5.Effect of shenxiong huayu capsule on cerebral ischemia/reperfusion injury and the expression of GAP43 in hippocampal CA1 of rats.

OBJECTIVETo explore the effect of Shenxiong Huayu Capsule (SHC) on the cerebral ischemia-reperfusion (IR) injury and the expression of growth associated protein 43 (GAP43) after total cerebral IR in the hippocampal CA1 region of rats.
METHODSTotally 100 male adult SD rats were randomly divided into five groups, i.e., the control group, the model group, the group A (by taking SHC once daily), the group B (by taking SHC twice daily), and the group C (by taking SHC thrice daily), 20 in each group. The total IR model was prepared by improved Pulsinelli's 4-vessel occlusion method. Morphological changes of the hippocampal CA1 region were observed by HE staining at day 1, 3, 7, and 14. The expression of GAP43 in the hippocampal CA1 region was detected using immunohistochemical assay at day 1, 3, 7, and 14. Meanwhile, the behavioral score was determined. The expression of GAP43 in the hippocampal CA1 region was detected using Western blot at day 14.
RESULTSCompared with the control group, the expression of GAP43 increased in the model group, the behavioral score was elevated, degenerated neurons increased, and survival neurons decreased in the model group (all P < 0.05). Compared with the model group, the expression of GAP-43 increased (with the most significant difference seen in the group C, P < 0.01), the behavioral score significantly decreased, degenerated neurons decreased, and survival neurons increased in each HSC group (all P < 0.05). Survival neurons obviously increased at day 14, of which, most number of survival neurons and highest contents of GAP43 protein could be seen in the group C, showing statistical difference when compared with those of the group A and the group B (P < 0.01).
CONCLUSIONSHC had protective effect on total cerebral IR in the hippocampal CA1, which might be associated with increased expression of GAP43.

6.Therapeutic effect of external - route microsurgery for rhegmatogenous retinal detachment

AlM: To study the therapeutic effect of external-route microsurgery forrhegmatogenous retinal detachment.
METHODS: ln 55 patients ( 55 eyes ) with rhegmatogenous retinal detachment, drainage of subretinal fluid, examination of locating the holes, sclera cryotherapy, scleral buckling, and vitreous cavity injection of filtrated air were performed under surgical microscope.
RESULTS:The retinal reattachment occurred in 50 cases after the primary surgery. The final rate of reattachment was 91% during 6 - 12mo follow - up. The retinal reattachment occurred in 1 case ( recurrent retinal detachment) after the secondary surgery and in 4 cases ( recurrent retinal detachment ) after vitrectomy. The eyesight was improved with different degrees in 55 cases.CONCLUSlON: The external- route microsurgery for rhegmatogenous retinal detachment is simple, safe and effective.
7. Medical protection patterns in preventing heat stroke during 5-km armed field examination in summer

Objective To discuss the role of medical protection patterns in preventing exertional heat stroke during 5-km armed field examination in summer. Methods We analyzed the original data collected from 501 soldiers who completed the 5-km armed field examination in the summer of 2016. The soldiers received medical protection during examination and were divided into fully-equipped group (loading 9-12 kg, n=270) and non-equipped group (n=231). The temperatures of the soldiers were detected before and after examination, and the temperature changes and the number of soldiers with hyperthermia after examination were compared between the two groups. Results A total of 501 soldiers took part in the 5-km armed field examination, and 51 (10.18%) soldiers were found with fever (temperature > 37 °C), with a highest temperature of 40.9 °C. Nine were put on a temporary stay in the field medical facility. Among them, four were observed because of the temperature maintaining at 38 °C for 15 min, and their temperatures dropped to below 37 C after cooling fluid infusion for 50-120 min; four were observed because of chest tightness, suffocation, and abdominal pain; one was observed because of faint; the latter five cases were improved without other treatments. No case was evacuated to the hospital. There was no significant difference in temperature between the fully-equipped and non-equipped groups before the examination, while the temperature in the fully-equipped group was significantly higher than that in the non-equipped group after the examination ([36.64±0.67] °C vs [36.48±0.38] °C, P<0.05). The number of soldiers with hyperthermia in the fully-equipped group was significantly more than that in the non-equipped group after the examination (16.67%[45/270] vs 2.60% [6/231], P<0.05). Conclusion The 5-km armed field examination in summer can elevate temperatures of the soldiers and has risk of inducing heat stroke. Establishing a targeted medical protection pattern during the examination may help to timely detect fever in the soldiers and give prompt treatment, so as to effectively prevent heat stroke.
8.Relationship between the morphological and functional alterations of pancreatic islets and peripheral insulin resistance in rats with long-term high-fat diet

