AIM: The synthesis of sarmentosin. METHODS: Condensation of butane-1,2,4-triol-1,2-acetonide(3) with α-D-glucopyranosyl bromide tetraacetate in the presence of Ag2O and molecular sieves gave the desired β-glucoside 4,which was transformed into sarmentosin via a reaction sequence of 7 steps. RESULTS: The overall yield is 5.8%. CONCLUSION: The first synthesis of sarmentosin(1),a potent natural GPT lowering agent,was achieved.
2.Expression of COX-2 and VEGF in non-hodgkin lymphoma and its clinical significance

Objectiye To explore the expression of COX-2 and VEGF and its clinical significance in non-Hodgkin lymphoma (NHL). Methods The expression of COX-2 and VEGF were detected by immunohistochemistry in 42 cases of NHL and 20 cases of lymph node with benign pathological change. Results The positive rate of COX-2 and VEGF was 45.24% and 73.81% in NHL respectively. The expression rate of VEGF was positively correlated with that of COX-2 in tissues of NHL ( x2 = 4. 63, P < 0. 05).The expression of COX-2 was related to clinical stage and histopathologic grade of NHL ( x2 = 5.43, P <0. 05), but it had no association with gender, age, B symptoms, and IPI. The expression of VEGF was significantly related with aggression, B symptoms and IPI ( x2 =8. 979, 8. 893,6. 434, P <0. 05), but it had no association with age, gender and clinical stages. Conclusion COX-2 and VEGF may be involved in NHL tumorgenesis, and COX-2 may accelerate angiogenesis by increasing VEGF expression. Specific COX-2 inhibitors may be a novel therapeutic approach for NHL.
3.Effects of fluvastatin on the tubulointerstitium in progressive diabetic kidney disease

Objective To investigate the effects of fluvastatin on the tubulointerstitium damage in progressive diabetic kidney disease. Methods A rat model of type 2 diabetic nephropathy (DN) was developed successfully by combination of dietary induced insulin resistance and low dose STZ induced hyperglycemia after unilateral nephrectomy. Female SD rats were randomly divided into three groups: control rats, type 2 diabetic rats and type 2 diabetic rats treated with fluvastatin (2mg/kg/d). After 6 weeks, blood glucose, serum insulin, serum triglyceride and cholesterol, serum creatinine, and urinary protein were measured respectively. The protein expressions of c Jun and tansforming growth factor (TGF) ? 1 were studied by immunohistochemistry. TGF ? 1 gene expression was studied with a RT PCR technique. 4.Establishment of Cloning and Sequencing Method for High-Resolution Human Leukocyte Antigen-B Genotype Assay

Ahigh-resolutionmethodforhumanleukocyteantigen-B(HLA-B)genotypingwasestablished based on the optimized polymerase chain reaction, cloning and sequencing technology. The exon2 and exon3 of HLA-B gene were amplified with primers based on the HLA-B gene sequence. These produced heterozygous alleles were effectively cloned into plasmid DNA based on the principle of plasmid incompatibility, and were followed by bacterial culture. Then Sanger sequencing was carried out and after analyzing the result by software ClustalX2 and IMTG/HLA database comparison, the HLA-B genotype of the samples was achieved. Seven clinical samples were detected, and the results were consistent with those of PCR-SBT genotyping method. The method was cost-effective, high-resolution and it did not require technical software. The use of universal primers simplified the cumbersome design and optimization process of specific primers.
5.Changes of myocardial membrane gene expression of natrium pump isoforms in rat with myocardial ischemia reperfusion injury

