AIM: To investigate the effects of 6, 7- dimethoxycoumarins on the proliferation and the cell cycle of human lung carcinoma cell( PA) in vitro METHODS: The effects of 6, 7- dimethoxycoumarins on the cytomorphology, growth and cell cycle of PA cells were evaluated by MTT assay and flow cytometry in vitro RESULTS: 6, 7- dimethoxycoumarins could inhibit the proliferation of PA cells in a dose- dependent manner The inhibition action of 6, 7- dimethoxycoumarins plateaued at 160μ g/ml(52 4% inhibition) The agent in dose of 80μ g/ml led to a decline in the proportion of cells to those in G2/M phase, made the cells remain in G0/G1 phase and reduced the proliferation index(PI) of PA cells CONCLUSION: In vitro, 6, 7- dimethoxycoumarins can inhibit the proliferation of PA cells, which leads to arrest of the cells in G0/G1 phase by way of inhibiting DNA synthesis

Objective To explore the reliability and validity of the Chinese revised version of the Intolerance of uncertainty scale(IUS).Methods A total of 930 college students completed the IUS.45 college students participated in the retest 3 weeks later. The self-rating anxiety scale (SAS) and the self-rating depression scale (SDS) were used as criterions for assessment of the validity of the Chinese revised version of IUS.Results The Chinese revised version of IUS contained 21 items.Exploratory factor analysis found a two-factor model (negative belief and emotional experience, behavioral dysfunction and negative self-reference) with a cumulative variance contribution rate of 42.09%.And the results of Confirmatory factor analysis (x 2/df =2.82,GFI =0.90,TLI =0.90,CFI =0.91,RMSEA =0.065) indicated that this model provided a reasonable good fit for Chinese college students. The Cronbach α of internal consistency reliability were good for IUS and two subscales, which were 0.929,0.885 and 0.864,respectively.The test-retest reliabilities for IUS and two subscales were 0.782,0.766 and 0.714 respectively. The IUS scores were positively correlated with the scores of SAS and SDS (r =0.489 ~0.575,P < 0.001).Conclusion It suggests that the Chinese revised version of IUS has good reliability and validity, which is qualified enough for relevant research and applications for Chinese adults.

Objective To explore the possible effects on differentiation of SHI-1 cells induced by puerariae radix flavones(PRF)in vitro. Methods SHI-1 cells were treated with PRF in various concertration, then the inhibitory effects of cell proliferation were detected by MTT assay, the cell cycles were analyzed by flow cytometry(FCM),the cells reduction rates were detected by NBT reduction test, and the expression of CD11b and CD14 were tested by FCM.Results 10-50 μg/ml PRF could inhibit the proliferation of SHI-1 cells in a time-and dose-dependent manner, and the cell cycles were arrested in S phase. When SHI-1 cells were treated with 10,30 and 50 μg/ml PRF in 48 houres respectively, the NBT reduction rates of cells were increased in a dose-dependent with PRF(P<0.05),and the expression of cells surface differentiation antigen CD14 was also increased along with the concentration of PRF.Conclusion The SHI-1 cells could be induced to differentiation partially after treated with 10,30 and 50 μg/ml PRF in vitro.

Objective curcumin can suppress the proliferation , induce apoptosis and partial differentiation , and inhibit the migration of many kinds of tumor cells .The aim of this study was to investigate the expressions of mitogen-activated protein kinase (MAPKs) and matrix metalloproteinases (MMPs) when the proliferation of human multiple myeloma RPMI8226 cells was inhibited by curcumin in vitro, and to reveal the antitumor molecular mechanism of curcumin . Methods RPMI8226 cells were treated with various concentrations of curcumin for different periods of times .The inhibitory rate of curcumin on cell proliferation was detected by MTT assay , the cell cycle analyzed by flow cytometry , the protein levels of MAPKs measured by Western blot , and the activity of MMPs analyzed by Gelatin zymography . Results Curcumin inhibited the proliferation of RPMI 8226 cells in a time-and dose-dependent manner , and the cell cycle was arrested in the G 2/M phase ([12.72 ±0.68]%vs [4.79 ±0.15]%).The expressions of JNK and p-JNK showed a con-centration-dependent increase in the RPMI8226 cells treated with curcumin at 6.25, 12.50, and 25.00 μmol/L, respectively (P<0.01) , but the expressions of ERK 1/2 and P38 MAPK did not change significantly compared with the control group (P>0.05).In addition, the activities of MMP-2 and MMP-9 were decreased in a dose-depend-ent manner in the supernatant of RPMI8226 cells ( P <0.01). Conclusion A certain concentration of curcumin could not only acti-vate the JNK signalling pathway of the MAPKs family and induce the apoptosis of RPMI8226 cells, but also inhibit the activity of MMPs and influence the invasion and metastasis of RPMI 8226 cells.

Objective To construct vector expressing soluble mPDL1-hIgGFc and study its effect on the proliferation and apeptosis of cells in vitro. Methods The extrncellular domain of mPDL1 gene was amplified from pmPDL1 vector by PCR and inserted into phIgGFc vector. The recombinant pmPDL1-hIgGFc was transfected into CHO cells by LipofectAMINETM2000, and the transfected cells were named as CHOp. The expression of mPDL1-hIgGFc in the culture supernatants of CHOp was assayed by ELISA and Western blot. The effects of CHOp culture supernatants on mixed lymphocyte culture(MLC) was analysed by Flowm-etry. Results The extracellular domain of mPDL1 gene were obtained from PCR. DNA sequencing and the identification of digestion by HindⅢ and KpnⅠ indicated the recombinant plasmid pmPDL1-hIgGFc was suc-cessfully constructed. ELISA and Western blot analysis proved that the CHOp could express mPDL1-hIgG-Fc. CHOp culture supernatants could inhibit lymphocyte proliferation and induce the apoptosis of the activa-ted T cells in MLC in vitro in a dose-dependent manner. Conclusion The mPDL1-hIgGFc protein could in-hibit lymphocyte proliferation and induce the apoptosis of the activated T cells.

Volatility can influence the lifetime of particles in the atmosphere, and provide useful information on the formation of secondary aerosol. The previous studies generally utilized thermodenuder ( TD ) to investigate the volatility behavior of particles. Using TD, semivolatile species are vaporized at different temperature, and the vaporized gas is adsorpted by activated charcoal. However, carbon might be emitted from activated charcoal under high temperature or activated charcoal ageing. In this study, a new method was developed for the measurement of particle volatility by coupling a thermodiluter system to an online single particle aerosol mass spectrometer ( SPAMS) . Aerosol particles were passed into two different channels, and then analyzed by SPAMS. Through Channel 1, aerosol particles were heated to different temperature by heating tube, then non-volatile particles and volatile gas entered into the diluter. After diluting and cooling by diluent air, the non-volatile particles were analyzed by SPAMS. Through Channel 2, aerosol particles were analyzed directly by SPAMS without the heating process. Particle volatility was obtained by comparing the information ( particle size, particle number and mass spectrum ) of particles through Channels 1 and 2. Laboratory tests showed that the diluter could avoid the re-condensation of volatiles to the particles. This developed method was applied in the real time measurement of individual particle volatility in the spring of Guangzhou. The results showed that these particles were primarily comprised of highly volatile and moderate volatile species.
