Objective To investigate the chnicopathogenical significance of c-jun N-terminal kinase(JNK) and multidrug resistance associated protein 1 (MRP1) expression in colorectal cancer. Methods The expression of JNK and MRP1 was detected in a tissue microarray containing 72 spots of colorectal cancer tissue and 40 spots of nor-mal colorectal tissue by immunohistochemisty. Results The positive expression rate of JNK and MRP1 expression was 47.22% and 51.39% respectively in colorectal cancer,significantly higher than that in normal colon tissue. JNK protein expression was not closely related to gender, age, tumor size and location (P>0.05), but closely related to tumor differentiation,lymph node metastasis,distsnt metastasis and Dukes staging (P<0.05). MRP1 protein expres-sion was not closely related to gender,age,tumor size,location and tumor differentiation(P> 0.05), but closely relat-ed to lymph node metastasis, distant metastasis, Dukes staging (P<0.05). Conclusions The JNK expression in cancer is corrlated with maligant biological behavior and may be involved in the chemotherapeutic resistance by upreg-ulating the expression of MRP1.

Objective To determine whether exposure to electromagnetic radiation results in behavioral effects of rat pups.Methods Wistar rats,either male or female,were divided randomly into four groups respectively:control(CTR),100 seconds(S1),1000 seconds(S10)and 3000 seconds(S30),then each of them were exposed to electromagnetic radiation of 100 kV/m field amplitude of corresponding time.For the day of exposure till 2 months later.they were mated with the rats in the same group and their offspring were divided into four groups(F-GTB,F-S1,F-S10,F-S30)correspondingly.Behavioral changes occur in 2-month old and 6-month old rat pups were found between radiation groups and control,and in Y-maze test,beth male and female pups in F-S10 significantly learned fewer times than their control(total study times:male 14.6±3.9 vs 21.1±7.8,female13.4±3.0 vs 25.8±8.8;false times:male 3.5±2.4 vs 7.8±5.4,female 3.4±2.6 vs 11.0±7.2).In open field test,both maze,male pups in F-S1 and F-S30 learned more times than control in total study time(24.2±8.9 vs 14.1±5.2.30.7±12.4 vs 14.1±5.2).In step throush test and open field test,no significant differences were found between radiation groups and control.Conclusion There was significant genetic effect exposed to electromagnetic radiation of 100 kV/m,mainly manifested in rat pups in growth period but without long-term effect.

Objective In order to analyze the pollution level of PM10 and PM2.5, as well as their sources in large and moderate scale shopping centres. Methods Using the portable aerosol monitor detected the PM10 and PM2.5 on different locale floors in Zhongyuan Supermarket, Commerce Building and Foodstuff Centre in Pingdingshan City area. Results The average mass concentration of PM10 in Zhongyuan Supermarket,Commerce Building and Foodstuff Centre were 0.212 mg/m3, 0.108 mg/m3, 0.056 mg/m3 and that of PM2.5 were 0.179 mg/m3, 0.092 mg/m3, 0.043 mg/m3 respectively. The pollution level of PM10 of all the shopping center didn't exceed the related standard. The average mass concentration of PM2.5 in Zhongyuan Supermarket,Commerce Building all exceeded the standard ,and the times of exceeding standard were 1.75 and 0.42. The people volume of flow was more masses, and the concentration of PM10 and PM2.5 were more higher. METHODS: AHNP was induced in 40 rats weighting 260-280 g by intraductal administration of 5% sodium taurocholate. The pathologic morphologic changes of liver and pancreas were observed under light microscope. The hepatocyte apoptosis was examined through TdT (terminal deoxynucleotidyl transferase) mediated dUTP nick end labeling (TUNEL) and the expression of TGF ?1 in hepatocytes was analyzed through immunohistochemistry. Objective To investigate the clinical significance of NaviCam magnetic-controlled capsule endoscopy (NMCE) system in the examination of upper gastrointestinal tract.Methods A total of 39 healthy volunteers were enrolled in the present study.NMCE system was used to examine upper gastrointestinal tract.The safety,gastric preparation,visualization and comfort of the subjects were evaluated.Results Visualization of the Z-line,gastric cardia,fundus,body,angulus,antrum and pylorus was subjectively assessed as more than 75％ mucosa in 19 (48.71％),37 (94.87％),25 (64.10％),30 (76.92％),39 (100.00％),39 (100.00％),and 39 (100.00％),respectively.The observation time was 1.5,3.0,8.0,17.0,3.0,3.0,5.0 min respectively.The capsule was driven into duodenum positively in 25 (64.10％).Seven subjects went into small bowel without control.The one-time visualization efficacy was 97.43％ (38/39).Only one subject felt foreign body sensation.All subjects extracted the capsule within 7 days.Conclusion Our study provides a preliminary assessment of the NMCE on its feasibility and safety.It is comfortable with no chance of cross-infection.NMCE system is a useful tool for upper GI examination and will have a good future.

