1.Effect of Serum Containing Bushen Prescription on Chondrocyte Proliferation

[Objective] To observe the effect of serum containing Bushen prescription (BP) on inhibition of chondrocyte proliferation and nitric oxide (NO) production in chondrocyte induced by interleukin-1 (IL-1). [Methods] Four New Zealand rabbits were administered BP 2.5mL (1.15kg/L) for 3 days for the preparation of BP-containing serum. Rabbit chondrocytes were cultured in-vitro and were added with DMEM culture containing IL-1 1, 10 and 20 ?g/L to examined the inhibition concentration of IL-1 for chondrocyte proliferation. After then, BP group was added with 20 ?g/L IL-1 and various concentrations of BP-containing serum and the model group with 20 ?g/L IL-1 and various concentrations of blank serum. Alter 3-day culture, chondrocyte proliferation were dectected by thiazolyl blue colorimetry (MTT) and NO content by Griess method. [Results] The inhibition concentration of IL-1 for chondrocyte proliferation was 20 ?g/L; various concentrations of BP-containing serum could counteract chondrocyte proliferation and inhibit NO pnxluction in chondrocyte induced by IL-1. [Conclusion] BP can counteract chondrocyte proliferation directly or indirectly by the mediation of NO, which may be one of its therapeutic mechanisms in treating osteoarthritis.
2.Detection of adhesion molecules in bone marrow metastatic tumors of digestive system.

Objective To evaluate the role of 4 types of adhesion molecules in digestive system tumors with bone marrow metastasis.Methods The fresh bone marrow slies of 72 cases of digestive system tumor with bone marrow metastasis,including 26 gastric cancer,25 liver cancer,21 colorectal cancer,were obtained.The tumor cell slides of 76 cases of digestive system tumor without metastasis,including 32 gastric cancer,20 liver cancer,24 colorectal cancer,were obtained.The expressions of 4 kinds of adhesion molecules in two groups were studied by ABC immunohistochemical methods,and positive rate was recorded and accumulated points.Results ICAM-1、E-cad and CEA positive rates/CD44V6 accumulated points were up-regulated in gastric cancer cells in bone marrow;ICAM-1、CD44V6 accumulated points/CEA positive rate were up-regulated in liver cancer cells in bone marrow,while expression of E-cad had no difference;ICAM-1、CD44V6 and CEA accumulated points/E-cad positive rate were up-regulated in large bowel cancer cells in bone marrow.Conclusion Different adhesion molecules play coordinative or resistant role in digestive system tumor with bone marrow metastasis.
3.Relationship between multiple adhesion molecules expression in gastric carcinoma and organic selection of gastric carcinomatous metastasis.

Objective To evaluate the effect of multiple adhesion molecules ICAM-1、CD_ 44 V6、CD_ 28 、E-cad and CEA in organic selection of gastric carcinomatous metastasis.Methods Eighty-four gastric carcinoma cases(32 cases without metastases,52 cases with transfer to liver、marrow、lymphonode、lung、kidney or belly cavity),were take operation sample or tissue under gastroscopy,being made cells single by primary culture,then were detected the expression of 5 kinds of adhesion molecules by ABC immunohistochemistry,positive rate was recorded.Results ICAM-1 had high expression in cases with marrow or lung metastasis,while low expression in cases with lymphonode metastasis.CD_ 44 v6 had high expression in cases with lymphnode or liver metastasis.CD_ 28 had high expression in cases with liver、lung or kidney metastasis.E-cad had high expression in cases with marrow metastasis,while low expression in cases with other metastasis.CEA had especially high expression in cases with liver、belly cavity or lymphnode metastasis.Conclusion High or low expression of adhesion molecules ICAM-1、CD_ 44 V6、CD_ 28 、E-cad and CEA plays a certain role in organic selection of gastric carcinomatous metastasis.
4.Disease constitution analysis of 4 590 children with abnormalities in the blood cells

