The chromosomal relationships of Simulium arakawae, a black fly of medicalveterinary
importance, are resolved relative to other members of the Simulium venustum
species group and to the standard reference map for the subgenus Simulium. Simulium
arakawae differs from the subgeneric standard by eight fixed inversions, apparently none of
which are shared with any of the other 17 chromosomally studied nominal members of the
group. The chromosomal features that indicate group membership might have been lost in S.
arakawae, particularly if they were polymorphic in an ancestor.
2.Rosiglitazone and all-trans retinoic acid inhibit human myeloma cell proliferation via apoptosis signaling pathway modulation.

OBJECTIVETo investigate the effects of artificial ligand of peroxisome proliferators activated receptors (PPARs), rosiglitazone (RGZ) and all trans-retinoic acid (ATRA) on human myeloma cell line growth in vitro and in vivo.
METHODSU266 and RPMI 8226 cells were treated with different concentration of RGZ in the presence or absence of ATRA and the results were studied by 3H-TdR thymidine incorporation (for cells proliferation), Annexin V-PI staining and caspase-3 activity assay (for cells apoptosis), RT-PCR (for FLIP, XIAP and survivin mRNA expression), and tumor formation test in BALB/c nude mice.
RESULTSExposure to RGZ induced proliferation inhibition in a dose-dependent manner in both U266 (r = 0.991, P < 0.01) and RPMI 8226 cells (r = 0.961, P < 0.01). A combination of RGZ with ATRA could enhance the inhibition effect (P < 0.001 in U266, P < 0.01 in RPMI8226). 10 micromol/L of RGZ induced apoptosis of (9.8 +/- 1.7)% in U266 cells and (10.7 +/- 3.3)% in RPMI8226 cells, in a time and dose dependent manner and combined with ATRA intensified the apoptosis induction effects (P < 0.01 in both cell lines). The FLIP, XIAP and survivin mRNAs were expressed in both cell lines and their levels decreased significantly after cultured with RGZ. The addition of RGZ + ATRA in the culture further decreased the levels. Caspase-3 activity increased substantially with the increase of RGZ concentration and the addition of RGZ + ATRA in the culture medium showed similar synergism effect on caspase-3 activation (P < 0.01). The xenograft of U266 cells in BALB/c nude mice were inhibited by RGZ and so did more by the combination of RGZ and ATRA (P < 0.01).
CONCLUSIONThe down-regulation of FLIP, XIAP and Survivin induced by RGZ can activate caspase-3, whereby induced apoptosis and proliferation inhibition in myeloma cells. ATRA can enhance these effects of RGZ.

3.Investigation and Study on Present Situation of Clinical Pharmaceutical Care in 39 Medical Institutions in Chong- qing

ABSTRACT OBJECTIVE:To investigate the present situation of clinical pharmacy in different levels of medical institutions in Chongqing,and to investigate the method for optimizing clinical pharmaceutical care. METHODS:By questionnaire survey,50 sample medical institutions(including 15 third level hospitals,25 second level hospitals and 10 first level hospitals)in Chongqing were selected according to the region,the level of medical institutions,comprehensive and specialized hospitals. The team of clinical pharmacists and the development of clinical pharmacy were investigated,and the results were statistically analyzed to find common problems and put forward relevant suggestions. RESULTS & CONCLUSIONS:Totally 45 questionnaires were collected, with a recovery rate of 90%,of which 39 questionnaires were qualified,with a qualified rate of 86.67%. There were 32 hospitals (82.05%)in 39 medical institutions with clinical pharmacy departments and 4 hospitals(10.26%)with clinical pharmacist training base. There were 239 full-time pharmacists in 39 medical institutions,with an average of 6.13 persons in each medical institution, including 4.5 full-time clinical pharmacists,and 2-5 full-time clinical pharmacists(25 hospitals)in most medical institutions. Top 5 clinical departments that clinical pharmacists mainly participated in were respiratory department,cardiovascular department, endocrinology department,ICU and oncology department. Most of the clinical pharmacists(16 hospitals,42.11%)worked in the clinic for more than 50% to no more than 80%. The number of medical institutions(28-37,>70.00%)carrying out ADR monitoring, rational drug use knowledge publicity, prescription and medical order comment, ward round, consultation, editing and publishing drug information was relatively high. 28 medical institutions(78.95%)did not carry out the test of therapeutic drugs;39(100%)medical institutions had carried out the education of patients’drug use,in the form of billboards(30 hospitals,78.95%),science lectures(28 hospitals,73.68%)and drug education forms(28 hospitals,73.68%)The contents of drug use education mainly included precautions(39 hospitals,100%),usage and dosage(37 hospitals,97.37%), ADR(36 hospitals,94.74%). 29 hospitals had carried out clinical pharmaceutical research(73.68%),mainly rational drug use research(28 hospitals,73.68%);5 medical institutions(12.82%)had set up joint pharmacy outpatient service,2(5.12%)had set up independent pharmacy outpatient service,the rest had not set up pharmacy outpatient service(26 hospitals,66.67%). In this survey,it is found that there are some problems,such as incomplete coverage of beds in the ward in pharmaceutical care,weak foundation of scientific research in pharmaceutical departments. It is suggested that the national and local health committees should strengthen the construction of multi-level clinical pharmacy talent echelon,explore and train resident clinical pharmacists,select and build key clinical pharmacy specialties in Chongqing,and establish incentive mechanism for clinical pharmacists. Medical institutions at all levels can adopt multiple ways to accelerate the training of clinical pharmacists,increase the publicity of pharmacists’outpatient service,change the working mode of the ward,cultivate special clinical pharmacists,improve the ways and paths of continuing education,and enhance the strength of scientific research to improve the level of clinical pharmaceutical care.
4.Dose-response relationship between maternal thyroid hormones in the first twenty weeks and physical and neuropsychological development of infants: A prospective cohort study in China.

