1.The analysis of clinicopathological characteristics and prognosis in Uygurs women with breast cancer of Xinjiang

Objective To explore the clinical trait and relationship between clinicopathological characteristics and prognosis in Uygurs women with breast cancer of Xinjiang. Methods The clinicopathological data of 74 cases with breast cancer of Uygurs women were analyzed, multivariate analysis was done by COX proportional hazard model. Results 74 cases have followed up: there-year and five-year survival rates were 75.00 %(48/64) and 43.50 %(20/46)respectively. Multivariable COX regression revealed that patient age, menstruation, status of involved lymph node, TNM and the expression of PR were five important and independent prognostic factors for breast cancer of Uygurs women, the hazard rations of these factors were 0.037, 0.103, 13.851, 18.877, and 0.046, respectively. Conclusions The proportion of breast cancer of Uygurs women was low in patient of Xinjiang, but patient age was young, age of onset were more than half between 36 and 50 years old and mainly belong to Ⅱ, Ⅲ stage of TNM, prognosis was poorly. Patient age, menstruation, status of involved lymph node, TNM and the expression of PR serve as independent prognostic indexes of breast cancer of Uygurs women. Early diagnosis and treatment are very important to improve the survival rate of breast cancer patients
2.Study of hemoglobin α-thalassemia screening of capillary electrophoresis

Objective To investigate hemoglobin by capillary zone electrophoresis in α-thalassemia screening application.Methods Blood specimens collected from 1000 early pregnancy women were analyzed by fluorescent PCR technique for αt-thalassemia genetic diagnosis.The samples were divided into thalassemia group and normal group according to the risk of having α-thalassemia related genes.Then compare hemoglobin ingredients difference between two groups.Results In thalassemia group,the mean and standard deviation of HbA2 respectively was 1.96 and 0.386,while HbF respectively was 0.01 and 0.098.In control,the mean and standard deviation of HbA2 respectively was 2.79 and 0.418,while HbF respectively was 0.01 and 0.105.There was significant difference on value of HbA2 among different groups (P < 0.05),but there was no significant difference on value of HbF.If the cut-off value of HbA2 was less than 2.2 in α-thalassemia screening,the detection rate of carrying α-thalassemia gene was 9/16,and false negative rate was7/16.If the cut-off value of HbA2 was less than 2.4,the detection rate of carrying α-thalassemia gene was 10/16,and false negative rate was 6/16.Conclusion The screening efficiency of α-thalassemia is less than β-thalassemia by capillary electrophoresis.But choosing a reasonable cut-off value and combination with other indicators are still clinically acceptable screening program.
3.Distribution and polymorphisms of host killing genes in the plasmids metagenome of Escherichia coli

Objective To investigate the genotypes of host killing genes and their single nucleotide polymorphisms (SNPs). Methods Three hundred and twenty strains of Escherichia coli that collected from the First Affiliated Hospital of Wenzhou Medical College were analyzed. The first sample ( E1 ) contains 160 strains isolated during the years from 2002 to 2003. The second sample (E2) contains 160 strains covering the years from 2008 to 2009. The plasmids of Escherichia coli were extracted by alkaline lysis method. Solexa/Illumina sequencing technology was used to sequence plasmids metagenome. Solexa Genome Analysis System and Soap programs were used to analyze gene distribution, SNPs and lineage-specific mutations. Results 11 077 768 reads were generated and 0. 045% of them can map to the reference sequences from El sample. Whereas 9 377 792 reads were generated and 0. 053% of which mapped to the reference from E2 sample. There are nine host killing genes identified in the two samples, of which hok gene is the most prevalent. A total of 29 SNP sites dispersed in five genes of the two samples. Approximately 33% of them were non-synonymous mutations. One position of A and G is the most prevalent polymorphism. Conclusion The known nine genotypes of host killing genes were all identified in plasmids of Escherichia coli in Wenzhou. hok gene showed the highest frequency. There were SNPs in five genotypes.
4.Observation on clinical efficacy of contact thrombolysis via radial artery or femoral artery pathway for treatment of patients with acute ischemic stroke

