype strains,a deletion of AGC at nucleotide position 421 of Rv0927c and a 127 G→A muta- tion in the upstream of Rv0927c.but not in non Beijing strains and H37Rv.Conclusion Characteris tic mutations of Rv0927c in Beijing genotype strains can be used as a novel genetic marker for rapid molecular typing of Mycobacteriuln tuberculosis Beijing genotype strains and non Beijing strains.
9.Randomized, double-blind, multi-center, positive parallel control clinical trial of compound Wuzhigan capsules on anemopyretic cold.

Compound Wuzhigan capsules is a compound preparation composed of Wuzhigan, Shidagonglao, Gangmei, Shanzhima. A Randomized, double-blind, multi-center, positive parallel control designed to evaluate the clinical efficacy and safety of compound Wuzhigan capsules on anemopyretic cold. One hundred and twenty anemopyretic cold patients were given compound Wuzhigan capsules (test group), 2 capsules one time, three times a day, 119 patients were given compound Wuzhigan tablets (control group) ,4 tablets one time, three times a day; three days of treatment The study showed, the markedly effective rate and total effective rate respectively were 63. 3% and 80% of the test group. For the control group, the markedly effective rate and total effective rate respectively were 72. 5% and 80. 7%. The difference was not statistically significant. Compound Wuzhigan capsules can reduce the dosage, and get better patient compliance.
