1.Effects of Endovascular Radiation on the Proliferation and Apoptosis of Vascular Medial Smooth Muscle Cells in Rabbits after Carotid Endarterectomy

Objective To observe the effects of endovascular radiation (ER) on the proliferation and apoptosis of medial smooth muscle cells (SMC) and to discuss the possible mechanisms of radiation in the prevention of vascular restenosis (RS) in rabbits after carotid endarterectomy (CEA).Methods Forty rabbits undergoing CEA were randomly divided into four groups (each group=10) and given a radiation dose of 0, 10, 20 and 40 Gy 32 P respectively. Rabbits were killed on the 3rd, 7th, 14th, 28th and 56th day after operation. The specimens were collected and histopathologic examinations were done.Results Proliferation apparently occurred in the intima and media of carotid the lumen became narrow in the control group on the 14 th, 28 th and 56 th day after operation. 2.The cytological examination of cancer cells in the peripheral blood of patients with gastric cancer

Objective The infiltration and micrometastasis of cancer cells via lymph vessel and blood is the important factors which influence the prognosis of patients with cancer. The present study was to establish the method of cytological examination of circulating gastric cancer cell in the peripheral blood and to explore its clinical significance. Methods The monocytes in blood samples, taken from 24 patients with advanced gastric cancer, were seperated by using Ficoll density gradient centrifugation and magnetic cell sorting system in which the magnetic microbeads were wrapped with cytokeratin 7 and 8. The samples were smeared and observed with HE stain. The expression of CD 34 , CD 45 , CEA and human telomerase reverse transcriptase (hTERT) of the cells were also examined with immunocytochemical and immunofluorescence staining methods. Results Gastric cancer cells were found in 10 out of 24 cases, and the positivity rate was 41.7%, which was significantly different from pathological differentiation of primary focus. Conclusion This method can be used to detect cancer cells from peripheral blood of patients with gastric cancer and therapeutic protocol and prediction of prognosis can be made based on it.
3.NEW DISCOVERY: REPORT OF A HUMAN SCHISTOSOME

Objective To delineate the m orp hology and ecology of a new human schistosome species. Methods (1)Adult schistosomes were obtained from infected mice and rabbi ts with cercariae shedding from snails which were infected with miracidia devel oping from the eggs in feces of acute schistosomiasis patients. (2)T he rabbits were dissected during 60-100 d infection period of cecariae, and thei r bladders were taken out for detecting eggs in the urine. (3)Schistosome eggs i n the feces of patients were examined with microscopy. Results (1)There were two types of male worms: one was with an average o f size 8.76 mm long and 0 43 mm wide while the other with an average size of 2 41 mm long and 0 091 mm wide and with undeveloped gynecophoral canal which ap peared in front of ventral sucker (2)There were two types of female worms: one was with an average of size 10 73 mm long and 0 74 mm wide while the other wi th an average of size 3 43 mm long and 0 12 mm wide and was fine, semitranspar ent and tape like with undeveloped ovary and vitellaria. Less than 10 eggs or none were found in t he uterus. (3)There were a lot of degenerated and black eggs in the feces of 2 p atients. (4) Schistosome eggs were detected in the urine and miracidia were hatc hed out from 8 out of 16 infected rabbits.(5) Others: 8 worms were detected in 1 mouse out of 12 dissected mice for evaluation of infectious water with sentry m ice in spite of that no Oncomelania snails were found in the area. Cercariae stayed on water surface in lying position like a c omma. The eggs returned forwardly a fter growing behind the ovary in female worm. ConclusionAccording to the primary investigation in morphology an d ecology, the new schistosome obtained is distinct from the 6 well defined ty p es of human schistosome. It is considered that a new schistosome species should be foun d and temporarily nominated as Schistosoma nanjingi( S.n.)
4.INDUCTION OF IMMUNE RESPONSES AND SAFETY PROFILES IN RHESUS MACAQUES IMMUNIZED WITH THE THERAPEUTIC DNA VACCINE AGAINST HEPATITIS B VIRUS

To evaluate the immune response and safety of the therapeutic DNA vaccine against the hepatitis B virus in rhesus macaques immunized by intramuscular injection of therapeutic DNA vaccine combined with electroporation. HBV DNA vaccine was administered to rhesus macaques by repeated intramuscular injections combined with electroporation in doses of 0 2,1 and 2mg, monthly for three consecutive months. The anti HBs antibody level was used to evaluate the efficacy of induction of humoral immunity. After that, the vaccine was intramuscularly injected daily in the same dose for six days to evaluate its safety. It was found that immunization with the therapeutic DNA vaccine against hepatitis B virus by means of intramuscular injections combined with electroporation induced a strong and specific anti HBs humoral immune response in rhesus macaques.Nine repeated intramuscular injections of the vaccine did not show adverse effects on behaviors, hematology, blood chemistry, and histopathology. No evidences of autoimmune mediated pathology and anti nuclear antibodies were observed in the rhesus macaques. These results suggested that electroporation significantly improves the efficacy of the therapeutic HBV DNA vaccine in rhesus macaques.The vaccine is safe and well tolerated.
5.A STUDY OF DISTRIBUTION IN THE TISSUES AND LONG-TERM TOXICITY OF THERAPEUTIC DNA VACCINE IN MICE

