1.Clinical study on effect of yufeng capsule on premonitory symptoms of apoplexy.

OBJECTIVETo observe the clinical effect of Yufeng Capsule (YFC), a Chinese herbal preparation with function of clearing Heat-toxin in treating premonitory symptoms of apoplexy in middle and old aged patients.
METHODSOne hundred and fourteen patients with premonitory symptoms of apoplexy were treated with YFC and compared with 57 patients treated with Venoruton for control. The changes of clinical symptoms, anti-oxidation capability, blood lipid, blood sugar and hemorrheological parameters were observed.
RESULTS(1) The therapeutic effect in the YFC group was cured in 42 cases (36.84%), markedly effective in 38 (33.33%), effective in 28 (24.58%) and the total effective rate was 94.74%, while in the control group, the corresponding numbers were 9 (15.79%), 8(14.04%), 20(35.09%) and 64.91% respectively, the difference between the two groups in cure rate, markedly effective rate and total effective rate was significant (P < 0.05); (2) The whole blood viscosity, plasma viscosity, hemagglutination index and red blood cell deformity index in the YFC group were significantly improved after treatment (P < 0.01) and the improvement was better than those in the control group (P < 0.05 or P < 0.01); (3) Levels of serum malonyldialdehyde (MDA), superoxide dismutase (SOD), blood lipids and glucose were markedly improved in the YFC group (P < 0.05) after treatment, and showed a significant difference to those in the control group (P < 0.05).
CONCLUSIONYFC has obvious effect in treating premonitory symptoms of apoplexy patients, it could markedly improve the hemorrheologic parameters, regulate blood lipids and blood glucose metabolism, and strengthen the anti-oxidation capability of patients.

2.Expression of CD_64 in Neonatal Infection Disease and Its Clinical Significance

Objective To study CD64 expression in neutrophilic granulocyte and its clinical effect in neonatal infection disease. Methods CD64 was detected among 59 neonatal patients(septicemia group 34 patients, local infection group 25 patients)hospitalized in our neonatal department diagnosed as neonatal infection disease in 48 h after hospitalized,2 weeks after therapy, then the results were compared with 27 patients as non - infection disease during the same period. Results CD64 in septicemia group was (6156. 21?3643. 32) molecula per cell,in local infection group was (2176.19 ? 946. 32)molecula per cell, in non- infection group was (2176. 19 ? 946. 32) molecula per cell.There were significant differences among three groups (all P0.05). Conclusions CD64 expression increases in bacterium infection disease. It is more obvious in widespread infection desease.and it can be the mark in early diagnosis of neonatal infection disease.
3.Application of acupuncture compound anesthesia in transvaginal ultrasound-guided oocyte retrieval.

OBJECTIVETo observe analgesic effect and safety of acupuncture compound anesthesia in transvaginal ultrasound-guided oocyte retrieval.
METHODSThree hundred and sixteen cases undergoing in vitro fertilization and embryo transfer (IVF-ET) were randomly allocated to an acupuncture compound anesthesia group (n = 146) and a simple Pethidine group (n = 170). They received respectively electroacupuncture combined with intramuscular injection of Pethidine and simple intramuscular injection of Pethidine 30 min before oocyte retrieval.
RESULTSThe acupuncture compound anesthesia group was significantly better than the simple Pethidine group in the pain rating and pain score (P < 0.01); the incidence rate of abdominal pain at 1 h and 2-5 h after oocyte retrieval in the acupuncture compound anesthesia group was lower than that in the simple Pethidine group (P < 0.01).
CONCLUSIONIn transvaginal ultrasound-guided oocyte retrieval, acupuncture compound anesthesia has the advances of safety, high effectiveness, rapid recovery after oocyte retrieval, and few side effects.

4.Effects of different time parameters on hypercholesterolemia treated with mild moxibustion.

