1.Effect of different instruments on glycosylated hemoglobin test

Objective To investigate the detection results of GHB with different instruments, and to provide reliable basis for improving the accuracy of GHB detection. Methods ADVIA 1650 biochemical analyzer and HA-8160 glycosylated hemoglobin analyzer were used. The accurate detection of glycosylated hemoglobin in 100 healthy people was carried out, and the related data were recorded and analyzed statistically. Attention should be paid to this group according to glycosylated hemoglobin detection value, that is, within 6.5% for A group, 6.5%-10.0% for B group, more than 10% for C group, and compare the above-mentioned different grouping of corresponding instrument glycosylated hemoglobin detection value. Results The analysis showed that there was no significant difference between the two groups of C instrument glycosylated hemoglobin levels, but the level of glycosylated hemoglobin detected by A in group HA-8160 and group B was significantly higher than that of ADVIA 1650(P<0.05). Conclusion Different instruments have some differences in the detection value of glycosylated hemoglobin. Clinicians should judge the disease comprehensively according to the actual situation and other indicators.

Objective To investigate the cell compatibility of polystyrene(PS) plate chemically modified with RGD peptides.Methods PS surfaces were carboxylated by permanganate oxidation in diluted sulfuric acid,and carboxyls were activated with water-soluble carbodiimide to graft with gelatin,collagen and RGD peptides.IR,X-ray photo-electronic spectroscopy (XPS) and dynamic contact angle were used to characterize the surface modification of PS surface.Results XPS results confirmed the existence of nitrogen element from protein molecules and the covalently binding of proteins to PS surfaces.Dynamic contact angle measurement indicated hydrophilicity of PS surfaces was improved obviously after grafting modification.The cell culture results showed that the cell adhesion and proliferation was better on modified surfaces than the initial.Conclusion The cell compatibility of PS surface was great improved after modification with RGD peptides,which would provide a potential strategy to improve the culture of purified endothelial progenitor cells isolated by immunomagnetic beads.
4.The anti-oxidative effect of deferiprone on ocular lens in diabetic rat

Background Oxidative stress is thought to be responsible to diabetes-complicated cataract.Our previous study demonstrated that as an iron chelator,deferiprone can protect lens from oxidative damage.Objective This further study aimed to investigate the role of deferiprone on the formation of diabetic-complicated cataract.Methods Forty 6-week-old Wistar rats were included in the study and randomized into 4 groups.Eight of them were used as the normal control group.Diabetes mellitus animal models were established in 22 rats by the carbonhydratediet and fat diet and the intraperitoneal injection of 40 mg/kg streptozocin (STZ).The deferiprone of 50 mg and 100 mg were intragastrically given in 8 model rats respectively after 3 days once a day for 8 weeks.The opacification of lenses was examined under the slit lamp weekly after treatment.The animals were sacrificed and the lenses were obtained at the eighth week of deferiprone injection.The concentrations of water-soluble protein ( WSP),urine-soluble protein (USP) and alkali-soluble protein (ASP) in rat lens suspension were detected by Bradford method.The super oxide dimutese (SOD),malondialdehyde (MDA) and glutathione (GSH) were determined spectrometically using xanthine oxidase,thiobarbituric acid,dithio bis-nitrobenzoic acid.Results No evidently differences were found in the content of the WSP,USP and ASP among the these groups( F=1.73,0.18,0.09,P>0.05).The contents of MDA in 50 mg deferiprone group and 100 mg deferiprone group were ( 1.05 ± 0.10 ) mmol/g and ( 1.05 ± 0.22 ) mmol/g respectively,showing a significant decline in comparison with diabetic model group (P<0.05).The SOD and GSH contents in lens were (321.29±16.57) U/mg,(322.07±22.16) U/mg and (7.83±0.65 ) mg/g,(7.70±0.77 ) mg/g respectively in 50 mg deferiprone group and 100 mg deferiprone group and were considerably elevated in comparison with ( 298.70± 14.69 ) U/mg and ( 5.47 ± 1.01 ) mg/g of diabetic model groups ( P<0.05 ).No significant differences were found in the indexes mentioned above between 50 mg and 100 mg deferiprone groups(P>0.05).Conclusions Deferiprone can reduce oxidative stress and improve the energy metabolism of the lens in diabetic rats.
5.Management of malignant biliary hilar obstruction with multiple stents

