Objective·To investigate the effects of insulin-like growth factor 1 on prion encoding gene (PRNP) expression and amyloid precursor protein (APP) metabolism of PC12 cells. Methods·After PC12 cells were treated with 20, 40, 80 ng/mL IGF-1 for 24 h, real-time PCR was used to detect the mRNA expression levels of PRNP, and Western blotting was used to detect the protein levels of AKT, phosphorylation of AKT (pAKT), ERK and phosphorylation of AKT (pERK). The level of β-amyloid 42 (Aβ42) in supernatant fluid was detected by ELISA. Results·Compared with the blank control group, the expression of PRNP mRNA in Alzheimer's disease (AD) model group was increased significantly (P<0.01). The expression of PRNP mRNA was significantly increased after cells were treated with 40 and 80 ng/mL IGF-1 for 24 h (P<0.01). There was no significant difference in APP protein expression among AD model group and three IGF-1 treatment groups (P>0.05). Compared with the blank control group, the level of Aβ42 in supernatant fluid of model group was decreased significantly with the increasing of IGF-1 concentration. The expression level of Aβ42 was decreased significantly in 40 and 80 ng/ml IGF-1 treatment group (P<0.05). The expression of AKT/pAKT, ERK/pERK in AD model group was significantly increased along with the increasing of IGF-1 concentration (P<0.05). Conclusion·IGF-1 could regulate the expression of PRNP gene and effect the metabolism of APP, which may be associated with PI3K/AKT, MAPK/ERK1/2 signaling pathway.

Objective To analysis the relation between diet,physical activities and chronic disease.Methods Collecting and analyzing the data of 3 673 Tianjin residences selected randomly as sample in 2002 China National Health and Nutrition Examination Survey.Dietary pattern and prevalence of chronic diseases were obtained,as well as the relative risk of diet,physical activities on chronic diseases including obesity,hypertension,diabetes and hyperlipidemia.Results The prevalent rates in Tianjin adults were 26.8% for obesity,3.07% for hypertension,32.2% for hyperlipidemia and 3.7% for diabetes.Main dietary problems among them were higher intake of oil and salt,lower intake of vegetable and fruit.The percentages of energy from fat ≥30% and watching TV≥3 hours/d were risk factors for overweight/obesity,hypertension,diabetes and hyperlipidemia with OR values as 2.04(95%CI:1.53～3.04),2.00(95%CI:1.13～3.57),2.22(95%CI:1.66～2.98),1.64(95%CI:1.18～2.31);1.89(95%CI:1.32～2.70),1.79(95%CI:1.48～2.16),1.47(95%CI:1.33～1.78)and 1.45(95%CI:1.28～1.65).Physical exercises ≥30 minutes /d was a protective factor for those four chronic diseases with OR values as 0.42(95%CI:0.28～0.68),0.60(95%CI:0.40～0.89),0.50(95%CI:0.35～0.80),0.61(95%CI:0.44～0.93)respectively.Intake vegetable and fruit ≥500 grams /d could reduce the risk for obesity and hyperlipidemia,OR values were 0.60(95%CI:0.40～0.89)and 0.47(95%CI:0.30～0.72).Conclusion There was a strong relation between diet,physical activities and chronic diseases in Tianjin sampled people.It should strengthen health promotion with balanced diet and good physical activity for the chronic diseases prevention.

Objective To analyze the epidemiological trend and distribution of stroke mortality in residents of Tianjin, China. Methods The study was based on 102 718 died cases with stroke in Tianjin between 1999 and 2006. The death cause and the stroke typing were coded according to the ICD -9 and ICD -10. Standardized mortality rates were calculated for stroke and its subtypes, adjusted for age and sex according to the world standard population of the Year 2000. The age, sex and geographic distribution of the stroke and its subtype mortality were analyzed. Chi-square tests were used to determine the statistical significance of mortality trends and differences. Results Stroke mortality rate in Tianjin declined from 133.52 / 100 000 / year in 1999 to 102.52 / 100 000/year in 2006. Cerebral infarction accounted for more than 50% of stroke mortality. Objective To know the primary effect of nutrition intervention in farmers. Methods The questionnaires were used to do baseline and evaluation survey. The intervention ways were used in trial group, including holding lectures, posting posters, sending out healthy material, quizzes with prizes, recommending salts low in sodium to each family, training to make healthy food by the chef; but there were no measures in control group. Results There were statistical significance in healthy awareness of nutrition, exercises and chronic diseases between two groups after intervention; However, the average intake amounts of vegetables did not reach the recommendation of diet guidelines; After intervention, the intake frequency of vegetables and fruits had statistical significance between before and after intervention in trial group, and also had statistical significance in two groups. Conclusion The awareness of healthy knowledge and attitudes has been obvious improvement, but the diets and life styles of farmers have no obvious change. The diet structure and behavior of farmers have been increasingly rational but not enough. The farmers have started to increase the intake times of vegetables and fruits and built up the conception of eating more vegetables and fruits.