Objective To investigate the effect of long-term high-fat diet on insulin resistance and the morphology and function of islets in rats and the relationship between them.Methods Thirty normal male Wistar rats (8 weeks old) were divided into two groups and fed either with normal chow (NC,n=15),or high-caloric and high-fat diet (HF,n=15).Insulin resistance was assessed by euglycemic hyperinsulinemic clamp technique. The insulin secretory function of islets was evaluated by intravenous insulin releasing test.Morphological and quantitative analysis of pancreatic tissues was performed by double-label insulin and glucagon immunohistochemistry.Proinsulin mRNA was detected by RT-PCR.Results The glucose infusion rate (GIR) in HF rats was significandy lower than that in NC rats [(5.83?0.79)mg?kg~(-1)?min~(-1) vs (7.60?1.29)mg?kg~(-1)?min~(-1),P<0.05].Immunohistochemistry showed that HF rats had larger islet size [(15168?1327)?m~2 vs (6264?1840)?m~2,P<0.01] and significantly reduced insulin relative concentration of?cells[(-5.15?0.03) vs (-4.81?0.17),P<0.01],as compared with NC rats.The islet relative?cell volume was decreased signifieandy (P<0.01),whereas the relative?cell volume was increased (P<0.01).So the ratio of?to?were lower in HF [(4.68?1.01) vs (11.84?3.82),P<0.05].The peak of insulin secretion in intravenous insulin releasing test in HF was at 10 min,whereas that in NC rats was at 5 min.AUC (area under curve) 10-60 rain of insulin in HF was higher than that in NC rats [(152.51?34.53)mIU?L~(-1)?min~(-1) vs (86.40?21.21) mIU?L~(-1)?min~(-1),P<0.01].There was no difference in proinsulin mRNA levels between two groups. Conclusion Long-term high-caloric and high-fat diet results in early impairment of islet morphology and function, as well as significant insulin resistance,suggesting that the compensation ability of islets has already been impaired in the early course of type 2 diabetes mellitus.
9.Expression and clinical significance of ezrin in breast carcinomas

Objective To study the expression and clinical significance of ezrin in breast carcinoma. Methods Immunohistuchemieal staining(S-P) was used to detect the expression of ezrin in 63 cases of breast car-cinoma samples. Results Among these 63 samples,the total expression rates of ezrin were 55.56% (35/63). With the increasing of axillary lymph nodes metastasis and clinical staging,the rates of expressing of ezrin elevated (P<0.05),which was remarkably lower in patients whose disease-free survival(DFS) was > 5 years,or those whose DFS≤5 years or died within five years(P<0.05) ,but the expression of ezrin was not correlated with tumor size,age and menopausal status (P>0.05). Conclusion The expression of ezrin may contribute to prognostic e-valuation for breast carcinoma.
10.Rat intestine absorption kinetics study on cucurbitacin B-sodium deoxycholate/phospholipid mixed nanomicelles with in vitro everted gut sacs model.

To investigate the absorption kinetics of Cu B-SDC/PLC-MMs in rat different intestinal segments and compared with the absorption of Cu B suspension. The in vitro everted gut sacs model was established to study the absorption characteristics of Cu B-SDC/ PLC-MMs in rat duodenum, jejunum, ileum and colon, and the content of cucurbitacin B was detected by HPLC method, and the effects of concentrations on intestinal absorption were evaluated as well. The results showed that the absorption of Cu B-SDC/PLC-MMs was linearity at different intestine segment and different concentrations (R2 > 0.9), which was consistent with zero order rate process. The Ka of different intestine segments showed a concentration-dependent increasing along with the raised concentration of Cu B-SDC/ PLC-MMs, indicating that it was likely to be a mechanism of passive absorption. The best absorption site of Cu B-SDC/PLC-MMs was ileum, and its absorptions in different intestinal segments were superior to cucurbitacin B suspension. SDC/PLC-MMs could significantly enhance the intestinal absorption of cucurbitacin B, and the study of intestinal absorption kinetics of Cu B-SDC/PLC-MMs had gave a support to its further reasonable solidfication.
