Multiple myeloma is a malignancy of plasma cells,which is still incurable now.Despite dramatic improvement of the response rate and overall survival have been made due to the application of target therapy and stem cell transplantation,the treatment effect is difficult to maintain,nearly all patients will eventually relapse and become refractory.At relapse,the challenge is to select the appropriate treatment for each patient.The decision will be both disease-dependent and patient-dependent.

Microdialysis probes were implanted into the rabbit cerebral cortex and caudate nucleus followed by perfusion with Ringer solution at a flow rate of 3.0?l/min. 4.The clinical research of the effect of live combined bacillus subtilis and enterococcus faecium granules with multivitamines and nonnutritive suck on the premature infant

Objective To study the effect of live combined bacillus subtilis and enterococcus faecium granules with multivitamines (Medilac-Vita) and nonnutritive suck on the premature infant.Methods Eighty premature infants without asphyxia and digestive tract malformation were randomly divided into control group(without Medilac-Vita and nonnutritive suck group) and observation group( Medilac-Vita and nonnutritive suck group) including 40 cases each.The gastrointestinal dysfunction and growth-development index were observed in two groups.Results In observation group,the occurrence rate of vomit,abdominal distention,gastric retention,hemorrhage of digestive tract,meconium peritonitis and necrotizing enterocolitis were all lower than that in control group.The cleaning lime of meeoninm in observation group was lower than that in control group [(3.20±1.01 )d and (6.23±2.56)d],the removing time of canalis ventriculi [(4.03±2.11 )d and (7.10±2.91 )d],the time of the jaundice fading away [( 11.10±5.36)d and ( 14.30±6.82)d ],the time of staying in hospital [(15.02±3.12 )d and (20.00 + 5.11)d],the growth of the head circumference and weight in seven and fourteen days were significantly different between two groups.The incidence rates of intracranial hemorrhage,apnea and death in observation group were 12.5% (5/40),10.0% (4/40) and 0 (0/40),which in control group were 45.0%(18/40),37.5% (15/40) and 5.0% (2/40).Conclusion The treatment of Medilac-Vita and nonnutritive suck on the premature infants can promote the maturity of stomach and intestine function,reduce the occurrence of complications,promote growth and development.
5.The relationship between serum osteopontin and disease activity in rheumatoid arthritis and the role in interstitial pulmonary fibrosis

ObjectiveTo investigate the association between serum level of osteopotin(OPN) and disease activity of rheumatoid arthritis (RA) patients, and explore the importance of OPN in the pathogenesis of interstitial lung disease (ILD) in RA. MethodsSixty-five RA patients and 20 healthy controls were pros-pectively enrolled. RA patients were divided into active group(n=43) and inactive group(n=22), and ILD groups (n=24) and non-ILD group (n=41). Enzyme-linked immunosorbent assay (ELISA) was used to detect the serum levels of OPN in patients with RA and healthy controls, and the relationship between OPN and other clinical and laboratory findings were analyzed.Results① Serum OPN tended to be significantly higher in RA patients (median, 18.0 ng/ml) than in the healthy controls (median, 14.3 ng/ml), P<0.01; ②The serum level of OPN in RA patients showed a significant positive correlation with the course of disease, numbers of tender joints , ESR and CRP, but no positive relationship was found in number of swollen joints; ③ The serum level of OPN was significantly higher in RA-ILD patients(median, 20.0 ng/ml) than that in non-lLD (median, 17.0 ng/ml, P<0.05). And there was remarkable negative correlation between the concentration of serum OPN and the value of PaO2, but no association was found with pulmonary function %VC and %DLCO. ④ Compared with the non-ILD group, the ILD group had more active disease in terms of tender joint counts and swollen joint counts, ESR, CRP(P<0.01) and the serum titer of RF-IgM,(P<0.05). ConclusionOPN plays a role in the pathogenesis of RA and is related to the disease activity. It may serve as an active disease inflammatory marker of RA . OPN may be involved in the pathogenesis of RA related ILD and is associated with the severity of pulmonary damage.

