1.Antidepressant Effects of Yiyukang in Mice and Rats

Objective To study the antidepressant effect of Yiyukang and to explore its possible mechanism.Methods By using the mice forced-swimming models,mice tail suspension models and mice stereotyped behavior models,the antidepressant effect of Yiyukang and its mechanism were observed.Result In the mice forced-swimming test,the im- mobility-time was significantly reduced by high and medium dosage Yiyukang with the reduced rate being 21.65% and 24.84%(P<0.01).The high dosage can also reduce the immobility time of tail-suspension mice with the rate of 29.58%(P<0.01).Yiyukang can strengthen the mice head shaking behavior and had synergistic action with 5- HTP on reducing stereotyped behavior in mice.Conclusion Yiyukang has certain antidepressant effect,and the mecha- nism may be associated with promoting the actions of 5-hydroxytryptamine and dopamine in mice brain.
2.Detection of hepatitis B virus pre-C region mutation in cases of chronic hepatitis B and its relationship with IFN treatment

Objective To investigate the relationship between hepatitis B virus pre-core region 1896 site mutation in cases of chronic hepatitis B and IFN-alpha treatment.Methods 200 patients with chronic hepatitis B were randomly divided into experimental group(n=100) and control group(n=100).The patients in experimental group were treated with regular liver-protective drugs and IFN-alpha,while Control group patients received only regular liver-protective drugs for 6 months.PCR-RFLP were used to detect hepatitis B virus(HBV)DNA with pre-C region 1896 site mutation in two grops pre and post treatment.Results The mutation rates before interferon treatment were 20%(20/100) and 18%(18/100) in experimental group and control group,respectly.After treatment,the effective rates in experimental group were 60%(12/20) and 50%(40/80) in Patients with Chronic Hepatitis B variant and wild strain virus Infection,respectly.The effective rate in experimental group was significantly higher than that in control group.Conclusion IFN-alpha therapy was not correlated with genomic variability of the core region.
3.Surgical management of liver carcinoma accompanied by portal hypertension

ObjectiveTo evaluate the effects of various surgical procedures on liver carcinoma accompanied by portal hypertension. MethodsOne stage surgical procedure was performed in 28 cases of liver carcinoma accompanied by portal hypertension in our department from January 1992 to June 2000. ResultsThere was no operative mortality. The postoperative complications developed in 39%(11/28). The postoperative survival rates of 1?2 and 3 years were respectively 91%, 61%, and 48%. Postoperative upper digestive tract hemorrhage developed in 9 cases, 13 cases died during follow-up period, of whom 6 cases died of the recurrence of liver carcinoma, 2 cases died of liver failure, 5 cases died of upper digestive tract hemorrhage.Conclusions The survival time can be prolonged and the postoperative complications reduced through prudent selection of surgical procedures in patients suffering from concurrent liver cancer and portal hypertension. Postoperative recurrent bleeding can be prevented by pericardial devascularization. In poor risk cases with huge carcinoma, simple surgical procedure should be applied,and with a discreet application of chemoembolization.
4.Correlation between serum 8-isoprostaglandin-F2α levels and clinical features in systemic sclerosis

Objective To investigate the oxidative status in systemic sclerosis (SSe) and explore the correlation between oxidative status and clinical features by measuring serum 8-isoprostaglandin-F2α (8-isoPGF2α) concentrations. Methods Serum 8-iso-PGF2α levels were detected by enzyme-linked immunosorbent assay (ELISA) in 51 SSe patients and 22 matched healthy controls. And in some SSc patients, serum homocysteic acid (Hey) concentrations were examined by cyclophorasc assay. Plasma yon Wilebrand factor (vWF) activity and serum immunoglobulin (lg) concentrations were detected by immunoturbidimetry.Antinuclear antibodies (ANA) and anti-endothelial antibodies (AECA) were detected by indirect immunofluorescence. Anti-sc170 antibody was detected by immunoblotting. Thirty-five out of 51 SSc patients were assessed for clinical features and laboratory parameters in order to analyze the correlations between 8-isoPGF2α levels and clinical features. Results Serum 8-iso-PGF2α levels were higher in sclerederma patients than in healthy controls. Values of 8-iso-PGF2α correlated with pulmonary involvement, such as diffusion capacity for carbon monoxide (DLCO) and pulmonary interstitial fibrosis by pulmonary high-resolution computed tomography (PhrCT), and correlated positively with renal vascular damage determined by the resistant index (RI) of renal glomeruli interlobular arteries. There was no correlation between 8-iso-PGF2αconcentrations and skin, peripheral vascular, heart, esophagus involvement and disease activity, diseasepattern, vWF,Hcy, lg, or autoantibedy profiles.Conclusion Increased Jevels of 8-iso-PGF2α, marker of oxidative stress, is correlated with pulmonary fibrosis and the extent of renal vascular damage in SSc and this supports the hypothesis that oxidative stress plays an important role in SSc pathogenesis.
5.The effect of microRNA-27a on U251 glioma cells

