Objective To study multiple-drug resistance expression of CNE1 and CNE2 cell lines induced by radiation and its reversal by cyclosporine A and interferon. Methods Drug-resistant CNE1 and CNE2 cell lines were induced by radiation, and expression of P-gp, MRP, TOPⅡ and GST-? on protein and mRNA level were analyzed by flow cytometry and reverse transcription-polymerase chain reaction methods before and after radiation. At the same time, effect of cyclosporine A and interferon on multiple-drug resistance expression induced by radiation was studied. Results The results showed that expression of P-gp, MRP and GST-? proteins on CNE1 and CNE2 cell lines were up-regulated by radiation, and cyclosporine A and interferon could reverse the up-regulation of P-gp and MRP proteins induced by radiation. Conclusion Radiation can induce multiple-drug resistance in CNE1 and CNE2 cell lines, and it can be reversed by cyclosporine A and interferon.

Journal of Applied Clinical Pediatrics 2004;0(09):-
Objective To explore the preventive and therapeutic effect of recombinant human interleukin-11(rhIL-11) on thrombocytopenia in children with non-lymphocytic leukemia after chemotherapy. Methods Sixteen children who had non-lymphocytic leukemia were divided into 2 groups by randomization, including a therapeutic group and a control group.RhIL-11[50 ?g/(kg?d)] was injected subcutaneously 24 h after chemiotherapy in the therapeutic group, and applied consecutively 10-14 days, and the control group was treated without RhIL-11.Duration of the thrombocytopenia, infusion of blood platelet, diversity of blood platelets counts and adverse effect were observed of the 2 groups.Differences between groups were examined using statistics analysis. Results There were 16 case-times(59.3%) in the therapeutic group that of could be cured without platelet transfusion, but that of control group only had 3 case-times(14.3%);the diffe-rence between 2 groups was significant(P

Objective To investigate the DNA typing, observe relationship between DNA fingerprinting patterns and serotypes of Cryptococcus neoformans, and find a suitable genotyping standard for Cryptococcus neoformans. Methods Three primers, including CN 1(GTG) 5, CN 2(GACA) 4 and CN 3(GATA) 4, were used to distinguish variations among strains of C.neoformans. Results The distinguishable fingerprinting bands for serotype of C.neoformans were yielded by primer CN 2. Using this primer, of 24 clinical and environmental isolates of serotype A of C.neoformans and 8 standard strains investigated by PCR, 20 strains produced complete identical fingerprinting patterns, the other 4 strains had different fingerprinting patterns. 2 strains of serotype B and C yielded indistinguishable fingerprinting patterns. Conclusions ①The majority of strains of serotype A had similar and stable fingerprinting patterns. ②Some strains of serotype B and C had an indistinguishable fingerprinting patterns. ③The same serotype strains from different sources may produce different DNA fingerprinting patterns. ④The DNA fingerprinting is a rapid, simple and feasible method for identifying Cryptococcus neoformans.

Objective To evaluate the viability of Cryptococcus in cerebrospinal fluid of patients with cryptococcal meningitis. Methods Electron microscopy, animal inocul ation and neutral red staining of the cere-brospinal fluid specimens were empl oyed. Results Transmission electron microscopy r evealed intact cells and budding cells of cryptococcus which appeared frequently during the early treatment.Edema of cytoplasm and d isar-rangement of structure of capsule we re often found during the later thera py.All mice inoculated experimenta lly with the cerebrospinal fluid specimens were positive on direct examination b ut negative in routine culture were i nfected.A definite number of deep blood-red fu ngal cells were observed in many spec imens. Conclusion These findings add a new approach for dynamic studying o f the viability of Cryptococcus in cerebrospinal fluid of patients with Crypto-coccal meningitis and provide an imp ortant parameter for evaluating therapeutic effect.

Objective To observe the changes of the outer ce ll wall in resting phase and during sw elling process of conidia of Aspergillus fumigatus, and the distribution of fibronectin receptors on conidia surface. Methods After being harvested with 3.5~4.0?m steel filters, the conidia were resuspended in RPMI 1640+10%FBS medium. The specimens were observed by the scanning electron microscopy after incubation for 0,2,4,6hours a t37℃respectively.The same amount of conidia was resuspendesd in PBS buffer, and put the suspension on 50?g /ml fibronectin coated microtitre plate, after incubation for 3hrs at 37℃,t he conidia were washed three times wi th PBS.Then rabbit antifibronectin antibody and fluorescein isothiocyanate(FITC)conjugated goat anti-rabbit immunoglob-ulin antibody were added, respectiv ely. After digesting with trypsin,t he conidia were washed again with PBS and examined by immuno-confocal microscopy. Results Acharacteristic echinulate pattern of protrusions was shown in the cell wall surface of resting coni dia.During swelling process, the ec hinulate layer of swollen conidia wa s gradually disintegrated, and almost complete ly shed 4hr later. Then,a smooth cell wall layer covered by some strands of fib-rillar material was shown.Binding o f fibronectin to the surface of conid ia was revealed by immuno-confocal m i-croscopy.FITC was homogeneously ad hered to the protrusions on the conid ial outer cell wall layer. Conclusion In resting phase the conidia of A.fumigatus express high levels of fibronectin r eceptors on their surface, they are h o-mogeneously distributed over the ex ternal echinulate layer of the conid ia,and shed gradually during the swe lling process.

Objective To compare the postoperative effect of pars plana vitrectomy, phacoemulsification and intraoeular lens (IOL)implantation with silicon oil removed, phacoemulsification and IOL implantation.Design A retrospective case-control study.Par- ticipants 91 cases(104 eyes)of proliferative diabetic retinopathy with tractive retinal detachment or vitreous hemorrhage were per- formed cataract surgery with IOL implantation. Methods All patients were followed up average 6 months,in whom 52 eyes with pars plana vitrectomy, phacoemulsification and IOL implantation(combined group),and the other 52 eyes with silicon oil removed,pha- coemulsification and IOL implantation(sequential group). The differences of the postoperative visual acuity and inflammatory reactions of the anterior chamber between the two groups were analyzed.Main outcome measures Visual acuity and inflammatory reactions of the anterior chamber. Results There were 14/52 eyes with best corrected visual acuity more than 0.1 in combined group and 8/52 eyes in the sequential group after the surgeries(X~2=-6.87,P=0.07) .Corneal edema happened in 16/52 eyes in combined group and 6/52 eyes in sequential group 1 week after the surgeries(X~2=11.53,P=0.001).Keratic precipitate happened in 20/52 eyes in combined group and 9/52 eyes in sequential group after the surgeries(X~2=5.79,P=0.019) .Conclusions The incidence of postoperative anterior chamber in- flammation is higher in Phacoemulsification combined with pars plana vitrectomy group.