To investigate the effects of antisense oligonucleotides directed at TNF-? on the intracranial pressure following craniocerebral explosive injury. Eighteen dogs were randomly divided into artificial cerebrospinal fluid (ACSF) group, sense group and antisense group. 8.Effects of Arsenic Trioxide on Mouse Cultured Melanoma Cloudman S91 Cells

Objectives To observe the effects of arsenic trioxide (As2O3) on the cell cycle, differentiation and proliferation of cultured mouse melanoma Cloudman S91 cells. Methods The cell viability was measured by MTT reduction assay. The morphological changes of the cells were examined by Wright-Giemsa staining. Clone forming was carried out to test the proliferation ability. Apoptosis rate and cell cycle of S91 cells were measured by flow cytometry. Results As2O3 in the concentrations of 0.031~0.25 ?mol/L promoted cell growth markedly and blocked the transformation of cells from G0 to G1. As2O3 in the concentrations of 0.5~8.0 ?mol/L inhibited the cell growth and induced apoptosis markedly. Conclusions As2O3 has marked effects on mouse melanoma S91 cells, which include inducing apoptosis and promoting differentiation and proliferation. These effects are dose- and time-dependent.
9.The expression of bone morphogenetic protein 4 and its effects in ischemia-anoxemia induced white matter injury model

Objective To investigate the ultrastructural alteration in brain tissues as well as the expression of bone morphogenetic protein(BMP) 4 and its effects on regulating myelination in the process of white matter injury development.Methods A total of 152 Sprague-Dawley newborn rats(3 days old) were randomly divided into white matter injury group(n=76) or control group(n=76).The white matter injury model was established by ligation of the right common carotid artery and hypoxic exposure(8% O2 and 92%N2),and samples were collected at 3d,7d,14d and 21d after operation.Morphological changes of the brain tissues were observed under a light microscope,while myelination was analyzed using a transmission electron microscope.The expression and location of BMP4 and myelin basic protein(MBP),a marker for myelination,was detected by immunohistochemistry staining,expression levels of BMP4 and MBP proteins were analyzed by Western blotting,and BMP4 mRNA expression was measured by real-time PCR.Results Observed under the light microscope,the cellular structure was clear,fibers arranged closely and orderly in the white matter of the control group.Whereas in the white matter injury group,sparse cells,loose mesh shaped white matter,and disorderly oriented fibers were observed.In the control group,myelin sheath had regular morphology,uniform density,and same thickness,observed using the transmission electron microscope.While in the white matter injury group,the myelin sheath was loosened,thinned,lamellar separated,and boundary obscured.Using immunohistochemistry staining,Western blot,and real-time PCR analyses,it was found that the protein and mRNA expression of BMP4 had no significant change with the increase of age in the control group,while it was rapidly increased with the extending of ischemic time in the white matter injury group.Comparing with the control group,the expression of BMP4 was significantly increased since 3d after operation in the white matter injury group(P<0.05),and the difference between two groups became more significant with the extending of ischemic time.The expression of MBP protein was analyzed by immunohistochemistry staining and Western blot,and a gradual increase was found in both groups with the increase of age.However,the expression of MBP protein was significantly decreased on 14d and 21d after operation in the white matter injury group compared with the control group(P<0.05).Conclusion Myelination disorders exists in white matter injury induced by ischemia-anoxemia.Meanwhile,the expression of BMP4 is significantly increased in the white matter injury group,indicating a possibility that BMP4 involves in the regulation of myelination disorders in white matter injury.
10.Fatigue rupture study of HL-3 Buccal Multiloops for simulation of oral movements

BACKGROUND:Concerning the problem of the ruptured phenomenon in clinic work of HL-1 and HL-2 Buccal Multiloops removable appliance, we invented HL-3 Buccal Multiloops.
OBJECTIVE:Through the Buccal Multiloops Fatigue Testing Machine which can imitate oral movements, to compare the fatigue strength of the HL-2 and HL-3 Buccal Multiloops in the same condition.
METHODS:A pair of the HL-2 or HL-3 Buccal Multiloops with the completely symmetrical size, symmetrical height and symmetrical direction was fixed on the Buccal Multiloops Fatigue Testing Machine which could simulate oral movements, then keep the Buccal Multiloops Fatigue Testing Machine working until any Buccal Multiloops broke. The cyclic loading times of this pair Buccal Multiloops was recorded. Scanning electron microscope was used to observe the appearance of broken end of the Buccal Multiloops.
RESULTS AND CONCLUSION:The cyclic loading times of HL-3 Buccal Multiloops were more than HL-2 Buccal Multiloops (P < 0.01), suggesting that HL-3 Buccal Multiloops has stronger fatigue strength than HL-2 Buccal Multiloops. Scanning electron microscope observation showed that the fractures of HL-2 and HL-3 Buccal Multiloops al had the typical fatigue fracture morphology, indicating that the rupture of HL-2 and HL-3 Buccal Multiloops belongs to fatigue fracture. However, HL-3 Buccal Multiloops exhibited bigger crack compared with HL-2 Buccal Multiloops, the presence of running-in region, suggesting that HL-3 Buccal Multiloops reduced the probability of fatigue fracture and prolonged the time of fatigue fracture.