Objective To compare the clinical efficacy of contact thrombolytic therapy between transradial artery intervention (TRI) and transfemoral artery intervention (TFI) pathways in patients with acute ischemic stroke (AIS). Methods The clinical data of 64 patients with AIS consistent with the indication of arterial thrombolysis in the People's Hospital of Huimin County of Shandong province from January 2010 to October 2017 were treated with arterial contact thrombolysis after exclusion of contraindications, including 42 cases treated with TRI (TRI group) and 22 cases treated with TFI (TFI group), and they were analyzed retrospectively. The differences in success rate of puncture, puncture time, the interval between the sheath set to the artery and the catheter reaching the diseased vessels, the re-canalization rate of occlusive vessels, surgical time, puncture site bleeding and intracranial bleeding complications and clinical outcomes in the patients were compared between the two groups. Results The comparisons of following indicators between the TRI group and TFI group were as follows: success rate of puncture [97.6% (41/42) vs. 100.0% (22/22)], puncture time (minutes: 5.5±2.0 vs. 5.4±2.3), the interval between sheath set to the artery and catheter reaching the diseased vessel (minutes: 6.2±3.8 vs. 6.7±3.9), occlusive vascular re-canalization rate [45.2% (19/42) vs. 40.9% (9/22)], operation time (hours: 1.50±0.38 vs. 1.45±0.32), incidence intracranial bleeding complications [9.52% (4/42) vs. 9.09% (2/22)], and the differences in above indicators in comparisons between the two groups had no statistical significance (all P > 0.05); however, the complication of bleeding at puncture site in TRI group was significantly lower than that in TFI group [0 vs. 22.7% (5/22), P < 0.05]. There was no statistical difference in clinical outcomes between the two groups. Conclusion It is safe and effective to treat patients with AIS by the TRI approach.
5.Overexpression of Uridine-Cytidine Kinase 2 Correlates with Breast Cancer Progression and Poor Prognosis.

PURPOSE: Uridine-cytidine kinase (UCK) 2 is a rate-limiting enzyme involved in the salvage pathway of pyrimidine-nucleotide biosynthesis. Recent studies have shown that UCK2 is overexpressed in many types of cancer and may play a crucial role in activating antitumor prodrugs in human cancer cells. In the current study, we evaluated the potential prognostic value of UCK2 in breast cancer. METHODS: We searched public databases to explore associations between UCK2 gene expression and clinical parameters in patients with breast cancer. Gene set enrichment analysis (GSEA) was performed to identify biological pathways associated with UCK2 gene expression levels. Survival analyses were performed using 10 independent large-scale breast cancer microarray datasets. RESULTS: We found that UCK2 mRNA expression was elevated in breast cancer tissue compared with adjacent nontumorous tissue or breast tissue from healthy controls. High UCK2 levels were correlated with estrogen receptor negativity (p<0.001), advanced tumor grade (p<0.001), and poor tumor differentiation (p<0.001). GSEA revealed that UCK2-high breast cancers were enriched for gene sets associated with metastasis, progenitor-like phenotypes, and poor prognosis. Multivariable Cox proportional hazards regression analyses of microarray datasets verified that high UCK2 gene expression was associated with poor overall survival in a dose-response manner. The prognostic power of UCK2 was superior to that of TNM staging and comparable to that of multiple gene signatures. CONCLUSION: These findings suggest that UCK2 may be a promising prognostic biomarker for patients with breast cancer.

A model of fluid dynamics related to the myocardial bridginged and mural coronary artery was designed and manufactured according to the physical principle and characteristic of the mural coronary artery. The model can imitate systematically well the effect of myocardial bridging on hemodynamic change of the mural coronary artery under different controlled experimental parameter. The methodology is proved to be feasible and has good prosperity of experimental study.

Expanded services such as clinical comprehensive evaluation of anti-tumor drugs and management of off-label drug use are an important development trend of oncology pharmacy.
