Objective To investigate the fluctuation of the iodine deficiency disorders(IDD) through continuous monitoring on it. Methods During 2005-2008, 10 monitoring countis (city,district) of IDD were selected in Jiangxi Province. Five townships were selected in each monitoring county, one school was selected in each townships, and 40 students in 8-10 years old from each school were selected to inspect their thyroid glands and determined the idodine content of salts from their home. Among the 40 students, 20 of which were selected to determine the intelligence quotient(IQ) and 6 of which were selected to determine urine iodine. Thirty pregnant women were also selected to determine the urine iodine in each monitoring county. Twenty students of 5th grade in each school and 5 housewives living nearby the school were selected to carry out the questionnaire survey of health education about IDD. Results Monitoring results showed the average goiter rote of 8-10 years old students was between 2.00% and 4.60% during 2005-2008. The rate of IQ of the students higher than 90 was all above 76% each year. The median of urine iodine of students and pregnant women was higher than 100 μg/L, and the proportion of people whose urine iodine content was lower than 50 μg/L was below 4% each year. The rate of inhabitant taking qualified idodine salt wasn't lower than 95%. The pass rate of the questionnaire survey of health education about IDD was between 11.67% and 43.50% in students and 92.50%-99.60% in housewives. Conclusions The situation of iodine deficiency in Jiangxi Province has been greatly improved, reaching the standard of eliminating IDD.

AIM:The present study was designed to establish H2O2-induced NRK52E cell apoptotic model and to investigate the effects and the mechanism of nmhaFGF on the NRK52E cell apoptosis induced by H2O2. METHODS: In vitro experiment, a apoptotic model of normal rat kidney epithelium (NRK52E) was made by MTT method, Hoechst33342 dyeing and flow cytosorting (FCR). NRK52E cells were cultured with different concentrations of nmhaFGF and haFGF for 24 h before H2O2 was added. The apoptotic rate was detected with FCR method. RESULTS: H2O2 at concentration of 0.4 mmol/L, the optimal condition to establish the apoptotic model, was used to treat NRK52E cells for 18h. Different doses of nmhaFGF (0.01, 0.03, 0.10, 0.30, 1.00 mg/L) reduced the apoptotic rates with the dose rising. However, the decreasing tends of apoptotic rates with dose increasing for haFGF was not so obvious. CONCLUSION: nmhaFGF protects the NRK52E cells against apoptosis. The mechanism might be connected with its non-mitogenic property.

RIG-I-like receptors (RLRs) belong to pattern recognition receptors, which perform significant roles in antiviral responses. RLRs can initiate a cascade of signaling transduction that induces the production of type I interferon and activates the interferon signaling pathway, ultimately resulting in antiviral responses. In the course of evolution, viruses have been constantly counteracting host immune systems to facilitate their own survival and replication, and have developed a set of antagonistic strategies. These mainly comprise elusion, disguise and attack strategies to eliminate the activation of RLRs. In virus-infected cells, RLRs recognize viral RNA and then induce antiviral responses. A better understanding of viral antagonistic strategies against RLRs will provide insights into the development of new antiviral medicines. This mini-review concludes that there are three main antagonistic strategies by which RNA viruses can counteract the activation of the RLRs pathway. It aims to provide references and insights for similar studies on viral antagonism in an array of RNA viruses.

