1.Sulfur mustard-induced changes of IL-2 and IL-6 concentrations and lymphocyte DNA damage in dog peripheral blood

Objective To study the changes of IL 2 and IL 6 levels and lymphocyte DNA damage in peripheral blood of dog intoxicated by sulfur mustard. Methods Chongqing dogs were subcutaneously injected with sulfur mustard at the dose of 16 mg/kg. Lymphocytes and serum were isolated from dog peripheral blood at different time points as designed. The IL 2 and IL 6 levels in blood serum were measured by radioimmunoassay and lymphocyte DNA damage represented as comet tail length was determined by single cell gel electrophoresis (SCGE). Results Four hours after administration of sulfur mustard, the serum IL 2 and IL 6 levels dogs injected with sulfur mustard decreased and reached the lowest level at 72 h. A significant recovery was found at the time point of 120 h. SCG revealed sulfur mustard induced DNA fragmentation (comet) in dog peripheral lymphocytes. The rate of the damaged lymphocytes and the degree of DNA fragment migration began to increase at 4 h and progressively increased from 24 h to 72 h after sulfur mustard treatment. Conclusion Sulfur mustard intoxication induces early and long lasting decreases in serum IL 2 and IL 6 levels and lymphocyte DNA fragmentation in dog peripheral blood.
2.Simulation of sparse views CT image iterative reconstruction based on defective projection data

Objective To reconstruct sparse views CT image based on defective projection data.Methods The position of bad bins in detector determined whether the linear interpolation was applied to the defective projection data.Moreover,reconstruction of air pixels in CT image was achieved rapidly and accurately.Results he experimental results showed that the proposed method could solve the problem from classical ART-TV method that the robustness was unstable due to the different positions of bad bin in CT detector.Conclusion Compared to analytical reconstruction methods,iterative methods can solve the reconstruction problems in this modality so that the radiologist is facilitated to perform image processing and quantitative analysis.
3.Integration and expression of HLA-B2704 gene in transgenic mice

Objective To study the integration and expression of HLA-B2704 gene in HLA-B2704 transgenic mice. Methods HLA-B2704 gene was introduced into fertilized eggs of C57BL/6?Kunming and Kunming?Kunming by microinjection. The founder mice were screened by PCR for integration of HLA-B2704 transgene and were then further confirmed by southern blot. RT-PCR and flow cytometry were carried out to detect the expression of HLA-B2704 gene at mRNA and protein level. Results Ten F0 hybrid mice carried HLA-B2704 gene in 101 F0 hybrid mice, 3 of 64 (4.7%) hybrid mice from Kunming?Kunming and 7 of 37 (18.9%) hybrid mice from C57BL/6?Kunming background carried HLA-B2704 gene. 5.Clinical value of transanastomotic pancreatic ductal stents placement after pancreaticoduodenectomy: a meta analysis

Objective To systematically review the clinical efficacy of transanastomotic pancreatic ductal stents placement after pancreaticoduodenectomy.Methods According to the Cochrane reviewers handbook (version 5.0 ),literatures were retrived from PubMed,Embase,Cochrane,VIP database,China Biology Medicine disc and CNKI database,and then the quality of the literatures was analyzed.Meta analysis was carried out using the RevMan software ( version 5.0.18 ).A random effects model was adopted,and the results of the meta analysis were presented with odds ratio (OR) and 95% confidence interval (95% CI).Results Four randomized controlled trials including 557 patients were retrieved.External stents were used in 160 patients and internal stents in 115 patients.The results of meta analysis showed no significant difference in the rate of fistula,overall postoperative morbidity and mortality between patients who did or did not receive pancreatic stents placement (OR =0.66,0.70,0.63,P > 0.05 ).There were significant differences in the rate of pancreatic fistula and overall postoperative morbidity between patients who received external pancreatic stents placement and those did not receive pancreatic stents placement ( OR =0.48,0.55,P < 0.05 ).There was no significant difference in the mortality rate between patients who received external pancreatic stents placement and those did not receive pancreatic stents placement (OR =0.71,P > 0.05 ).There were no significant difference in the incidence of pancreatic fistula,overall postoperative morbility and mortality between patients who received internal pancreatic stents placement and those did not receive pancreatic stents placement ( x2 =0,0.75,2.11,P > 0.05 ).Conclusions External pancreatic stents placement after pancreaticoduodenectomy can reduce the incidence of postoperative complications.The effects of internal pancreatic stents placement need to be proved by further highquality prospective randomized trials.
6.The effects of mild stress to gastrointestinal motility and oxytocin in hypothalamic paraventricular nucleus of rats