Objective To investigate the relationship between infarct location and prognosis after emergency percutaneous coronary intervention (PCI) treatment in patients with acute myocardial infarction.Methods Consecutive 121 patients with ST elevation myocardial infarction (STEMI) who underwent primary PCI were divided into 3 groups according to the infarct location.Their clinical,ECG,echocardiography and coronary angiography features were analyzed,and the follow-up was made. Results Compared with inferior wall infarction group and inferior wall infarction combined with right ventricular infarction group,anterior wall myocardial infarction group had higher CPK-MB and cTnT,less frequent collateral,reduced ST-segment resolution,higher mortality rates in hospitalization and one year after discharge.On the other hand,inferior infarction combined with right ventricular infarction group had more occurrences of shock and atrial vetricular block or ventricular arrhythmia,more incidence of multivessel disease and thrombus,and higher readmission rate.Conclusion Patients with anterior wall myocardial infarction and inferior wall in

AIM: To establish a method for determing camphor and eucalyptol in Shidi Water(Camphor,Rhizoma Zingiberis,Fructus Foeniculi,Cortex). METHODS: The chromatographic condition,DB-WAX capillary column(30 m?0.53 mm,1 ?m).The column temperature programming was from 65 ?C to 155 ?C(at rate of 6 ?C/min).FID Detection.The temperatrue was at 230 ?C. RESULTS: The linear range of camphor was within 0.18 mg/mL-5.78 mg/mL(r=1.000 0,n=6).The average recovery was 98.05%,RSD=0.94%.The linear range of eucalyptol was within 0.06 mg/mL-1.92 mg/ml(r=(0.999 7),n=6).The average recovery was 98.99%,RSD=1.31%. Objective To observe the curative effect of transplantation of mesenchymal stem cells (MSCs) transfected with recombinant lentiviral vectors carrying brain-derived neurotrophic factor (BDNF) gene on intracerebral hemorrhage in rats.Methods MSCs were isolated from the rat bone marrow,cultured and transfected by recombinant lentiviral vectors carrying BDNF gene.Intracerebral hemorrhagic models were constructed and randomly divided into 4 groups:phosphate buffered saline transplanted (PBS) group,MSCs group,mesenchymal stem cells transfected with empty lentiviral vectors transplanted (MSCs-EGFP) group and mesenchymal stem cells transfected with recombinant lentiviral vectors carrying brain-derived neurotrophic factor gene transplanted (MSCs-EGFP-BDNF) group.PBS and MSCs were transplanted according to the groups 72 hours after the establishment of models.The improvements of the neurological function were recorded of each group 7 d,14 d,and 21 d after the transplantation.Double labeling immunofluorescent staining were used to detect the migration and the differentiation of transplanted MSCs.Results MSCs-EGFP-BDNF group had significant higher levels of BDNF gene and protein expression than MSCs group and MSCs-EGFP group.All MSCs transplanted groups (MSCs groups:7 d:1.6 ±0.2,14 d:1.2 ±0.3,21 d:0.8 ±0.2; MSCs-EGFP groups:7 d:1.6 ±0.3,14 d:1.1 ±0.2,21 d:0.8 ±0.3; MSCs-EGFP-BDNFgroup:7 d:1.2 ±0.3,14 d:0.6 ±0.1,21 d:0.2±0.2) had more improvements in the neural function (F=6.667,18.417,20.882,all P ＜0.05) than PBS group(7 d:2.0 ±0.4,14 d:1.7 ±0.2,21 d:1.3 ±0.2),and MSCs-EGFP-BDNF group had the most significant improvement.With double labeling immunofluorescent staining,the MSCs-EGFP-BDNF group had significantly higher positive rates of glial fibrilary acidicprotein,neuron specific nuclear protein,2',3 '-cyclic nucleotide 3'-phosphodiesterase than MSCs group and MSCs-EGFP group,while there was no significant differences between the latter two.Conclusions The expression levels of gene and protein are higher for the MSCs modified with recombinant lentiviral vectors carrying BDNF gene.The modified MSCs can migrate to the perihematomal brain issue of intracerebral hemorrhage,express the characteristic molecules of neurons and improve the neural function after intracerebral hemorrhage.

