1.Comparison of morphology, pathogenic ity and proteomics of granuloma- and tinea corporis-derived Trichophyton mentagrophytes

Objective To screen differentially expressed proteins between granuloma- and tinea corporis-derived Trichophyton mentagrophytes strains,and to explore the pathogenesis of T.mentagrophytes in deep and superficial infection.MethodsFour T.mentagrophytes isolates from granuloma and 4 isolates from tinea corporis were cultured in agar plates and small steel rings at 27 ℃ and 37 ℃ respectively, followed by morphologic observation.Eight guinea pigs were immunocompromised by glucocorticoids,and superficially and subcutaneously inoculated with the same amount of fungal suspension to develop an animal model of tinea corporis and granuloma,respectively.Two weeks later,the infection of guinea pigs was confirmed by microscopy,fungal culture and histopathology.Proteins were extracted from a highly toxic granuloma-derived T.mentagrophytes strain and a lowly toxic tinea corporis-derived T.mentagrophytes strain,and subjected to twodimensional electrophoresis,mass spectrometry and identification by using the National Center for Biotechnology Information (NCBI) database.ResultsThe granuloma-derived T.mentagrophytes isolates grew better at 37 ℃ than tinea corporis-derived T.mentagrophytes isolates did,while no significant difference was observed in the morphology of colonies between the two kinds of T.mentagrophytes isolates at 27 ℃.Tinea corporis models were successfully established in guinea pigs with the 8 T.mentagrophytes strains,and the granuloma-derived isolates induced a more intense inflammation than tinea corporis-derived isolates.Granuloma model was constructed only with 3 granuloma-derived strains,which was proved by microscopy,fungal culture and histopathology.A total of 463 ± 20 and 398 ± 17 protein spots were detected,with 62 and 21 upregulated proteins,from granuloma-derived and tinea corporis-derived T.mentagrophytes strains,respectively.Bioinformatics analysis revealed the following meaningful proteins from differentially expressed proteins in granuloma-derived T.mentagrophytesstrains, includingenolase, heatshockprotein, serine/threonineprotease, cellularsignal transduction proteins,energy metabolism-related proteins,cytoskeleton proteins and some hypothetical proteins.ConclusionsThe granuloma-derived T.mentagrophytes strains are more heat-resistant with a higher potency to cause infection compared with tinea corporis-derived T.mentagrophytes strains.Differences exist in the expression of proteins between granuloma- and tinea corporis-derived T.mentagrophytes strains.
2.HPLC Characteristic Fingerprint of Jiuwei Qianghuo Granules

Objective:To provide scientific basis for the quality control of Jiuwei Qianghuo granules by establishing an HPLC char-acteristic fingerprint. Methods:The HPLC analysis was performed on a Sunfire C18 chromatographic column (250 mm × 4. 6 mm, 5μm) , and the mobile phase was acetonitrile-0. 1% phosphoric acid with gradient elution at the flow rate of 1. 0 ml·min-1 . The de-tection wavelength was 270 nm and the temperature of column was 30℃. Results:The common pattern of HPLC characteristic chroma-tographic profile was established. There were 15 common peaks,and four of which were identified in the pattern. The similarities of 12 batches of Jiuwei Qianghuo granules was evaluated,and all of them were greater than 0. 90. Conclusion:The analysis method of HPLC characteristic chromatographic fingerprint is simple and reproducible, which can provide scientific basis for the identification and quali-ty evaluation of Jiuwei Qianghuo granules.
3.Determination of Sulfide in Source Water by Ion Chromatography-Pulsed Amperometric Detector

Objective To develop a new method for the determination of inorganic sulfide in source water by ion chromatography combined with pulsed amperometric detector.Methods The source water samples were injected into ion chromatography system after being filtrated with 0.22 ?m nylon membrane.100 mmol/L of NaOH-250 mmol/L of the NaOAc-0.5% (V/V) ethylenediamine was used as the eluent,special polarizable anions separation column IonPac 3AS7 was selected and sulfide was separated from other anions by isocratic eluent following by three-waveform pulsed amperometric detection.Results The liner range of determination for sulfide was 5-1 000 ?g/L,the equation of linear regression was y=0.325 4x-1.305 3.The detection limit was 0.5 ?g/L,the recovery rates were in the range of 90.8%-96.7%,and the relative standard deviation was 1.32%.Conclusion The method is simple with good selectivity,high sensitivity and good reproducibility and is applicable to the determination of inorganic sulfide in source water.
4.Effects of truncated type Ⅱ receptor of TGF-?_1 expressing plasmid on rat liver fibrosis

