1.Galvanic corrosion between Ti implants and implant superstructure dental alloys

BACKGROUND: Most of the dental metal repair materials contacting Ti implants will cause galvanic corrosion and local environment with positive electricity on the implant interface, which directly affects the status of the tissues, especially bone resorption.OBJECTIVE: To evaluate the galvanic corrosion of commercially pure Ti (TA2 type) coupled with Au alloy, CoCr alloy, Ti alloy, and NiCr alloy in vitro. METHODS: Circuit of commercially pure Ti contacting with Au alloy, CoCr alloy, Ti alloy, and NiCr alloy was simulated in vitro in artificial saliva, and the common potential and galvanic current in 15 hours were recorded to draw current-time curves. RESULTS AND CONCLUSION: The galvanic current became stable after 8 hours of contacting. The galvanic current was the greatest in Ti/Au, followed by Ti/CoCr, Ti/Ti alloy, and then Ti/NiCr. Results indicated that the galvanic corrosion of the couple of Ti and Au alloy is the lowest. Au alloy is the most suitable material for implant superstructure. The galvanic corrosion of the couple of Ti and NiCr alloy is the highest, so NiCr alloy is not acceptable for implant superstructure.

This information management system standardizes management for qualification documents of medical instruments, and ensures that qualification documents of medical instruments are available and can be traced. Besides, it can reduce the amount of work for medical instruments management.

5.Effects of electro-acupuncture on Jiaji acupoints combined with treadmill training on tibia bone mass of rabbit with sciatic nerve injury

Objective To observe the effects of electro-acupuncture on Jiaji acupoints combine with treadmill training on tibia bone mass of rabbits with sciatic nerve injury,so as to provide the theoretical support for better project to maintain normal metabolism of bone after peripheral nerve injury.Methods A total of 24 New Zealand rabbits were randomly divided into four groups (n =6),namely a model group,a sham group,a treadmill group and an electro-acupuncture + treadmill group according to a random number table.The sciatic nerve of rabbits in the model,treadmill and electro-acupuncture + treadmill group was clamped using hemostatic forceps to establish the model of sciatic nerve injury,while that of the sham group was not clamped,just cut soft tissue around the sciatic nerve.Three days after modeling,the rabbits in treadmill group and the electro-acupuncture + treadmill group were given treadmill training and electro-acupuncture combine with treadmill training respectively,lasting 4 months,whereas those in the sham and the model groups were not given any intervention.Right after modeling,the behavior of rabbits in four groups was observed.Four weeks after treatment or feeding,their bone mineral density and bone strength of tibia were measured.Results Four weeks after treatment,there was significant reduction in bone density and bone strength of tibia in the model,the treadmill and the electro-acupuncture + treadmill groups compared with those in the sham group(P < 0.05) ; the bone mineral density and bone strength of tibia in the electro-acupuncture + treadmill group were 0.17 ± 0.01 g/cm2 and 161.92 ± 43.27 N respectively,significantly higher than those in both the model and the treadmill groups (P < 0.05).Conclusion Electro-acupuncture combined with treadmill training can improve tibia bone mineral density and bone strength after sciatic nerve injury and facilitate the normal metabolism of the bone and to maintain its normal function.
6.Effects of acute knockdown of hepatic insulin receptor on secretion of apolipoprotein B and triglyceride in mice

Objective To analyze the effects of acute depletion of liver-specific insulin signaling on secretion of apolipoprotein B (apoB) and triglyceride (TG).Methods Based on Cre-LoxP principle,a promoter of hepatic tissue specific albumin gene was inserted into upstream of the cre recombinase gene.Albumin-Cre adenovirus (Ad-CRE) and GFP adenovirus (Ad-GFP) were amplified in 293A cells and purified before intravenous administration to mice.After adenovirus infection for 2 days,4 days and 6 days,blood samples from mice were collected and hepatic tissues were frozen.The secretion rates of hepatic newly synthesized apoB and very low density lipoprotein (VLDL)-TG were determined by injection of Triton WR-1339.The levels of plasma cholesterol (TC) and TG were measured.The expressions of insulin receptor and other lipoprotein metabolism related proteins in hepatic tissues were analyzed by Western blot.Results After 2 d,4 d and 6 d of the Ad-CRE injection into mice,insulin receptor expression was reduced by 30.50% (P<0.05),60.12% (P< 0.01) and 99.54% (P<0.001),and VLDL-TG secretion rate was decreased by 20.43% (P<0.05),33.63% (P<0.05) and 44.21% (P<0.01),respectively.Expressions of sterol regulatory binding proteins 1,fatty acid synthase,and the related proteins of VLDL-formation were decreased,but there were no changes in hepatic secretion of apoB100 and hepatic lipids.The hepatic secretion of apoB48 was increased by 35.07% (P<0.05) 6 d after Ad-CRE injection.Conclusions Acute depletion of hepatic insulin receptor might reduce VLDL-TG secretion in manner of time-dependent,and increase the assembly and secretion of smaller apoB-containing lipoproteins in mice liver,which is probably associated with decreased lipogenesis.
7.Establishment of quantitative measurement method of cell migration in vitro and it's application

AIM: To establish a quantitative measurement method of ECV-304 cell migration in a scratch wound model in vitro. METHODS: The method of ECV-304 cell scratch wound model was improved. The ECV-304 cell migration was measured quantitatively using a computer-assisted video microscopic method with image-analysis system. The effects of heparin and thrombospondin-1 on ECV-304 cell migration were observed in the scratch wound model in vitro. RESULTS: The average width and length of cell scratch wound was (0.95?0.08) mm and (51.20?3.40) mm respectively in 120 experiments. The peak of cell migration reached about 24 h after culture. The stimulated effect of heparin and inhibited effect of thrombospondin-1(TSP-1) on cell migration were observed. CONCLUSION: A quantitative measurement method of ECV-304 cell migration in vitro is established. The stimulated and inhibited effects on ECV-304 cell migration were proven respectively using this method.
8.Diagnosis and treatment of 81 patients with primary gastrointestinal lymphoma