Objective To seek the specific receptors associated with hepatitis B virus (HBV) adhesion by separating the binding protein of the HBV preS1 region in HepG2 and performing the mass spectrometry .Methods The immunomagnetic bead method was adopted to separate HepG2 membrane protein combined with preS1 peptide fragment and the binding protein was separated by the sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) ,then the destination strips was analyzed by LC-MS/MS mass spectrometry and retrieved by the database .Results 16 bands were separated from HepG2 membrane proteins combined with preS1 by SDS-PAGE ;14 kinds of proteins were identified from 6 bands with better repeatability separated from HepG 2 membrane proteins combined with preS1 .Conclusion Protein analyzed by the mass spectrometry is mainly related with the material transport , cellular signal transduction ,antigen presentation ,immune regulation and energy metabolism .

Objective To develop an evidence-based community extended care model for patients with asthma.Methods Mixed research methods were applied to integrate best evidences:80 literatures were included in systemic review; 394 medical records were analyzed; 112 nurses admitted a test and a questionnaire to assess their competence on asthma care; clinical observation to both hospital and community centers were conducted; 6 multidisciplinary experts were consulted; and 100 patients admitted a questionnaire on patient needs and 17 of them were further interviewed.Results The nursing care model with the feature of hospital and community health service center collaborating to provide extended care for patients with asthma was established.Conclusions The development of a nursing care model for practice should be based on the current best evidence generating from the best research evidence,professional judgment on clinical practice requirement and patients' needs.

Objective To explore the expression change of intestinal epithelial tight junction (IJ)protein claudin-1 in mice with fulminant hepatic failure (FHF).Methods FHF was induced with a method that combined intraperitoneal injection of lipopolysaccharide (LPS,10 mg/kg) and D-galactosamine (GalN,800 mg/kg).Control saline (2 ml/kg,ip),LPS (10 mg/kg,ip) and GaIN (800 mg/kg,ip) were also detected.The effect of administration of anti-tumor necrosis factor alpha (TNF-α) IgG antibody (anti-TNF-α IgG,100 μg/per) on the level of TNF-α was assessed before administration of D-galactosamine/lipopolysaccharide.At the 2nd h,6th h,9th h,12th h,24th h after injection in FHF group,the 9th h after injection in control groups and 9th h after injection in anti-TNF-α IgG group,the mice were killed for the collection of large intestine specimens.Claudin-1 was analyzed with immunohistochemistry,Western blotting,and real-time quantitative PCR.Results Tight junction protein claudin-1 was localized along the apical region of the lateral plasma membrane representing the region of tight junctions in surface and crypt epithelial cells.Weakly distributed density of claudin-1 in intestinal mucosa was found in mice with FHF from the 9th h after injection.Compared to saline group,Western blotting analysis demonstrated markedly reduced claudin-1 expression in mice with FHF at the 6th h and 9th h after injection (6th h:0.8600±0.0208 vs 1.0,P ＜0.05; 9th h:0.6633 ±0.0328 vs 1.0,P ＜0.01).Furthermore,the expression of claudin-1 mRNA was markedly reduced at the 6th h,9th h,and 12th h after injection in mice with FHF (6th h:0.3067 ±0.1291 vs 1.0,P ＜0.05; 9th h:0.2233 ±0.1155 vs 1.0,P ＜0.01 ; 12th h:0.5275 ±0.1222 vs 1.0,P ＜0.05).Compared to saline group,no significant difference in claudin-1 expression was found with prophylactic treatment with anti-TNF-α-IgG antibody in mice with FHF at the 9th h after injection (protein:0.9533 ±0.0186 vs 1.0,P ＞0.05; mRNA:0.85 ±0.1437 vs 1.0,P ＞0.05).Conclusions The expression of tight junction protein claudin-1 was reduced at both protein and mRNA levels in intestinal epithelial cells that were induced by TNF-α in mice model of FHF.