Objective To observe the effects of truncated type Ⅱ receptor of TGF ? 1 expressing plasmid on rat liver fibrosis. Methods An eukaryotic expression plasmid of a truncated type Ⅱ receptor of TGF ? 1 was constructed by recombinant DNA technique. Sequence analysis of this recombinant plasmid revealed that it contained the complete extracellular and transmembrane domains of TGF ? 1 Ⅱ receptor but lacked most of the cytoplasmic kinase domain. The recombinant plasmid encapsulated by liposome was transferred into the rats by intraperitoneal injection and its effects on the grade of the pathologic fibrosis and the levels of HA,LN,PCⅢ,Ⅳ C were observed. Results The levels of serum HA,LN,PCⅢ,Ⅳ C were lower in the early treatment group than those in the model control group ( P 0.05). 5.Correlation between reduction quality of femoral neck fracture and femoral head necrosis evaluated by three-dimensional measurement method

BACKGROUND:Reduction quality is essential for the healing and function of limb. Traditional X-ray and CT scans have some limitationsin quantitatively and visualy observing the reduction quality of the remnant fracture in three-dimensional displacements. Mimics have advantages in bone rapid modeling and three-dimensional measurement, which offers the chance in quantifying the reduction quality of the displacement in three-dimensional methods.
OBJECTIVE:To measure residual displacement in femoral neck fracture patients with satisfactory reduction by using three-dimensional measurement method, and to evaluate the association between residual displacement and femoral head necrosis.
METHODS:We enroled 141 patients with femoral neck fracture, who were treated in the Third Department of Joint, Affiliated Hospital of Logistics University of People’s Armed Police Force from January 2012 to December 2013. Al patients were divided into undisplaced fracture group (n=44; Garden1&2) and displaced fracture group (n=97; Garden3&4). Based on DICOM data of CT files, three-dimensional models were constructed in each patient. Three parameters were measured: displacement of femoral head’s center, fossa capitis femoris and rotational displacement. Femoral head necrosis was folowed up. The correlation between residual displacement and femoral head necrosis was explored.
RESULTS AND CONCLUSION:(1) The duration of folow-up in those patients ranged from 25 to 43 months. Femoral head necrosis occurred in 7 cases from the undisplaced fracture group and 21 cases from the displaced fracture group. (2) The three parameters (displacement of femoral head’s center, fossa capitis femoris and rotational displacement) were respectively (7.44±2.84) mm, (11.75±4.12) mm and (17.68±8.99)° in the undisplaced group, and (6.51±3.75) mm, (9.34±5.27) mm and (14.67±7.43)° in the displaced group. Except displacement of femoral head’s center, significant differences in the other two parameters were detected between the two groups (P< 0.05). (3) These findings suggest that poor reduction quality is the main factor causing femoral head necrosis. X ray is insufficient to describe the spatial shift and the angle, and has limitations in assessing the reductionquality of femoral neck fracture. Three-dimensional measurement has certain advantages to assess quality of fracture reduction.
6.Injection of human umbilical cord derived mesenchymal stem ceils into the vitreous of rabbits

Objective To observe the survival of human umbilical cord derived mesenchymal stem cells (hUC-MSCs) after injection into the vitreous of rabbits,and the animal safety under those procedures.Methods Twenty-seven pigmented rabbits were randomly divided into 3 groups (intravitreal injection 1 week group,2 weeks group and 4 weeks group),each with 9 rabbits.For each animal the right eye was the experimental eye receiving hUC-MSCs injection,while the left eye was the control eye receiving cuhure medium.The rabbit eyes were examined by slit-lamp microscope,indirect ophthalmoscopy,fundus photography,fundus fluorescence angiography(FFA)and Tono-pen tonometer before and after injection.hUC-MSCs were labeled by CM-Dil in vitro,and their survival status was measured by eonfocal fluorescence microscopy,light microscope and transmission electron microscope at 4 weeks after injection.Results Four weeks after injection,a large number of the hUC-MSCs were still alive in the vitreous cavity.The overall condition of those rabbits was good.The anterior segment and retina of experimental eyes were normal,without hyperfluorescence,hypofluorescenee and leakage in the retina at 1,2 and 4 weeks after injection.There was no significant difference on lOP before and after injection at different time points (P>0.05),and no obvious changes at cornea,anterior chamber angle,lens,retinal structure by.light microscope and transmission electron microscope examination.Conclusion hUC-MSCs can survive in the rabbit vitreous for four weeks;intravitreal injection of hUC-MSCs was safe and feasible.
7.Development of Recognized Software on Multi-tumor Protein Chip Analysis System V1.0

