Objective:To study the effect of airway IL-4RA gene transfer on asthma-associated expression of STAT6 in the mice of allergic asthma. Methods:Forty BALB/c mice were randomly divided into four groups:blank control, asthma model, asthma model administered by pLNC-laz,and asthma model administered by glucocorticoid and pLNC-IL-4RA.The ten mice in each groups were sensitized by i.p.injection of 10 ?g of OVA emulsified in 40 mg of aluminum hydroxide in a total volume of 200 ?l on the days of 1st,7th and 14th.On the 15th day,the mice were challenged via airways with OVA for 7 days,20 min a day, blank control were sensitized by i.p.injection of 200 ?l pH=7.4 PBS on days 7 and 14.Asthma model was administered by pLNC-laz or pLNC-IL-4RA for 3 days beginning at the 12th day.The mice of the group with glucocorticoid was administered with glucocorticoid on the 15th day for 7 days.24 hours after the last challenge, the mice were sacrificed by bloodletting.Serum OVA-specific IgE and blood Eosinophils were measured. The lungs were fixed by immersion in 10% formalin.Expression of STAT6 was identified by immunohistochemistry. Results:It was shown that pLNC-IL-4RA was integrated to the genomic DNA of pneumonic tissues.Retrovirus-mediated delivery of IL-4RA to airways of the asthma mice weakened airway eosinophilia triggered by either IL-13 or IL-4.Furthermore,IL-4RA delivered by retrovirus was expressed in airways of the mice with allergen sensitization, resulting in significiant reduction of expressing level of asthma-associated STAT6 in the experimental mice of allergic asthma. Conclusion:Retrovirus mediated delivery of IL-4RA to airways reduces expression of STAT6 in pneumonic tissues of asthmatic mice. Thus, the gene theraphy can be a potential therapeutic option to treat and control chronic airway inflammation and asthmatic symptoms.

Objective To explore the application of domestic bladder capacity scanner in intermittent catheterization in patients with neurogenic bladder. Methods 40 patients with neurogenic bladder accepted intermittent catheterization. Their bladder volume was tested with domestic and imported bladder capacity scanner (Padscan and Bladderscan) and catheterization respectively. Results The bladder volume was (262.592±6.3012) ml tested by Padscan, and was (262.570±6.3006) ml by Bladderscan (P>0.05). The bladder volume tested by bladder catheterization was (245.549±6.0511) ml, which was significantly lower than the other methods (P<0.001), but the D-value was just 17 ml. Conclusion The accuracy of bladder volume tested by domestic bladder capacity scanner (Padscan) was very similar with the imported one (Bladderscan), and the difference between them and the volume tested by bladder catheterization was meaningless in clinical.

Objective To study the immunosuppressive mechanism of alkaloid sinomenine (SIN) by observing the effects of SIN on the proliferation and intracellular protein expression levels of nuclear factor of activated T cells (NF-AT) and interferon-gamma (IFN-γ) in CD4+ T lymphocytes of human periphery blood. Methods CD4+ T lymphocytes were isolated from PBMC suspensions with immunomagnetic beads and divided into five groups to culture. (1) Negative control group: no medicine was added to cell culture medium; (2) Positive control group: CsA solution (final concentration: 50ng/ml) was added to cell culture media; (3) Low-concentration SIN group (L-SIN): low-concentration SIN solution (final concentration: 10 μmol/L) was added to cell culture media; (4) Middle-concentration SIN group (M-SIN): middle-concentration SIN solution (final concentration: 200 μmol/L) was added to cell culture media; (5) High-concentration SIN group (H-SIN): high-concentration SIN solution (final concentration: 1000 tmol/L) was added to cell culture media. The proliferations of CD4+ T lymphocytes were observed. Western blotting was performed to detect the protein expression levels of NF-AT. FCM was used to determine the levels of IFN-γ. Results Compared with negative control group, the cell proliferation was significantly inhibited in H- and M-SIN groups (P<0. 01 ). SIN concentration-dependently inhibited the protein expression levels of NF-AT and IFN-γ in CD4+ T lymphocytes of human periphery blood (P<0.01). The protein expression levels of NF-AT and IFN-γ were lowest in positive control group. There was a close negative correlation between intracellular levels of NF-AT and cell proliferation inhibition ratio in CD4+ T lymphocytes of human periphery blood (rs = - 0. 969, P = 0. 000). Conclusion SIN can inhibit the protein expression of NF-AT and IFN-γ in CD4+ T lymphocytes of human periphery blood probably by decreasing protein levels of NF-AT to inhibit the activity and proliferation of CD4+ T lymphocytes.
