1.A Study on Echographic Dimensions of Normal Liver in School-Children

Objective To determine the echographic dimensions range of normal liver in chinese school children.Methods The echographic dimensions of normal liver in 972 chinese school-age children were measured by means of random sampling including the maximum oblique dimension right lobe,the transverse dimension of right lobe,the anterior-posterior dimension of right lobe,the thickness and length of left lobe,the thickness and length of right lobe below the costal margin in the midcalvicular line in inspiratory and expiratory phase respectively.The range of normal values was defined according the different age class and sex.Results The nine liver dimensions were all correlated to age,height,weight and bodysurface area.Each echographic dimension showed skew distribution.Conclusion It is necessary to determine the normal dimensions range of liver in chinese school children according to their sex and age.

Objective To investigate the levels of serum tumor necrosis factor-alpha (TNF-?琢) and soluble tumor necrosis factor receptor (sTNF-R) and sTNF-R/TNF-?琢 ratios in patients with severe lupus nephritis (SLN) and the impact of double pulse therapy with methylprednisolone (MP) combined cytoxan (CTX). Methods Serum TNF-?琢, soluble tumor necrosis factor receptor type Ⅰ(sTNF-RⅠ) and soluble tumor necrosis factor receptor type Ⅱ (sTNF-RⅡ) levels were determined in 38 cases of SLN patients and 35 health controls by double antibody sandwich enzyme-linked immunosorbent assay (ELISA) before and after the double pulse therapy with MP and CTX. Anti-dsDNA antibody was detected by ELISA. Antinuclear antibody (ANA) was detected by indirect immunofluorescence experiments. Complement C3 and complement C4 were detected by rate nephelometry. Results The serum levels of TNF-?琢, sTNF-RⅠ, sTNF-RⅡand ratios of TNF-?琢/sTNF-RⅠ, TNF-a/sTNF-RⅡ were significantly higher in SLN patients than those in normal control group (P

6.Changes and clinical significance of serum cytokines in patients with bee-sting poisoning

Objective To investigate changes and clinical significance of serum granulocyte-macrophage colony stimulating factor(GM-CSF),tumor necrosis factor-alpha(TNF-?),interleukin-6(IL-6)and interleukin-8(IL-8)level in patients with bee-sting poisoning.Methods One hundred and nine patients with bee-sting poisoning were also divided into slight poisoning group,moderate poisoning group,and server poisoning group and also divided into dead group and survival group according to the prognosis.Thirty healthy persons were selected as control group.GM-CSF and TNF-?were measured using radioimmunoassay,IL-6 and IL-8 was measured using enzyme linked immunoadsorbent assay(ELISA),and they were compared with control grouup.Results The levels of serum GM-CSF,TNF-?,IL-6,IL-8 in slight poisoning group were increased a little,and there were no significant difference compared with control group(P>0. 05).The levels of serum GM-CSF,TNF-?,IL-6 and IL-8 were significantly higher in moderate poisoning group and severe poisoning group than those in the control group(P<0.01),but the severe poisoning group were much higher than the moderate poisoning group(P<0.01).They were significantly higher in survival group than those in dead group(P<0.01).APACHEⅡscores were obvious correlated with the levels of serum GM-CSF,TNF-?,IL-6 and IL-8(P<0.05 or P<0.01). Conclusion GM-CSF,TNF-?,IL-6 and IL-8 may participate in the occurrence and progress of patients with bee-sting poisoning.Dynamic monitoring of serum GM-CSF,TNF-?,IL-6,IL-8 and APACHEⅡscores plays an important role in diagnosis of the severity of patients with bee-sting poisoning and its prognosis.The use of antagonist and inhibitor of cytokines may be helpful to treat patients with bee-sting poisoning.
7.Experimental study of ultrasound microbubble-mediated HGF gene transfer on liver fibrosis in rats

Objective To investigate the efficiency of ultrasound microbubble-mediated HGF gene transfer on liver fibrosis in rats. Methods The eukaryotic expression vector pMD18-T/HGF was constructed. The rats were randomly divided into 5 groups: normal group, model group, low dose group,middle dose group and high dose group. The rat liver fibrosis were induced. At day 14 after treatment, the rats were sacrificed, the ALT lever were evaluated, pathological evaluation of liver fibrosis were made,immunohistochemical evaluation of HGF protein expressions were observed, HGF mRNA expression were detected by RT-PCR. Results Liver 1function after the treatment showed that the treatment groups were better than model group ( P < 0. 05). The difference between low dose group and normal group showed significant difference ( P <0.05), and the difference between middle/high dose groups and normal group showed no significant ( P >0. 05). Pathological results showed that the treatment groups could reduce liver fibrosis;treatment groups had the positive HGF expression by immunohistochemistry and the expression level increased with the increasement of the treatment dose ( P <0.05). RT-PCR showed HGF mRNA expression in the treatment groups were higher than in model group ( P <0.05). Conclusions Ultrasound microbubbles which carrying HGF can inhibit the process of hepatic fibrosis.
8.Pathogenesis of renal lesion in Fechtner syndrome

