1.Applied Anatomy of Reverse Transposition of Vascularized Proximal Ulnar or Radical Periosteal Flap

Objective This paper provides an anatomical and theoretic basis for the reverse transposition of proximal ulnar or radial periosteal flap to repair non-union in distal humerus.Methods The origin,course,meanwhile,the outside diameter and length of relevent vessels were measured,on 30 upper limb cadaveric specimens.Results The recurrent interosseous,radial recurrentand the posterior branch of radial collateal arteries,their origin outside diameter and length from the origin to the humerus extermal epicondyle were (1 47?0 2)mm,(65 1?9 5)mm,(2 1?0 3)mm,(63 7?0 8)mm,(1 48?0 22)mm and (104 5?8 9)mm,respectively.Anastomoses pattern existed 3 types: in vicinity of the external epicondyle"Reverse Y"73 3%(22 cases)."Meeting in one point 13 3%(4 cases)"."Arteries networks"13 3%(4 cases).Conclusions It's possible that the reverse transposition of proximal ulnar or radial periostea flap pedicled with recurrent interosseous membrane or radial recurrent vessel for repairing non-union in distal humerus.
2.The relationship between NDRG1 gene and differentiation level and metastasis in prostate cancer

Objective To investigate the expression of NDRG1 and its relationship to differentiation level and metastasis in prostate cancer. Methods The expression NDRG1 was examined by immunohistochemistry LSAB method in 86 samples of prostate cancer and benigh prostate hyperplasia. Results The NDRG1 was shown to be highly expressed in 30(73.3%)of prostate cancer and 70.0%of benign prostate hyperplasia.There were no evident difference between prostate cancer and prestate hyperplasia(P>0.05).The expression of NDRG1 was gradually reduction with the Gleason score from 2-4 to 5-7 to 8-10 in prostate cancer, which was statistical significance(P<0.05).The expression of NDRG1 in lymph node or bone metastasis was significantly lower than that in no metastasis of prostate cancer. There was a negative correlation between the expression of NDRG1 and lymph node or bone metastasis. The age had no correlation with the expression of NDRG1(P>0.05).Conclusion NDRG1 is involved in the genesis and development of prostate cancer, which may be related to the differentiation level of prostate cancer and serve as a useful marker for early diagnosis of cancer and metastasis.

Objective To investigate the expression of Aiolos transcription factor in peripheral blood mononuclear cells (PBMCs) from patients with systemic lupus erythematosus (SLE),and to explore their clinical significance.Methods Western blot was performed to detect the protein expression of Aiolos transcription factor in PBMCs from 19 patients with active SLE,13 patients with stable SLE and 30 healthy volunteers.The relationship between the expression of Aiolos transcription factor and SLE disease activity index (SLEDAI) was analyzed.Results A significant difference was noted in the relative expression of Aiolos transcription factor between SLE patients and normal controls(0.56±0.17 vs 0.81±0.09,P<0.01) and between patients in active stage and those in stable stage (0.52±0.14 vs 0.65±0.19,P<0.05).In addition,the expression level of Aiolos protein was negatively correlated with SLEDAI in patients (r=-0.65,P<0.01).Conclusion A decrease is observed in the expression of Aiolos transcription factor in SLE patients.which is correlated with the disease activity in SLE.
4.Clinical significance of cell factors in serum samples of hand-foot-mouth disease