Objective To investigate the significance of bone marrow cell morphology on clinical diagnosis of children hematopoietic diseases.Methods The data of bone marrow cell morphology in the bone marrow puncture specimens from 4 590 children admitted to Children' s Hospital of Shanxi Province from January 2009 to December 2014 were retrospectively analyzed.Results The proportion of infancy patients was the highest in 4 590 bone marrow specimens, accounting for 29.0 % (1 333/4 590), then that of the toddler age patients was second highest, accounting for 26.7 % (1 224/4 590).The constituent ratio of thrombocytopenic purpura (ITP) at the different ages was the highest.The most common diseases in bone marrow cell morphology diagnosis were in order of ITP, iron deficiency anemia (IDA), infectious bone marrow, leukemia (acute lymphoblastic leukemia, acute myeloid leukemia), aplastic anemia and so on.Conclusions Accurate analysis of bone marrow cell morphology is still the most basic and rapid approach in children with hematopoietic system disease, which has important value.Except hematopoietic system diseases, once fever of unknown origin, hepatosplenomegaly and enlargement of lymph nodes the patients should be early given bone marrow cell morphology check, early diagnosis and therapy.
5.Retinoic acid incubation effect on proliferation of U251 cell line and effect on MAPK signal pathway

Objective To investigate the retinoic acid incubation effect on proliferation of U251 cell line and effect on MAPK signal pathway. Methods ATRA solution of different concentration on the U25 1 glioma cells were incubated,the influence of ATRA on the proliferation of U25 1 cells were detected,and the proteins of MKPs and MAPK signaling pathways were detected by qRT-PCR and Western blot.Using Graph Prism 5 software for quantitative analysis of experimental results.Results Compared with control group,ATRA could effectively inhibit the proliferation of U25 1 glioma cells, in a concentration dependent manner.QRT-PCR results showed that,different concentrations of ATRA after incubation for 48 hours,the expression of MKPs mRNA changed,but the changes of MKP-5 and expression of 67LR was different,explained the main differences between the two methods of the MAPK signaling pathway was the regulation of MKP-5.Western blot results showed that the ATRA,after 48 hours of incubation,the protein MAPK pathway had changed in phosphorylation, which showed that ATRA protein in the MAPK signaling pathway through control of the degree of phosphorylation on U25 1 cell line regulation.Conclusion Retinoic acid and retinoic acid receptor play its physiological effects and regulate human glioma cell line U25 1 proliferation through different combination.Retinoic acid could not only reduce the expression of phosphorylated ERK1/2 to inhibit tumor proliferation,but also regulate three kinds of protein phosphorylation,therefore its mechanism will be more complex,at the same time that the MAPK signaling pathway plays a crucial role in tumor proliferation process.
6.Detection of specific antibody IgM of Lyme disease by ELISA using recombinant outer surface protein C of borrelia burgdorferi

Objective To detect specific IgM of Lyme disease indirect ELISA using recombinant outer surface protein C(OpsC)of Borrelia burgdorferi in vitro was established. Methods Coated concentration of recombinant OspC and dilution multiple of serum anol concentration of enzyme secondary antibody were determined by block,and degree of percision.specificity interference and interruption test were performed. Results Best concentration of OspC was 150 μg/L.within-run CV was 6.3% between-run CV was 11.8%.Confimred 33 Lyme patients in clinic and 57 controls were examined meanwhile by this method and import ELISA kit,coincidena rate was 97.8%.Conclusion This ELISA using recombinant OspC was a good method for early diagnostic of Lyme disease.
7.Analysis of treatment model for stroke in 1200 elderly inpatients

Objective To compare the differences in curative effect between general treatment model and stroke unit model in the treatment of stroke in the elderly inpatients, to explore the standardized management model, for improving efficacy and reducing the degree of invalidity and mortality. Methods A total of 1200 cases with acute cerebrovascular disease were admitted into our department from April 2002 to April 2007. All patients were randomly assigned either into stroke unit or general ward to receive treatment, and the data of two groups were compared. The key indexes included mortality, hospitalization time, Barthel Index (BI), National Institutes of Health Stroke Scale (NIHSS) and Oxford Handicap Scale (OHS). All the patients or their family members were asked to have the Visual Analogue Scales (VAS). The data of cost and effect were analyzed with SPSS software. Results The complication rate and mortality rate were significantly lower in stroke unit group than in general ward group (P< 0.01). The scales of BI, NIHSS, OHS and VAS were significantly better in stroke unit group than in general ward group when discharged (P<0.05). The hospitalization time was shorter in stroke unit group than in general ward group (P<0.05). There was no significant difference in the costs of hospitalized patients between the two groups (P>0.05).Conclusions Stroke unit model can improve the physical activity and life quality of patients and reduce disease mortality without increasing the cost of patients.
8.Extraction Technology Optimization of Total Flavonoids in Geum Aleppicum by Box-Behnken Design and Response Surface Methodology

