1.Advances in Study on Gastrointestinal Mucosal Low-grade Inflammation in Functional Dyspepsia

Functional dyspepsia( FD)is a common functional gastrointestinal disorder,its pathogenesis has not been fully clarified. Recent studies showed that gastrointestinal mucosal low-grade inflammation had been found in patients with FD,and impaired mucosal integrity,psychological factor and abnormal brain-gut axis may promote the inflammation formation,which may also contribute to the onset and persistence of FD. This article reviewed the advances in study on gastrointestinal mucosal low-grade inflammation in FD.

3.Clinical Observation of Centro-square Needling at Shangxing (GV23) for Anxiety After Cerebral Stroke

Objective To observe the clinical efficacy of centro-square needling at Shangxing (GV23) (so called Ghost Hall point) in treating anxiety after cerebral stroke as a major treatment.Method Sixty patients were randomized into a treatment group and a control group. The two groups were both given Flupentixol and Melitracen tablets (Deanxit). In addition, the treatment group received centro-square needling as the major treatment, and the control group received ordinary needling. Hamilton Anxiety Scale (HAMA), Zung's Self-rating Anxiety Scale (SAS), and Barthel Index (BI) of Activities of Daily Living (ADL) were observed before and after the treatment for evaluating the therapeutic efficacy.Result There was a significant difference in comparing the HAMA score between the treatment group and control group after the treatment (P<0.05). There were no significant differences in comparing the total effective rate and recovery rate between the two groups (P>0.05).Conclusion The treatment group shows a significant efficacy in improving the anxiety symptoms; the effect in improving the anxiety symptoms is insignificant in the control group; the recoveryrate and total effective rate are equivalent between the two groups.
4.Prevalence rate of dyslipidemia,hyperglycemia,hypertension and metabolic syndrome in a large occupational population in Beijing

Objective To investigate the prevalence of dyslipidemia,hyperglycemia,hypertension and metabolic syndrome(MS)and compare the prevalence rate of dysilipidemia obtained in this study with the previous study during 1984--1986.Methods A total of 16 344 adults in Beijing who attended for the medical examinations were recruited in the study.Subjects were asked to complete questionnaires regarding personal health,lifestyle and family health history.The physical examination emphasized measurement of height,weight and blood pressure.Blood samples were collected after an overnight fast and subjected to serum glucose,TC,HDL-C,LDL-C and TG measurements.MS was diagnosed according to Diabetes Society of Chinese Medical Association(CDS)criteria.Results Dyslipidemia occurred in 36.9%(6 034)of subjects.948 hyperglycaemia cases were found.The prevalence rate of high blood glucose was 5.8%. Hypertension occurred in 17.1% of subjects.Obesity/Overweight occurred in 5 252 subjects with the prevalence rate of 32.1%.On the basis of the diagnostic criteria for MS in China.MS occurred in 12.2% of subjects(14.2% for males and 10.0% for females).In both genders,the rate increased along with the increment of age.65.5% participants had at least one of the metabolic disorders.The prevalence of hypercholesterolemia,hypertriglyceridemia,and abnormally low HDL-C were higher than previous analysis during 1984-1986 in all age groups.Conclusions The prevalence of MS in this large occupational population in Beijing was quite high. The prevalence of hypeflipemia had increased considerably over the past 20 years.Balanced nutrition and reasonable consumption should be undertaken to modify these situations.
5.High efficient expression of recombinant human cystatin C in Pichia pastoris

Objective To construct human Cystatin C expression vector and make primary protein purification. Methods A 380 bp cDNA fragments encoding Cystatin C were isolated by RT PCR from human placenta tissue and then inserted into pPIC9 vector. Recombinant pPIC9 Cystatin C was transformed into Pichia pastoris strain GS115. Secreted soluble Cystatin C was produced by induction of the AOX1 promoter with methanol. Recombinant Cystatin C was purified by Hitrap SP cation exchange chromatography. Results The expression level of Cystatin C reached 16 mg/L. The stability of Cystatin C is 3 days in expression system. SDS PAGE revealed that Cystatin C was expressed up to 60% of the total soluble protein. After purified by Hitrap SP cation exchange, the recovery rate was 40.5%. Conclusion Cystatin C has a high secretion in Pichia pastoris yeast and could be recognized by polyclonal antibody of native human Cystatin C. After purification,this protein can be used as a standard and antigen to develop immunoassay for human Cystatin C in serum.
6.The effect of one station style in the reform of out-patient service flow