Gentianae Urnulae Herba, dried whole herb of Gentiana urnula,is a commonly used Tibetan medicine. However, only the character identification is used as quality control standard officially at present. As a part of project for the Chinese Pharmacopoeia (2015 edition), the quality standard of this species was established in this study. The tests of water content, total ash, acid-insoluble ash and ethanol-soluble extractives of the crude drugs were carried out following the methods recorded in appendix of Chinese Pharmacopeia (2010 edition, volume 1). The TLC identification method was established by using gentiournoside A as reference substance, and a mixture of ethyl acetate-methanol-water-formic acid(7:1. 5:1: 0. 2) as the developing solvent system on silica gel G TLC plate. The content of gentiournoside A was assayed by HPLC on an Agilent Zorbax SB-C18 (4.6 mm x 250 mm,5 μm) column, using acetonitrile-water (0.1% phosphoric acid) (26:74) as the mobile phase at a flow rate of 1.0 mL x min(-1). The column temperature is at 30 degrees C and the detection wavelength is at 240 nm. As a result, gentiournoside A and the other constituents were separated and presented the same fluorescence light comparing with the reference substance on TLC detected under the UV light(366 nm). The methodology validation for the assay of gentiournoside A showed that it was in a good linear correlation in the range of 0.009 95-0.398 g x L(-1) with the regression equation of Y = 1 467.1X +41.407(r = 0.999 9), and the average recovery was 98. 3% (RSD 2.2%). The mass fractions of gentiournoside A, water content, ethanol-soluble extractives of 15 batches samples were varied in the ranges of 0.175% -1.83%, 8.60% - 9.93% and 29.2% - 35.2%, respectively. Total ash and acid-insoluble ash were 10.2% - 17.2% and 5.26% - 10.8% detected from 10 batches samples. The recommended standards of quantitative indexes are that the mass fractions of gentiournoside A and extractives are not less than 0.80% and 26.0%, respectively; the water, total ash and acid-insoluble ash are not more than 12.0%, 15.0% and 8.0%, respectively.

Red in vivo recombination is a new kind of genetic engineering technique based on homologous recombination. In this work, plasmid pKD46 which expresses Red recombination proteins is transferred into Escherichia coli strain DH5?.The kanamycine resistant gene is generated by PCR by using primers with homology to hisDCB gene of E.coli chromosome. Thus, the hisDCB gene was replaced with kanamycine resistant gene by the plasmid recombination system, then the resistant gene was eliminated by a helper plasmid encoding the FLP recombinase. At last, a E.coli histidine auxotroph which is sensitive to kanamycine was got. The results indicate that Red in vivo recombination is a convenient method to construct auxotrophs.

Objective To understand the situation of iodized salt consumption at the household level and non-iodized salt distribution in those areas with low iodized salt coverage.Methods In 2010,iodized salt was monitored in 31 provinces and Xinjiang Production and Construction Corps in accordance with the Monitoring Program of the National Iodine Deficiency Disorders (Trial) (hereinafter referred to as the Program) requirements.Under the jurisdiction of counties (cities,districts,banners) with more than 9 townships (towns,street offices),based on the location of east,west,south,north and center,9 townships (town,district offices) were selected using simple random sampling method; 4 administrative villages (neighborhoods) were selected in each township (town,district office); and 8 residents in each administrative village (neighborhood) were selected.Under the jurisdiction of counties (cities,districts,banners) with less than 9 townships (towns,street offices),based on the location of east,west,south,north and center,1 township(town,district office) was selected using simple random sampling method; 4 administrative villages(neighborhoods) were selected in each township(town,district office);and 15 residents in each administrative village(neighborhood) were selected.Iodized salt coverage rate,qualification rate of iodized salt and consumption rate of qualified iodized salt were calculated in various provinces.The salt samples were tested by semi-quantitative method on the spot and then tested with quantitative method in laboratories.The standard of qualified iodized salt was set as 20-50 mg/kg and that of non-iodized salt was set as ＜ 5 mg/kg (GB/T 13025.7-1999).Results In 2010,a total of 2862 counties(districts,cities and banners) and 14 divisions of Xinjiang Production and Construction Corps,reported the monitoring results,and the monitoring coverage rate was 99.79％(2876/2882).A total of 826 696 copies of edible salt samples were tested,the coverage rate of iodized salt was 98.63％,the consumption rate of qualified iodized salt was 97.95％,and the coverage rate of qualified iodized salt was 96.63％.At province level,only in Tibet iodized salt coverage rate was ＜ 90％.At county level,2755 counties qualified iodized salt coverage rate was ≥90％,and 33 counties iodized salt coverage rate was ＜ 80％.The counties with qualified iodized salt coverage rate of 90％ or more accounted for 96.63％(2785/2882) of the total counties.Conclusions The counties where non-iodized salt coverage is higher than 20％ mainly distributed in the western or coastal areas and adjacent areas with higher iodine.These areas need policy and funding support from governments at all levels to reducc the gap between these areas and other areas.