Objective To develop the system of tumor recognized tool software based on the examination results of multi- tumor protein chip. Methods On the basis of the diagnostic recognized equation, the examination results were comparison by HTA combining VB script and Javascript language program. Results The functions of the software include dates input of tumor protein chip,recognized results, dates interrogation and record printing of 10 kinds of common tumor. Conclusion The software can be diagnosed automatically with the sorts of tumor, which is a scientific, convenient and efficient tool for tumor clinician.
8.Influence of katsutoxin and its extract on bone marrow colony-forming unit-granulocyte and monocyte following ~(60)Co gamma ray radiation

The experiment was performed at Basic Medical College and First Affiliated Hospital, Zhengzhou University from September 2004 to December 2005. Totally 60 Kunming mice were divided into 5 groups randomly: ①blank control group (n =15) and simple radiation group (n =15). The mice were given 0.2 mL sterile saline by intraperitoneal injection. ②antineoplastic polypeptide from Buthus Martensii Venom (APBMV) group (n =10) and APBMV plus radiation group (n =10) received 0.2 mL APBMV according to prepared concentration by intraperitoneal injection. ③Katsutoxin extract Ⅲ plus radiation group (n =10) received 0.2 mL katsutoxin extract Ⅲ by intraperitoneal injection every other 5.5 hours for 7 days. After 24 hours from the last injection, the mice were endured 60Co g ray radiation (80 cm, 7.5 Gy irradiation dose, 0.27 Gy/min dose rate). Then katsutoxin extract Ⅲ was given same as above for 7 days. Then bone marrow was extracted to be cultured to colony-forming unit-granulocyte and monocyte (CFU-GM). 9.Meta analysis of clinical efficacy of epilepsy after stroke treated with combination of traditional Chinese and Western medicine

Objective To evaluate clinical effects of integrated traditional Chinese and western medicine therapy for epilepsy after stroke. Methods Literatures about the treatment of epilepsy after stroke with the combination of traditional Chinese medicine and western medicine which come from the database such as CNKI, VIP, WanFang and Pubmed, were studied and all the randomized controlled trials on comparing therapeutic effects of treating epilepsy between combined treatment of TCM and western medicine and western medicine were enrolled. The Cochrane Collaboration's software RevMan 5 was used for meta-analysis. Results 7 articles were included in the study, there is homogeneity among these articles(P>0.05), The fixed effect model was used for the meta analysis. the combined results were OR=5.17, 95%CI(3.02, 8.82), the overall effect of test Z=6.01, P<0.01, the difference between the treatment group and the control groups was significant, that means the overall effect of the combination of traditional Chinese medicine and Western medicine treatment was better than western medicine treatment; Funnel plot graphic displayed asymmetrically, which indicated the literatures publication bias. Conclusion The current data indicate that the curative effect of TCM and west medicine combined treatment is better than pure western medicine therapy for epilepsy after stroke, but as the literature publication bias, clinical studies need more high quality large samples to prove this conclusion.
10.Study on extraction process of chlorogenic acid in Flos Lonicerae by enzymic treatment

Objective: To optimize the extraction process of chlorogenic acid in flos lonicerae.Methods: The flos lonicerae was treated by means of cellulase and/or pectolase before extracted by ethanol reflux, studying the enzyme dosage, treatment time, treatment temperature how to influence the percentage of extraction of flos lonicerae and the amount of chlorogenic acid in extrction.Results: The percentage of extraction of flos lonicerae and the amount of chlorogenic acid in extrction could be obviously increased by the cellulase treatment, 8.15g chlorogenic acid could be extracted from 100g flos lonicerae. The optimum temperature of treatment was 40℃~50℃; The percentage of extraction was higher by the combined treatment of cellulase and pectilase than by cellulase only, but the amount of chlogenic acid was not obviously different between these two methods.Conclusion: The amount of chlorogenic acid in extrction could be increased by enzymic treatment.