OBJECTIVETo probe into the relevance between the parameters of mild moxibustion (persistent time, interval time, treatment session) and the efficacy, as well as the optimum treatment program of mild moxibustion in hypercholesterolemia.
METHODSThe patients with hypercholesterolemia were selected as the observation objects. Mild moxibustion was applied to Shenque (CV 8) and Zusanli (ST 36). The total cholesterol (TC) was taken as the index. The orthogonal experimental design was adopted for paired combination of 3 factors (persistent time, interval time and treatment session) that affected TC reducing in mild moxibustion. Moreover, the changes in blood lipid were observed to choose the optimum treatment program.
RESULTSThe importance of the factors of mild moxibustion that induced TC reducing in the patients with hypercholesterolemia was interval time > treatment session > persistent time. The effect of 10 min moxibustion on each point was better than that of 5 min moxibustion. The effect of once moxibustion every other day was better than that of once every day; and the effect of 6-week treatment was better than that of 12-week treatment.
CONCLUSIONThe interval time of mild moxibustion is the chief factor of the efficacy on hypercholesterolemia. The optimum treatment program is mild moxibustion, 10 min on each point, once every other day and lasting 6 weeks.

5.Inhibitory effect of Panax notoginseng saponins on alveolar epithelial to mesenchymal transition.

In the study, the effects of Panax notoginseng saponins (PNS) on alveolar epithelial to mesenchymal transition (EMT) and extracellular matrix degradation were observed in a type of human alveolar epithelial cell, A549 cells, stimulated by TGF-beta1. Firstly, MTT method was applied to evaluation of cellular proliferation and found that PNS from 12.5 mg x L(-1) to 200 mg x L(-1) dosage could not inhibit significantly cellular proliferation. Then, cells were divided into five groups, normal group, TGF-beta1 group, TGF-beta1 + 50 mg x L(-1) PNS group, TGF-beta1 + 100 mg x L(-1) PNS group and TGF-beta1 + 200 mg x L(-1) PNS group. Normal cells were not stimulatec by TGF-beta1; TGF-beta1 cells were only stimulated by TGF-beta1 and the other cells were stimulated by TGF-beta1 with different doses of PNS, respectively. After stimulation, cells and supernatants were collected for assays. Cellular roundness was applied to quantitative evaluation of morphological change. Immunocytochemistry was applied to examine E-cadherion, a-SMA and FN proteins expression in the cells. Enzyme linked-immunosorbent assay was applied to MMP-9 and TIMP-1 levels. The results showed that EMT of A549 cells was induced by TGF-beta1, showing significant change of roundness, E-cadherion, alpha-SMA and FN (P < 0.05, P < 0.01). Compared to TGF-beta1, PNS significantly inhibited the changes of roundness (P < 0.05), FN and alpha-SMA (P < 0.05, P < 0.01) and not significantly inhibited the change of E-cadherion. Furthermore, MMP-9 levels were significantly increased by TGFbeta1 stimulation (P < 0.05), without significant change of TIMP-1. Compared with TGF-beta1, PNS could significantly increase MMP-9 level (P < 0.05) and decrease TIMP-1 levels (P < 0.05, P < 0.01). In conclusion, PNS could inhibit alveolar epithelial cell EMT induced by TGF-beta1, with increase of extracellular matrix degradation ability, which showed anti-fibrosis of lung ability.

6.RP-HPLC determination of mangiferin in the leafs of Folium mangiferae sampled in different months and regions.

OBJECTIVETo develop a new method for determination of Mangiferin in the leafs of Folium mangiferae. By this new method, Mangiferin in F. mangiferae sampled in different months and in different regions was determinated.
METHODA RP-HPLC method was set up, using Shim pack CLC-ODS column, methanol-0.05 mol.L-1 H3PO4(65:134, pH 3.5) as mobile phase, with 258 nm of detection wave, at room temperature, 1 mL.min-1. F. mangiferae sampled in Nanning, Qinzhou and Tianyang, Guangxi province and sampled respectively in January to December were determinated.
RESULTThe average recovery of the RP-HPLC was 99.2%, RSD = 1.05% (n = 5). The content of Mangiferin in F. mangiferae was statistically different due to different sample-regions or sample-time.
CONCLUSIONThis RP-HPLC method is simple, specific and exact. The contents of Mangiferin in the leafs of F. mangiferae sample in Nanning and Tinayang were statistically similar, but higher than that in Qinzhou. The contents of Mangiferin in the leafs of F. mangiferae sampled in July to October were higher than that in the other months. The content in September was the highest, the content in February was the lowest.

