2.Development and verification of whole column imaging detection-capillary isoelectric focusing method for determination of isoelectric point of pertactin

Objective To develop and verify a whole column imaging detection-capillary isoelectric focusing(WCID-CIEF)method for the determination of isoelectric point(pI)of pertactin(PRN).Methods The WCID-CIEF method for the determination of PRN antigen was developed by optimizing the parameters such as the focusing time and final concentration of samples in the WCID-CIEF process,and verified for the specificity,accuracy,repeatability,intermediate precision,durability and inter-batch consistency.Results The optimal focusing time of WCID-CIEF for the determination of PRN antigen pI was 1 min at 1 500 V and 3 min at 3 000 V. The optimal final concentration of PRN antigen was 300 μg/mL. The PRN antigen pI was about 6. 035,and the blank matrix showed no interference peak in the position of each peak of antigen. The method had good specificity,accuracy,repeatability,intermediate precision,durability and consistency among batches.Conclusion The developed WCID-CIEF method is suitable for the pI detection and charge heterogeneity analysis of PRN antigen,which can provide basis for the characterization of PRN antigen and reference for the quality control in the process of development and production of related vaccines.
3.Surgical treatment of secondary hyperparathyroidism in patients with chronic renal failure

With the development of the dialysis technology,the survival time of patients with chronic renal failure is prolonged,while more complications which affect the quality of life or even threat the life of patients are followed.Secondary hyperparathyroidism(SHPT)with a certain incidence and mortality is one of the common complications,and its main characteristic performances ale hyperphosphatemia,hypocalcemia and high level of parathyroid hormone.Most patients Can be cured by the treatment of medicine while surgical treatment is still required by some cases in which the disorder of the parathyroid function can not be well regulated through medicine treatment.The purpose of this paper is to make a review of the recent studies of surgical treatment in patients with SHPT.
4.Effects of intensive insulin therapy on the level of inflammatory factors and prognosis in severe multiple trauma patients

Objective To observe the influence of intensive insulin therapy on inflammatory fac-tors and prognosis in severe multiple trauma patients.Methods A total of 53 cases of severe multiple trauma were randomly divided into the treatment group(n =27)and the control group(n =26).Besides basic treatment,patients in the treatment group received additional intensive insulin therapy by micro-pump.The level of blood glucose in the control group was controlled under 11.1 mmol/L.Levels of TNF-α,IL-1β,IL-6,and CRP were tested before and after treatment.Multiple organ dysfunction syndrome,noso-comial infection rate,and mortality rate were also observed.Results The levels of TNF-α,IL-1β,IL-6, and CRP in the treatment group were significantly lower than those of the control group(P <0.05 or P <0.01).The incidence of multiple organ dysfunction syndrome,nosocomial infection,and mortality rate in the treatment group was lower(P <0.05).Conclusion Intensive insulin therapy can effectively decrease the expressions of inflammatory factors in patients with severe multiple trauma,improve the prognosis,re-duce the incidence of nosocomial infection and mortality.
5.Effects of repetitive transcranial magnetic stimulation on post-stroke depression, sleep quality and activities of daily living

Objective To explore the effects of repetitive transcraninal magnetic stimulation (rTMS) on depression,sleep disorder and activities of daily life (ADL) in post-stroke depression (PSD) patients.Methods A total of 90 PSD patients were divided into a control group,a treatment group and a combined treatment group according to a random number table,30 cases in each group.All patients were given basic treatment and comprehensive rehabilitation training.Moreover,the control group was additionally received escitalopram treatment,the treatment group rTMS treatment,the combined treatment group the above both,lasting 8 weeks.Before and after the treatment,the Hamilton depression (HAMD) scale,the Pittsburgh sleep quality index (PQSI),the Chinese stroke scale (CSS) and the modified Barthel index (MBI) were employed to evaluate depression,quality of sleep,neurological impairment and the activities of daily living (ADL) of patients in three groups.Results Before treatment,there was no significant difference in the HAMD,CSS,PSQI and MBI score among the 3 groups.After 8 weeks' treatment,the HAMD,PSQI and CSS scores of three groups had significantly decreased,but their MBI scores had significantly increased.Compared with the control group,no significant differences were observed in HAMD,CSS,PSQI and MBI scores of the treatment group and the combined treatment group (P > 0.05).After treatment,there was no significant difference in the above four values between the combined treatment group,and the treatment group.Conclusion The rTMS can significantly improve PSD patient's depressive symptoms,relieve sleep disorders and neurological deficit situation,and effectively improve their ADL.
6.Influence of neurtophil elastase on sperm quality in male infertile patients