Objective To explore the effect of the diet recovery time on postural hypotension after gynecological laparoscopic operation. Methods A total of 300 patients undergoing gynecological laparoscopy were randomly divided into observation group and control group according to the order of admission,150 cases in each group. Patients in observation group took liquid food since recovery from anesthetic, their appetite and dietary levels restored to preoperative level within 24 h after surgery from liquid diet to regular diet. Patients in control group were given routine nursing care of diet after belly operation. Results The diet recovery time and the anal exhaust time of patients in observation group were (21.2±3.5) h and (15.5±4.7) h, the control group were (46.8±5.4) h and (23.4±5.4) h, there was significant difference between two groups (t=7.543,5.126, P<0.05).The incidence rate of orthostatic hypotension in observation group was 9.3% (14/150), the control group was 33.3% (50/150), there wassignificant difference between two groups (χ2=8.907, P<0.05). Conclusions Patients undergoing gynecological laparoscopy took food as soon as possible, their appetite and dietary levels restored to preoperative level within 24 h after operation. It didn′ t increase abdominal distension, but helped the intestinal function recovery and helped to reduce the incidence of postoperative orthostatic hypotension.

Objective To explore the causes of death and life expectancy after elimination of main causes of disease in residents of Tianjin. Methods The death registry data of Tianjin residents in 2014 were collected and coded in“international classification of disease, 10th edition”. The crude death rate and life expectancy after elimination of main causes of disease were calculated, respectively. Results In 2014, the crude death rate in Tianjin residents was 70.708 per million, while in male and female were 78.728 and 62.637 per million respectively. The main cause of death in Tianjin residents was non-communicable disease. The top four death causes were heart disease, cancer, cerebrovascular disease and respiratory disease, accounting for 31.5%, 23.6%, 22.2% and 8.3% of the total death. The top four life expectancy lost diseases were heart disease, cerebrovascular disease, cancer and respiratory disease, with a 6.46 year, 3.28 year, 3.11 year and 1.25 year life increase respectively. Conclusion Non-communicable diseases are the major reason of death and life expectancy lost disease in Tianjin residents, which needs urgent effective intervention to control.

Objective: To explore the acute effect of air pollution on mortality for patients with cardio cerebral vascular disease and to provide the basis for disease prevention and control.
Methods: The Mortality for patients with cardio cerebral vascular disease from 2001-01 to 2009-12 was from Tianjin Centers for Disease Control and Prevention, the meteorological and air pollution data were from Tianjin meteorological bureau and Tianjin environmental monitoring station respectively. The Single and multiple generalized additive model (GAM) extended poisson regression analysis was performed to calculate the relationship between air pollution and cardio-cerebral vascular disease mortality by controlling the time trends, weather, the day of week and air population.
Results: Results: Single GAM analysis indicated that when the effect of SO2, NO2 and PM10 in the air reached the maximum in the day, the RR values for the mortality in patients with cardio-cerebral vascular disease increased 1.13%[95%CI (0.76-1.51)%], 0.78% [95%CI ( 0.41-1.15)%] and 0.61% [95%CI ( 0.51-0.71)%] respectively; when the average concentration of SO2, NO2 and PM10 increasing 10μg/m3 per day, after 0-5 days, the RR values for the mortality elevated 0.70% [95%CI (0.47-0.94)%], 0.51% [95%CI (0.27-0.74)%] and 0.16% [95%CI (0.06-0.27)%] respectively. Multiple GAM analysis presented that when SO2, NO2 and PM10 increasing 10 μg/m3 per day, the RR values for the mortality elevated 0.77% [95%CI (0.58-0.97)%], 0.41% [95%CI (0.05-0.78) %] and 0.38% [95%CI (0.12-0.64%)%] respectively.
Conclusion: The air pollution could increase the mortality risk in patients with cardio-cerebral vascular disease, it is necessary to establish the prevention system in order to decrease the mortality risk in those patients.