Objective To study the changes of gastrointestinal movement function in rats with chronic unpredictable mild stress(CUMS) and explore the mechanisms underlying it.Methods The rats were divided into stress model group and control group.The stress model rats were induced by 21-day chronic unpredictable mild stress as well as social-isolated fed.The rate of ink propulsion of gastrointestinal tract and the contraction of intestinal canal in rats were observed.Immunohistochemistry was adopted to detect the expression of OT in rats.Results (1) After the models were induced,weight-gain and sucrose preference of model group ((69.97 ± 9.81) g,(49.05± 5.98) g) were lower than those in control group ((116.27 ± 13.60) g,(83.51 ± 3.08) g) (P < 0.001),and both the crossing-score and rearing-score ((24.00 ± 13.52),(3.90 ± 2.51)) were lower than those in control group ((53.60 ± 27.98),(11.50 ± 8.85)) in the open-field test.(2) The rate of ink propulsion of model group ((67.33 ± 6.24) %) was decreased when compared to the control group ((76.83 ± 10.00) %) (P < 0.05),and the intestinal canal contraction amplitude and contraction frequency ((1.37 ± 0.18) g,(0.58 ± 0.02) S-1) were lower than those in control group ((1.88 ± 0.13) g),(0.62 ± 0.04) S-1) (P < 0.05).(3) Compared with the control group (6.07 ± 3.71),OT immunoreactive substance was increased in model group (59.17 ± 16.08) of rats (P<0.001).Conclusion Chronic stress can cause the decrease in gastrointestinal movement function of rats.These changes may be related to the increased expression of OT in paraventricular nucleus.
7.Association of single nucleotide polymorphism of xeroderma pigmentosum group C and smoking with genetic susceptibility to pancreatic cancer

Objective To investigate the correlation of single nucleotide polymorphism of rs3731055 and rs2607775 of xeroderma pigmentosum group C (XPC) and smoking with genetic susceptibility to pancreatic cancer.Methods The clinical data of 214 patients with pancreatic cancer who were admitted to the First and Third Affiliated Hospitals of Xinjiang Medical University from January 2009 to June 2011 and 214 healthly individuals were retrospectively analyzed.The samples of venous blood of 214 patients with pancreatic cancer (case group) and 214 healthy individuals (control group) were analyzed by the Multiplex SNaPshot method.The count data were analyzed using the chi-square test.The association between the single nucleotide polymorphism of rs3731055 and rs2607775 with genetic susceptibility to pancreatic cancer was analyzed using the Logistic regression method.Results Four hundred and twenty-three samples of gene were successfully typed,including 210 in the case group and 213 in the control group.The frequency of G allele of XPC rs3731055 was 75.95% (319/420) in the case group and 77.00% (328/426) in the control group,with no significant difference between the 2 groups (x2 =0.12,P > 0.05).The frequencies of genotypes GG,GA and AA were 58.57% (123/210),34.76% (73/210) and 6.67% (14/210) in the case group,and 60.09% (128/213),33.80% (72/213) and 6.10% (13/213) in the control group,with no significant difference between the 2 groups (x2=0.12,P > 0.05).The frequency of C allele of XPC rs2607775 was 87.86% (369/420) in the case group and 93.43% (398/426) in the control group,with significant difference between the 2 groups (x2=7.75,P < 0.05).The frequencies of genotypes CC,CG and GG were 77.62% (163/210),20.48% (43/210) and 1.90% (4/210) in the case group,and 86.85% (185/213),13.15% (28/213) and 0(0/213) in the control group,with significant difference between the 2 groups (x2=8.54,P < 0.05).Patients with rs2607775 GC genotype were associated with a significantly increased risk of pancreatic cancer compared with patients with rs2607775 CC genotype (adjusted OR =1.81,95% CI:1.06-3.10,P < 0.05).Patients with rs2607775 GC + GG genotype were associated with a significantly increased risk of pancreatic cancer compared with patients with rs2607775 CC (adjusted OR =1.98,95% CI:1.16-3.36,P < 0.05).The ratio of patients in the case group who smoked cigarettes ≥ 17 pack years was 25.24% (53/210),which was significantly higher than 13.15 % (28/213) of the control group (x2 =11.37,P < 0.05).The results of univariate analysis showed that patients who smoked cigarettes ≥ 17 pack years had higher risk of getting pancreatic cancer (adjusted OR =2.82,95% CI:1.27-6.29,P < 0.05).Patients who smoked cigarettes ≥ 17 pack years and with rs2607775 CC also had higher risk of getting pancreatic cancer (adjusted OR =2.87,95% CI:1.18-6.99,P <0.05).No significant gene-environment interaction was observed between rs2607775 GC + GG and smoking ≥ 17 pack years (adjusted OR =3.65,95% CI:0.67-20.03,P > 0.05).Conclusions The polymorphisms of XPC rs2607775 may play a role in the onset of pancreatic cancer.Patients who smoke cigarettes ≥ 17 pack years are more easily to have pancreatic cancer.There is no interaction between smoking and XPC rs2607775 in influencing the progression of pancreatic cancer.

