1.A study of FUT2 gene point mutation in the Chinese Han Population

Objective To study the point mutation of FUT2 gene in Chinese Han population.Methods Using direct sequencing,molecular cloning techniques and the comparing with the gene sequence reportedby Kelly, the FUT2 gene structures of 41Chinese Han individuals have were studied.Results The G428A mutations of FUT2 gene was not found,but the A385T and C357T mutations were found in the 41 Chinese Han individuals.Among the 41 individuals,24 had A385T mutation and 17 had no A385T mutation.The neutral mutation C357T was found in all 41individuals.Conclusion The G428A point mutation of FUT2 which is commonly found in non secretor of Africans and Caucasian was not found in Chinese population.There are A385T and C357T point mutations which were found in 41 Chinese Han individuals.The present stady shows the difference between Chinese and Caucasian,and other non secretor mutations will be revealed by further investigation
2.Study on the FUT2 gene structure of Xinjiang Uighur people of China

Objective To study the Secretor gene (FUT2) molecular structure of Uighur population in Xinjiang area,China. Methods DNA was extracted from 40 Uygur unrelated donors' blood and sequence analysis of FUT2 genes was performed. Results Four mutations in the FUT2 genes of Uighur donors have been identified. The frequencies of mutations were 71.25% for 357T, 28.75% for 357C,77.50% for 385A,22.50% for 385T,70% for 428G,30% for 428A,72.50% for 739G and 27.50% for 739A. Conclusion Based on the characteristics of FUT2 gene structure of Xinjiang Uighur,it cauld be thought that there are some relationships between Xinjiang Uighur, Taiwanese of China and Caucasiany.
3.Protective effects of riboflavin on diabetic nephropathy in STZ-induced diabetic rats

AIM:To explore the protective effects of riboflavin on the kidney in streptozotocin (STZ)-induced diabetic rats.METHODS:Male Sprague-Dawley rats were randomly divided into 3 groups:normal control group,diabetic model group and riboflavin-treated group.Diabetes was induced by a single injection of STZ (dissolved in 0.01 mol/L citrate buffer,pH 4.5,65 mg/kg,ip) in rats.The biochemical methods were used to measure the contents of urine protein and malondialdehyde in the kidney,and the activities of superoxide dismutase (SOD) and catalase (CAT) in serum and renal tissues.Furthermore,the protein expression of TGF-?1 and plasminogen activator inhibitor-1(PAI-1) in renal cortex was detected by Western blotting.The morphological changes of renal tissue were observed under microscope.RESULTS:Compared to the diabetic model group,riboflavin significantly increased the activities of SOD and CAT (P
4.Quantitative Study of MRI and~1H MRS in Normal Adult Brain

Objective:To evaluate magnetic resonance imaging(MRI) and 1 H magnetic resonance spectroscopy( 1HMRS)in the study of normal biochemical process of the brain as well as differentiation of normal senile brain from cerebral diseases related to senility.Methods:180 cases of healthy adults were selected to perform MR examination,in which,none of the subjects had the history of neurological and psychotic diseases according to MRI and clinical results.Meanwhile,60 healthy subjects were selected to perform 1 HMRS examination.The ages ranged from 18 to 80 years.They were divided into six age groups purposely.Point resolved spectroscopy sequence was required for 1 HMRS.The metabolites in the spectra included:N-acetylaspartate(NAA),choline compounds(CHO),creatine compounds(CR),myo-inosito(MI),glutamate and glutamine(Glu-n).Results:(1)In 180 cases of MRI,T 2 relation time was lowest in the deep gray matter in the same age group;T 1relation time was in low-high order,while T 2 relation time was decreased with the increase of age in the different age group.(2)The amplitudes in high-to-low order were as follows in 60 cases of 1 HMRS:NAA、CR、CHO、MI、Glu-n.No prominent difference of shape and peak arrangement was seen at the different ipsilateral site in the same age groups;while slight difference at the same site in the different age groups was present.The ratio of NAA/Cr and Glu-n CR was higher in senile age group;while that of MI/Cr was lower.The ratio of CHO/CR was in low-to high order with the difference of age.The ratio of NAA/CR and MI/CR was gradually lower from anterior to posterior part of the brain;the ratio of CHO/CR was highest in occipital cortex however,no definite changing rule was observed in the ration of Glu-n/CR.Correlation of T 1 relation time and partial metabolite ratio with age was present in gray matter.Conclusion:Quantitative study of MRI and 1 HMRS is essential for determination of normal myelinization and neuronal integrity and age-related biochemical changes in the brain.
5.Effects of hydrogen sulfide on wound healing of skin ulcer in diabetic rats