Objective To discuss the effect of microRNA -27a on U251 glioma cells.Methods Over-expression or inhibition of miR -27a in U251 glioma cells were done by transient transfection of miR -27a mim-ics or AMO-27a in vitro.Cell viability was detected by MTT assay .Invasion ability of U251 was detected by tr-answell invasion assay.The level of miR-27a and PPARγwere detected by real -time PCR.Results Under in-hibition of miR-27a condition,the proliferation and invasiveness of U 251 glioma cells were decreased .The level of PPARγwas significantly increased ,whereas the level of miR-27 a was decreased .Overexpression of miR-27 a increased the proliferation and invasiveness of U 251 glioma cells and decreased the level of PPARγ.Conclusion Inhibition of miR-27a is benefit for inhibiting the proliferation and invasiveness of U 251 glioma cells.

Forkhead box A1 (FOXA1) is expressed in various organs,including breast,liver,pancreas,bladder,prostate,colon,ovary,lung and esophagus.The FOXA1 expression is associated with the growth and carcinogenesis of these organs.It is recently demonstrated FOXA1 plays a significant role in different biological processes including cell proliferation,differentiation,devolopment,carcinogenesis and the process of epithelial-mesenchymal transition (EMT) etc.Its expression is positively correlated with ER and PR expression,and it is demonstrate to be one of the most important favorate prognostic factors of breast cancer.
7.Optimization of Extraction Process for Paeoniae radix Alba by Central Composite Design-Response Surface Methodology

Objective To optimize the extraction process of Paeoniae radix alba. Methods Extraction rates of paeoniflorin and total glucosides from Paeoniae radix alba and yield of dry extract were served as research indexes. Extraction process was optimized by central composite design-response surface methodology. Results Optimum condition of extraction technology was to extract 2 times with 9-fold of 85.00% ethanol for 100 min each time. Results of the three batches of confirmatory test were accordant with the predicted values. Conclusion The observed and predicted values were close to each other,and the optimum formulation by CCD-RSM was successful. The established model has a reliable predictability. The optimum extraction process is reasonable,stable and feasible.
8.Multi-slice spiral computerized tomography findings of colorectal cancer with chronic schistosomiasis

Objective To retrospectively analyze computerized tomography (CT) imaging features of colorectal cancer with chronic schistosomiasis in order to improve the diagnostic accuracy.Methods Eighty patients whose diagnosis was pathologically confirmed as colorectal cancer with schistosomiasis were collected from Tongji Hospital and Kunshan Chinese Medicine Hospital from January 2007 to December 2012.All the patients underwent abdominal plain CT and contrast-enhanced CT scan.The lesion location,morphology,size,calcification features,enhancement patterns and cancer metastasis were evaluated and compared by two radiologists who were blind to the diagnosis.Twenty colorectal cancer cases without schistosomiasis from the same area were also collected as controls.Results CT imaging showed that the tumors all occurred in the colon and rectum in 80 patients,mainly in colon descendens,colon sigmoideum and rectum.The lesion was characterized by irregular bowel wall thickening with soft tissue masses,and the average length of impaired intestine was (21.35 ± 4.50) cm.The lesions were solitary in 68 cases (85.0%) and were multifocal in 12 cases (15.0%).Linear,spotty and small patchy calcifications were seen in all the patients,with margins unclear in 59 patients (73.8%) and margins clear in 21 patients (26.2%).Early phase enhancement was seen in 68 cases (85.0%),of which 45 cases (56.2%) had homogenous enhancement and 23 cases (28.8%) had heterogeneous enhancement.Late phase enhancement was seen in 73 cases (91.2%) and necrosis was hardly seen.Only two cases (2.5%) had liver metastasis without lymphatic metastasis.All the 20 colorectal cancer cases without schistosomiasis presented with single lesion,which was characterized by irregular bowel wall thickening and local ulcer.The average length of impaired intestine was (6.90±3.40) cm.No calcification was seen in these lesions.All cases had early phase lesion with remarkable heterogeneous enhancement with low density necrotic area.The late phase enhancement was decreased.Four cases (20.0%) had liver metastasis.Compared to cases without schistosomiasis,cases with schistosomiasis tended to have multifocal (x2=100,P=0.000) and longer lesion (t=45.506,P=0.000),and more calcification (x2=100,P=0.000).The early phase heterogeneous enhancement was less frequent (x2 =88,P=0.000) and late phase enhancement was more frequent (x2=100,P=0.000).The liver metastasis rate was significantly lower (x2 =8.688,P =0.014).Conclusions The CT imaging of colorectal cancer with schistosomiasis is characterized by calcifications inside the tumor with obscured margins and multiple intestinal segments involvement.Hematogenous metastasis and lymphatic metastasis are rarely seen.
9.Effect of neural stem cells transplantation on protein expressions of doublecortin and nestin in hippocampal regions in aPP/PS1 mice

