1.Expression of FAM134B in liver cancer and its association with prognosis: An analysis based on data mining

ObjectiveTo investigate the expression of FAM134B in liver cancer and its association with clinicopathological features and prognosis. MethodsData mining was performed for Oncomine, Human Protein Atlas, TCGA, and Kaplan Meier-Plotter databases to analyze the expression of FAM134B in liver cancer, and the association of the expression of FAM134B with clinicopathological characteristics and prognosis was analyzed. The chi-square test was used for comparison of categorical data between groups, and the Kaplan-Meier method was used to investigate the association between FAM134B expression and prognosis of liver cancer. ResultsAs for normal tissue, the highest expression level of FAM134B was found in the kidney, and the expression of FAM134B in the liver ranked 9th; as for tumor tissue, the highest expression of FAM134B was found in renal cancer, and the expression of FAM134B in liver cancer ranked 11th. A total of 147 studies found a significant difference in FAM134B expression, and there was a significant difference in FAM134B expression between various tumors. FAM134B expression in liver cancer tissue was lower than that in normal liver tissue. FAM134B protein was localized in the endoplasmic reticulum, and its expression was associated with age of onset of liver cancer patients, liver cirrhosis, alpha-fetoprotein, race, and degree of tumor differentiation (all P<0.05). FAM134B expression was associated with the overall survival rate of liver cancer patients (hazard ratio [HR]=0.67, 95% confidence interval [CI]: 0.47-0.95, P=0.026) and had significant influence on the prognosis of the Asian population (HR=0.40, 95%CI: 0.22-0.74, P=0.003), while it had no significant influence on the prognosis of the Caucasian population (HR=0.65, 95%CI: 0.40-1.05, P=0.076). ConclusionThere is a significant difference in the expression of FAM134B between tumors, with a low expression in liver cancer. FAM134B protein is localized in the endoplasmic reticulum, and its low expression is associated with several malignant phenotypes of liver cancer and the low overall survival rate.

5.Congenital vascular rings: a rare cause of respiratory distress in infants and children.

BACKGROUNDCongenital vascular rings may often cause unexplained respiratory symptoms in infants and young children. Their diagnosis and treatment are often delayed. Few studies of vascular rings have been reported in China. The aim of this study was to describe the clinical presentation, diagnosis and surgical management of infants and children with congenital vascular rings.
METHODSClinical histories, physical examinations, investigations, image studies and surgical interventions were retrospectively evaluated in 7 children (age range: 2 months-4 years, mean 7 months) with congenital vascular rings. Chest radiography was performed in all patients. Echocardiography and computed tomography (CT) with 3-dimensional (3D) reconstructions were performed in 6 patients. Esophagography, cardiac catheterization and angiography, and bronchoscopy were performed in 1, 1 and 4 children, respectively.
RESULTSSix of the 7 patients had respiratory symptoms, including recurrent cough, stridor and wheeze. Age at onset of symptoms ranged from 1 month to 11 months. Chest X-ray showed nothing important on the vascular rings, besides bronchitis and pneumonia. Contrast-enhanced CT diagnosed vascular rings in 6 patients. Four patients had double aortic arches, two had balanced arches and two were right arch dominant. One patient had a right aortic arch with left ligament and 1 patient had a pulmonary artery sling. Echocardiography failed to diagnose vascular rings in 2 patients. The esophagogram of 1 patient showed esophageal compression. Bronchoscopy of 4 patients showed compression of the distal trachea. Five of the 7 patients underwent surgical division of the vascular rings. Surgical observation confirmed the CT findings in each patient.
CONCLUSIONSPatients, especially infants or young children, with recurrent respiratory symptoms such as chronic cough, stridor and wheeze, should be examined for the possible presence of congenital vascular rings. Contrast-enhanced CT can clearly show the anatomy of vascular rings. As a noninvasive technique, echocardiography is helpful for diagnosis. Early surgical management in symptomatic patients is effective.

6.Effects of Huafenqinutang and vardenafil for treatment of chronic prostatitis/chronic pelvic pain syndrome with concomitant erectile dysfunction.

OBJECTIVETo evaluate the effects of Huafenqinutang combined with vardenafil for the treatment of chronic prostatitis/chronic pelvic pain syndrome (CP/CPPS) with erectile dysfunction.
METHODSOne hundred and thirty-eight cases diagnosed as CP/CPPS with erectile dysfunction were randomized into trial group (70 cases) and control group (68 cases) for treatment with Huafenqinutang for 8 weeks, and in the former group, vardenaffil was added since the fifth weeks. All the cases were evaluated according to NIH-CPSI and IIEF-5 scores at 4 weeks and 8 weeks, respectively.
RESULTSAt the end of the fourth weeks, NIH-CPSI score was 13.1-/+4.7 in the trial group and 13.3-/+4.5 in the control group, which were comparable between the two groups (P>0.05) but significantly decreased compared with the pre-treatment scores in both groups (P<0.01). IIEF-5 score was also similar between the two groups (14.1-/+3.3 vs 14.3-/+5.0, P>0.05) but significantly increased compared with the pre-treatment scores in both groups (P<0.01). At the end of the eighth week, NIH-CPSI score was 7.8-/+2.2 and IIEF-5 score 20.1-/+4.4 in the trial group, which were significantly different from those at the end of the fourth week (P<0.01). In the control group, NIH-CPSI score was 12.7-/+2.3 and IIEF-5 score 14.3-/+4.5 at the eighth week, similar to those at the end of the fourth week (P>0.05). There were significant differences in NIH-CPSI and IIEF-5 scores between the 2 groups (P<0.01), and the change of NIH-CPSI score was negatively correlated with IIEF-5 score in the trial group (r=-0.89, P<0.01).
CONCLUSIONFor patients with CP/CPPS with erectile dysfunction, Huafenqinutang treatment in combination with vardenafil can effectively improve the erectile functions and decrease the NIH-CPSI score to favor the recovery from CP/CPPS.

