1.Expression of COX-2 in Bladder Transitional Cell Carcinoma and its Association with Tumor Cell Proliferation and Apoptosis

Objective To investigate the expression level of cyclooxygenase-2 (COX-2) and its effects on cell proliferation and apoptosis in bladder cancer. Methods The immunohistochemical method was used to determine the expression of COX-2 and Ki67 in 54 cases of bladder cancer, 29 cases of adjacent tumor tissue and 10 cases of normal bladder specimen respectively, and the apoptotic levels of the obove three tissues were assessed by counting the number of the cells labeled by TUNEL method. Results The positive rate of COX-2, the mean labelling index of Ki67 and the cellular apoptosis index in the cancer tissues were significantly higher than those in adjacent tumor tissues and normal bladder specimens(P

BACKGROUND: Titanium cage of cervical vertebral fusion is often used during a surgical operation for cervical syndrome; however, it has a bad impact on the operative effect because of submerging.OBJECTIVE: To observe the biocompatibility and stability of cervical fusion cage of high molecular polyethylene in animals.DESIGN: A randomized grouping and observational control experiment.SETTING: The Animal Experimental Center of Zhengzhou University.MATERIALS: Twenty female goats without pregnancy at 1.1-1.6 years of age.METHODS: The experiment was conducted at the Animal Experimental Center of Zhengzhou University between June and November 2001. Twenty goats were equally randomized into experimental group and control group.Self-made cervical fusion cage filled with autogenous spongy bone was implanted into the goats in the experimental group, only autogenous iliac bone was implanted in the control group. X-ray for observing the changes in the height of intervertebral space was performed in two groups 6 weeks later. All the animals were killed under anesthesia to extract cervical vertebra samples, which were placed on YJ-14 Biomechanical Tester to measure anti-compression ability and height of intervertebral space and to observe pathological changes.space: Compression loading in the experimental group was significantly higher than that in the control group [(358.64±15.63),(268.82±11.36)N,P < 0.05]; however, there was no difference in the height of interverteA great amount of osteoblasts were found in the tissues around the cage in the experimental group, but there was no difference between the two groups.CONCLUSION: The self-designed cervical vertebral fusion cage possesses high upholding strength, good biocompatibility and stability and can play a role in stabilizing the height of intervertebral space.
3.Fluorescence in situ hybridization analysis of HER-2/neu oncogene amplification in osteosarcomas

Objective To investigate whether HER-2/neu oneogene amplification exists in 08- teosarcomas. Methods Fluorescence in situ hybridization(FISH)and DNA probes for HER-2/neu and centromere 17 were used to examine the HER-2/neu gene amplification in samples taken from 23 patients with osteosarcoma. Results None of the 23 osteosarcoma samples showed the HER-2/neu gene amplification.Condusion Since HER-2/neu gene amplification is rare in osteosarcomas, HER-2/neu target medicine should not be applied to patients with osteosarcoma
4.Effect of timing of initiation of renal replacement therapy on prognosis in septic patients with acute kidney injury:a Meta analysis

ObjectiveTo systemically review the effects of timing of initiation of renal replacement therapy (RRT) on prognosis in septic patients with acute kidney injury (AKI).Methods A systematic search for randomized controlled trials (RCTs) and other clinical studies focused on comparing varied timing of initiation of RRT in septic AKI patients was performed in English or Chinese from PubMed, Web of Science, EMBASE, CNKI, Wanfang data, VIP from January 2000 to July 2014. Data screened were extracted with Cochrane systemically review method, and enrolled literature was collected for Meta analysis with RevMan 5.2 software. Total mortality, 28-day mortality, the total length of hospital stay and intensive care unit (ICU) stay in septic AKI patients with early or late initiation of RRT was analyzed. Funnel plots were drawn to detect publication bias.Results Five retrospective comparative studies with a total of 885 patients were enrolled. Random effect model in Meta analysis showed that there was no significant difference in total mortality between early RRT group and late RRT group [65.7% (226/344) vs. 68.7% (239/348), odds ratio (OR) = 0.71, 95% confidence interval (95%CI) = 0.38-1.31,P = 0.27]. The funnel plot demonstrated publication bias. Fixed effect model showed that there was significant difference in 28-day mortality between early RRT group and late RRT group [43.4% (66/152) vs. 55.3% (94/170),OR = 0.59, 95%CI = 0.36-0.94,P= 0.03]. The funnel plot demonstrated publication bias too. The data of total length of hospital stay and ICU stay could not be Meta-analyzed because of different data distribution, but no differences in total length of hospital stay and ICU stay between early and late RRT groups for septic AKI patients were found.ConclusionEarly RRT could reduce the 28-day mortality in patients with septic AKI compared with late RRT, but it did not lower the total mortality.
5.Expressions of CaSR and Claudin-14 in Renal Calcium Oxalate Stone Model of Rats