Objective To explore the effect of neural stem cells (NSCs) transplantation on neurogenesis in an alzheimer' disease mouse model and its related mechanism.Methods A total of 16 12-month old aPP/PS1 double transgenic aD mice were randomly divided into two groups:NSC group (receiving NSCs transplantation in the bilateral hippocampi,n=8) and PBS group as the negative control group (receiving an equal quantity of 0.01M/L phosphate buffer saline,n=8).another 8 wild type mice without any treatment were selected as the positive control group (Wt group).at five weeks after transplantation,the expressions of doublecortin (DCX) and nestin in the hippocampal dentate gyrus (DG) and subgranular zone (SGZ) were analyzed by immunofluorescence staining,the number of positive cells in these regions were counted by confocal microscopy,and Morris water maze (MWM) test was used to assess cognitive function in all mice.Results NSC group showed the enhanced spatial learning and memory ability in Morris water maze (MWM) as compared to PBS group,but it was still lower than that in Wt mice (both P<0.05).Immunofluorescence staining showed NSCs induced the increased positive cells expressing DCX and nestin in the NSC group as compared with PBS group (F=85.400 and 55.687,both P<0.001).However,the number of DCX positive cells was still lower in NSC group than in Wt group (P<0.05).There was no significant difference in the number of Nestin-positive cells between the NSC group and the Wt group (P> 0.05).Conclusions NSCs transplantation can promote endogenous neurogenesis via the increased expression of DCX-and Nestin positive cells in the hippocampal DG/SGZ in aD mice,which improves the cognitive abilities in aD mice in some extent.
10.Efficacy of ultrasound-guided continuous brachial plexus block for analgesia after fixation of upper extremity fractures in pediatric patients

Objective To evaluate the efficacy of ultrasound-guided continuous brachial plexus block for analgesia after fixation of upper extremity fractures in the pediatric patients.Methods Sixty American Society of Anesthesiologists physical status Ⅰ pediatric patients of both sexes,aged 3-10 yr,weighing 13-46 kg,with body height of 97-152 cm,scheduled for open reduction and internal fixation of distal humeral fractures,were selected and divided into 2 groups (n =30 each) using a random number table:intravenous analgesia group (group Ⅴ) and continuous brachial plexus block group (group B).Surgery was completed under brachial plexus block combined with general anesthesia in the two groups.At the end of surgery,patient-controlled intravenous analgesia was used in group Ⅴ,and continuous brachial plexus block was performed with 0.1% ropivacaine 250 ml (background infusion 0.1 ml · kg-1 · h-1,bolus dose 0.2 ml/kg,lockout interval 30 min) in group B.Tramadol was given as rescue analgesic when necessary.Ramsay sedation scores were assessed and recorded at 2,4,8,12,24,36 and 48 h after surgery,and the development of over-sedation was recorded.The require,nent for rescue analgesics and development of respiratory depression,dizziness,nausea and vomiting,pruritus and urinary retention during analgesia were recorded.The development of adverse reactions such as vascular and nerve injury,local hematoma and pneumothorax was recorded in group B.Family's satisfaction with analgesia was assessed and scored.Results Compared with group Ⅴ,Ramsay sedation scores at 2-12 h after surgery,the incidence of oversedation,nausea and dizziness and requirement for tramadol were significantly decreased (P<0.05),and no significant change was found in the degree of family's satisfaction with analgesia in group B (P> 0.05).The adverse reactions such as vascular and nerve injury,local hematoma or pneumothorax were not found in group B.Conclusion Ultrasound-guided continuous brachial plexus block can be safely and effectively used for analgesia after fixation of upper extremity fractures in the pediatric patients.