To evaluate the safety of the therapeutic DNA vaccine against the hepatitis B virus. Plasmid DNA was detected by PCR in tissues of BALB/c mice immunized with the plasmids combined with electrotransfer in doses of 10 and 50?g. In long term toxicity study, HBV DNA vaccine was administered by repeated intramuscular injections combined with electrotransfer of pDNA to NIH mice in doses of 30, 60 and 120?g, twice a week for four consecutive weeks. Morbid manifestations, behaviors, hematology, blood chemistry, anti nuclear antibodies, and histopathology were analyzed. Results showed that plasmid DNA was detected primarily in the muscle at the site of injection, where it remained for up to 8 weeks. Eight repeated intramuscular injections of HBV DNA vaccine showed no adverse effects on behaviors, hematology, blood chemistry, and histopathology. No evidences of autoimmune mediated pathology and anti nuclear antibodies were observed in the mice. These results suggested that the therapeutic HBV DNA vaccine was safe and well tolerated.

Objective To study the correlation of the number and ratio of involved lymph nodes and the prognosis of gastric carcinoma .Methods 174 patients with positive gastric lymph nodes were included,they were treated by radical gastrectomy and lymph nodes dissection (D2 or D3)in our department. The number and ratio of metastastic gastric lymph nodes were calculated. Their influence and their interrelation with pathogenic parameters were analyzed. Results The overall 5 year survival rate was 29.59%, and the five year survival rates of patients with 1 20 per cent and more than 20% positive lymph nodes were 70.58%,11.86% respectively, one to five positive lymph nodes and five more were 46.47%,17.35% respectively, the five year survival rate was decreased as the ratio and number of involved lymph nodes increased. Studies also indicated that with the increase of depth of cancer invasion ,the number and ratio of involved lymph node elevated.Conclusions The results shows that the number and ratio of involved lymph nodes correlated to tumour progression and the five survival rate.Perigastric lymph node status can be considered as a simple and effective prognosis factor.
8.Relation Between CD44v6 and Lymph Node Metastasis and Prognosis of Gastric Carcinoma

Objective To investigate the relation between CD44v6 and lymph node metastasis and prognosis of gastric carcinoma. Methods The expression of CD44v6 in 100 cases of advanced human gastric carcinoma was detected by immunohistochemical two-step method. Its relation with clinical pathology and prognosis was analyzed. Results CD44v6 expression occurred in 64%(64/100) of all gastric carcinomas; with the increase of depth of cancer invasion and local lymph node metastasis, the expression of CD44v6 elevated. Conclusion CD44v6 plays an important role in lymph node metastasis of human gastric carcinoma. It may be used as a new indicator to predict metastatic potential and prognosis of gastric carcinoma.
9.Experience in diagnosis and treatment of digestive tract bleeding with gastrointestinal vascular malformation

Objective To summarize the experience of diagnosis and management of bleeding with GIVM.Method The clinical data of 26 patients admitted from 1989 to 1999 were analyzed retrospectively.Results GIVM with bleeding were revealed by endoscopies (42%),by ECT (55%),by selective arteriography (83%),by barium radiography (0%),and by exploratory laparotomy(100%).1 case was cured by conservative treatment.Bleeding intestinal segment was resected in 14 cases.4 cases were cured by interventional therapy.The others were cured by sclerosing agent injection,or by suture ligation of the dilated vessel or arterial gastroduodenalis.Conclusions Endoscopy is the first choice in the bleeding from GIVM.ECT may beused later if endoscopy is useless.Selective arteriography is the most valuable method in GIVM diagnosis.If necessary, exploratory laparotomy and intraoperative endoscopies may be used.Resecting bleeding segment is the most effective treatment.Other methods such as interventional therapy and suture ligation of dilated vessel are also effective.
10.Expression of bone morphogenetic proteins-4 in human gastric cancer

Objective:To determine the expression level of bone morphogenetic proteins-4 (BMP-4) and the relationship between the expression of BMP-4 and the clinical/pathological parameters in patients with gastric cancer. Methods:Using im-munohistochemical staining technique ,expressions of BMP-4 were investigated in different tissues from 90 gastric cancer specimens and 30 specimens from normal gastric mucosa as control.