AIM: Changes of endoxin level, ATPase activities, intramitochondrial Ca2+ concentration, and gene expression of Na+-K+-ATPase isoforms in myocardium of rats with MIR and effect of verapamil were observed, in order to investigate mechanism of endoxin mediating intracellular calcium overload of myocytes. METHODS: Twenty four male Sprauge Dawley rats were randomized into 3 groups. Sham operation group: silk suture was threaded the left anterior descending coronary artery without ligature; MIR group (MIR): left anterior descending coronary artery was subjected to 30 min ligation followed by 45 min reperfusion; verapamil group: MIR model was given 5 mg/kg verapamil. Verapamil was injected via femoral vein 5 min before reperfusion. Left ventricle myocardium samples were processed immediately after reperfusion in order to measure the activities of Na+-K+-ATPase and Ca2+-Mg2+-ATPase, endoxin level, and intramitochondrial Ca2+ concentration. The levels of ?1, ?2, ?3 and ?1 isoforms of Na+-K+-ATPase were measured by immunohistochemical assay. RESULTS: After MIR, the level of endoxin in myocardium was substantially increased; the activities of Na+-K+-ATPase and Ca2+-Mg2+-ATPase in myocardial membrane were significantly decreased while the concentration of intramitochondrial Ca2+ was increased; the levels of the ?1, ?2, ?3 and ?1 isoforms of Na+-K+-ATPase were reduced markedly. Verapamil had only effect on reducing the concentration of intramitochondrial Ca2+. CONCLUSION: MIR increases endoxin secretion. The latter may depress the activity of Na+-K+-ATPase by changing the gene expression of ?1, ?2, ?3 and ?1 isoforms of Na+-K+-ATPase in myocardial membrane, inducing intramitochondrial Ca2+ overload.
6.Endoxin antagonist up-regulates gene expression of sodium pump isoforms in rats with myocardial ischemia reperfusion injury

AIM: To observe the effect of endoxin antagonist,anti-digoxin antiserum,on endoxin level,ATPase activities,intramitochondrial total calcium concentration and gene expression of sodium pump isoforms in myocardium of rats with myocardial ischemia reperfusion(MIR).METHODS: Fifty-six male Sprauge Dawley rats were randomly divided into 7 groups.Sham operation group: silk suture threading the left anterior descending coronary artery without ligature;MIR group: left anterior descending coronary artery was subjected to 30 min ligation followed by 45 min reperfusion;normal saline group: MIR model was given 5 mL/kg normal saline;verapamil group: MIR model was given 5 mg/kg verapamil;low dose antidigoxin antiserum group: MIR model was given 8.6 mg/kg antidigoxin antiserum;middle dose antidigoxin antiserum group: MIR model was given 17.3 mg/kg antidigoxin antiserum;high dose antidigoxin antiserum group: MIR model was given 34.5 mg/kg antidigoxin antiserum.All drugs were injected into vessel via femoral vein within 5 min before reperfusion,respectively.After reperfusion,left ventricle myocardium samples were processed immediately in order to measure the activity of Na+-K+-ATPase and Ca2+-Mg2+-ATPase,endoxin level,intramitochondrial total calcium concentration and the experssion of ?1,?2,?3 and ?1 isoforms of sodium pump on mRNA and protein levels by RT-PCR and Western blotting and immunohistochemical assay,respectively.RESULTS: After MIR,the level of endoxin in myocardium was obviously increased.The activities of Na+-K+-ATPase and Ca2+-Mg2+-ATPase in myocardial membrane were significantly decreased while intramitochondrial total calcium concentration increased.The gene expression of the ?1,?2,?3 and ?1 isoforms of sodium pump at both mRNA and protein levels were reduced markedly.Only the effect of verapamil on reducing intramitochondrial total calcium concentration was observed.Antidigoxin antiserum significantly reduced the level of endoxin in myocardium,restored the activities of Na+-K+-ATPase and Ca2+-Mg2+-ATPase,reduced intramitochondrial total calcium concentration,and up-regulated the expression of ?1,?2,?3 and ?1 isoforms of sodium pump at both mRNA and protein levels.CONCLUSION: MIR results in increase of endoxin secretion.The latter depresses the activity of Na+-K+-ATPase by down-regulating the gene expression of ?1,?2,?3 and ?1 isoforms of sodium pump in myocardial membrane,and also induces intramitochondrial calcium overload,thereby mediates MIR injury.
7.Genotyping of Alcohol Dehydrogenase Gene by Pyrosequencing Coupled with Improved Linear_after_the_Exponential Polymerase Chain Reaction Using Human Whole Blood as Starting Material

