1.Development of vocational interest scale of university students

Objective To develop a Vocational Interest Scale of University Students which fits the contemporary current university students, and it could give directions to university students when they were choosing a vocation. Methods Taking Holland's theory as the guide and taking 65 items of Self-Directed Search (SDS) for reference, then adding 28 items which standed for contemporary current university students' ideas of vocational interest to develop original Vocational Interest Scale of University Students. Results The formal scale included 7 tendency(subscales) and 61 items,7 subscales were technology tendency, administration tendency, business trade tendency ,art tendency ,investigation tendency,transaction tendency. The reliability of the scale was very good, the Cronbach's coefficients and spit-half reliability were over 0.7. The factor analysis abstracted 7 factors and the total variance explained reached 45%. The factor analysis results indicated that the scale had a good structure.Conclusion This scale make vocational interest as measuring object and could be a useful tool to give directions to university students who are choosing some vocation.
2.Personality traits on higher vocational students with mathematics learning difficulties

ObjectiveTo explore the personality traits on higher vocational students with mathematics learning difficulties(MD).MethodsAll students of three grades in their first year were divided into MD students ( n =486) and outstanding students ( n =407 ) according to their final math scores.Cattell's 16 PF Questionnaires was used to assess the personality.ResultsStudents with MD got significantly higher records in personality factors of aggressiveness (6.59 ± 1.94 ),excitability ( 7.29 ± 2.24 ),sensitivity ( 5.85 ± 1.98 ),suspicion ( 3.51 ±2.03 ),fantasy(5.76 ± 1.86 ),urbane ( 5.81 ± 1.75 ),tense ( 5.63 ± 2.05 ),adapting and anxiety type ( 4.49 ±2.23 ),introverted and expert-oriented type(7.59 ± 2.43 ),and cowardice and decisive type ( 5.04 ± 1.62 ) than outstanding students(P<0.05) ;however,they got lower records in personality factors of steadiness(5.89 ±2.19),perseverance (4.41 ± 1.98 ),independence ( 3.74 ± 1.77 ),emotional with unruffled alert type ( 5.51 ± 1.81 ),professional achievements (52.72 ± 10.66),growth ability in new environments ( 18.78 ± 3.91 ) (P < 0.05 ).ConclusionHigher vocational students with MD have sensitive,suspicious,nervous,anxious,exoscopic,excited,fantasy personalities,lacking self-control,elasticity,carefulness and constancy.
3.Preliminary development of scale on work values for college students

Objective To develop the scale on work values for college students. Methods 63 items were determined through reviewing items in related literatures and scales, experts consulting and panel discussion.528 students from 3 universities in Shanghai were enrolled in the study. The data were analyzed through exploratory factor analysis,discrimination analysis,reliability analysis and other statistical analysis methods. Results College students' work values comprises six dimensions: self-value dimension ( including two sub-dimensions: self-esteem dimension and self-realization dimension), social-value dimension, dimension of the working environment, wages and welfare dimension , job stability dimension, and the dimension of social status. 43 items were reserved after exploratory factor analyses. The coefficient of reliability of the 43 items was 0.943, and the item loading of the 43 items was 0. 321 ~ 0. 792. Conclusion The discernment, internal consistency and validity of the scale accord with psychometric demands.

5.Simulation study of canceling technology for strong clutters in life detector based on radar

Strong clutters are produced by the echo waves from the debris when the life detector based on radar locates the living human subject under the debris, which makes the life detector work abnormally because the amplifier is saturate. A canceling technology for the strong clutters is provided to solve the problems discussed above in this paper. The canceling technology for the strong clutters can provide methods and theories in practice by constructing and simulating the mathematic model in the computer, analyzing and discussing the results.
6.Preliminary study on relationship between gene polymorphisms of interleukin-23 receptor and inflammatory bowel disease

