1.Application of chitosan-gold nanoparticles hybrid film biosensor in glucose measurement

BACKGROUND:At present,syndromes happening frequency of diabetes mellitus can be controlled through regulating glucose concentration in blood.Therefore,a fast,steady,accurate glucose biosensor has been an intense area of focus for investigators.OBJECTIVE:To observe the effect of chitosan-gold nanoparticles hybrid film on electrochemical glucose biosensor.DESIGN,TIME AND SETTING:The observation experiment was performed at the Department of Medicine Laboratory,General Hospital,Guangzhou Military Command of Chinese PLA from February to April 2009.MATERIALS:Based on the immobilization of glucose oxidase (GOD) with cross-linking in the matrix of chitosan on an Au electrode by glutaraldehyde,a novel glucose biosensor was constructed with gold nanoparticles by electrodeposition.METHODS:The properties of CHIT/AuNPs hybrid film modified electrode were characterized by cyclic voltammetry (CV) in 20 mmol/L K3Fe (CN)6-0.1 mol/L KCI system by using the three-electrode system and the response to glucose was investigated by linear scan voltammetry (LSV).The optimal experiment conditions,stability and reproducibility about the glucose biosensor were also studied.MAIN OUTCOME MEASURES:The cyclic voltammetry properties of different modified electrode,as well as relation between glucose concentration and response current.RESULTS:Glucose concentration showed a linear correlation with response current in range of 0.001-6 mmol/L.The biosensor exhibited excellent performances of producibility,stability and the ignorance interferences of reducing substances,with the CONCLUSION:The glucose biosensor possesses excellent reproducibility and stability with a fast response time to glucose,which can be used in detection of glucose concentration in human serum.

Objective To construct a kind of novel electrochemical glucose biosensor for clinical detection.Methods Multiwall carbon tube(MWCNTs),NanoAu and glucose oxidase(GOD) were immobilized on the Au electrode by monolayer self-assemble.Cyclic voltammetry and linear scan voltammetry were used to investigate the assembling process and the chemical characteristics of the glucose biosensor.Results The linearity of glucose in the range of 10~1 400 mg/L with the correlation coefficient of R2=0.990 9 was obtained by linear scan voltammetry and the detection limited reached 0.000 1 mg/L.The four criterion samples were measured with 3 different biosensors in same conditions and the RSD
4.Investigation of optimum exposure dose for chest imaging using CR and amorphous silocon DR system

Objective To compare the difference of entrance dose between CR and amorphous silocon DR system in chest imaging, and to discuss their optimum exposure dose. Methods For CR and DR, different entrance dose was measured by dosimeter in chest phantom. The value of IQFinv was analyzed by CDRAD2. 0 software. Image quality difference between CR and DR was assessed by group t-test. The relationship between image quality and entrance dose was tested by using Pearson correlation analysis. The best IQFinv values in CR and DR system were achieved via ROC curve analysis, and the exposure dose was then calculated. Results There were direct correlation values between entrance dose and the value of IQFinv in CR and DR system, respectively( r =0. 893 ,0. 848 ,P < 0. 01 ) . The linear regression equation for DR was IQFinv =0. 0050 +3. 359, and for CR was IQFinv =0. 005D + I. 651 , where D was entrance dose. The difference of IQFinv value between CR and DR was significant(t = 5. 455 ,P < 0. 05). The best IQFinv value of the two groups from ROC analysis was 3.55. Conclusions With the entrance dose increased, the detection ability of contrast-detail was elevated in the two digital radiography systems. With equal entrance dose, the detection ability of DR in contrast-detail was superior to CR. With equal image quality, DR obviously decreased the radiation dose to the patients.
5.The expressions of CDC4 and c-Myc in gastric cancer and their clinical signifieance

Background and purpose:The gastric cancer is the highest incidence of malignant tumors in the world. The main treatment methods for gastric cancer are operation and chemotherapy. But the effect is not good. With the rapid development of economy and molecular biology, early diagnosis and molecular targeted therapy for gastric cancer has become a research hotspot. The oncogene overexpression and the anti-oncogene lower expression are closely related with gastric cancer.CDC4/FBXW7 is an anti-oncogene, butc-Myc is an oncogene. The previous research showed that CDC4 affected the expression of many oncogenes, such as Cyclin E. This study aimed to investigate the expression of CDC4 and c-Myc in gastric cancer and to elucidate the potential relationship between their expressions and clinical pathological characteristics.Methods:Semi-quantitative reverse transcription polymerase chain reaction (sRT-PCR), immunohistochemistry and Western blot method were used to determine the mRNA and protein expressions of CDC4 and c-Myc in 40 specimens of gastric carcinoma tissues, corresponding adjacent tissues and normal mucosal tissues. The expressions of CDC4 and c-Myc and the clinical pathological characteristics were analyzed.Results:The protein expressions of CDC4 in gastric cancer tissues were signiifcantly lower than those in adjacent tissues and normal mucosal tissues (P<0.05), whereas the protein expression of c-Myc in gastric cancer tissues was signiifcantly higher than that in adjacent tissues and normal mucosal tissues (P<0.05). The protein and mRNA expression of CDC4 and c-Myc were correlated with differentiation, TNM stage, lymph node metastasis, inifltration, but not with patients’ gender, age and site of cancer (P<0.05). There was a signiifcant negative correlation between CDC4 and c-Myc at the mRNA and protein expression levels (P<0.05).Conclusion:The lower expression of CDC4 is correlated with differentiation, TNM stage, lymph node metastasis and inifltration. c-Myc overexpression is likely to be the CDC4 loss. It suggests that the loss of CDC4 may be a valuable marker for assessing the diagnosis and treatment and the prognosis of gastric cancer.
6.Venous Catheter Infection in Malignancy Patients and Its Countermeasures