Pyrosequencing is one of the important genetic polymorphism detection methods currently, but the complicated pretreatment procedure limits its application in clinical test. To simplify the whole process of pyrosequencing, on the basis of the linear_after_the_exponential_polymerase chain reaction ( LATE_PCR) , we improved the primer design method of LATE_PCR, increased the length and the concentration of the excess primer, applied direct amplification technology with whole blood, and established a whole blood_imLATE_PCR method based on common rTaq polymerase and “HpH Buffer” ( High pH buffer ) . The amplification system was optimized, and the influences of blood anticoagulant and the amount of whole blood template were investigated. The single stranded template for the pyrosequencing was obtained by PCR amplification using a single tube in one_step process, and the alcohol dehydrogenase gene polymorphisms of 24 clinical blood samples were then detected successfully. The results could be used to guide clinical individualized medication. The genotypes of ADH1B locus of 24 samples were 6 cases of AA homozygote, 14 cases of AG heterozygote, and 4 cases of GG homozygote. The genotypes of ADH1C were 20 cases of GG homozygote, 4 cases of AG heterozygote, and no cases of AA homozygote.
8.Effects of nasofacial sulcus reversed fasciocutaneous flaps on medial canthal skin defects

Objective To discuss the clinical effect of medial canthal skin defects reparation with nasofacial sulcus reversed fascio-cutaneous flaps.Methods Medial canthal skin defects in 10 patients were repaired with nasofacial sulcus reversed fascio-cutaneous flaps in the hospital.These patients included 3 cases of pigmented nevus,6 cases of skin tumors,1 case of scar.The size of defects varied from 0.8 cm× 1.0 cm to 1.8 cm×2.2 cm.Results In these 10 cases,the maximum area of fascio-cutaneous flaps was 2.0 cm×3.0 cm,and the minimum area was 1.2 cm× 1.5 cm.All the flaps survived with primary healing postoperatively.Incision line scar was inconspicuous and the color and texture of flaps were similar to those of surrounding tissues.Conclusions To maintain and restore the integrity of medial canthal by using nasofacial sulcus reversed fascio-cutaneous flaps would lead to a good aesthetic effects.
9.Clinical efficacy and strategy of complications of blepharoplasty for double eyelids

Objective To investigate the causes,the prevention and the treatment of the complications of blepharoplasty for double eyelids.Methods The clinical data,the surgery methods and the causes were analyzed in 100 cases of blepharoplasty from July 2011 to December 2017.Results All of these patients underwent these perfect procedures with satisfied anesthesia.Incisions were all healed in one stage with no infection.Double eyelid width was (7.0± 1.2) mm with natural and smooth lines after 3 months operation,86 patients (86%) had double eyelid with natural and smooth lines,slight scars and good bilateral symmetry.14 patients (14%) required surgical revision for complications,as asymmetry of supratarsal folds (7 %),over narrow of supratarsal folds (4 %),over width of supratarsal folds (2 %),disappearance of supratarsal folds (2 %),multiple eyelid (3 %) and ptosis (1%),and the effects were satisfactory.Conclusions In order to achieve the natural and smooth double eyelids,it is important for plastic surgeons to have the comprehensive grasp of overall outline and operational details in blepharoplasty for double eyelids.Preoperative design should be accurate,intraoperative procedures should be careful,and postoperative care should be standardized.
10.Application of orbital septum fat reset for tear though deformity and palpbromalar groove in lower blepharoplasty

Objective:To explore the clinical application of orbital septum fat reset for tear though deformity and palpbromalar groove in the lower blepharoplasty.Methods:From March 2015 to March 2018, 72 cases of lower eye baggy deformity with tear though deformity and palabromalar groove were treated with the procedures of orbital septum fat reset, relaxing the tears ligament and orbicularis retaining ligament; the orbital fat that was released from orbital septum was repositioned at the inferior orbital and fixed on periosteum.Results:The incisions of 72 cases were healed successfully, the shape of the eyes was naturally symmetrical, the palpebral and buccal parts were excessive, and the contour lines formed a monoconvexcurve, with no obvious scar. All the cases were available for 6 to 24 months follow-up. 66 cases (91.67%) were satisfied with the cosmetic results.Conclusions:Orbital septum fat reposition is an effective method for correction of tear though and eyelid sulcus deformity.