The human-based and patient-centered notion is a significant breakthrough of the management conception of modernized hospitals.The service mode of one station style was first adopted in health examination center in the reform of out-patient service flow.Based on the humanized service notion,we can provide health examination service with high quality and efficiency to various patients by taking multifarious measures such as reforming the department locations,optimizing the health examination course and setting convenience-stations.The service mode of one station style was demonstrated to have not only elevated the quality and efficiency of health examination service,but also increased both the patients satisfaction and the profit and reputation of the hospital.
7.Effect of dexmedetomidine on proliferation and differentiation of rat embryonic neural stem cells in vitro

Objective To investigate the effect of dexmedetomidine (DEX) on proliferation and differentiation of rat embryonic neural stem cells in vitro. Methods Embryonic neuraI stem cells of fetal SD rats were separated from rats with gestational age of 14-16 days , and underwent primary culture for nestin expression. Cells were divided into control group, low-dose DEX group, and high-dose DEX group, which were cultured with 0,1 and 10 ng/mL DEX respectively. Cell viability was detected by MTT assay;the proliferation rate was estimated by BrdU incorporation; the phosphorylation of ERK1/2 in the proliferation phases was detected by western blot; the differentiation of neuronal cells,astrocytes,oligodendrocyte and neural stem cells were assessed by immunocytochemistry of cell specific markers. Results More than 95% of the embryonic neural stem cells in primary culture were Nestin-positive. Cell viability , BrdU-positive cells and phosphorylation of ERK1/2 in low-dose DEX group increased significantly (P<0.05), and decreased significantly in high-dose DEX group compared with control group and low-dose DEX group (P < 0.05). NeuN-, GFAP- and MBP-positive cells increased (P < 0.05), while Nestin-positive cell decreased (P<0.05) in both low and high dose DEX group compared with control group. However, no significant difference was found in low and high dose DEX group (P > 0.05). Conclusions The proliferation of neuraI stem cells can be promoted by low-dose DEX and depressed by high-dose DEX , as well as regulated by DEX of the phosphorylation of ERK1/2. DEX induces neural stem cells non-selective differentiation.

9.Analysis and intervention of ethical problems in new technique therapy of wound care

Objective Medical ethical problems have become underlying risks since the introduce of Negative pressure wound therapy ( NPWT) in Wound Care Center .The article was to analyze ethical problems in applying new technology in wound care and take intervention countermeasures to investigate the effects . Methods Patients treated by NPWT from January 2009 to Decem-ber 2012 were collected as objectives of investigation from the indexes of compliance rate , cure rate and adverse event rate .The ethical problems related to efficacy ,safety and conflicts of patients′economic interests during the application of NPWT were analyzed .On the basis of the ethnical core principle which was harmless , beneficial, respectful and justifiable , the details of effectiveness and safety of NPWT technique , informed consent form and emergency response protocols of the adverse events were designed according to interna -tional guidelines .Nurses received standardized training to learn new technology . Results 78 cases with chronic wounds accepted negative pressure wound therapy and all cooperated to complete treatment , compliance rate was 100%, no adverse events , the average cure rate was 91 .03%.The treatment costs are less than international and national costs from 75%to 66%. Conclusion Following the ethical principle , informed consent , good communication , strict entry system and standardized operation process make sure the safety and effectiveness of new technology and optimize patients′benefits .
10.Establishment of ovarian SKOV3 cell line stably expressing PES1 by Tet-on inducible system

Objective To further research the biological functions of PES1,the ovarian SKOV3 cell line with inducible stable PES1 expression is established by using Tet-on system.Methods PES1 was cloned into pTRE-Tight vector via PCR and its expression was identified. After transfected the regulating plasmid pTet-on,SKOV3 cells were screened with G418 and re-transfected pTRE-Tight-PES1.The positive cell clones were screened out with hygromycin and were induced by doxycycline (Dox) to definite the best induction concentration.Growth velocity of SKOV3 cells stably expressing PES1 induced by Dox was detected with viola crystallina.Results The SKOV3 cells with inducible PES1 expression were screened out after the cells were transfected pTRE-Tight-PES1 constructed.Dox could dose-dependently induce the PES1 expression with the concentration under 2 mg/L,and 2 mg/L of Dox induced the highest PES1 expression.Growth velocity of SKOV3 cells transfected pTRE-Tight has no significant difference between the SKOV3 cells transfected nothing induced with Dox.However,the SKOV3 cells transfected pTRE-Tight-PES1 grew faster than the cells transfected pTRE-Tight or without transfection in the fourth day (P =0.001 ).Conclusion The inducible stable PES1 expression SKOV3 cells are successfully established and could be used to be an effective cell model to research the biological functions of PES1.The expression of PES1 could promote the growth of SKOV3 cells.