Objective To analyze the status quo of the diagnosis and treatments of primary gastro-intestinal lymphoma (PGIL) in order to improve it. Methods Eighty-one patients with PGIL were ana-lyzed retrospectively including clinical manifestations, endoscopic features, pathological features, HP in-fection, treatment, and prognosis. Results The age of patients with gastric lymphoma was (52.84±15.33) years. The age of patients with intestinal lymphoma was (42.09±15.28) years. Common symp-toms included abdominal pain (76.5%), gastrointestinal bleeding (55.6%), anemia (54.3%), abdominal mass (25.9%), hypoproteinemia (40.7%), bowel obstruction (11.1%), abdominal dis-tension, vomiting, and other non-specific gastrointestinal symptoms (32.1%), weight loss (33.3%); fever (8.6%), diarrhea (7.4%), digestive tract perforation (1.2%), constipation (1.2%), and dysphagia (1.2%). Endoscopic appearances were as follows: tumor type (67.7%), ulcer type (27.7%), and diffuse type (4.6%). Clinical diagnosis rate and endoscopic biopsy confirmation rate were 30.9% and 73.8%. MALT lymphoma accounted for 61.7% of the patients. HP detection rate was 39.5 % and positive rate was 37.5 %. A total of 69 patients received surgeries: 3 had preoperative chem-otherapy, and 34 had postoperative chemotherapy. Twelve patients had non-surgical treatment, 6 patients of whom had simple chemotherapy and HP eradication therapy, and the other 6 gave up during the treat-ment. There was no significant difference in the survival rate of Stage Ⅰ~Ⅱ patients in the surgery alone group, surgery plus chemotherapy group, and chemotherapy and HP eradication therapy group (P>0.05). The survival rate of Stage IIIⅢ~Ⅳ patients in the surgery alone group was lower than that in the other 2 groups (P<0.05). The 5-year, 3-year, and 1-year survival rate was 55.87%, 70.96%, and 96.39%, respectively. Conclusion There are no specific clinical and endoscopic features in PGIL, so the misdiagnosis rate is high. Multi-site biopsy or repeated biopsies and immunohistochemical methods can be used to raise the pathological diagnosis rate. Chemotherapy and HP eradication are recommended.
9.Ultrasonographic evaluation of osteosarcomas.

The ultrasonographic appearances of osteosarcomas and the roles of ultrasonography in the diagnosis and surgical staging of osteosarcomas were investigated. A comparative study was performed on 45 cases of osteosarcomas by ultrasonography and radiography. Bony changes, periosteal reaction and soft tissue mass were evaluated for each lesion. The results showed that ultrasonography revealed a solid mass around bone in 42 patients, bone destruction in 24 patients and periosteal reaction in 16 patients. Plain radiographs showed bony changes in 44 patients and no bony change in remaining one patient, shadowing of soft tissue swelling in 30 patients, and pulmonary metastases in 3 patients. Surgical biopsy and pathological examination confirmed osteosarcoma in all 45 patients. Soft tissue mass was confirmed in 42 patients surgically. The diagnostic accuracy of soft tissue masses by ultrasonography and radiography was 100 % (42/42) and 71.4 % (30/42), respectively. The positive rate of ultrasonography and radiography in displaying bony changes was 53.3 % (24/45) and 97.8 % (44/45), respectively. In conclusion, in the detection of soft tissue mass of osteosarcoma, ultrasonography is superior to radiography, and in displaying bony changes of osteosarcomas, radiography is superior to ultrasonography. So it may come to a conclusion that plain radiography combined with ultrasonography can completely display the bony and soft tissue lesion of osteosarcomas.
10.11 beta-hydroxysteroid dehydrogenase type 1 involved in tumor necrosis factor-a-induced insulin resistance

Objective To evaluate the molecular mechanism of 11 beta-hydroxysteroid dehydrogenase type 1 (11 β-HSD1)in tumor necrosis factor-α (TNF-α) induced insulin resistance.Methods The optimal concentration and incubation time of TNF-α for treating HepG2 cells were selected.Real-time polymerase chain reaction(PCR)and Western blot analysis were used to explore the effect of TNFα on the mRNA and protein expression levels of 11β-HSD1,glucocorticoid receptor (GR),phosphoenolpyruvate carboxylase(PEPCK)and glucose 6-phosphatase(G6Pase).Glucose(GO) Assay Kit was used to determine the level of gluconeogenesis.Liver/Muscle glycogen assay kit,PAS/Glycogen Stain kit were used to detect glycogen synthesis.The expression of AKT/GSK pathway was analyzed by Western blot analysis.In addition,HepG2 cells were pre-treated with BVT2733,a specific inhibitor of 11β-HSD1,before exposure to TNFα The expression of AKT/GSK pathway,the level of gluconeogenesis and glycogen synthesis were analyzed.Results The expression of 11β-HSD1 was increased along with the increasing concentration of TNF-α,showing a dose-dependent effect.When HepG2 cells treated with the optimal dose of TNF-α 10 μg/L for 24 hours,the levels of GR,gluconeogenesis and gluconeogenesis-related genes PEPCK and G6Pase were increased,while conversely,the levels of phosphorylation of AKT and GSK,and glycogen synthesis were decreased.However,all the above effects induced by TNF-α in HepG2 cells were greatly reversed by BVT2733 pre treatment.Conclusions 11 β-HSD1 participates in TNF-α-induced insulin resistance in HepG2 cells,and BVT2733 could significantly improve insulin sensitivity.