Objective To investigate the association of two single nuclear peptides(SNPs)polymorphisms(rs11209026 and rs11805303)which lies in interleukin-23 receptor(IL23R)gene with susceptibility to inflammatory bowel disease(IBD).Methods The target SNPs were directly sequenced by polymerase chain reaction(PCR)and gene polymorphisms of 50 healthy and 81 patients with IBD (Crohn's disease in 41 patients and ulcerative colitis in 40 patients)were analyzed using chromassoftware.Results The geno-type frequency and allelic frequency of rs11209026 were 7.3%and 3.7%in patients with Crohn's disease respectively,15.0%and 7.5%in patients with ulcerative colitis respectively as well as 14.0%and 7.0%in normal population respectively(all P value>0.05).The geno-type frequency and allelic frequency of rs11805303 were 22.0%and 52.4%in patients with Crohn's disease respectively,15.0% and 41.2% in patients with ulcerative colitis respectively as well as 34.0%and 59.0%in normal population respectively(all P value>0.05).But in allelic frequency there was significant difference between ulcerative colitis patients and normal population(P=0.018).The polymorphisms of rs11805303 loci did not correlate with age,gender,disease duration.activity and site in patients with ulcerative colitis.Conclusions IL23R gene polymorphism is not associated with the susceptibility to Crohn's disease.rs11805303 allele may be related with susceptibility to ulcerative colitis.But no correlation was found between the SNP polymorphisms and the clinical characteristic of ulcerative colitis.
7.Effect of polymorphisms of Crohn disease related NOD2 gene and human beta-defensin 2 on expres-sion of human beta-defensin 2

Objective To explore the effects of polymorphisms of Crohn's disease related NOD2 gene and human beta-defensin 2 (hBD-2) on transcription of hBD-2 gene and its mechanism. Methods HEK293T cells were transfected with hBD-2 gene and NOD2 eukaryotic expression plasmid, and were then stimulated with LPS, TNF-α, or BAY 11-7082 (antagonist of NF-κB), respectively. Transcriptional activity of hBD-2 was detected afterwards. Results LPS could suppress transcription of hBD-2 (P=0. 020), which was increased by TNF-α in a dose-dependent manner (P =0. 004). In the presence of LPS, there was sig-nificant difference in transcriptional activity of hBD-2 between wild-NOD2 transfected group and mutated NOD2 (P268S) transfected group (P=0. 008), but there was no significant difference between wild hBD-2 transfected group and mutated hBD-2 transfected group (P=0. 053). With the stimulation of TNF-α (5 ng/ml), there was a significant difference between mutated hBD-2 transfected group and wild hBD-2 transfected group (P=0. 006), but no significant difference between wild-NOD2 transfected and mutated NOD2 transfected group was defected (P = 0. 064). Pretreatment with BAY 11-7082 before TNF-α (5 ng/ml) significantly inhibited the transcriptional activity of hBD-2 (P < 0. 001). Conclusion The poly-morphism of NOD2 affects the innate expression of hBD-2, the polymorphism of site in hBD-2 promoter (-233) may lead to significant decline of the inducible expression of hBD-2, and NF-κB might be a key pathway that NOD2 protein mediates the expression of defensin.
8.Construction of recombinant adenovirus and mediated reported gene expression in the guinea pig cochlea.