Objective To evaluate the efficacy of the treatment of hilar biliary malignant obstruc tion with multiple stent drainage.Methods One hundred and twenty-seven patients with malignant biliary hilar obstruction were enrolled.The obstructions at the common hepatic duct within less than 1 cm to the junction of the left and the right hepatic duct were found in 66 cases,at the proximal common hepatic duct and the left and the right hepatic ducts in 45 cases,at the right hepatic duct in 5 cases and at the both left and right hepatic duets in 11 cases.Sixty-six patients received stent placement through the right biliary ducts and the common bile duct by puncturing the right mid-axillary line.The other 37 patients received 2 stents placement (disposed"Y"style) through the left and the right hepatic duct punc turing routway.Seven patients received 2 stents placement (disposed"┌"style) through the right hepatic duct and the common bile duct with a stent placed between the left and the right hepatic duct.Three patients had right hepatic duct stent placed first,followed by right hepatic duct and common hepatic duct stent. Twelve patients had stents placed in the right hepatic duct with external drainage from the left hepatic duct. Two patients had multiple strictures at the right hepatic duct,who got multiple external drainages.The total serum bilirubin levels were measured pre-and post-operatively.Results One hundred and twenty-seven patients with bi[iary obstraction had internal stents placed for drainage.The average total bilirubin levels among 121 patients were (283.4?175.4 )?mol/L pre-operation and (63.2?11.8)?mol/L post-operation (P
6.A comparison study of interbody cage in lumbar degenerative disease.

OBJECTIVETo compare the advantages and disadvantages of TFC/BAK, Prospace and Brantigan I/F carbon cages and summarize their applications.
METHODSOne hundred and twelve cases of Brantigan I/F, 123 cases of TFC/BAK and 45 cases of Prospaces were used in the treatment of degenerated lumbar disorders. The clinical results and complications were observed and analyzed.
RESULTSAll devices restored the height of intervertebral space and achieved immediate stability after insertion. One hundred and twelve cases of Brantigan I/F cages were solid fused, the clinical results were satisfactory. Three cases of TFC/BAK cages subsided into the vertebra and 2 cages retropulsed slightly during follow-up. There was no serious complications of nervous system, but the operation time and amount of bleeding in Brantigan I/F group increased.
CONCLUSIONSCarbon fiber intervertebral cages are an effective method in treating degenerated lumbar disorders. To avoid complications, the indications and techniques must be selected carefully.

7.Application of intervertebral carbon fiber cage in degenerated lumbar disorders.

OBJECTIVESTo evaluate the effect of intervertebral carbon fiber cages in the treatment of degenerated lumbar disorders.
METHODSOne hundred and twelve patients of degenerated lumbar disease were analyzed retrospectively with regard to the effect of posterior lumbar interbody fusion (PLIF) using intervertebral carbon fiber cage.
RESULTSIn the 112 patients, disc height was 7 mm (3 to 11 mm) on average before operation and 11.2 mm (8 - 14 mm) after operation. Cobb angle was 32 degrees (21 degrees to 42 degrees ) on average before operation and 18.9 degrees (13 degrees to 21 degrees ) after operation. After follow-up for 7.8 months (4 to 13 months) on average, all operative disc spaces showed solid arthrodesis through X-ray images. Disc height was 10.9 mm (8 - 14 mm) on average, and Cobb angle was 18.5 degrees (13 degrees to 21 degrees ) on average. During the follow-up, endplate absorbing, pseudo-articulation, cage displacement, subsidence, and breaking were not found.
CONCLUSIONImplantation of intervertebral carbon fiber cages is an effective method for the treatment of lumbar degenerative disease. Accurate indications and skilled techniques are required to avoid complication.

10.Occurrence of supernumerary upper incisor teeth in Pax6-/- mouse fetuses.

OBJECTIVETo investigate the occurrence of supernumerary upper incisor teeth in Pax6-/- mouse fetuses and to provide a model to explore the role of Pax6 in the upper incisor development and the mechanism of supernumerary teeth involving Pax6.
METHODSTwenty Pax6-/- mouse fetuses of strain DEBA were isolated on E18.5 (embryonic day). The fetuses were sectioned serially in coronal plane and stained with haematoxylin and erosion, then the presence of supernumerary teeth in the upper anterior area was examined histologically, and also the number, morphology and structure of lower incisor germs and the first and second molar germs in the maxilla and mandible were observed histologically. Eighteen E18.5 mouse fetuses of strain DEBA with Pax6+/+ genotype were used as control.
RESULTSOf the 20 Pax6-/- fetuses examined, four possessed a single supernumerary tooth in the upper incisors' region. No supernumerary upper incisor teeth were observed in any of the 18 Pax6+/+ fetuses examined. In the regions of lower incisors and the first and second molars of the maxilla and mandible, no significant difference was observed between Pax6-/- and Pax6+/+ fetuses regarding the number, morphology and structure of tooth germs.
CONCLUSIONSThe results suggest that Pax6 played an important role in the development of upper incisor teeth in mice.
