3.Establishment of euglycemic clamp technique in beagle dogs

Objective To establish a euglycemic clamp technique in beagle dogs. Methods The euglycemic clamp technique was applied in healthy beagle dogs and the blood glucose, insulin, C-peptide, insulin and glucagon were monitored during the clamp. Results The blood glucose was controlled within the basal level.The coefficient of variation was less than 5% during the clamp. The serum insulin concentration finally reached(40.0±3.8)mIU/L stably and a significant inhibition was shown in endogenous insulin by the determination of C-peptide. But there was no significant increase in serum glucagon compared with basal values.Conclusion Methodology confirmed that the euglycemic clamp technique is successful in beagle dogs and can be applied in the study of pharmacodynamics of insulin preparations.
4.The Effect of bFGF on Xeograft of Rat.

The purpose of this study was to assess the effect of basic-fibroblast growth factor (bFGF) on the xenograft for radial bone defect in adult rats. Twenty adult Wistar rats weighing between 200 and 250g were divided into two groups; bFGF group (10 rats, 20 radiuses) and non-bFGF group (10 rats, 20 radiuses). A segmental bone defect measuring 5mm in length was made at both radiuses 10mm proximal to the wrist joint. The defect was filled with xenograft (Lubboc) measuring 3x3x5 mm in size. The xenograft was treated with 400 ng/ml bFGF (recombinant human basic-fibroblast growth factor ) mixed with liquid collagen (Vitrogen 100) in the bFGF group, but with collagen alone in the non- bFGF group. At 6 and 12 weeks after the treatment, the radiuses were harvested and examined histologically and the area of the remained xenograft not replaced by host bone was measured by image analysis system (VIDAS) on mid-sagittal section. The new bone formation and maturation from the differentiation of osteoblast was more remarkable in the bFGF group than in the non-bFGF group. At the time of 6 and 12 weeks after the treatment, the mean of the remained xenograft area was significantly smaller in the bFGF (9.16+/-0.49mm2, 6.20+/-0.30mm2) than in the non-bFGF group (12.16+/-0.59mm2, 10.07+/-0.48mm2) (P<0.001). In comparison with the areas of new bone replacemet area between 6 week and 12 week groups, sigificant and remarkable new bone replacement area could be achieved in the bFGF group than in the non-bFGF group (P<0.05). From these results, it is suggested that bFGF might increase the replacement of xenograft by new host bone and accelerate the healing process for the segmental bone defect of the radius in adult rats.

5.Discussion on Common Problems in Inspecting New Drug Qualification

OBJECTIVE:To strengthen the inspection for qualification of new drugs when introduced in order to guarantee the quality of new drugs and the safety of drug use.METHODS:The problems existing in the inspection for qualification of new drugs were exemplified and classified.RESULTS&CONCLUSION:There existed the problems in the respect of validity,authenticity,effectiveness,and integrality.The good maintainance of the database of SFDA plays a important role in in-specting the new drugs when introduced,while on the other hand such issues as establishment and use of credibility file in medical agencies are worth discussing.
6.Change of Prescription Administrative Policy vs. Its Trial Edition and Its Problems

OBJECTIVE:To facilitate the in-depth implementation of Prescription Administrative Policy("New Method" in short).METHODS:The change of the Prescription Administrative Policy and its problems were analyzed through comparison between the "New Method" and the trial edition.RESULTS & CONCLUSIONS:Improvements of Prescription Administrative Policy such as the introduction of prescription comment system,clarifying supervising subjects and strengthening requirement on prescription dispensing rights etc play a role in tightening comprehensive management on prescriptions and promoting rational drug use.However,the defects of Prescription Administrative Policy such as the absence of the regulations unsuitable for medical orders in retail pharmacy and endemic areas and the unidentified prescription forms need to be improved step by step in the practice.
7. Charge storage stability of electret film made of polypropylene/porous polytetrafiuoroethylene/polypropylene composite

Objective: To study charge storage stability of electret film made of polypropylene (PP)/porous polytetrafiuoroethylene(PTFE)/polypropylene composite. Methods: The PP/porous PTFE/PP composite electret film was prepared with porous PTFE, polyethylene(PE) and PP by heat melting technique. The charge storage stability of the electret film and transportation were evaluated by using the methods of isothermal surface potential attenuation and open-circuit thermally stimulated discharge (TSD) current spectra. Results: (1) The charge storage stability of PP/porous PTFE/PP composite electret was superior to that of PP electret made under ordinary temperature, because the space charge of PP/porous PTFE/PP electret was transferred from deep trap to flat trap during the heat melting process. (2) Satisfactory charge storage stability was observed in PP/porous PTFE/PP electret when under the condition of high humidity. Conclusion: PP/porous PTFE/PP electret film has satisfactory charge storage stability and can be used as a novel enhancer in transdermal delivery of drugs.
8. Synthesis of isavuconazonium sulfate: Research advances

Isavuconazole is a broad-spectrum triazole antifungal agent, which is administered as a water-soluble prodrug-isavuconazonium sulfate. Recently, its oral and intravenous formulations are available. Isavuconazole inhibits fungal cytochrome P45014DM, thus interferes with the biological function of cell wall formation and plays the antifungal role. In this paper, we summarize the synthetic methods of isavuconazole, water-soluble segments of side chain and isavuconazonium sulfate and evaluate their advantages and disadvantages, which lays a foundation for isavuconazonium sulfate technology development.