Objective To study the clinical significance of cell factors in plasma samples of hand?foot?mouth disease. Methods Sixty?nine cases hand?foot?mouth diseases children who were treated in Fifth People’ s Hospital of Wuxi from July to December 2014 were selected as research objects,and divided into normal hand?foot?mouth disease group with 48 cases and severe hand?foot?mouth disease group with 21 cases. Twenty healthy children were selected as healthy control group. Serum samples were collected from 3 groups of subjects. The lev?el of IgA,IgG and IgM were detected by immune turbidity method. The level of IFN?γ, IL?10, IL?13 and IL?2 were detected by flow cytometry. Results The level of serum IgG,IgA of severe hand?foot?mouth disease group patients were (12. 33±8. 58) g/L,(0. 78±0. 55) g/L,significantly lower than control group((7. 09±6. 36) g/L,(0. 61±0. 36) g/L) and the normal hand?foot?mouth disease group((7. 41±5. 07) g/L,(0. 57±0. 41) g/L),the differences were significant(P<0. 05),while there was no significant difference between normal hand?foot?mouth disease group and control group( P>0. 05) ,and there was no significant difference in term of the level of IgM among the three groups( P>0. 05) . The level of serum IFN?γ,IL?2,IL?13 of severe hand?foot?mouth dis?ease group were (0. 95±0. 67) ng/L,(1. 07±0. 64) ng/L,(1. 25±0. 46) ng/L,significantly lower than normal hand?foot?mouth disease group((2. 72±1. 15) ng/L,(2. 07±1. 53) ng/L,(2. 31±1. 35 ) ng/L) and the control group((1. 83±1. 12 ) ng/L,(1. 61±1. 37) ng/L,(1. 99±0. 75) ng/L),the differences were significant(P<0. 05). The level of serum IL?10 of normal hand?foot?mouth disease group was (2. 69±1. 33) ng/L,significant?ly higher than severe hand?foot?mouth disease group ( ( 1. 54 ± 1. 23 ) ng/L ) and control group ( ( 1. 35 ± 1. 12 ) ng/L) ,the differences were significant( P<0. 05) . Conclusion The level of serum cytokines can reflect the ge?netic profile status,understand the development trend of the disease,provide a reliable index for early clinical in?tervention.
5.Effect of TRPV5 and TRPV6 channels on the biological behaviors of SW480 colon cancer cell line

Objective:To investigate the role of TRPV5 and TRPV6 in intracellular calcium regulation and biological behaviors of SW480 colon cancer cells. Methods:qRT-PCR, Western blot, and immunocytochemistry were applied to determine the mRNA and protein ex-pression levels of TRPV5 and TRPV6 in SW480 colon cancer cell line upon treatment with TRPV5 and TRPV6 agonist, 1-25(OH)2D3, and inhibitor, CuCl2. The change of intracellular Ca2+level was examined with a confocal laser scanning microscope. Scratch test, MTT, and TUNEL assays were used to analyze the cell migration, proliferation, and apoptosis, respectively. Results:As an agonist of TRPV5 and TRPV6, 1-25(OH)2D3 significantly up-regulated the mRNA and protein expression levels of TRPV5 and TRPV6 in SW480 cell lines. On the other hand, CuCl2, being an inhibitor of TRPV5 and TRPV6, effectively down-regulated the TRPV5 and TRPV6 mRNA and protein expres-sion levels (P<0.05). The intracellular calcium concentration in SW480 cell line significantly increased upon treatment with 1-25 (OH)2D3, and significantly decreased with CuCl2 treatment (P<0.05). 1-25(OH)2D3 promoted cell proliferation and migration, and inhibit-ed apoptosis of SW480 cell in a time-and dose-dependent manner (P<0.05). However, CuCl2 significantly repressed cell proliferation and migration and induced apoptosis (P<0.05). Conclusion: TRPV5 and TRPV6 can affect the biological behaviors of colon cancer SW480 cells by regulating intracellular Ca2+level.
6.The expression of NCX1 and its effect on proliferation and migration of hepatocellular carcinoma cells through regulation of intracellular Ca2+

Background and purpose:Previous studies have suggested Na+-Ca2+ exchanger isoform 1 (NCX1) as a key component of calcium homeostasis was involved in the tumorigenesis. However, the role of NCX1 and calcium signal in tumorigenesis of hepatocellular carcinoma (HCC) has not been explored. This study aimed to investigate the effect of NCX1 on cell proliferation and migration of HCC HepG2 cells in vitro and the possible mechanism.Methods:Both the real-time fluorescent quantitative polymerase chain reaction (RTFQ-PCR) and Western blot were applied to assess the expression of NCX1 mRNA and protein in normal hepatic cells (LO2), HCC cell line (HepG2), human normal hepatic tissues and hepatocellular carcinoma tissues. The change of intracellular calcium signal in LO2 and HepG2 cells via acti-vated NCX1 channel in the presence or absence of Na+ was examined by a confocal laser scanning microscope. The effects of NCX1 special inhibitor KB-R7943 on cell proliferation and migration of HepG2 cells were measured by MTT and cellscratch test.Results:Both mRNA and protein expression of NCX1 were higher in HCC tissues and cell line HepG2 than in the normal tissues and cell line LO2 (P<0.05). The activation of NCX1 channel induced a slight rise in cytoplasmic Ca2+concentration ([Ca2+]cyt) in normal cells, but caused a marked increase in cancer cells. And the NCX1 activation induced intracellular calcium increase was significantly reversed by NCX1 inhibitor KB-R7943 (P<0.05). Both NCX1-mediated proliferation and migration of HepG2 were also significantly attenuated by the KB-R7943 (P<0.05).Conclusion:NCX1 is up-regulated in HCC cells and tissues. The activation of NCX1 mediates intracellular calcium homeostasis. The inhibition of NCX1 activity can suppress the proliferation and migration of HepG2 cells. It is suggested that NCX1 may be involved in the development and progression of HCC.