7.Evaluation of early cognitive ability of infants born preterm by near-infrared spectroscopy.

OBJECTIVETo compare the differences in cerebral oxygenation responses between the infants born preterm and full-term infants and to evaluate the early cognitive ability of infants born preterm.
METHODSCerebral oxygenation after light stimulation was detected by near infrared spectroscopy (NIRS) in preterm infants at 3 or 6 months corrected gestational age (GA). The results were compared with those of age-matched infants born at term.
RESULTSThe start and peak response time of cerebral oxygenation occurring after light stimulation in preterm infants at 3 months corrected GA was 17.2 +/- 5.2 and 38.4 +/- 9.6 seconds respectively, which were significantly longer than in age-matched term infants (13.1 +/- 2.7 and 28.9 +/- 5.0 seconds respectively) (P < 0.05). The maximum response value of hemoglobin, oxyhemoglobin and regional oxygen saturation of the preterm infants at 3 months corrected GA was (1.2 +/- 0.5)%, (1.5 +/- 0.6)%, and (1.3 +/- 0.4)% respectively , which were significantly lower than that of the term infants [(2.3 +/- 0.3)%, (2.8 +/- 0.3)% and (2.4 +/- 0.5)% respectively] (P < 0.05). Cerebral oxygenation responses to light stimulation in preterm infants examined at 6 months corrected GA were not significantly different from age-matched term infants.
CONCLUSIONSCerebral oxygenation responses to light stimulation in infants born preterm at 3 months corrected GA are not as good as age-matched term infants, but were close to the level of age-matched term infants at 6 months corrected GA. This suggests that the early cognitive ability of preterm infants before 3 months corrected GA might fall behind age-matched term infants.

9.Differential proteomic analysis in human umbilical cord mesenchymal stem cells induced by cobalt chloride.

OBJECTIVETo analyze the differential proteomics in human umbilical cord mesenchymal stem cells (MSC) induced by chemical hypoxia-mimetic agent cobalt chloride (CoCl(2)) by two-dimensional gel electrophoresis (2-DE) and mass-spectrometry.
METHODS2-DE was performed to separate proteins from treated and untreated human umbilical cord MSC with CoCl(2). 2-DE images were analyzed by ImageMaster 2D Platinum software 6.0. The differential expressed proteins was identified by MALDI-TOF-MS. The differential proteins were classified based on their functions.
RESULTS2-DE reference patterns of CoCl(2) treated human umbilical cord MSC were established. A total of twenty-six differential proteins were identified, of them eleven proteins were up-regulated and fifteen down-regulated. Their biological functions involved in carbohydrate metabolism, protein metabolism and modification, lipid metabolism, coenzyme and prosthetic group metabolism, cell cycle, immunity and defense, cell structure and motility, signal transduction, protein targeting and localization, neuronal activities, muscle contraction, etc. Peroxiredoxin1 (Prdx) was down-regulated, whereas alpha-enolase (ENO1) and vesicle amine transport protein 1 homolog (VAT1) up-regulated.
CONCLUSIONThe effects of hypoxia on human umbilical cord MSC were participated by multiple proteins and involved in multiple functional pathways.

10.Cytotoxicity evaluation of three kinds of perforation repair materials on human periodontal ligament fibroblasts in vitro.

OBJECTIVETo select three kinds of perforation repair materials, mineral trioxide aggregate (MTA), Z350, amalgam. And to evaluate the cytotoxicity of three kinds of perforation repair materials on human periodontal ligament fibroblasts (HPDLF) in vitro.
METHODSThe proliferation of HPDLF to three perforation repair materials were examined by methyl thiazolyl tetrazolium (MTT) assay at 1, 3 and 5 days. The mRNA expression levels of bone-associated alkaline phosphatase (ALP) and osteocalcin (OC) were determined using a real-time quantitative polymerase chain reaction (PCR).
RESULTSMTA shew almost no inhibition to HPDLF, the expression of ALP mRNA and OC mRNA in the HPDLF cultured on MTA were higher. Z350 induced a slight inhibition to HPDLF, and the expression of ALP mRNA but there was no difference in the expression of OC mRNA. Cell proliferation was significantly impaired by amalgam with grade 3, and the expression of ALP mRNA and OC mRNA were significantly reduced.
CONCLUSIONMTA have minimum cytotoxicity on HPDLF and can promote cell differentiation and regenerate of periodontal tissue. Z350 have lower cytotoxicity on HPDLF. Amalgam show highest cytotoxicity on HPDLF in the three materials and inhibit cells differentiation.