Objective To investigate the expressions of calcium sensitive receptor (CaSR) and tight junction protein (Claudin)-14 in renal calcium oxalate stone rat model. Methods Thirty male SD rats were randomly divided into control group (n=15) and model group (n=15). The rat model of renal calcium oxalate stone was established by gavaging 1%glycol (2 mL/d) and 2% ammonium chloride. The expression of CaSR protein was detected using immunohistochemical assay. RT-PCR was used to detect the Claudin-14 mRNA expression. The expression levels of CaSR and Claudin-14 protein were de-tected by Western blot assay respectively. The full automatic biochemical analyzer was used to detect rat renal function and changes of blood and urine biochemical indices. Results A large stone crystallization was observed under light microscope in model group. The serum levels of Cr, BUN, 24-h urine calcium and urine volume were significantly higher in model group than those of control group (P<0.01). There were no significant differences in serum calcium level and urinary pH value be-tween two groups. The expression levels of Claudin-14 mRNA and CaSR protein were significantly higher in model group than those of control group (P<0.01). The Claudin-14 protein expression was specifically higher in renal tissues of model group. There was no Claudin-14 protein expression in control group. There was a positive correlation between CaSR and Claudin-14 protein expression in renal tissues of model group. Also, CaSR and Claudin-14 protein expressions were posi-tively correlated with the 24-h urine volume. Conclusion The increased expressions of CaSR and Claudin-14 involved in the formation of kidney stone by increasing the urinary calcium excretion.
6.Comparison of sequence between adenoviral DNA and Escherichia coli DNA

Objective To identify the putative CpG-N ODNs in adenovisus 2 DNA (Adv2 DNA) and Adv5 DNA by comparing the sequence difference among Adv2, 5, 12 DNA and E.coli (EC) DNA. Methods Sequences of Adv2, 5, 12 DNA and EC DNA were obtained from the Entrez Nucleotides database at NCBI. The specific CpG motifs of Adv2 DNA and Adv5 DNA were identified after above sequences were analyzed and compared by softwares such as DNATools, BioEdit, and so on. All the 12-ODNs with specific CpG motif core were searched from Adv2 DNA and Adv5 DNA. Results 19 specific CpG motifs were ascertained and 504 12-ODNs were detected in Adv2 DNA and Adv5 DNA. Conclusion 504 12-ODNs were putative CpG-N ODNs in Adv2 DNA and Adv5 DNA.
7.Screening of bacterial DNA antagonizing CpG oligodeoxynucleotides

Objective To obtain a potent bacterial DNA antagonizing CpG oligonucleotides (CpG-N ODN) from the structures of Adv2, 5 DNA. Methods Ten putative CpG-N ODNs were synthesized and investigated. Their abilities to inhibit the TNF-? release from hPBMC were observed. Based on the above results, the putative CpG-N ODN was redesigned according to the relationship between the structure and free energy using RNA structure software (version 3.71). Eleven putative CpG-N ODNs were synthesized and screened. Results Out of the ten initial CpG ODNs, ODN101 was the only CpG-N ODN with weak activity to inhibit TNF-? release from hPBMC induced by CpG-N ODN. After redesign, five CpG-N ODNs with strong activity were confirmed. Conclusion Six CpG-N ODNs were obtained with activity to inhibit TNF-? release from hPBMC induced by CpG-N ODN.
8.Effects of different catheterization on patients with oral poisoning gastric lavage:a meta-analysis with trial sequential analysis

