1.Investigation on molecular mechanism of angiotension II induced inflammatory activation of pancreatic stellate cells

Objective To investigate the impact of angiotension II (Ang II ) on the activation of nuclear factor-kappaB (NF-?B) signaling transduction pathway in cultured rat pancreatic stellate cells (PSCs). Methods Growth-arrested rat PSCs were incuhated for indicted time intervals with increasing concentrations of Ang II . The DNA binding activity of NF-?B was determined using electrophoretic mobility shift assay (EMSA). Western blot analysis was used to examine the degradation of inhibitory proteins of NF-?B (I?Bs). Expression of monocyte chemoattractant protein-1 (MCP-1) mRNA and protein were assessed by Northern blot and enzyme-linked immunosorbent assay (ELISA), respectively. Results Treatment of cells with Ang II led to a biphasic activation of NF-?B, which peaked at 15 min and 6 h later, and it was correlated with differential degradation of I?B? and I?B?. Ang II -induced NF-?B activation was greatly inhibited by antioxidants pyrrolidine dithiocarbamate ( PDTC), diphenylene iodonium. and N-acetylcysteine, suggesting the involvement of oxidative stress in this process. In PSCs, Ang II induced a dose-dependent increase in the expression of MCP-1 and this effect was markedly inhibited by blocking NF-?B activation with MG132 and PDTC, indicating that Ang II -induced MCP-1 gene expression was NF-?B-dependent. Conclusion The present results suggest that Ang II , through an oxidative stress-dependent mechanism, may activate a functional NF-?B signaling pathway in PSCs, through which it may participate in pancreatic inflammation and fibrosis.

3.Cleavage of Oncogene ki-rasG12V mRNA by Ribozyme and It' s Specificity

Objective: To design and confirm the cleavage activity of ribozyme Rz217 to oncogene ki-rasG12V messenger RNA and search for a new method for gene therapy targeting oncogene ki-ras. Methods: According to Symon' s principle,design an ribozyme specific for ki-rasc12v mRNA, both the constructs for transcription in vitro of ribozyme Rz217 and ki-ras exonl and the mammalian expression constructs of ribozyme Rz217 were constructed by DNA recombinant technique,ribozyme Rz217 and ki-ras exonl mRNA was obtained by transcription in vitro with T7 and SP6 RNA polymerase. Pancre atic carcinoma cell line PaTu8988 and human hepatocellular carcinomacell line BEL7404 were transfected with Rz217 mammalian expression constructs and the level of endogenous ki-rasG12V mRNA or ki-ras mRNA was determined by semiquantitative RT-PCR. Results: Not only in vitro but also in vivo, ribozyme Rz217 can cleave the mRNA of oncogene ki-ras (G12V) in site-specific manner and can not cleave the mRNA of wild-type ki-ras. Conclusion: Ribozyme Rz217 can cleave oncogene ki-rasc12v mRNA and the cleveage is specific for ki-rasG12V mRNA.
4.Comparison of fine-needle aspiration cytology, ultrasound and thyroglobulin examination for the differential diagnosis of enlarged neck lymph nodes in patients with differentiated thyroid cancer after radioactive iodine remnant ablation

Objective To compare the value of fine-needle aspiration cytology (FNAC),Tg,TgAb and ultrasonography (US) for the differential diagnosis of lymph node metastasis in patients with DTC after operation and radioactive iodine remnant ablation.Methods A total of 61 DTC patients with enlarged lymph nodes detected by US or physical examination after operation and radioactive iodine remnant ablation were included.FNAC was performed on the lymph nodes under US guidance within one week and the results were compared with those of Tg,TgAb and US.Final results were confirmed by comprehensive FNAC,posttreatment whole,body scan (Rx-WBS) and clinical follow-up.x2 test and Fisher's exact test were performed.Resuits Fifty-eight DTC patients had successful FNAC results,and 40 patients were confirmed as malignant and 18 as benign after overall assessment.FNAC identified 39 malignant and 19 benign cases.The coincidence rates of FNAC with postoperative pathological diagnosis and Rx-WBS for malignant cases were 100% (20/20) and 78.9% (15/19),respectively.For the benign cases,the coincidence rates of FNAC with clinical follow-up and Rx-WBS were 93.3% (14/15)and 4/4,respectively.The sensitivity,specificity and accuracy were 97.5%(39/40),100%(18/18) and 98.3%(57/58) for FNAC; 87.5%(35/40),55.6%(10/18) and 77.6%(45/58) for US; 92.5%(37/40),72.2%(13/18) and 86.2%(50/58) for Tg combined with TgAb (Tg/TgAb).The accuracy of FNAC was higher than that of US and Tg/TgAb(x2 =4.336,11.697,both P<0.05),while US and Tg/TgAb showed no significant difference (x2 =1.450,P>0.05).Tg/TgAb and US results were consistent in 39 cases with a diagnostic accuracy of 97.4% (38/39) verified by FNAC,while the other 19 cases with inconsistent Tg/TgAb and US results were verified as malignant in 4 cases and benign in 15 cases.Conclusions FNAC is superior to US and Tg/TgAb in the evaluation of lymph node metastasis in DTC patients after operation and radioactive iodine remnant ablation.FNAC should be recommended particularly when Tg/TgAb and US had inconsistent results.
5.Role of bcl-2 and bax expression in apoptosis of pulmonary artery smooth muscle cells of rats induced by NHE-1 inhibition in vitro