7.Changes of Pituitary Magnetic Resonance Imaging and Clinical Manifestations in Short Stature Children with Growth Hormone Deficiency

Objective To study the relationship between the clinical manifestations and changes of pituitary magnetic resonance imaging(MRI) in short stature children with growth hormone deficiency(GHD).Methods The pituitary MRI finding in 38 cases of short stature children diagnosed as GHD(males 23,females 15;5-14 years old,10 children were in pubertas and Tanner Ⅱ-Ⅲ) were analyzed,and the pituitary morphology,size,signal and pituitary stalk's shape and location were observed.SPSS 12.0 soffware was used to analyze the data.Results The forms of pituitery were plaque in 20 children(53%),cupped in 17 children(45%),and carinate in 1 children.In the 22 cases of completely GHD,18 cases had different levels of anterior pituitary dysplasia,abnormal pituitary stalk and/or pituitary signal changes,5 cases without posterior lobe disappeared high signal and 4 cases with pituitary stalk interruption syndrome;the other 4 cases had completely normal pituitary.In the 16 cases of partially GHD,7 cases had varying degrees of pituitary size and/or abnormal pituitary stalk,8 cases had completely normal pituitary,and 1 case had pituitary adenoma.Conclusion Pituitary MRI could assist diagnosis and evaluate pituitary function in short stature children.
8.Progress on the Study of Sex-specific Membrane Proteins in Mammalian Spermatozoa

The expression of specific genes in sex chromosomes is the basis of sex-specific membrane protein in mammalian spermatozoa. The gene expression products are shared among spermatozoa through intercellular bridges, however, the phenomena of male transmission-ratio distortion and sex ratio distortion proved that differential proteins exist between X and Y spermatozoa. In addition, the existence of sex-specific proteins was confirmed by the separation experiment of X/Y chromosome bearing spermatozoa and the detection result of sex specific proteins. At the same time, it was also confirmed that the difference of the sex-specific protein is weak . The advance of separation techniques as well as the integration and optimization among these techniques has made it possible to separate sex-specific membrane proteins in mammalian spermatozoa.
10.Blockage of Th17 cells differentiation exacerbated mouse acute intestine graft-versus-host disease following allogeneic bone marrow transplantation.

OBJECTIVETo study the role of Th17 cells in acute intestine graft-versus-host disease following allogenetic bone marrow transplantation(allo-BMT).
METHODSMice were split randomly into five groups: normal control, irradiated, allo-BMT, allo-BMT + DMSO and allo-BMT + Halofuginone (HF) groups. HF was given intraperitoneally at a dose of 5 µg per mouse from -1 d to 10 d after allo-BMT. aGVHD symptoms were followed-up to perform clinical and pathogenic scores. The levels of Th1/Th17, interleukin-17 and interferon-γ were measured by flow cytometry at day 7 d. mRNA expressions of T-bet, RORγT, CXCL9, CXCL10, CXCL11 and CCL20 in intestine were evaluated by real-time PCR.
RESULTSIntestinal damages in allo-BMT-HF mice was more serious than in normal control and allo-BMT groups at day 14 after transplantation. At day 7, Th17 ratio in allo-BMT + HF group was significantly lower than in allo-BMT group. IL-17A was not detected, but Th1 ratio was higher in allo-BMT + HF. There was a similar increment in the relative expressions of T-bet in both allo-BMT and allo-BMT + HF groups. Expressions of CXCL9 and CXCL10 elevated in allo-BMT + HF group, which were significantly higher than those in allo-BMT group (P < 0.01). CCL20 expression significantly increased in allo-BMT group, but it was not detected in allo-BMT + HF group.
CONCLUSIONBlockage of th17 cells differentiation exacerbated acute intestine graft versus-host disease.