Objective: To study the dose-response relationship between maternal thyroid hormone levels in the first twenty weeks of pregnancy and the infant physical and neuropsychological development. Methods: In this prospective cohort study, a total of 945 women and their children were included. Maternal serum samples during first half of the pregnancy were collected and analyzed for levels of thyroid hormones by using the electro-chemiluminescence immunoassay. Maternal social demographic information was collected by using the a self-administered questionnaire. Physical measurements of newborns and neuropsychological evaluation of infants were performed by doctors of maternal and child health care. Results: The differences in newborns' birth length and head circumference were significant among the newborns of mothers with different percentiles of maternal serum (thyroid-stimulating hormone, TSH) levels (P<0.05). Newborns with maternal TSH level ≥P(95) or <P(5) had significantly lower birth length and birth head circumference, compared with the newborns with maternal TSH level between P(25)-P(75) (P<0.05). Newborns' birth head circumferences showed an inverted U-shaped association with maternal serum TSH level (Y=33.940+0.003X-0.109X(2), F=4.685, P=0.009). The difference in mental development index (MDI) of the infants at 18-30 months were significant among the infants of mothers with different percentiles of maternal serum TSH level (P<0.05). Infants with maternal TSH level ≥P(90) showed lower MDI (6.39, 95%CI: 2.29-10.49, P=0.002) compared with the infants with maternal TSH level between P(25)-P(75). Infant's MDI at 18- 30 months also showed an inverted U-shaped association with maternal serum TSH level (Y=103.249-1.524X-0.939X(2), F=6.616, P=0.001). Conclusions: Maternal TSH level was associated with newborn's birth length, birth head circumference and infant's MDI at 18-30 months. Newborn's birth head circumference and infant's MDI at 18-30 months showed an inverted U-shaped association with maternal serum TSH-Z score.

5.Molecular characterization of ascaris from Tibetan pigs by three mitochondrial markers of nad1, cox1 and cox2

Ascaris is a helminthic parasite, which infects a wide range of host species
causing ascariasis, a predominant disease worldwide. This parasite causes significant economic
losses to the pig industry. The current study was designed to determine the Ascaris nematode
by the genetic characterization of three mitochondrial (mt) genes namely NADH dehydrogenase
subunit 1 (nad1), cytochrome oxidase subunit 1 (cox1) and cytochrome oxidase subunit 2
(cox2). A high infection rate of Ascaris nematode has been found in Tibetan pigs at the
slaughter houses in Tibet Autonomous Region of China. The nad1, cox1 and cox2 genes
sequences collected from adult Ascaris individuals were amplified by polymerase chain
reaction. The cloned-amplicons and the positive products were sequenced and phylogenetic
analysis was performed. The results indicated that the Ascaris infecting the Tibetan pigs
were Ascaris suum (A. suum). This is the first report on the isolation, identification and
genetic characterization of three mitochondrial genomes (nad1, cox1, and cox2) of A. suum
originated from Tibetan pigs at high altitudes in Tibet.
6.Nonalcoholic fatty liver disease versus metabolic-associated fatty liver disease: Prevalence, outcomes and implications of a change in name

Nonalcoholic fatty liver disease (NAFLD) affects about a third of the world’s adult population and is a major public health concern. NAFLD is defined by the presence of hepatic steatosis and the absence of other causes of liver disease. As NAFLD is closely associated with the presence of the metabolic syndrome, several experts have called for a change in nomenclature from NAFLD to metabolic-associated fatty liver disease (MAFLD) to better reflect the underlying pathophysiology of NAFLD as a metabolically driven disease and shift to a “positive” diagnostic criteria rather than one of exclusion. Recent studies have suggested that the global prevalence of MAFLD is higher than that of NAFLD, and patients with MAFLD have more metabolic comorbidities compared to those with NAFLD. Emerging data also suggest that all-cause and cardiovascular mortality may be higher in MAFLD compared with NAFLD. In this synopsis, we discuss differences in clinical features, prevalence and clinical outcomes between NAFLD and MAFLD. In addition, we highlight the advantages and disadvantages of a name change from NAFLD to MAFLD from the perspective of the scientific community, care providers and patients.
8.Differentiating effect of PPARgamma ligand rosiglitazone and all trans-retinoic acid on myeloma cells and its possible mechanism.