Objective To assess on the effect of the different catheterization on patients with oral poisoning gastric lavage systematically and objectively. Methods A computerized search of PubMed, the Cochrane Library, EMBASE, Springer Link, Science Direct,China Biology Medicine (CBM),China National Knowledge Internet(CNKI), Wanfang Data and VIP database was performed for relevant randomized controlled trials (RCTs) which investigated the effects of the oral intubation gastric lavage on patients with oral poisoning relative to nasogastric lavage, retrieval to January 2016, and back into the study of references. According to include and exclude standard to screening literature, literature which met inclusion criteria was selected for quality evaluation and data extraction. Meta-analysis and trial sequential analysis were performed by using Rev-Man 5.3 and TSA soft-ware to estimate the required information size (RIS). Results 13 studies incorporated into Meta-analysis including a total o 1 296 patients. Meta-analysis results showed that the oral intubation gastric lavage group was better than nasogastric lavage in shortening the time of intubation [MD=-9.05, 95% CI(-12.86--5.23), P=0.00], improving the rate of intubation for the first time [MD=1.36, 95% CI (1.14-1.62), P =0.00], mucosal bleeding complication [MD=0.11, 95% CI (0.04-0.64), P=0.010], nausea and vomiting [MD=0.60, 95%CI (0.37-0.98), P=0.04], kinking [MD=0.14, 95% CI (0.02-0.80), P=0.03], reflex cardiac arrest [MD=0.24, 95% CI (0.08-0.71), P=0.01] and asphyxia [MD=0.45, 95% CI (0.26-0.80), P=0.007], the difference were statistically significant. But no significant difference existed in gastric tube fall off. Conclusions The oral intubation gastric lavage could shorten intubation time of patients, improve the first time intubation rate and reduce certain complications. It suggests that the oral intubation be extensively applied to care patients with oral poisoning.
9.The study of transfection through perfusing bladder of guinea pig with lentivirus

Objective To observe the effects of green fluorescence protein mediated by lentivirus in bladder, and to de?termine the amount of virus obtained good transfection effects. Methods lentivirus carring GFP gene was perfused using transurethral approach into bladder of guinea pigs. Samples of bladder, liver, kidney and lungs were collected for frozen sec?tions after feeding seven days. The distribution of green fluorescence was observed using laser confocal microscopy. Re?sults The titer of lentivirus was 4 × 108. GFP was found under the mucosa when the amount of lentivirus transurethral was 30μL. GFP was distributed widely in muscle layer using 40μL lentivirus. GFP was detected even stronger in muscle layer when the amount was 50μL. GFP was found in muscle layer when 25μL lentivirus was injected intravenously. GFP was not found in other tissues than in bladder via transurethral perfusion. There was higher GFP in liver, lungs and other organs than in bladder via intravenous injection. Conclusion Lentivirus can mediate GFP transfecting bladder of guinea pig successful?ly and escape the distribution of GFP all over the body intravenously, which will bring new research direction and method for clinical treatment of diseases in bladder.
10.Clinical analysis of cerebral angio spasm after aneurysm rupture subarachnoid hemorrhage by Fasudil

Objective To explore the clinical curative effect of cerebral angio spasm after aneurysm rupture subarachmoid hemorrhage by Fasudil, Methods The clinical data of cerebral angio spasm after aneurysm rupture subarachnoid hemorrhage were retrospectively analyzed, which was divided into detection group and control group.Results The GCS score of detection group was better than control group;the average blood flow rate of arteria cerebri media after curing in detection group the difference had statistical significance( all P <0. 05 ). The mutilation rate、rate of death of detection group were lower than control goup, the difference had statistical significance ( P < 0. 05 ).Conclusion The clinical curative effect was good of cerebral angio spasm after aneurysm rupture subarachnoid hemorrhage by Fasudil, and prognosis was good.
