4.Clinical research of sentinel lymph node imaging for contrast-enhanced ultrasonography in colorectal cancer during operation

Objective To study the feasibility of contrast-enhanced ultrasonography (CEUS) in detecting sentinel lymph node(SLN) in patients with colorectal cancer during operation and their clinical significance.Methods Twenty-one patients who underwent radical resection of a primary colorectal cancer were enrolled.During operation,1 % methylene blue was injected around the tumor,SLN mapping was performed.Then the SonoVue was injected at the same sites,CEUS was performed to observe changes of SLN in real time,and marked them.The SLN detection rate and accuracy of CEUS were compared to those of conventional dye guided method.Results The rectal cancer of below peritoneal reflex,the detection rate of CEUS for SLN was 92.31 %,and the accuracy was 88.89%.The detection rate of dye guided method for SLN was 84.62%,and the accuracy was 93.10%,there was no statistically significant difference between CEUS and dye guided method (P >0.05).Conclusions When injected SonoVue in submucosal layer,the good mapping of SLN of patients with rectal cancer of below peritoneal reflex can be obtained,it will play an important role in clinical application to map SLN of patients with rectal cancer of below peritoneal reflex.
6. Protective effects and mechanism of Naoxintong Capsules on rat brain microvascular endothelial cells (rBMECs) induced by oxygen-glucose deprivation

Objective: To investigate the protective effects and mechanism of Naoxintong Capsules (NXT) on primary cultured neonate rat brain microvascular endothelial cells (rBMECs) induced by oxygen-glucose deprivation. Methods: The primary cultured rBMECs model was established and the identification of rabbit anti-rat VIII factor was carried out. MTT was used to screen out the concentration range of NXT intestinal absorption solution to pretect rBMEC in vitro, three doses were selected for experiment. The experimental groups were divided into control group, model group, nimodipine group (200 μg/mL, NXT intestinal absorption solution group (62.50 mg/L, 125.00 mg/L, and 250.00 mg/L), and NXT intestinal absorption solution (250.00 mg/L) and LY294002 (20 μmol/L, PI3K/Akt pathway inhibitor) co-administration group. The morphology of rBMECs was observed under inverted microscope. The expression of lactate dehydrogenase (LDH) and matrix metalloproteinase 9 (MMP-9) in the cell supernatant was detected by ELISA kit. The apoptosis was observed by Hoechst33342 staining fluorescence microscope. The early apoptotic rate of rBMECs in each group was detected by FCM, and the expression of PI3K/Akt signaling pathway key proteins was detected by Western blotting. Results: Compared with model group, the administration of NXT could significantly improve the morphology of rBMECs, decrease the intracellular levels of LDH and MMP-9, significantly reduce the number of apoptotic cells and early apoptotic rate of rBMECs, and inhibit the expression of p-Akt, Bcl-2 upregulation, decrease the expression of Bax, and inhibit caspase-3 activity. The addition of LY294002, a specific inhibitor of PI3K/Akt signaling pathway, blocked the signal transduction of this pathway and significantly reduced the protective effect of NXT. Conclusion: NXT have protective effects on rBMECs induced by oxygen-glucose deprivation, and its mechanism is related to the PI3K/Akt signaling pathway.
7. Identification of the CYP enzymes mediated the metabolism of dipfluzine hydrochloride

OBJECTIVE: To identify the specific cytochrome P450 (CYP) enzymes involved in the metabolism of dipfluzine hydrochloride (Dip) in the rat liver microsomes. METHODS: The rat liver microsomes were prepared and incubated with Dip. The Dip metabolites (M1, M2, M4 and M5) were identified by LC-MS/MS, and the CYP isoenzymes were identified by the combination of the selective CYP inhibitor study, correlation analysis and a panel of recombinant rat CYP expereiment, respectively. RESULTS: The results from the experiments of selective CYP inhibitors, correlation analysis and recombinant rat CYP isoenzymes indicated that CYP2A1, CYP3A and CYP2C11 contributed to the formation of M1 and M5 in the rat liver microsomes. CYP3A, CYP2A1, CYP1A2, CYP2C11 and CYP2E1 metabolized Dip to M2. CYP3A, CYP2A1, CYP2E1 and CYP2C11 contributed to M4 formation. And the recombinant rat CYP researches further indicated that CYP3A2 exhibited more activity than CYP3A1. CONCLUSION: CYP3A and CYP2A1 are the major CYP isoenzymes responsible for catalyzing Dip to the four metabolites formation in the rat liver microsomes.
8. In vitro anticancer activity of doxorubicin-loaded heparinized magnetic mesoporous silica nanoparticles

OBJECTIVE: To prepare doxorubicin-loased heparinized magnetic mesoporous silica nanoparticles (MMSNs-HP) and investigate their drug release profile and anticancer activity in vitro. METHODS: Amino-modified MMSNs was synthesized by combining phase transfer method with sol-gel method firstly. Then heparin was conjugated with the above nanoparticles via carbodiimide chemistry to form MMSNs-HP. Finally, the following experiments were performed, such as loading/release of doxorubicin into/from MMSNs-HP in vitro, cellular uptake of MMSNs-HP by hepatoma cell HepG2 and cell cytotoxicity of doxorubicin-loaded MMSNs-HP. RESULTS: MMSNs-HP was able to delay the release of doxorubicin significantly, penetrate into tumor cells, kill HepG2 cell, and inhibit the proliferation of HepG2 cells induced by basic fibroblast HepG2 cells (bFGF). CONCLUSION: MMSNs-HP is a potential drug carrier for the delivery of antitumor drugs.
9.Analysis of Target Organ Damage in Youth with Pre-hypertension

Objective To explore target organ damage in the youth with pre-hypertension. Methods 77 consecutive youth visiting our institution form 2008 to 2010 were divided into pre-hypertension group (n=32) and control group (n=45). The relevance of left ventricular mass index (LVMI), left ventricular end-diastolic diameter (LVDd), interventricular septum thickness (IVST), left ventricular posterior wall thickness (LVPWT), carotid plaque intima-media thickness (IMT) and glomerular filtration rate (GFR) were compared. Results The blood pressure of pre-hypertension group was higher than that of control group, and the pre-hypertension group followed higher family hypertension history (P<0.05). There was no difference in LVDd, IVST, LVPWT, carotid plaque IMT and GFR between 2 groups (P>0.05). Compared with the control group, LVMI increased (P<0.05) and the E/A ratio decreased (P<0.05). Conclusion Target organ damage in pre-hypertension youth is salient compared with changes in renal function and damage of arteriosclerosis, and heart damage is more often occurred in the obese youth with pre-hypertension.
