2.Factors affecting the prognosis of ruptured intracranial aneurysm

Objective To investigate factors related to the prognosis of patients with ruptured intracranial aneurysm.Methods Clinical data of 165 patients with ruptured intracranial aneurysm treated with surgery were retrospectively analyzed.The age,gender,PAASH scale on admission,smoking,hypertension,hyperlipemia,cerebral vasospasm,timing of surgery and mode of surgery,etc.were analyzed by the simple and multiple Logistic regression models.Results Simple Logistic regression analysis indicated that age,PAASH scale on admission,smoking,hypertension,timing of surgery and mode of surgery were influencing factors for the prognosis of patients with ruptured intracranial aneurysm(all P<0.05).Multiple Logistic regression analysis showed that age,PAASH scale on admission(OR=2.542),smoking(OR=5.772),hypertension(OR=7.150),timing of surgery(OR =2.908) and mode of surgery(OR =0.161) were independent risk factors for the prognosis of patients with ruptured intracranial aneurysm(all P<0.05).Conclusions Accuratel and systemsatic assessment of patient conditions and the optimal timing and mode of surgery are the key to improving the prognosis of patients with ruptured intracranial aneurysm.
3. Mechanism of inhibitory effect of miRNA-33a on proliferation of HCT-116 cells

Objective To explore the mechanism of chemically synthesized miRNA-33a inhibiting proliferation activity of colon cancer cells HCT-116. MethodsHCT-116 cells were transfected with miRNA-33a mimics, miRNA-33a inhibitor and miRNA-33a negative control (NC). The contents of miRNA-33a in the transfected cells and changes of Twist protein expression were detected by real-time PCR and Western blotting respectively 48 hours after transfection; the changes in cell proliferation activity were measured using CCK-8 method 72 hours after transfection. Bioinformatic software was used to predict the binding site of miRNA-33a with Twist, and a luciferase reporter method was used to verify whether there is the target site of miRNA-33a in Twist gene. Results Transfection of HCT-116 cells with miRNA-33a mimics significantly increased miRNA-33a mRNA and decreased Twist protein expression (P<0.05), but the transfection with miRNA-33a inhibitor significantly decreased miRNA-33a mRNA and increased Twist protein expression (P<0.05); moreover, miRNA-33a mimics significantly inhibited proliferation activity of HCT-116 cells and miRNA-33a inhibitor significantly enhanced cell proliferation activity 48 and 72h after transfection compared with the control group (P<0.05). The binding site of miRNA-33a in targeting gene Twist was successfully predicted and verified. Conclusion miRNA-33a inhibits the proliferation activity in HCT-116 cells by silencing the expression of its target gene Twist.

5.Approaches for developing a system model for health evaluation based on body constitutions of traditional Chinese medicine and order parameters.

From the point of view of systems science, human body can be considered as a complex system, and the human health system is a subsystem of it. Systems science conducts investigation in a holistic manner. As a theoretical method, it deals with the operation and evolution of systems from the macroscopic perspective, so this theory is similar to phenomenological theory of traditional Chinese medicine (TCM) in methodology. Naturally, numerous theories of systems science can be used in research of the human health systems of TCM. In this paper, the authors introduced synergetics, a theory of modern systems science, and its slaving principle, and in particular, analyzed the concept of order parameters related to the slaving principle and the relationship between body constitutions of TCM and order parameters. The body constitution of TCM can be treated as a slow variable in the human health systems. By using synergetics, the authors established a model of the human health system based on body constitutions of TCM. As an application of the model, the authors illustrated the argumentations in the theory of constitution being separable, the theory of a relationship between constitution and disease, and the theory of a recuperable constitution. To some extent, this work has made links between the TCM theory of body constitution and modern systems science, and it will offer a new thought for modeling the human health system.

10.Melatonin combined with compound betamethasone for articular cartilage lesions in rats with knee osteoarthritis

BACKGROUND: Due to lack of the distribution of vessels and nerve, self-repairing capability of articular cartilage tissue is poor after inflammatory erosion.OBJECTIVE: To evaluate the effects of melatonin combined with compound betamethasone on the articular cartilage of osteoarthritis (OA) in rats.METHODS: Thirty Sprague-Dawley rats received intra-articular injection of papain solution for establishing knee OA models.Meanwhile, 20 of them underwent constant intensive light condition for establishing pinealectomy models. Ten rats that under pinealectomy were administered melatonin combined with compound betamethasone. Another 10 normal control rats receiving no treatment served as controls. After 4 weeks of treatment, serum melatonin concentrations at 2 a.m. (highest melatonin concentration within circadian rhythms) and 2 p.m. (lowest melatonin concentration within circadian rhythms) were detected by ELISA. At the same time, all rats were sacrificed to collect femoral condyle cartilage for gross observation.After decalcification and toluidine blue staining, articular cartilage lesions were evaluated based on Mankin scores.RESULTS AND CONCLUSION: After OA model was created, cartilage surface was uneven, lost their luster, the chondrocytes were poorly arranged, severe loss of staining was observed, serum level of melatonin was decreased, and circadian change was unobvious. Constant intensive light condition further aggravated cartilage damage. After treatment by melatonin combined with compound betamethasone, softened cartilage disappeared, there were more regular chondrocytes arrangement, and dispersed chondrocytes and loss of staining were gradually decreased. In addition, there was significant difference in Mankin scores of toludine blue staining among groups (P < 0.05). These findings indicate that melatonin combined with compound betamethasone can restrain the progression of cartilage damage.
