1.Distinction Between Adult T-cell Leukemia/Lymphoma and Cutaneous Lymphoma

Adult T-cell leukemia/lymphoma (ATLL) and cutaneous T-cell lymphoma (CTCL) have similar clinieopathology and immunophenotype, in order to differentiate the two diseases, 4 eases of ATLL associated cutaneous lesions and 18 eases of CL were studied for elinieopathology, immunophenotype and HTLV-I provirus DNA. Two eases of actinic reticuloid and two eases of lymphocytic infiltration of the skin were selected as negative control. The results showed that four patients of ATLL manifested cutaneous lesions, at same time, they had additional systemic diseases, such as generalized lymphadenopathy, increased levels of LDH and IL-2R, rosette-like cells in their peripheral blood and abnormal bone marrow. The HTLV-I provirus DNA was detected in the peripheral blood, bone marrow, cutaneous lesions and lymph node biopsy specimens of the four patients by PCR amplification of specific HTLV-I DNA fragment. 18 cases of CL were negative for HTLV-I. The study indicates that ATLL may be diagnosed if the patients are associated with CTCL-Iike cutaneous lesions, characteristic histopathological pattern and immunophenotype, rosette-like cells in the peripheral blood and positive HTLV-I provirus DNA.
2.The Changes of Serum Tumor Necrosis Factor and Epidermal Growth Factor in Rats with Experimental Liver Fibrosis.

Objective To study the the changes of serum tumor necrosis factor-?(TNF-?)and epidermal growth factor (EGF) in rats with experimental liver fibrosis and its significance. Methods All the SD rats were randomly divided into the normal group and the model group. The rats in the model group were subcutaneously injected with 40%CCl4 according to 0.3ml/100g body weight twice one week for the first four weeks, the initial dose was double. In the following five weeks, the rats were injected 40% CCl4 of 0.2ml/100g body weight twice one week.The rats in the normal group were subcutaneously injected with equal dosage of saline,mentioned aboveo.At the end of the ninth week,the serum was obtained,the levels of serum TNF-?, EGF were detected with radioimmunity, with HE stains, the severity of inflammation and fibrosis in all liver tissuses were measured by semiquantitative scoring. 3.The distribution and it's clinical significance of hepatitis B core antigen in hepatocytes of chronic hepatitis B virus infected patients

Objective To explore the distribution and clinical significance of hepatitis B core antigen( HBcAg) in the hepatocytes of chronic hepatitis B virus infected patients. Methods Paraffin sections were made from 41 liver biopsy samples obtained from chronic hepatitis B virus infected patients. The immuno-fluorescence confocal technique was utilized to analyze the expression level and localization of HBcAg in hepatocytes. The data were analyzed by using Kruskal Wallis test and chisquare test. Results HBcAg expression were detected in 36 (87. 8%) patients, among whom 23 cases had moderate abnormal liver function, 10 with mild abnormal liver function and 3 with normal liver function. Among the cases with moderate abnormal liver function, 6 cases showed the simple membrane-type HBcAg expression, 17 cases showed mixed cytosolic-type and membrane-type HBcAg expression without the nuclear-type expression. Twelve cases with mild abnormal liver function only showed simple cytosolic-type HBcAg expression without membrane-type or nuclear-type expression. In the three patients with normal liver function, HBcAg was expressed in cytoplasm and nuclear but not on membrane. The correlation between HBcAg expression pattern and liver function was statistically significant (χ2 =10. 60, P<0.01). Conclusion HBcAg expression is directly correlated with liver injury in chronic hepatitis B virus infected patients, which indicates that membrane expressed HBcAg is the target antigen during the immuno-attack of liver.

6.Effect of rutin on proliferation of HepG2 cells

Objective To observe the effect of rutin on growth and proliferation of human hepatic cancer line(HepG2).Methods HepG2 cells were cultured in vitro,then cocultured with 50 to 250 ?mol/L rutin for 24 h.The inhibition rate of rutin on growth and proliferation of HepG2 was determined by MTT,~(3)H-TdR,and apoptotic cells were observed in fluorescent staining by Olympus fluorescent microscopy,and cell cycle was analysed by flow cytometry.Results Rutin inhibited HepG2 cells from growth and proliferation,and evoked apoptosis.Flow cytometry showed that 50 to 250 ?mol/L rutin caused an increase at G_(0)/G_(1) phase and a decrease at G_(2)/M phase and arrest at G_(0)/G_(1) phase in the cell cycle.Conclusion Rutin markedly inhibits the proliferation of HepG2 cells and induces apoptosis in a concentration-dependent manner.

8.Oxygen-induced retinopathy in newborn mouse

Objective To estabolish an optimal animal model of oxygen-induced retinopathy(OIR) suitable for examining pathogenesis and therapeutic intervention for retinopathy of prematurity(ROP). Methods Fifty-four 7-day-old C57BL/6J mice were devided into two groups. Twenty-seven mice in hyperoxic group were exposed to 75% oxygen for 5 days and then to room air for another 5 days. Twenty-seven mice in normoxic control were exposed to room air for 10 days. The proliferative neovascular response was estimated by observing the vascular pattern in adenosine diphosphate-ase(ADPase) stained retina flat-mounts and quantitated by counting the number of new vascular cell nuclei extending into the internal limiting membrane in cross-sections. Results Angiography in ADP ase stained retina flat-mounts delineated the entire vascular pattern. Hyperoxia-induced neovascularization occurred at the junction between the vascularized and avascular retina in the mid-periphery in all mice exposed to hyperoxia. After 5 days of exposure to hyperoxia at postnatal day 12(P12),the larger central radial vessels became tortuous and constricted and central perfusion became decreased obviously. After return to room air for 2 days at P14,neovascularization was seen. This response was maximal at P17. There was a mean of 44 neovascular nuclei per cross-section extending into the vitreous in hyperoxia compared to less than 2 nuclei in the normoxia control ( P
9.Inhibitory effect of aminoglucomannan on human respiratory syncytial virus in vivo

