Objective:To determine the promoting effect of pyrrolidine dithiocarbamate(PDTC),an inhibitor of NF-?B,on the sensitivity of human ovarian cancer cell line SKOV-3 to chemoagent cisplatin and the possible mechanisms.Methods:SKOV-3 cells were treated with various concentrations of cisplatin and PDTC combination for different periods.Then the growth inhibition of SKOV-3 cells were observed by MTT assay,expression of I-?B? and P65 protein by Western blotting assay,and apoptosis of cells by flow cytometry.Results:Separate application of both cisplatin(0.1-10 ?g/ml)and PDTC(10-40 ?mol/L L)significantly inhibited the growth of SKOV-3 cells and caused cell apoptosis(P
2.Air Pollutants Change in Shenzhen, China in 2002-2005

Objective To study the change tendency of air pollutants in Shenzhen, China in 2002-2005, further, to explore the relationship between disease spectrum and air pollution. Methods The daily mean concentration of SO2, NO2 and PM10 in 2002-2005 in eight environmental monitoring sites were determined according to Ambient Air Quality Standard. Results Air concentrations of SO2, NO2 and PM10 increased yearly in 2002-2004, from 0.018 3 mg/m3 to 0.023 4 mg/m3, 0.050 1 mg/m3 to 0.072 5 mg/m3, 0.061 0 mg/m3 to 0.075 7 mg/m3, respectively, however, the concentrations of the three indexes decreased in 2005. In the past four yeas, average levels of SO2 were under the limit of grade 2 of GB3095-1996, as for PM10, under the limit of grade 2, and most of the concentrations of NO2 were under the limit of grade 3, but in the other two monitoring sites, the concentrations of NO2 in 2004 exceeded the limit of grade 3. The year average concentrations of SO2, NO2 and PM10 in 2002-2005 in the outer region were higher compared with those in the inner region of the special economic zone, except the level of NO2 in 2002. As for the quarter average concentrations of SO2, NO2 and PM10, 1st and 4th were higher compared with 2nd and 3rd. Conclusion The air quality in Shenzhen is better in China, however, more attention should be paid to the pollution of nitrogen oxides and inhalable particle matters.

4.Comprehensive Intervention on Behavior Problem of Elementary School Children

Objective:To study the effect of comprehensive intervention on behavior problems of elementary school children.Method:By means of cluster sampling, 1482 students from two elementary schools in Lixia district of Jinan were investigated with CBCL (Child Behavior Checklist) and a self-made mental health inventory for children. One of the two schools were selected as intervention site with 738 students, 744 students of the other school as control. The comprehensive intervention lasted for two years.Result:At the baseline, the rate of behavior problems in the intervention group was 14.8% (109/738), that of the control group was 15.2% (113/744), there was no significant difference between the two groups. After two years, the rate of the intervention group was 8.8%, that of the control group was 14.1%, there was significant decrease of behavior problems in the intervention group. After 2 years, the couple relationship of parents, parent-child relationship and education style of the family with child in intervention group improved than before.Conclusions:Comprehensive intervention focusing on behavior problems can help school children and their family.
5.Impact of NF-?B on P-glycoprotein-mediated multidrug resistance in ovarian cancer cells

Objective To investigate the expression of multidrug resistance gene 1 product P-glycoprotein(P-gp) when nuclear factor kappa B is induced or inhibited.Methods Ovarian cancer cells SKOV-3 were treated with Docetaxel or combinded with pyrrolidine dithiocarbamate(PDTC),the inhibitor of NF-?B.Western blot assay、 flow cytometry assay and MTT assay were used to measure P65 protein,P-gp,apoptosis of cancer cells,and the survival rate respectively.Results Docetaxel increased P65 protein and P-gp expression,while combind use of PDTC reversed this function.Both Docetaxel(≥(1 ?g/mL))and PDTC(≥(10 ?mol/L)) significantly inhibited the growth of SKOV-3 cells,and caused apoptosis.The combined use of Docetaxel and PDTC at low concentrations significantly inhibited the growth of cancer cells and caused apoptosis as compared with those treated with Docetaxel only.Conclusion The expression of P-gp may be ralated to the activation of NF-?B;Inhibition of NF-?B can enhance the chemosensitivity of ovarian cancer cells to Docetaxel.
6.Study of Arsenic Trioxide and Cisplatin on Human Ovarian Cancer Cell.