OBJECTIVE To make the patients using the central venous catheter safely when given chemotherapy and intravenous nutritional therapy,and to decrease the catheter infection by establishing effective infection-preventing systems. METHODS Many infection-preventing actions were carried out,including a three-level management,improving the workers′ training in catheter operation and maintenance,and health education. RESULTS Comparing with the matched group,the infection rate of the experimental group was much lower after the actions.The actions prevented the increase of infection effectively. CONCLUSIONS Building and carrying out the systems are proved to be important measures to decrease the catheter infection and ensure the quality of life among patients during the therapy in hospital.
7.IMMUNOHISTOCHEMICAL STUDY ON G AND D CELLS IN PANCREATIC ISLETS OF RAT DURING EXPERIMENTAL GASTRIC ULCER

Objective Studies on the changes of gastrin and SS immunoreactive cells in pancreatic islets during experimental gastric ulcer. Methods The immunohistochemical ABC technique was used. Results The gastrin immunoreactive cells were located in most of the pancreatic islet. The mumber of G cells in experimental gastric ulcer group were higher than that of control group on the 4th and 10th day after operation.The D cells raised on the 10th day.Conclusion The present work provides the evidence that the G and D cells of pancreatic islets might be involed in the self-healing process of the experimental gastric ulcer by endocrine or paracrine regulation.
8.Action of Tuina on Retro-positioning of Skull Spatial Offset in Patients with Cervical Vertigo

Objective:To investigate the onset mechanism of cervical vertigo from the proprioceptive sensation and provide clinical basis for its treatment.Methods:Among the 121 cases that conformed to the diagnostic criteria of cervical vertigo,70 cases who presented with negative neck-rotation test and cervical vertigo without obvious vascular factors by transcranial Doppler(TCD)were assigned to the observation group,while 51 cases who presented with positive neck-rotation test and cervical vertigo due to spasm of vertebral basal artery or insufficient blood supply by TCD were assigned to the control group.The cases in the two groups were treated once every day,5 days make up one treatment course and the resuIts were statistically analyrzed after one treatment course.Results:The skull triaxial spatial offset of the cases in the two groups were significantly reduced after the treatment (P<<0.01).However, there was no statistically significant difference between the skull triaxial spatial offset between the two groups (p>0.05).It is not conclusive that the therapeutic effect in the two groups was significantly different after one treatment course.Conclusion:Tuina manipulation therapy can improve the skull spatial ofrset repositioning ability of the Patients.
9.Treatment of tibial fractures through compression reduction and osteosynthesis with common plate

Objective To evaluate clinical effects of the treatment of tibial fractures through compression reduction and osteosynthesis with common plate. Methods From March 2001 to September 2003, 48 cases of tibial fractures were treated through compression reduction and osteosynthesis with common plate. Results All the cases were followed up for 6 to 24 weeks (averaging 14 weeks). The healing time was 12 to 16 weeks (averaging 5 weeks). No delayed union, non union, infection, or ineffective internal fixation occurred. Conclusion Compression reduction and osteosynthesis with common plate to treat tibial fractures is a kind of biomechanical fixation that facilitates fracture healing.
10.Expression of soluble PD-1 in the sera of patients with recurrent genital herpes and its immunologic significance

Objective To determine the concentration of soluble PD-1 (sPD-1) in the sera of patients with recurrent genital herpes (RCH), and to explore the significance of abnormal expression of sPD-1 in RCH. Methods Serum samples were obtained from 88 healthy blood donors, 74 patients with RCH including 34 cases of outbreak-stage RCH and 40 cases of stable-stage RCH. The serum level of sPD-1 was measured by monoclonal antibody labeling and enzyme linked immunosorbent assay (ELISA). Results A significant difference was observed in the serum level of sPD-1 between the patients with RGH and the blood donors (33.06 ± 17.5 μg/L vs. 53.07 ± 26.3μg/L, P < 0.01) and between the patients with outbreak-stage RGH and those with stable-stage RGH (27.47 ± 12.9 μg/L vs. 37.71 ± 19.6 μg/L, P< 0.01). Conclusions There is a low expression of sPD-1 in patients with RGH, which may be one of the mechanisms underlying the escape of HSV- Ⅱ from immunologic surveillance and development of immunological tolerance.