OBJECTIVETo investigate the effects of the quinoline derivative PQ1 combined with cisplatin on the proliferation and gap junction communication of prostate cancer PC3 cells.

METHODSWe cultured in vitro prostate cancer PC3 cells and divided them into DMSO blank control, cisplatin control, and cisplatin (10 mg/ml) plus PQ1 (1, 2, 5, 10, and 15 μmol/L) groups. We measured the proliferation of the prostate cancer PC3 cells, determined the expressions of the connexin 43 (Cx43) mRNA and protein by RT-PCR and Western blot, and compared the indexes among different groups.

RESULTSCisplatin combined with PQl at 1 - 10 μmol/L significantly inhibited the proliferation of the PC3 cells and the inhibition rate rose in a concentration- and time-dependent manner, from (48.72 ± 0.98)% vs (50.33 ± 0.62)% at 0 μmol/L to (77.38 ± 1.12)% vs (83.50 ± 1.05)% at 15 μmol/L at 24 and 48 hours (P < 0.05). Compared with the cisplatin control, cisplatin combined with PQ1 at 1, 2, 5, 10, and 15 μmol/L increased the expression of Cx43 mRNA from 0.379 ± 0.113 to 0.669 ± 0.031, 0.831 ± 0. 127, 0.769 ± 0.100, 0.532 ± 0.086, and 0.475 ± 0.134, respectively (P < 0.05), and cisplatin combined with PQ1 at 1, 2, 5, and 10 μmol/L elevated that of Cx43 protein from 0.138 ± 0.146 to 0.263 ± 0.111, 0.306 ± 0.152, 0.415 ± 0.280, and 0.643 ± 0.310, respectively (P < 0.05).

CONCLUSIONThe quinoline derivative PQ1 can promote the gap junction communication of prostate cancer PC3 cells and enhance the killing effect of cisplatin on PC3 cells by upregulating the expressions of Cx43 mRNA and protein.

The tests of water content, total ash and ethanol-soluble extractives of the crude drugs were carried out based on the methods recorded in appendix of Chinese Pharmacopeia (2010 edition, volume 1). The TLC method was established by using reference drug and gentiournoside A as reference substance, and a mixture of ethyl acetate-methanol-water-formic acid (7: 1.5: 1: 0.2) as the developing solvent system on silica gel G TLC plate. The content of gentiournoside A was assayed by HPLC on a Ultimate XB-C18 (4.6 mm x 250 mm, 5 μm) column, using methanol-water (0.02% phosphoric acid) (52:48) as the mobile phase at a flow rate of 1.0 mL x min(-1). The column temperature is 25 degrees C and the detection wavelength is at 240 nm. As a result, gentiournoside A and the other constituents were separated and presented the same fluorescence light comparing with the reference substance on TLC detected under the UV light(366 nm). The methodology validation for the assay of gentiournoside A showed that it was in a good linear correlation in the range of 10.01-400.32 mg x L(-1) with the regression equation of Y = 1 539.5X - 33.339 (r = 0.999 7), and the average recovery was 99.68% (RSD 1.92%). The mass fractions of gentiournoside A, water content, ethanol-soluble extractives of 19 batches samples were varied in the ranges of 14.48-31.51 mg x g(-1), 11.25% -12.74% and 24.21% - 31.60%, respectively, and total ash was 4.64% - 6.12% detected from 10 batches samples. The recommended standards of quantitative indexes are that the mass fractions of gentiournoside A and extractives are not less than 15.0 mg x g(-1) (1.5%) and 21.0%, respectively; the water and total ash are not more than 13.0% and 6.0%, respectively.