Objective To investigate the pathogenesis of renal lesion in Fechtner syndrome . Methods Pathological characteristics of kidney tissues from Fechtner syndrome patients were explored by HE staining, immunochemistry, immunofluorescence and electron microscopy . Results Immunochemistry analysis showed that non-muscle myosin heavy chain IIA (NMMHC-IIA)was expressed in podocytes of giomeruli and distal convoluted tube, and was faintly expressed in the brush border of proximal tube . Histological examination demonstrated glomerulosclerosis and decreased expression of NMMHC-IIA in abnormal podocytes . Through standard immunofluorecence, the expression of NMMHC-IIA in patient's podocyte was higher than that in normal pedocytes . The fusion of foot process and microvillus were detected by electron microscopy . Conclusion Abnormal NMMHC-IIA aggregates in the glomeruli podocyts and foot process fusion accompanied with appearance of microvillus leads to renal lesion in Fechtuer syndrome .
9.Study on the correlation between ultrasound imaging and pathology in acute lung injury of rats

Objective To evaluate the correlation between ultrasonography and histopathology in acute lung injury of rats.Methods Twenty male Wistar rats were randomly divided into control group (4 rats) and experimental group (16 rats),in the experimental group acute lung injury models were step-by-step induced by oleic acid (OA) and sequentially by OA and lipopolysaccharide (LPS).After the end of each step of models,ultrasound examination was applied,and the rats were sacrificed to take the lung specimens to be observed.The ultrasonography and pathological results were analyzed,and the numbers of alveolar were counted under microscope (200 times).Results With the aggravation of acute lung injury,the sonographic findings showed unclear or disappeared pleural line,close-set of B-lines,and pulmonary consolidation with air bronchogram.For control group,the ultrasonographic score was 0.25 ± 0.50,lung injury pathological score was 0.50 ± 0.58,and the numbers of alveolar were 25 ± 3.For OA group,the ultrasonographic score was 2.86-± 1.35,lung injury pathological score was 6.28-± 0.76,and the numbers of alveolar were 10-± 2.For OA + LPS group,the ultrasonographic score was 5.83-± 2.32,lung injury pathological score was 9.83 ± 0.98,and the numbers of alveolar were 6-± 2.All 3 groups were significant different on ultrasonographic score,pathological score and alveolar counts (P < 0.01).Conclusions The ultrasonography image of acute lung injury of rats were positively correlated with its histopathological alteration.The ultrasonography can effectively evaluate the degree of acute lung injury of rats.
10.Study on antigen-specific T cells in chronic hepatitis B patients accepting antiviral therapy

Objective To explore the responses of antigen-specific T cells stimulated by hepatitis B virus(HBV)-specific proteins in chronic hepatitis B patients accepting antiviral therapy. Methods Seventeen patients with chronic hepatitis B (CHB) accepting antiviral therapy were included in this study. The peripheral blood monocular cell ( PBMC) were separated from the whole blood collected at the three different time of before and one and three months after accepting antiviral therapy. ELISPOT assay was used to detect the frequency and strength of secreting IFN-γ cells of PBMC stimulated by HBsAg, HBcAg and HBeAg. HBV virus loading, HBsAg, HBeAg, ALT and AST in serum were detected at the same time. Results After three months therapy, ALT, TBiL were improved in all patients, and HBV DNA level were dropped and undetectable in 11 cases. The rates of T cell response in patients to HBV specific proteins were 64. 7% , 76. 5% and 82. 4% at the time of before and one and three months after accepting antiviral therapy, respectively. The frequency of responses of antigen-specific T cells stimulated by HBcAg was higher than that stimulated by HBsAg or HBeAg, and the frequency was enhanced after antiviral therapy. The average response magnitude was expressed as spot forming cells (SFC) per million input cells. SFC of T cell responses to HBcAg was also higher than to HBsAg or HBeAg. There was no significant difference in SFC of T cell responses to HBsAg or HBeAg at the time of before and after antiviral therapy, but there were significant difference in SFC of T cell responses to HBcAg at the time of before and after antiviral therapy. SFC of T cell responses to HBcAg was negatively associated with HBV DNA, and no associated with level of ALT in serum. Conclusion The responses of antigen-specific T cells were improved in CHB patients accepting antiviral therapy which associated with the decrease of HBV DNA. It suggested to investigate HBV specific T cell responses was important.