Objective To explore the relationship between sperm quality and neurtophil elastase (NE) in male infertile patients . Methods the NE concentration was detected in1 30 male infertile patients ,the research subjects were classified into 3 groups based on the neurtophil elastase concentration :neurtophil elastase concentration more than 1 000 ng/mL (group A ) ,250-1 000 ng/mL (group B) less than 250 ng/mL (group C) .Then sperm basic parameters and biochemical markers (phosphatase ,citrate ,zinc and fructose) were detected .Results (1) The sperm density and motility showed statistically significant differences among the group A ,B and C(P<0 .05)moreover neutrophil elastase concentration was negatively correlated with sperm density (r= -0 .43) and sperm motility(r= -0 .37) .Sperm volume and morphology showed no statistically significant differences among three groups (P>0 .05) .(2) There were statistically significant differences in the levels of phosphatase ,citrate and zinc among three groups (P<0 .05) ,moreover the neutrophil elastase concentration had significantly negative correlation with the concentrations of phosphatase , citrate and zinc(r= -0 .19 ,r= -0 .17 ,r= -0 .23) ,while the fructose concentration had no statistical difference among three groups(P>0 .05) .Conclusion The NE level in semen has significant reverse correlation with sperm density ,sperm motility and bi‐ochemical markers (phosphatase ,citrate and zinc) ,implying that semen NE has significantly negative effect on the sperm quality , thus affecting male fertility ability .
7.Progress of mitochondrial aldehyde dehydrogenase in cardioprotection

Mitochondrial aldehyde dehydrogenase(ALDH2),one of the isoforms of aldehyde dehydrogenase,has multiple enzymatic functions including the activity of dehydrogenase and esterase.The metabolisms of ethanol,amino acids,biogenic amine,vitamin or steroid in the body produce various substances of aldehyde.With the help of co-factor NAD(P)+,ALDH2 can convert aldehydes into corresponding carboxylic acid,which plays a key role in reducing toxic effects of aldehydes on the body.It does not need co-factor when ALDH2 works as esterase.It can convert carboxylic ester or other acids into corresponding carboxylic acids or alcohols.Recently,it has been shown that the decrease of ALDH2 activity exacerbates multiple factors(such as ethanol,ischemia)-induced myocardial injury and accelerates the development of nitroglycerin tolerance.Therefore,the development of specific agonists of ALDH2 may provide a novel approach to the therapy and prevention of heart diseases.
8.Protective effect of extract from Shuilu Erxiandan on oxidative injury of structure and function of human sperm membrane

Objective To explore the mechanism of Shuilu erxiandan against male′s infertility and asthenoospermia .Methods Human sperm with normal physiological function were collected ,and then sperm suspension were divided into the normal group ,the model group ,Vitamin C group and the groups of large ,medium and small dose of Shuilu erxiandan .The reactive of species was made from hypoxanthine-xanthine oxidase .In the oxygen environment ,Vitamin C(0 .25 g/L) and different content (1 .0 ,0 .5 ,0 .25 g/mL) of extract from Shuilu erxiandan were hatched with sperm .Then the function of sperm membrane in every group was evalu-ated using hypoosmoticswelling .Meanwhile the content of MDA and SOD were detected .Results Expansion of sperm and sperm motility in all the groups of vitamin C and large ,medium and small dose of Shuilu erxiandan′s extract were significantly higher than that in the model group at time point of 1 ,3 and 6 h (P< 0 .05) .Also ,medium dose of Shuilu erxiandan (0 .5 g/mL) displayed more effective on expansion of sperm and sperm motility compared with that in the vitamin group C (P< 0 .05) .All the groups of vitamin C and large ,medium and small dose of Shuilu erxiandan′s extract significantly indicated descreased MDA content and improved SOD activity compared with that in the model group (P< 0 .05) .Also ,this effect of medium dose of Shuilu erxiandan (0 .5 g/mL ) on MDA and SOD activity were superior to that in vitamin C group ,which showed significant difference (P< 0 .05) .Conclusion Ex-tract from Shuilu erxiandan could significantly protect structure and function of sperm membrane ,which is correlated with interve-ning its lipin peroxidation .
9.Dynamic changes of calcium ions in retinal ganglion cells exposed to hypoxia and their relationship with TNF-? and lipid peroxidation

Objective To investigate the changes of retinal ganglion cells (RGCs) exposed to hypoxia and the mechanism. Methods RGCs were isolated from the retina of neonatal Long Evans rats aged 1 day and cultured, then divided into normal control group and hypoxia group (cultured in incubator containing 1 O_2, 5 CO_2 and 94 N_2). At 1, 3, 12 and 24 h after incubation, the calcium ion level by laser scanning confocal fluorescence microimaging system, the activity of SOD and the content of MDA by biochemistry technology, TNF-? by ELISA were detected. 10.Clinical analysis of phacoemulsification on patients with cardiovascular disease

Objective To investigate the changes of systolic pressure,diastolic pressure,oculocardiac reflex(OCR) and vision of patients suffering from cardiovascular disease when they applied phacoemulsification.Methods Totally 168 cases(182 eyes) who had underwent phacoemulsification were analyzed in random selection.Of all the cases,97(106 eyes) were combined with cardiovascular disease,the others were without any general disease.The changes of blood pressure,OCR,ECG and vision were observed in two groups.Results Compared with preoperative period,the mean systolic blood pressure increased significantly during operation,but the differences of diastolic pressure,OCR,ECG and vision were not significant between the two groups.Conclusion The safety and reliability of the surgery depend on reasonable medication,careful surgical procedure and monitoring for the patients with cardiovascular disease in the peri-operation period.