2?mol/L) was obvious and the inhibitive degree was nearly 100% when the concentrations of arsenic trioxide and cisplatin were 12?mol/L and 4?mol/L. 2.Higher concentrations of arsenic trioxide and cisplatin perturbed cell through S-phase and up-regulated p53 gene expressions.Conclusions The inhibitive effect of arsenic trioxide combined with cisplatin was more obvious than either one used separately; the apoptosis mechanism of ovarian cancer cell line HO-8910 is related with perturbing of S phase and up-regulating of p53 gene expression.
7.Application of transcranial doppler ultrasonograpy on brain death in severe craniocerebral injury

Objective To investigate and evaluate the diagnostic value of transcranial doppler ultrasound (TCD) on brain death in patients with severe craniocerebral injury.Methods Forty-two cases of severe craniocerebral injury from Affiliated Hospital of Jining Medical University were divided into brain death group and survival group,according to the clinical prognosis.TCD was conducted to examine brain blood flow numerical and changed characteristics of the spectrum.Average blood flow velocity (Vm),pulse index (PI),diastolic blood flow in reverse (reverberating flow) and small systolic peaks in early systole (wave of spikes)were documented.TCD parameters were compared between the two groups with t test.Results In the brain death group,frequency of reverberating flow was 86.67% and frequency of wave of spikes was 13.33%.In survival group,frequency of reverberating flow was 14.81% and no wave of spikes was recorded.The difference in frequencies of reverberating flow and wave of spikes between the two groups was significant(x2 =28.25,P <0.01).The average speed of bilateral middle cerebral artery blood flow in brain death group((20.02±13.96) cm/s) was significantly reduced compared with survival group((56.81± 16.84) cm/s,t=2.30,P<0.05).Pulse index (PI) values in brain death group (4.02±3.49)were significantly increased compared with survival group (1.24±1.03) (t=2.10,P<0.05).Conclusion Reverberating flow or wave of spikes is an important marker for brain death in patients with severe craniocerebral injury.TCD may be applied to clinically confirm the diagnosis of brain death.
8.Construction, expression, purification and polyclonal antibody preparation of Fas extracellular region gene

Purpose To construct expression vector of Fas extracellular region gene(eFas) ,to express and purify recombination protein and to prepare polyclonal antibody, which have laid a foundation of studying its function. Methods The eFas gene encoding sequence was acquired through overlapping PCR, and pET-22b ( + )/eFas expression vector was constructed. Then this vector was transformed into E. coli Rosetta-gami. Re-combinant protein was expression being induced by IPTG,and was purified using Ni-NTA matrix of affinity chromatograph. The purity of recombination protein was identified by SDS-PAGE. Hereafter, the purified eFas recombinant protein was immunized to New Zealand white rabbit in order to prepare polyclonal antibody. The titer of polyclonal antibody was determined by ELISA. Results The encoding sequence and expression vector of eFas was obtained while the interest protein was mainly expressed in the inclusion body. The eFas fusion protein's expression quantity accounts for more than 30% proportion of total E. coli protein. The eFas protein we obtained was provided with the purity of at least 95 % . Conclusion The successful constrution, expression and purification of FasL fusion protein and preparation of polyclonal antibody will provide some material for further studies of Fas.
9.Mathematical model of regulating competitive dysbacteriosis with antibiotics

Objective To explore the conditions for the restoration of competitive dysbacteriosis with antibiotics. Methods The mathematical model of the two competitive floras was analyzed by the Qualitative Theory of Ordinary Differential Equations. Results Three different types of dysbacteriosis and their restoring conditions were obtained. Conclusion Different restoring schemes should be applied for the regulation of different types of dysbacteriosis. Misuse of antibiotics can not result in satisfactory therapeutic effect.

Acute kidney injury ( AKI) is a clinical syndrome associated with increased morbidity and mortality, and early recognition is critical .In an attempt to engage laypeople and non-experts,the term kidney attack has been introduced to refer to acute kidney injury (AKI),and AKI with functional loss and tubular damage can be diagnosed both with functional criteria ( CRIAKI, creatinine increase AKI ) and damage criteria ( NCRIAKI, non-creatinine increase AKI ) according to the most recent ADQI consensus .This comprehensive definition of Acute Kidney Syndromes may be a major advancement in terms of early referral , prevention of progression and therapeutic strategies for AKI .