Objective:To analyze and optimize the extraction technology of Geum aleppicum by Box-Behnken design and response surface methodology. Methods:With the content of total flavonoids as the index, the effects of three factors including the solvent-solid ratio, ethanol concentration and extraction time on the content of total flavonoids were studied by single factor experiment, and the ex-traction technology was optimized by Box-Behnken design and response surface methodology. Results: The optimal extraction condi-tions of Geum aleppicum were as follows:the solvent-solid ratio was 4. 2, the ethanol concentration was 50% and the extraction time was 80 min. Under the above conditions, the average yield was 12. 590 0 mg·g-1 . Conclusion: Optimizing extraction technology of the total flavonoids in Geum aleppicum by Box-Behnken design and response surface methodology is reasonable, and the mathematical mod-el is consistent with the experimental data, which has good predictability.
9.Effects of electroacupuncture at points selected by orthogonal experiment on the extracellular signal regulated kinase signal pathway in a rat model of cerebral ischenia-reperfusion injury

BACKGROUND:Acupuncture point matching is little reported in the electroacupuncture for cerebral ischemia-reperfusion injury.
OBJECTIVE:To explore the influence of electroacupuncture acupoints selected by orthogonal experiment on rats with cerebral ischemia reperfusion and extracel ular signal regulated kinase (ERK) signal pathway.
METHODS:150 Sprague-Dawley rats were randomly divided into five groups:sham operation group rats were exposed the middle cerebral artery only;models of cerebral ischemia reperfusion were established in the rats in model and orthogonal groups. Then rats in the orthogonal groups 1 (Baihui (DU 20), Quchi (LI 11), Zusanli (ST 36), and Chize (LU 5)), 2 (Baihui (DU 20), Zusanli (ST 36), Chize (LU 5), and Sanyinjiao (SP 6)) and 3 (Hegu (LI 4), Zusanli (ST 36), Chize (LU 5) and Sanyinjiao (SP 6)) were given electroacupuncture at acupoints selected by orthogonal experiment, respectively.
RESULTS AND CONCLUSION:Compared with the model group, in al orthogonal groups, the neurological function deficit scores were decreased, infarct volume was smal er, the number of microvessel density in the ischemia region and the p-ERK expression level in the cortex and hippocampal CA1 region of ischemic hemisphere were increased, and significant changes were found in the orthogonal group 3. These findings indicate that electroacupuncture at points of Hegu (LI 4), Zusanli (ST 36), Chize (LU 5) and Sanyinjiao (SP 6) selected by orthogonal experiment can significantly reduce the infarct volume of cerebral ischemia-reperfusion injury in rats possibly by regulating the ERK signaling pathway.
10.Concentration Screening and Antimicrobial Effectiveness of Antimicrobial Preservative for Fluconazole Eye Drop

Objective:To explore the reasonable amount of bacteriostatis agent in fluconazole eye drop. Methods: According to the method of bacteriostatis effect test in 2015 edition of Chinese Pharmacopoeia (ChP), and Staphyococcus aureus, Pseudomonas aeruginosa, Escherichia coli, Candia albicans and Aspergillus niger as the test strains, the bacteriostatis effect of fluconazole eye drop containing 5%, 1%, 0. 3% and 0. 1% benzalkonium chloride was determined. Results: The bacteriostatis effects of fluconazole eye drop containing 5%, 1% and 0. 3% benzalkonium chloride on the five species of tested bacteria complied with the regulations of ChP. The bacteriostatis effects of 0. 1% benzalkonium chloride on Pseudomonas aeruginosa didn't meet the requirements, therefore, 0. 3%was the minimum inhibitory concentration ( MIC) of benzalkonium chloride. Conclusion: The best inhibitory concentration of benza-lkonium chloride is 1%.