OBJECTIVETo investigate the effects of PPARgamma ligand (rosiglitazone, RGZ) as well as combined with all trans-retinoic acid (ATRA) on human myeloma cells and try to explore the possible mechanism.
METHODSHuman myeloma cell lines U266 and RPMI-8226 cells were treated with RGZ in the presence or absence of ATRA. Cell proliferation was evaluated by [(3)H] thymidine incorporation, cell cycle distribution and CD49e expression were analyzed by flow cytometry, morphology changes were evaluated by Wright-Giemsa staining, and p27(Kip1) and p21(Waf1) expression was detected by Western blotting.
RESULTSThe exposure to RGZ induced proliferation inhibition in both cell lines in a dose-dependent manner. After cultured with 5 micromol/L RGZ, the proportion of U266 and RPMI-8226 cells in phase G(0)/G(1) was (45.2 +/- 6.7)% and (40.3 +/- 7.3)%, respectively (P < 0.05). The proportion of the cells in phase G(2)/M and S was (52.2 +/- 7.4)% and (57.4 +/- 9.5)%, respectively (P < 0.05). These changes were more evident when the RGZ concentration was increased to 10 micromol/L. A combination of RGZ with ATRA enhanced the growth inhibition and cell cycle arrest effects of RGZ. The RGZ-treated myeloma cells displayed morphological characteristics of cell differentiation, and more evident signs of differentiation were observed when RGZ was combined with ATRA. These changes were confirmed by the detection of CD49e expression. The expression of p27(Kip1) and p21(Waf1) in myeloma cells was up-regulated by RGZ and this change was more apparent when RGZ was used in combination with ATRA.
CONCLUSIONRGZ can induce cell cycle arrest and cell differentiation in myeloma cells which maybe caused by up-regulation of p27(Kip1) and p21(Waf1) expression. ATRA can enhance these effects of RGZ on multiple myeloma cells and combined use of these two drugs may show a synergistic effect on myeloma cells.

9.Epidemiological and pathogenic characteristics of mumps in Fujian province, 2005-2017.

Objective: To understand the epidemiological and etiological characteristics of mumps in Fujian province, 2005-2017. Methods: All the reported mumps cases were collected through the National Notifiable Disease Information Management System, 2005-2017. Active search and interviews were conducted to collect the information on vaccination of mumps. Throat swab specimens were collected for cells culture, genotyping and gene sequence analysis on mumps virus (MuV). Results: A total of 83 959 cases of mumps were reported in Fujian province from 2005 to 2017, with an average annual incidence of 17.6 per 100 000. Since 2007, the incidence appeared increasing but then decreasing, reaching the lowest level (7.5 per 100 000), after the setup of a monitoring program. Annually, the onset time of mumps showed an obvious two seasonal peaks, one from April to July, with a weakening trend, and the other from October to January with a rising trend. Most of the mumps cases occurred among students, kindergarten and scattered children (89.2%, 5 814/6 517), children aged 5-9 years (38.8%, 2 527/6 517), with cases reported from every region. Program from the pathogen surveillance showed that the transmission chain of G genotype mumps virus did exist in Fujian. Data from the sequence analysis revealed that mutations in the nucleotide of G genotype strain in 2015 had led to mutation of 6 amino acid sites in the SH gene coding region, resulting in the differences appearing in both nucleotide and amino acid homology with type A vaccine strain. Conclusions: The incidence of mumps decreased annually, in Fujian. Prevention programs should focus on primary and secondary school students. In Fujian province, we also noticed the transmission chain of mumps G genotype with some amino acid mutations in the SH gene coding region. Monitor programs on both epidemiologic and etiology, should be strengthened.

10.WNKs: protein kinases with a unique kinase domain.

WNKs (with-no-lysine [K]) are a family of serine-threonine protein kinases with an atypical placement of the catalytic lysine relative to all other protein kinases. The roles of WNK kinases in regulating ion transport were first revealed by the findings that mutations of two members cause a genetic hypertension and hyperkalemia syndrome. More recent studies suggest that WNKs are pleiotropic protein kinases with important roles in many cell processes in addition to ion transport. Here, we review roles of WNK kinases in the regulation of ion balance, cell signaling, survival, and proliferation, and embryonic organ development.