Objective To evaluate the anti-human respiratory syncytial viurs ( hRSV ) effect of aminoglucomannan ( AGM) in mice.Methods BALB/c mice infected with hRSV were randomly divided into AGM and konjac glucomannan ( KGM) groups with 54 in each.Then, mice in AGM group and KGM group were subdivided into three groups and treated with different doses of AGM or KGM as 2.5 μmol/L, 0.25 μmol/L or 0.025μmol/L.Each subgroup was further divided into 3 groups with 6 in each according to administration intervals of AGM or KGM as 8 h, 12 h or 24 h.All the mice are sacrificed at 72 h.The general condition of the mice was observed everyday.Reverse transcription-polymerase chain reaction ( RT-PCR) was used to detect the expression of hRSV fusion protein ( F) mRNA in the lung tissue of the mice. HE staining method and immunohistochemistry technique for intercellular cell adhesion molecule-1 ( ICAM-1) expression in the lung tissue were used to evaluate the inflammation in the lung tissue.ANOVA for randomized block design was used to examine the influence of dose and administration intervals on the expressions of hRSV F mRNA and ICAM-1.Results Reduced activity and asthma were observed in 16 mice in the KGM group, but not in the AGM group.And two mice in the KGM group died at d3, but there was no death in the AGM group.RT-PCR showed that the levels of hRSV F mRNA in lung tissues were 0.49 ±0.21 in AGM group and 0.88 ±0.06 in KGM group (t=6.71, P<0.05).HE staining of lung tissue showed that the inflammation in KGM group was more serious than that in AGM group.The expression of ICAM-1 in AGM group was much lower than that in KGM group, and there was statistical difference between two groups (t=13.88, P<0.05).In AGM group, the levels of RSV F mRNA and ICAM-1 decreased with the increase of AGM’s concentration and rised with longer administration intervals (P<0.05), but these were not observed in KGM group (P>0.05).Conclusion AGM can alleviate the inflammation of lung tissue in mice infected with hRSV in a dose and time-depandent manner.
10.Computer analysis of acoustic features of consonants before and after restoration with complete denture

BACKGROUND: Traditional acoustic examination is to make subjective descriptions of abnormal acoustic features, or carry out quantitative analysis of vowels with sensitive consonant utterance only subjected to qualitative analysis.OBJECTIVE: To make quantitative analysis of the acoustic features of sensitive consonants before and after restoration with complete denture. DESIGN: Randomized controlled experiment.SETTING: Department of Prosthodontics; Department of Oral & Maxillofacial Surgery, Ninth People's Hospital of Shanghai Second Medical University.PARTICIPANTS: Totally 30 edentulous patients who received treatment in the Department of Prosthodontics, Ninth People's Hospital of Shanghai Second Medical University, between November 2001 and March 2003-03were enrolled in this study. They were divided into plastic base group and metal base group with 15 patients in each group according to the denture base; the two groups of patients were matched in baseline data such as sex and age. Inclusion criteria: had no history of denture repairing, aged 45-75years, had received certain years of education, could speak Mandarin with out obvious dialect, had no obvious dysaudia (≤ 20dB), and had not received acoustic therapeutic training.METHODS: Plastic base group: the plastic base was 2.0-2.5 mm thick,with smooth anterior palate polished surface. Metal base group: the metal base was 1.0-1.5 mm thick, covering entire palate and forming palatal rugae, which transformed to net base at alveolar ridge side, and cheek base was plastic. The scope of base, the posterior bank of denture, overbite of anterior teeth, coverage degree, occlusal surface and 1/3 vertical range beneath surface were designed conventionally. Utterance of/zi/ and/ji/ by edentulous patients was detected before restoration, at the beginning and after restoration 1, 2, 4, 8 weeks respectively, with the results analyzed with the assistance of computer acoustic analysis system, the linear forecast technology and fast Fourier transformation were used to record the 1st and 3rdformants, the computer acoustic analytic sampling frequency was 10 000 Hz and he step number of linear forecast technology was 12.MAIN OUTCOME MEASURES: The differences in acoustic parameters between the detected utterances/zi/and/ji/.RESULTS: According to the intention-to-treat analysis, 30 patients were enrolled in this experiment and all of them entered the result analysis. ①When utterance/zi/was detected, most of 1st formants were found gradually reduced in plastic base group and metal base group after restoration compared to that before restoration, most obviously at week 8 [(353.67±35.65),(322.60±44.03); (332.27±37.10), (302.20±40.39); P < 0.05]. However,the 3rd formant variance was different between the two groups, presented by gradually ascending in plastic base group and gradually descending in metal base group. ② When the detected utterance was/ci/, changes of the 1st formants in two groups were basically the same as /zi/; moreover the changes of the 3rd formants also displayed different tendency in the two groups.CONCLUSION: Reducing the basal thickness of complete denture and reconstructing palatal rugae will be helpful for the acoustic functional restoration in patients with dentition defects. The computer acoustic analysis system can be used for quantitative analysis of acoustic features of patients before and after restoration with complete denture.