AIM: To study the effect of hydrogen sulfide on wound healing of skin ulcer in diabetic rats . METHODS:Male SD rats were randomly divided into 3 groups, including non-diabetic control (NDC) group, untreated diabetic control ( UDC) group, and treated diabetic administration ( TDA) group.Diabetic rats were induced by intraper-itoneal injection of streptozotocin (STZ).After 1 week, wound healing model was prepared by making a round incision ( 2.0 cm in diameter ) on the dorsal skin in full thickness .The rats from TDA group received 2%sodium bisulfide ointment on the skin ulcer wound , and the animals from UDC and NDC groups received control cream .After 21 d of treatment with sodium bisulfide , blood samples were collected for biochemical analysis , including prothrombin time ( PT) , thrombin time (TT), and fibrinogen (FIB) in plasma, as well as the activity of superoxide dismutase (SOD) and the content of malondi-aldehyde ( MDA) in the serum.White blood cells ( WBC) and lymphocytes were also counted .Granulation tissues from the wound were processed for histological examination and Western blot analysis was used to detect heme oxygenase -1 ( HO-1) and tumor necrosis factor α(TNF-α) expression.RESULTS:Compared with UDC group, sodium bisulfide treatment accelerated wound healing of skin ulcer (P<0.01), and increased the activity of SOD in serum (P<0.01) in the diabet-ic rats.The declined number of WBC and lymphocytes , prolonged PT and TT , and decreased FIB levels in rats treated with sodium bisulfied were also confirmed .Pathological section showed that there were inflammatory cell infiltration , and irregu-lar and loose fibril alignment in the granulation tissue of rats from the UDC group , but there were regular fibril alignment and increased angiogenesis in the granulation tissue of rats from the TDA group (P<0.05).Furthermore, sodium bisulfide treatment raised HO-1 protein expression , and decreased TNF-αprotein expression in the diabetic rats .CONCLUSION:Hydrogen sulfide accelerates the wound healing of skin ulcer in the rats with diabetes .The beneficial effect of H 2 S may be associated with formation of granulation , anti-inflammation , and antioxidation .
6.Video assisted small incision in treatment of infected pancreatic necrosis

Objective:To study the indications and clinical efficacy of video assisted small incision in treatment of infected pancreatic necrosis.Methods:A retrospective study was conducted on 27 patients with infected pancreatic necrosis treated by video assisted small incision at the Department of Pancreatic and Splenic Surgery, Hunan Provincial People's Hospital, from January 2018 to December 2019. The surgical approach, operation time, intraoperative blood loss, postoperative hospital stay and complications were analysed. Postoperative follow-up was carried out at outpatients’ clinic, and the patient's time to full recovery and long-term complications were studied.Results:The 27 patients successfully underwent the operations. There were 22 males and 5 females, aged (50.6±6.2) years. The treatment results were analyzed according to the different surgical approaches: the retroperitoneal approach group ( n=4); the omental sac approach group (n=14); the intercostal space approach group ( n=2); and the combined approach group ( n=7). The operation time was (85.3±31.6)min. The intraoperative blood loss was 65.0(45.2, 121.4)ml. The postoperative hospital stay was 23.0(12.5, 36.1)days. The incidence of complications (Clavien-Dindo grade Ⅲ and above) was 14.8%(4/27). There were 2 patients in this study who were admitted to the intensive care unit due to postoperative hemorrhage: 1 patient responded well to conservative treatment and the remaining patient required interventional treatment. Another patient because of poor results, underwent debridement by laparotomy 2 weeks after the operation. There was 1 patient who developed grade C pancreatic fistula which was cured by surgical treatment 6 months later. On follow-up, 2 patients developed colonic fistula 2 weeks after surgery and 2 patients gastric fistula 1 week and 3 weeks after surgery. These patients responded to conservative treatment. Conclusion:With proper case selection, video assisted small incision could safely and effectively be used to treat infected pancreatic necrosis.
7.A novel O~1 variant allele at the ABO blood group locus