AIM:To investigate the role of bcl-2 and bax expression in apoptosis of pulmonary artery smooth muscle cells(PASMC) of rats induced by Na +/H + exchanger isoform-1(NHE-1) inhibition in vitro. METHODS:Intracellular Ca 2+ concentration ([Ca 2+ ]i) was measured using Fura-2/AM. The expression of bcl-2 and bax mRNA in cells were detected by sqRT-PCR, and the expression of Bcl-2 and Bax protein were examined immunohistochemically. RESULTS:The expression of bax mRNA and Bax protein and [Ca 2+ ]i in cells transfected with NHE-1 ribozyme gene increased significantly compared with those of cells transfected with pLXSN and nontransfected control. Meanwhile, the expression of bcl-2 mRNA and Bcl-2 protein in cells transfected with NHE-1 ribozyme gene decreased significantly. CONCLUSION:Apoptosis of pulmonary artery smooth muscle cells induced by inhibiting NHE-1 may have relevance to the increase in [Ca 2+ ]i and bax expression and the decrease in bcl-2 expression.

Objective :
To investigate the prevalence of comorbid chronic obstructive pulmonary disease (COPD) and osteoporosis (OP) and its influencing factors among residents aged 40 years and older in Zhangjiagang City, Jiangsu Province, so as to provide insights into prevention of comorbid COPD and OP.
Methods:
Permanent residents aged 40 years and older were sampled using a multi-stage stratified cluster random sampling method in Zhangjiagang City in 2019, and their pulmonary functions and bone mineral density were measured. The prevalence of comorbid COPD with OP was estimated. The demographic features, smoking, diet, exercises and physical examinations were collected, and factors affecting the prevalence of comorbid COPD with OP were identified using a logistic regression model.
Results:
Totally 3 140 subjects were enrolled, including 1 315 men (41.88%) and 1 825 women (58.12%), and a mean age of (62.89±9.13) years. A total of 137 subjects were detected with comorbid COPD and OP, with prevalence of 4.36%. The patients with COPD alone included 19 cases with mild, 133 cases with moderate, 87 cases with severe and 26 cases with very severe COPD, and the patients with comorbid COPD and OP included 10 cases with mild, 56 cases with moderate, 56 cases with severe and 15 cases with very severe COPD. The grade of pulmonary functions was higher in patients with comorbid COPD and OP than in patients with COPD alone (Z=-12.304, P<0.001). Multivariable logistic regression analysis identified ages of 65 years and older (OR=2.703, 95%CI: 1.862-3.923), women (OR=2.897, 95%CI: 1.915-4.384) and physical labor (OR=1.540, 95%CI: 1.083-2.189) as factors affecting the development of comorbid COPD and OP.
Conclusion
The prevalence of comorbid COPD and OP was 4.36% among residents at ages of 40 years and older in Zhangjiagang City, and moderate and severe degree was the predominant grade of pulmonary functions. The elderly, women and physical labors are at a high risk of developing comorbid COPD and OP.

Objective To investigate the expression of neuronal nitric oxide synthase(nNOS) and heme oxygenase-2(HO-2) in colon after stress in rats. Methods Water immersion-restraint stress(WRS) model was performed.The expressions of nNOS and HO-2 in colon of rats were examined by immunohistochemical staining and analysed by image analysis system. Results In control group rat,nNOS immune-positive substance mostly expressed in neurons of submucous plexus and myenteric plexus and HO-2 mostly expressed in lamina propria mucosal muscles of colon mucosa,circular muscles of colon and the endothelium and smooth muscle cells of submucosal bloodvessels.Compared with control group rat,the average gray value of nNOS positive neurons in submucous plexus and myenteric plexus and the average number of nNOS positive neurons increased significantly,the average gray value of HO-2 positive mucosal muscles and the positive unit(PU) of circular muscles increased in stress group rat,moreover,nNOS expression in mucosal epithelial cells and lamina propria lymphocytes and HO-2 expression in some large intestinal glands were observed.Compared with stress group rat,the average gray value of nNOS positive neurons in submucous plexus and myenteric plexus and the average number of nNOS positive neurons decreased in stress +L-NAME group,the average gray value of HO-2 positive mucosal muscles and the PU of circular muscles decreased in stress +ZnPP group.Conclusion Both nitric oxide(NO) and carbon monoxide(CO) are important gaseous signal molecules and neurotransmitters in colon of rats and may play a cooperative role in colonic functional disorder induced by WRS.

Objective To investigate clinical characteristics of reflux esophagitis ( RE) in Chinese population according to the retrospective analysis of RE in the past 14 years.