Objective To optimize separation and purification technology of Zhizichi decoction by macroporous resina . Methods The content of total iridoid glycosides and total isoflavones were determined by ultraviolet spectrophotometry .Tak-ing the content of total iridoid glycosides and total isoflavones as indexes ,the effect of the concentration of sample solution , medicinal herbs on the amount of sample ,concentration and volume of eluent was investigated by single factor test .Results The optimum separation and purification technology was as following :the concentration of sample solution was 0 .1 g/ml ,the volume ratio of resin to material drug was 2∶1 ,the adsorption time was 2 h ,and the sample was firstly eluted with 1 BV wa-ter ,then 6 BV 20% ethanol and 60% ethanol ,and the eluent was collected .Conclusion This optimized separation and purifi-cation technology was reasonable and stable ,and it could be extended to large-scale production applications .

Aim To investigate the effects of diterpene ginkgolides meglumine injection (DGMI ) on amino acids and monoamine neurotransmitters in rats with cerebral ischemia/reperfusion injury.Methods In-traluminal suture was applied to establish middle cere-bral artery occlusion (MCAO/R)model with ischemia for 1.5 h and reperfusion for 24 h.After the adminis-tration of DGMI (i.v.),the levels of amino acid and monoamine neurotransmitters in brain tissue were de-tected through HPLC-ECD.Results DGMI down-reg-ulated the concentrations of aspartic acid, glutamic acid,glycine and γ-aminobutyric acid which were in-creased in MCAO/R group.DGMI also reduced the levels of norepinephrine epinephrine,glyoxylic acid, serotonin and 5-HIAA in cortex and hippocampus,and increased adrenaline content compared to the model group.Conclusion DGMI exhibits a protective role in rats with cerebral ischemia /reperfusion injury through regulating amino acids and monoamine neuro-transmitters.

Objective:
To report the effects of operative treatment of Sneppen Ⅴ talus fracture through the approach for malleolus medialis Ⅴ osteotomy plus hollow compression screw fixation.
Methods:
From January 2015 to January 2019, 16 patients with Sneppen Ⅴ talus fracture were treated at Department Ⅱ of Hand & Foot Microsurgery, The Second Affiliated Hospital to Inner Mongolia Medical University. They were 14 men and 2 women with a mean age of 38.4 years (range, from 20 to 55 years). All fractures were fixed with hollow compression screws through the approach for malleolus medialis Ⅴ osteotomy. The ankle and hindfoot functional scoring system developed by American Orthopaedic Foot and Ankle Society (AOFAS) was used to evaluate the clinical outcomes.
Results:
All patients were followed up for a mean time of 12.6 months (range, from 6 to 30 months). The mean operation time was 68.4 minutes (range, from 52 to 96 minutes); the mean amount of hemorrhage during operation was 96.8 mL (range, from 48 to 122 mL); the mean period of bone union was 4.8 months (range, from 3 to 8 months). The postoperative mean AOFAS score was 75.3 points (range, from 43 to 91 points). Complications occurred in 4 cases, including one case of talus ischemic necrosis, one case of partial talus ischemic necrosis accompanied by tibial arthritis, one case of subtalar arthritis, and one case of combined tibial, talar and subtalar arthritis. All incisions obtained primary healing, with no complications like infection, screw breakage, delayed union or nonunion.
Conclusion
Operative treatment of Sneppen Ⅴ talus fracture through the approach for malleolus medialis Ⅴ osteotomy plus hollow compression screw fixation can provide sufficient operative exposure to facilitate reduction and fixation of the talus fracture so that the ischemic necrosis of the talus and traumatic arthritis can be effectively reduced.