0.05).Conclusions The reduced expression of HLA-G on placenta in ICP patients may alter the maternal-fetal immune response and thus be involved in the pathogenesis of this disorder. Dexamethasone can upregulate the expression of HLA-G on placenta.The 14 bp deletion polymorphism in exon 8 of HLA-G gene might not have a significant influence on the development of ICP.

Objective To investigate the influence of motor impersistence on functional recovery and prog- nosis of patients with left hemiplegia. Methods A total of 64 in-patients with left side hemiplegia who were able to understand the oral instructions were assigned into a motor impersistence group (n = 31 ) and a control group (n = 34) , respectively, according to the assessment with Scale of Motor Impersistence developed by Joynt. Both groups were comparable in terms of the basic clinical characteristic and motor function of patients except motor impersis- tence. The conventional rehabilitation training programs were adopted for all patients in this study. Brunnstrom func- tional category, Ueda Satoshi hemiplegic finger function assessment scale, upper limb function assessment scale, Berg balance scale, Functional ambulatory category, Modified Barthel Index were employed to evaluate the subjects before and after 1 and 2 months of treatment, with regard to the motor function of their upper limbs, walking ability, activities of daily living performance and balance ability. Results After 1 month of treatment, the patients in the M1 group were significantly improved with regard to the motor function of upper limbs, hand function, standing ba- lance and activity of daily living performance( P0.05 ) , while those in the control group were improved with all the parameters except the lower limb function. 10.Screening, Identification of a Chitinolytic Bacterium Strain SWCH-6 and Its Optimal Chitinase Fermentation Conditions

A bacterium, having high chitinolytic activity, was isolated from sediment of Shantou Bay, named SWCH-6. According to its physiological and biochemical characteristics and 16S rDNA sequence, it was identified as Aeromonas hydrophlilla. The optimal chitinase fermentation conditions of strain SWCH-6 were conformed by single-factor experiments and orthogonal experiments, they were colloidal chitin 25.0 g/L, tryptone 10.0 g/L, seawater 1 L, pH 8.5, 32 ℃ , 150 r/min for 72 h. In these conditions, its chitinase activity reached 0.39 U/mL. In addition, at 40℃ and pH 5.0, its chitinase performed the highest catalytic activity and its chitinase activity could be enhanced by Cu2+, Fe3+ and surfactant toween-80; weakened by Zn2+, Mn2+ and surfactants SDS, detergent powder, and there were some differences from orther chitinases.