Objective To identify novel ABO allele in Chinese population. Methods The ABO blood group was tested by serological method, and then genotyped by sequence-specific primer (PCR-SSP) , gene cloning and sequence analysis. Results A healthy blood dornor who was diagnosed as having A2 subgroup and A2O1genotype was subjected to ABO gene cloning and sequence analysis. The haplotype-specific sequence analysis indicate that two single-base deletions, where G-deletion at nucleotide position 261 and A-deletion at nucleotide position 496 were determined in the O1 allele. The nucleotide sequence of the novelO1 allele were identical to ABO 0101 allele except for A-deletion at nucleotide position 496 in exon7 of ABO locus. Conclusion We defined this 0 allele as a novel O1 variant allele, and its registered number by GenBank is AY374123.
8.Mutational analysis of FUT1 and FUT2 genes in para-Bombay individuals

Objective To study the mutation of FUT1 and FUT2 genes in para-Bombay individual.Methods Direct DNA sequencing of FUT1 and FUT2 gene coding region were analyzed in two individuals with para-Bombay phenotype.Results One individual lost one of the three AG repeats located at nucleotides 547~552 of the FUT1 gene, whereas two of the three T repeats located at nucleotides 880~882 were deleted in the other.Conclusion Two frame-shift mutations of FUT1 gene are responsible for the H antigen deficiency
9.Surgical experience of refractory cholelithiasis:a report of 521 cases

Objective To summarize the experience in operation manner and surgical technique of refractory cholelithiasis.Methods A total of five hundred and twenty one patients with refractory cholelithiasis admitted to Hunan Provincial People's Hospital from Jan.1990 to Dec.2007 were involved in this study for retrospective analysis.Results All patients in this group accepted surgery.Apart from three cases of perioperative death with liver and kidney failure,the remaining five hundred and eighteen cases had no serious complications,were cured and discharged.The imaging examination showed residual stone in seventy cases,accounting for fifteen percent.Four hundred and twenty one patients were followed up.The mean time of follow-up was seven years and six months (range 5 months - 17 years).Good result rate was 90.1%(381/423).Conclusions Most intractable cholelithiasis can be cured radically.Individual surgery programme,fine and standard surgical procedure are the key to treatment effect.
10.Expression of FBXW7 in a murine model of T-cell acute lymphoblastic leukemia

Objective To investigate the expression of FBXW7 during the development of Notch1induced murine leukemia.Methods Notch1 over-expressing murine model of T-cell acute lymphoblastic leukemia was used to study the expression of FBXW7 by real-time PCR methods.Bone marrow mononuclear cells (BMNC) were isolated on different days after transplantation and CD45.2+ GFP+ leukemia cells were sorted by flow cytometry at late stage.The expression changes of FBXW7 were tested by real-time PCR.Results The mouse bone marrow cells both from leukemia and control groups expressed FBXW7.Different expression patterns of FBXW7 were observed during the development of leukemia. The expression of FBXW7 was gradually increased in control group, whereas the expression level of FBXW7 in leukemia group was decreased steadily and reached one-sixth of that in control group on 12th day.Furthermore,lower expression level of FBXW7 was observed in CD45+.2 GFP+ leukemia cells.Conclusion Decreased expression of FBXW7 is observed in Notch1-induced mouse leukemia model,suggesting that the abnormal ubiquitin degradation pathway mediated by FBXW7 might contribute to the leukemogenesis in Notch1-induced murine leukemia model.