OBJECTIVE:
To purify P0 protein from guinea pig's inner ear by preparative SDS-PAGE and study the possible role it may play in the etiology of autoimmune inner ear disease.
METHOD:
A mixture of membraneous proteins of inner ear was separated by preparative SDS-PAGE. The corresponding band at 30kd was cut and electrically eluted. The protein collected was identified by analytical SDS-PAGE and Western blot assay. A group of 20 guinea pigs were immunized with P0 protein emulsified in complete Freund's adjuvant, another 10 guinea pigs were immunized with complete Freund 's adjuvant only as control. The guinea pigs' hearing thresholds, serum IgG level and morphological changes in the inner ear were investigated. The distribution of P0 protein in the cochlear was detected by immunohistochemical technique.
RESULT:
The purity of the protein was demonstrated by a single band at the 30 kD site in SDS-PAGE, which was identified as P0 protein by western blot analysis assay. About 17.5% P0-immunized guinea pigs showed increased hearing thresholds, elevated IgG level (F =6.48, P <0. 01), as well as a decreased number of spiral ganglion cells and inflammatory cell infiltration in the cochlear nerve region. The P0 protein is distributed in the cochlear nerve and spiral ganglion only.
CONCLUSION
P0 protein from guinea pig's inner ear can be successfully purified by preparative SDS-PAGE and an animal model of experimental autoimmune inner ear disease induced by P0 protein is successfully established.

9.Comparison of lentivirus and adenovirus vector mediated gene transfer into cultured spiral ganglion cells.

OBJECTIVE:
We aimed to compare the characteristics between lentivirus and adenovirus vector mediated gene transfer into cultured spiral ganglion cells (SGCs).
METHOD:
SGCs from newborn rats were cultured and exposed to lentivirus-GFP and adenovirus-GFP vectors. GFP expression and the cell morphology were evaluated under epi-fluorescence microscope at 3 days and 7 days after exposure. Survival number of SGCs was counted, and the average percentage of SGCs with GFP expression was calculated, and axon length was measured by ImageJ software.
RESULT:
Cultured SGCs were transfected by either adenovirus or lentivirus vector successfully. The adenovirus vector presented an instant and efficient transfection. However, the expression of GFP went down after 7 days. In lentivirus-GFP group, GFP expression was detected at 7 days after exposure, and the number of cells with GFP expression increased gradually in the following days. Statistical analysis revealed that there were no differences in survival number of SGCs and average axon length among lentivirus-GFP group, adenovirus-GFP group and control group.
CONCLUSION
Cultured SGCs can be transfected by either lentivirus vector or adenovirus vector safely and efficiently. SGCs are more susceptible to adenovirus vector, but GFP persists for a longer period after the lentivirus-mediated gene transfer.

10.Effect of the intense noise on hearing function and cochlea morphology in rat.

OBJECTIVE:
To observe the changes of auditory electrophysiology and inner ear pathology in rat cochlea after the noise exposure, and to offer the experimental data for exploring the mechanism of noise-damaged cochlea.
METHOD:
The rats in the study group were exposed to a intense narrow band noise centered at 4 kHz at the leave of 120 dB (SPL) for 4 h. The exposed cochleae were collected at various intervals (1 or 21 days) after the noise exposure. Auditory function was monitored by measuring thresholds of auditory brain stem responses (ABR). The morphological changes in rat cochlear hair cell (HC) were examined by HC nuclei stained with Propidium iodide (PI), a fluorescent dye specifically labelling the nuclear DNA and scanning electron microscopy (SEM). The number of spiral ganglion cells was calculated using pathologic technique.
RESULT:
The thresholds of ABR in the study group were significantly greater than that in the normal control group (P < 0.01). Examined at 1 day after the noise exposure, normal, apoptosis, necrotic and missing out hair cell (OHC) could be distinguished with PI staining, whereas the apoptosis OHC were not found at 21 days. Significant OHC loss was found in as compared to the normal control group (P < 0.01). There was not significant difference in the calculation of spiral ganglion cells (P > 0.05). SEM revealed the injured stereocilia of OHC (disarrangement, collapse) and OHC loss in the study group, which was more severe in OHC3 than the other two rows of OHC.
CONCLUSION
The intense noise used in our study could injure the rat cochlea and bring permanent threshold shift (PTS). Under this condition, the death modes of HC in the cochlea include apoptosis and necrosis in the fore part, whereas necrotic is the major mode in the evening of exposure. The injured stereocilia of OHC and OHC loss could remain the most consistent correlate of PTS.
